1.Application value of extravascular lung water index and pulmonary vascular permeability index in patients with acute respiratory distress syndrome

Objective To investigate the value of extravascular lung water index (EVLWI) and pulmonary vascular permeability index (PVPI) in the diagnosis and treatment of acute respiratory distress syndrome(ARDS).Methods From February 2015 to October 2016,selected 80 patients with ARDS in Danzhou Municipal People's Hospital,including 19 cases mild patients,28 cases moderate cases and 33 cases severe cases.The patients' EVLWI,PVPI,acute physiology and chronic health score system Ⅱ (APACHE Ⅱ)score,lung injury score (LIS) were detected.Results The score of APACHE Ⅱ in severe group was 22.81(21.91,25.40) points,significantly higher than that in mild group (19.81 (18.12,21.10) points) and moderate group(20.07 (19.01,22.02),P =0.002).The LIS,EVLWI and PVPI in Severe group were 2.01(1.83,2.11) points,17.01 (14.82,23.02) ml/kg and 3.82 (3.01,5.01),significantly higher than that in mild group (1.01 (0.98) points,1.7412.71 (10.89,13.67) ml/kg,2.71 (2.36,2.94)) and moderate group (1.52 (1.36,1.91) points,14.21 (13.10,16.60) ml/kg,3.01 (2.52,3.03),P < 0.05),LIS,EVLWI and PVPI in moderate group were significantly higher than the mild group (P<0.05).The mortality rate of 28 d in moderate and severe group was 32.14% (9/28) and 51.52% (17/33),which was significantly higher than that in mild group 0.00%(0/19) (P<0.05).EVLWI and PVPI were 19.12(17.22,22.96) ml/kg and 3.71(3.08,5.22) in patients with death,which were significantly higher than those in survival 14.19 (11.20,16.59) ml/kg and 2.97(2.31,3.10) (P=0,021 and 0.016);EVLWI was positively correlated with PVPI,LIS and APACHE Ⅱ score(rs =0.411,0.323 and 0.304,P< 0.001).PVPI was positively correlated with LIS and APACHE Ⅱ score (rs =0.346,0.297,P<0.001).No correlation between LIS and APACHE Ⅱ score (P>0.05).Conclusion EVLWI and PVPI have certain apphcation value in the diagnosis and treatment of ARDS,and it is related to the patient's condition and prognosis.
2.Effect of compound-fluoride preparation (Co-F) on synthetic hydroxyapatite

砄bjective: To compare the effects of compound fluoride preparation(Co F) on the inorganic phases of the teeth with those of ZnF 2 .Methods: Synthetic hydroxyapatite(HA) was immersed into Co F containing ZnF 2, SrF 2. LiF and CaF 2, and with F - concentration of 3 500 mg/L, or into saturated ZnF 2 solution with F - concentration of 3 200 mg/L or into NaF solution with F - concentration of 3 500 mg/L for 1 and 10 days respectively. Fluoride absorption into HA was measured by X ray diffraction analysis. Results: F - absorption (?g/g) of HA in NaF solution for 1 day and 10 days were 781.81?94.38 and 805.25?87.11; that in ZnF 2 5 764.93?726.58 and 7 177.33?1 498.37, in Co F 4 901.68?680.02 and 7 136.19?461.56, respectively. KOH F(?g/g) of HA in NaF solution for 1 and 10 days were 94.96?8.28 and 151.10?19.79, that in ZnF 2 230.8?25.08 and 6 576.89?1 440.17, in Co F 1 168.18?69.42 and 4 336.72?363.16, respectively. Conclusion: ZnF 2 contained in the Co F is very important in respect of the increase of fluoride absorption.
3.Influence of Changing the Parameters on the Wave of NRT

Objective To inevestigate the changes of the responses by changing the parameters in the neural response tetemety(NRT).Methods In this project,twenty patients who were implanted in Nucleus C124M were measured the electrically evoked compound action potential(ECAP)using the NRT system.It was expected to observe the changes of the responses by changing the parameters in the NRT 2.04 software.Results It was found that the parameters that made great influences to the responses were current level,stimulate pulse width,gain,delay and masker advance.Conclusion The parameters that made great influences to the record time were stimulate rate and number of sweeps.
4.miRNA-126 inhibits proliferation, migration and invasion of human lung cancer A549 cells via EGFR/AKT/mTOR pathway

