Objective To investigate the distribution of T follicular helper(Tfh)cell subsets in hepa-titis B patients at different immune stages and to clarify the relationships between the level of CXCL13 and the distribution of Tfh cell subsets. Methods Flow cytometry analysis was performed to detect the distribution of Tfh cells. Enzyme-linked immunosorbent assay(ELISA)was performed to measure the levels of CXCL13 in ser-um samples collected from hepatitis B patients. RT-PCR and Western blot assay were used to analyze the expres-sion of CXCL13 in HepG2 and HepG2. 2. 1. 5 cells. Results The percentages of Tfh1 cells were significantly up-regulated at the immune activation(IA)stage,while those of Tfh2 cells were significantly raised at the im-mune tolerance(IT)stage. The percentages of Tfh17 cells in patients at the stage of IT were similar to those in patients at the stage of IA,but were higher than those in responders with HBsAg seroconversion(RP)or healthy controls(HC). The expression of CXCL13 was positively correlated with the percentage of Tfh2 cells. More over,hepatitis B virus(HBV)enhanced the expression of CXCL13 at both transcriptional and translational lev-els. Conclusion HBV might up-regulate the percentage of Tfh2 cells through promoting the expression of CXCL13,which resulted in the induction of immune tolerance. Elucidating the functions of Tfh1,Tfh2 and Tfh17 cells and understanding the type conversion mechanism among the three subsets are important for further researches on HBV-induced immunosuppression.

Human interleukin-15 (hIL-15) is an important cytokine to activate endothelial cells and can be regulated by many other cytokines. The aim of this study is to examine the ability of interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha) to induce the production of human interleukin-15 (hIL-15) and IL-15 receptor (IL-15Ralpha) by human umbilical vein endothelial cells (HUVECs). The data are summarized as follows: 1. Northern blot revealed that IL-15 mRNA was up-regulated by IFN-gamma and TNF-alpha. 2. Intracellular IL-15 protein was visualized by fluorescence microscopy, whereas the expression of IL-15 on the surface of HUVECs was detected by fluorescence activated cell sorting (FACS), and no detectable IL-15 in the medium was verified by ELISA. 3. IL-15Ralpha was detected on the surface of HUVECs by FACS after IFN-gamma and TNF-alpha stimulation, whereas Western blotting revealed that the elevated expression on surface IL-15Ralpha was not due to the increased protein expression. The conclusion demonstrated from our results is that IFN-gamma and TNF-alpha play an important role in regulating the expression of IL-15 and IL-15Ralpha on the surface of HUVECs.

Objective An HPLC method was established for determination of paeoniflorin in plasma after ig compound decoction of Radix Paeoniae Rubra with Fructus Piperis to mice. Methods The conditions of chromatography: Kromasil C 18 column (250 mm ? 4.6 mm, 7 ?m) was used with a mobile phase of CH 3OH-H 2O ( 38∶ 62); flow rate was 0.5 mL/min; the detecting wavelength was 230 nm; external standard method was quantitative analysis method. Results Paeoniflorin was fully separated from the other components in plasma. The linear range was 5.0—250.0 ng/?L (r= 0.999 9 ), the lowest detectability was 1.49 ng/?L, the average recovery was higher than 90%. Conclusion This method specially provides an accurate and sensitive way in detecting the in vivo blood concentration of paeoniflorin in plasma.

Objective To establish an analytical method for determination of ferulic acid inDanggui HujiaoRadix Angelicae Sinensis and Fructus Piperis]) Decoction in mouse plasma.Methods HPLC method was used.The conditions of chromatography: Kromasil C 18250 mm?4.6 mm, 7 ?m) was used with a mobile phase of CH3OH-H2O-CH3COOH (36.4∶63∶0.6).Flow rate was 1.0 mL/min.The detecting wavelength was 322 nm.External standard method was quantitative analysis method.Results The ferulic acid could be totally separated from other ingredients in plasma.The linear range was 1.88—188.00 ng/?L (r=0.999), the lowest detectability was 0.47 ng/?L, and the average recovery was 94.85%.Conclusion This method provides an accurate and sensitive way in detecting blood concentration of ferulic acid and studying in pharmacokinetics.