AIM:To investigate the effects of microRNA ( miRNA)-126 on the proliferation , migration and in-vasion of human lung cancer cell lines , and to explore its mechanism .METHODS:The A549 cells were transfected with miRNA-126 agomir by Lipofectamine 2000.The expression of miRNA-126 was detected by real-time PCR.The cell activity was detected by MTT assay .The number of viable A549 cells was counted by the method of Trypan blue exclusion .The cell colony-forming capability was determined by cell colony formation test .The cell migration and invasion abilities were assayed by wound healing and Transwell methods , respectively.The protein levels of p-EGFR, EGFR, p-AKT, AKT, p-mTOR and mTOR were determined by Western blot .RESULTS:The expression level of miRNA-126 was significantly in-creased in the A549 cells compared with negative control ( NC) group and control group ( P<0.01 ) .The proliferation of A549 cells was decreased extremely after transfected with the miRNA-126 agomir (P<0.01), so did the result of the cell colony-formation test.The migration and invasion abilities of the lung cancer cells were also significantly inhibited .The protein levels of p-EGFR, p-AKT and p-mTOR were significantly down-regulated compared with NC group and control group ( P<0.01) .CONCLUSION:Over-expression of miRNA-126 significantly inhibits the proliferation , migration and invasion ability of human lung cancer A 549 cells by down-regulation of EGFR/AKT/mTOR pathway .
5.Insulin promotes rhesus retinal vascular endothelial cells angiogenesis via VEGF-A/VEGFR2 pathway

Rhesus retinal vascular endothelial cell line RF/6A cells were treated with human insulin. Cell proliferation, migration, and lumen formation, as well as the expressions of vascular endothelial growth factor-A ( VEGF-A ), VEGF-A receptors, and phosphorylated receptors were measured. Insulin promoted RF/6A cell proliferation, migration, and lumen formation ( all P<0. 01 ). Insulin increased the expression of VEGF-A mRNA and improved its protein activity ( all P<0. 05 ), and promoted the expression of VEGFR2 mRNA and its phosphorylation ( both P<0. 01 ). There was no significant difference in the expression of VEGFR1 mRNA among the groups ( P>0. 05 ).
6.Artificial trachea with pedicle rib cartilage and cilia endothelial wall

[ABSTRACT]OBJECTIVETo explore the method on reconstruction of long and special tracheal defects which can mostly match with natural airway: pedicle rib cartilage with cilia endothelial wall.METHODS8 experimental model of rabbits were trained with cervical double belt blood supply fascia embedding and autologous costal cartilage and nasal septum mucos in the first period, then followed by transplantation in the second period. After the operation, we would assess the physiology, breathing and histopathology index of the rabbits, etc. After the animal experiment, we tried to apply the method to an appropriate clinical case.RESULTS8 cases of experimental rabbits dead after the second period operation with the average survival time of 21.9 days and caused by asphyxia. Histopathological results: rib cartilage and trachea ring up of cartilage cells and fibers have high similarity in histology; cartilage of all cases under the cultivation of the pedicle fascial package has not been absorbed; all cases' nasal septum mucosa in the body and blood supply to differentiation under fascia nutrition. Then we applied the method on a clinical case.CONCLUSIONTrachea ring rib cartilage had a higher similarity to the tracheal cartilage on the histology and biological characteristics that can be used as the preparation of artificial trachea shaping material or cell culture to tissue engineering materials. Package of rib cartilage pedicle fascial can provide adequate blood supply to make up for a free training rib cartilage defect to its absorption. Nasal septum mucosa of pressure in the body and blood supply of the fascia nutrition can simulate the trachea ciliated epithelium, which can play its biological characteristics similar to the inner wall of the trachea.
7.Comparison of the Effect of Imipenem and Cilastatin Sodium and Meropenem in the Treatment of Acute Leukemia Granulocytopenic Phase Combined with Severe Lung Infection

OBJECTIVE:To compare the clinical efficacy and safety of imipenem and cislastatin sodium and meropenem in the treatment of acute leukemia granulocytopenic phase combined with severe lung infection. METHODS:A total of 64 patients with acute leukemia granulocytopenic phase combined with severe lung infection were selected from our hospital during Jul. 2015-Jul. 2016 as study objects. They were divided into trial group(odd number)and control group(even number)according to admis-sion order,with 32 cases in each group. Control group was given Meropenem for injection 1 g+0.9% Sodium chloride injection 100 mL,ivgtt(about 30 min),q8 h. Trial group was given Imipenem and cislastatin sodium for injection 1 g+0.9% Sodium chlo-ride injection 100 mL,ivgtt(about 30 min),q12 h. Both groups were treated for 14 d. Clinical efficacies as well as blood gas pa-rameters [p(O2),p(CO2),SaO2] and pathogenic clearance were observed in 2 groups,and the occurrence of ADR was recorded. RESULTS:The total response rate of trial group(78.13%)was significantly higher than that of control group(71.88%),with sta-tistical significance (P<0.05). Before treatment,there was no statistical significance in blood gas parameters or the number of detected pathogenic strains between 2 groups (P>0.05). After treatment,the levels of p(O2) and SaO2 in 2 groups were in-creased significantly,while the level of p(CO2) was decreased significantly;there was statistical significance compared to be-fore treatment(P>0.05). There were 27 cases of pathogenic clearance in trial group(clearance rate of 84.38%),which was sig-nificantly more than control group (26 cases,clearance rate of 81.25%),without statistical significance (P>0.05). The inci-dence of ADR in trial group(9.38%)was significantly lower than control group(15.63%),without statistical significance(P>0.05). CONCLUSIONS:Imipenem and cislastatin sodium and meropenem show good clinical efficacy for acute leukemia granu-locytopenic phase combined with severe lung infection,blood gas parameters improvement and pathogenic clearance effect,both of them have good safety.