One hundred and ten patients with primary hepatocellular carcinoma underwent hepatectomy from 2005 to 2010.Different methods of hepatic exclusion were applied in the surgery,including 54 patients with total hepatic vascular exclusion,22 with half hepatic vascular exclusion and 34 with regional hepatic vascular exclusion.The recovery of postoperative liver function was retrospectively analyzed.The results showed that the postoperative liver function of regional hepatic vascular exclusion [ALT(311 ± 80) U/L,total bilirubin (TB) (22.2 ± 8.3) μmoL/L at 1 st day and ALT (58 ± 17) U/L,TB (11.3 ± 3.1)μmol/L at 7th day] was better than that of total hepatic vascular exclusion [ALT(874 ±299)U/L,TB (42.9± 19.1) μmol/L at 1st day (P＜0.05) and ALT (108-±52)U/L,TB (14.6±9.2) μmol/L at 7th day] (P ＜ 0.05,＞ 0.05),indicating that regional hepatic vascular exclusion can effectively reduce hepatic injury during the operation and promote recovery of liver function after operation.

Human interleukin-15 (IL-15) is a proinflammatory cytokine to suppress neutrophil apoptosis, which is a potential therapeutic agent. The modulatory effect of TNFα was investigated in IL-15-induced suppression of human neutrophil apoptosis. TNFα was shown to reverse the ability of IL-15 to delay neutrophil apoptosis within certain time course. Moreover, this reverse effect by TNFα might be associated with a reduction of the expression of the anti-apoptotic Bcl-Xl protein detected by Western blotting. It is concluded that TNFα can be used to modulate IL-15-induced suppression of neutrophil apoptosis within certain time course.

OBJECTIVETo study the influence of Chinese herbs for inner-warming on the bioavailability of paeoniflorin (PF) and its mechanism.

METHODSChinese herbs (pepper fruit, evodia fruit, cassia bark, fennel fruit and prickly-ash peel) were separately used in combination with PF for gastrogavage to mice. Reversed phase high-performance liquid chromatography was used to determine the plasma concentration of PF in mice after medication. The bioavailability of PF was calculated and compared, taking single use of red peony root for control.

RESULTSThe pharmacokinetics of PF in mice was conformed to the one-compartment model, as combined use with Chinese herbs for inner-warming, the relative bioavailability of PF was 137.22% for pepper fruit, 123.62% for evodia fruit, 108.39% for cassia bark, 226.02% for fennel fruit and 116.73% for prickly-ash peel, there were difference of Cmax and AUC(0-infinity) in comparison of these data with the control group (P < 0.05), but with no difference of tmax (P > 0.05).

CONCLUSIONThe Chinese herbs used in this experiment in combination with red peony root could enhance the bioavailability of PF, which illustrated the scientific meaning of the recipe combination of Chinese herbs for activating blood circulation and inner-warming viewing from pharmacodynamics.

The author discussed the statistics and management of medical records from the perspective of big medical data through on-the-spot investigation and literature review, explained the significance of using big data in statistical management of medical records, introduced the current situation and existing problems of information management of medical records in hospitals, and explored the development of statistical information from the perspective of big medical data. It is pointed out that informationized means are necessary for increasing the specialized institutions for information management of medical record and managing medical record rooms. And also, the cultivation of quality of staff in the hospital is a problem to be solved, and the prospects of development have been predicted.

OBJECTIVETo study the characteristics of soil microbial variation during Salvia miltiorrhiza crop rotation.

METHODthe conventional cultivating microbial method was used to study the microbial number and communities structure and soil microbial biomass phosphorus (SMBP) was determined by chloroform vapor extraction method. The data was then analyzed by SPSS software.

RESULTWith the increase of the crop rotation years, the numbers of bacteria and actinomycetes in soil also, but the fungi and SMBP decreased.

CONCLUSIONMicrobial mechanism of crop rotation of the planting S. miltiorrhiza is the regulation of microbial number and bacteria physiological communities, the process rebuilds the soil ecological system balance. Microbial communities in soil need at lest 2 years to get to restore, after planting S. miltiorrhiza, which consisting with traditional planting experience.