9.Curcumin inhibits migration and invasion of lung cancer PC-9 cells via down-regulation of nectin-4 expression

AIM:To investigate the effects of curcumin on the abilities of migration and invasion in the lung cancer PC-9 cells, and to observe the relationship between curcumin and nectin-4 expression.METHODS:The viability, migration and invasion of lung cancer PC-9 cells treated with curcumin or transfected with siNectin-4 were measured by MTT assay, wound healing test and Transwell assay , respectively.The protein levels of nectin-4, p-AKT and AKT in the PC-9 cells treated with curcumin or transfected with siNectin-4 were detected by Western blot .RESULTS:Curcumin in-hibited the viability of PC-9 cells.The wound healing rates and the numbers of the transmembrane cells in curcumin 10μmol/L and 20 μmol/L groups were decreased compared with control group without curcumin treatment .The expression level of nectin-4 was reduced after curcumin treatment for 24 h.The viability of the PC-9 cells was significantly inhibited after transfected with siNectin-4 for 48 h or 72 h (P<0.01), and the wound healing rates was decreased in siNectin-4 group compared with NC group (P<0.01).The numbers of the transmembrane cells in siNectin-4 group was significantly reduced (P<0.01).Curcumin and knockdown of nectin-4 suppressed the activation of AKT pathway in PC-9 cells.In si-Nectin-4+curcumin group , the cell viability reduced compared with curcumin group , and wound healing rates , cell inva-sive ability and AKT phosphorylation levels were decreased .CONCLUSION:Curcumin inhibits migration and invasion of the lung cancer PC-9 cells via down-regulation of nectin-4 expression and inhibition of AKT pathway .
10.Microsatellite instability and abnormal mismatch repair in laryngeal squamous cell carcinoma

OBJECTIVE The aim of our study was to evaluate the significance of the frequency of microsatellite instability (MSI) and its relationship to mismatch repair gene (MMR) in laryngeal squamous cell carcinoma.METHODS We investigated the expression frequency and clinical significance of MSI and MMR in 50 laryngeal squamous cell carcinoma (LSCC) patients from Beijing Tongren Hospital. The status of MSI was evaluated using microdissection - polymerase chain reaction (PCR) - single strand length polymorphism (SSLP) - silver staining.Five markers on chromosomes 1p, 3p, 5q, 9p, 17p, which were adjacent to BCAR3 (breast cancer anti-estrogen resistance 3), FHIT, APC, CDKN2A (p16), TP53 respectively, were used. Two of the six components of MMR -hMLH1 and hMSH2- were investigated by an immunohistochemical approach because of the high frequency of their downregulation in head and neck tumors. RESULTS The informative case number of the five markers (D17S796, D3S3544, D5S656, D1S375, D9S162) were 44, 42, 45, 44 and 40 respectively. The incidence of MSI was lower than the frequency of loss of heterozygosity (LOH). The incidence of MSI on D17S796 (TP53)was 20.5% (9/44),on D3S3544 (FHIT) was 14.3 % (6/42), on D5S656 (APC) was 31.1% (14/45), on D1S375 (BCAR3) was 20.5 % (9/44), and on D9S162 (CDKN2A) was 15.0 % (6/40). Though there was no relationship between MSI status and age, gender, smoke history, tumor location, tumor differentiation and T stage (P>0.05),there was a strong correlation between MSI and relapse condition (P<0.01). Also, MSI status correlated with MMR expression to some degree (P<0.01), but it was common for negative and positive staining of MMR to coexist on the same slide. CONCLUSION Microsatellite instability and abnormal mismatch repair may contribute to the carcinogenesis of a subset of laryngeal squamous cell carcinoma. Microsatellite instability may be a characteristic signal of tumor recurrence.
