1.Analysis of Lymph Node Metastasis and Clinical Characteristic in Rectal Cancer(Report of 79 Cases)

Objective To investigate the correlation among lymph node metastasis and clinical features,postoperative survival rate in rectal cancer.Methods Seventy-nine patients who had accepted total mesorectal excision(TME) were collected,and the correlation among their clinical features(including gender,age,tumor size,gross type,depth of infiltration,histology type,differentiated degree and the level of blood serum CEA),lymph node metastasis,and postoperative survival rate were analyzed.Results There was significant correlation between six factors(namely the tumor size,gross type,depth of infiltration,histology type,differentiated degree and the level of blood serum CEA) and lymph node metastasis in single factor analysis.However,multivariate analysis showed that only gross type of tumor and depth of tumor infiltration were related to lymph node metastasis.The postoperative survival time of 43 non-metastasis cases was remarkably longer than that of 33 cases with lymph node metastasis(?2=18.806,P=0.000),and it was longer in 22 cases with
2.AN ANALYSIS OF X-RAY PICTURES OF PULMONARY EDEMA OF HIGH ALTITUDE IN CHILDREN

A retrospective analysis was made of 56 children with a definite diagnosis of pulmonary edema,in whom x-ray films were taken before and after treatment.Four patients had been in Tibet for five months to 6 years.In 50 patients,pulmonary edema occurred 6~72 hours after they flied into Tibet.Two of them died during treatment because of delayed treatment and critical conditions.All 56 patients were of Han nationality,aged 5 months to 12 years.Childhood acute pulmonary edema accountes for 69% of all high altitude pulmonary edema cases.Childhood pulmonary edema had an acute onset,and it ran a fulminating course.chest films usually showed disseminating alveolar changes,and the diagnosis may be misleading.X-ray physicians should get familiar with the diagnosis to avoid a delayed diagnosis and treatment.
3.The effects of social factors on anxiety by logistic regression analysis in undergraduate students

ObjectiveTo explore the prevalence of anxiety and its related social factors in undergraduate student,in order to provide scientific evidence for the prevention of anxiety in undergraduate.MethodsThe cluster random sampling was adopted for this study,and 965 college students from Zhengzhou University were investigated by Personal Report of Communication Apprehension,Simplified Coping Style Questionnaire,Toronto Alexithymia Scale,Perceived Social Support Scale and Self-Rating Anxiety Scale.Results①The anxiety detection rate of respondents was 19.3%.There were significant differences in the detection rate and severity of anxiety between genders(Male:27.0%,Female:10.2% ) and grades( Freshman:16.4%,Junior:23.8% ) in the college students (P < 0.05).There were not significant difference in different family residence students anxiety detection rate and level(P > 0.05 ).②The scores of communication apprehension ( ( 69.31 ± 12.32),( 65.25 ± 12.56) ),positive coping style( (20.84 ±5.10),(23.99 ±5.18) ),negative coping style( ( 11.03 ±4.15),(09.18 ±3.96) ),Toronto alexithymia( (74.97 ± 6.93 ),(70.31 ± 7.98 ) ) and perceived social support ( (53.14 ± 5.78 ),(57.02 ± 5.79) ) of the anxiety group were significantly different with those of the normal group (P < 0.05).③The significant relationship was found among anxiety emotions and gender,positive coping style,negative coping style,Toronto alexithymia and perceived social support by using Logistic regression analysis (P< 0.05 ).ConclusionsThe prevalence of anxiety in undergraduate students is relatively high and it is related with multiple factors.In addition,social support,alexithymia and coping style have a close correlation with anxiety in undergraduates.
4.Cloning of trichosanthin gene and its induction effects on the apoptpsis of colorectal carcinoma LoVo cell

Objective To clone cDNA of trichosanthin (TCS) and purify TCS, and to study its influence on apoptosis and growth inhibition of colorectal carcinoma LoVo cells in vitro. Methods MTT assay was adopted to measure the growth inhibition ratio of LoVo cells treated with TCS, and apoptosis was assayed by agarose gel eletrophoresis. Results The results showed that the higher concentration of TCS and the longer testing time, the stronger growth inhibition of LoVo cells. DNA agarose gel eletrophoresis showed a gradient, which confirmed that TCS could induce the apoptosis of LoVo cells. Conclusions TCS can inhibit the growth of LoVo cells in vitro and induce its apoptosis. Our study provides evidence for the application of TCS in clinical treatment of human colorectal carcinoma.
5.Application of ultrasound-guided transversus abdominis plane block for operations of inguinal region in infants

One hundred and fifty ASA Ⅰ-Ⅱ infants scheduled to undergo surgery on unilateral inguinal region,were randomly assigned to 3 groups with 50 in each group.In groups Ⅰ and Ⅱ puncture was performed at traversus abdominis plane under ultrasound-guidance,0.4 or 0.5/kg of 0.25% ropivacaine was injected respectively.In group Ⅲ the triangle of Petit was positioned through palpating and then 0.5 ml/kg of 0.25% ropivacaine was injected.HR,RR,SpO2,PETCO2,expired sevoflurane concentration and BIS value at the time of entering the operation room (T1),incision of skin (T2),pulling hernia sac (T3),ending the surgery (T4),waking (T5),the number of pressing the analgesia pump after operation were recorded.The results showed that HR at T2 and T3 of groups Ⅰ and Ⅱ was lower than group Ⅲ (P < 0.05).The were significant differences in case numbers of insufficient intraoperative analgesia among 3 groups(x2 =10.500,P =0.005).The CHEOPS scores and the number of pressing analgesia pump after operation in groups Ⅰ and Ⅱ were lower than those in group Ⅲ (x2 =7.230,P =0.027).Results indicate that ultrasound-guided transversus abdominis plane block is safer and more effective than conventional method for operations of inguinal region in infants; it may reduce dose of local anesthetics and postoperative use of analgesics.
6.Hinokitiol induces bladder cancer J82 cells apoptosis via autophagy induction

Background and purpose: Bladder cancer is the most common urological malignancy in our country which seriously threatens human health, and its incidence increased year by year. This study aimed to investigate the effects of hinokitiol on the proliferation, apoptosis and autophagy in human bladder cancer cell lines. Methods:CCK-8 assays were performed to analyze the effects of hinokitiol on the proliferation of J82 cells. Apoptosis rate was determined by lfow cytometry. Cleaved caspase 3, LC3 and P62 protein expression was determined using Western blot. EGFP-LC3 microscopy assay was performed to assess autophagy. Results: Hinokitiol significantly inhibited the proliferation of J82 cells and induced cell apoptosis via caspase pathway. The apoptosis effect of hinokitiol could be partially antagonized by Z-VAD-FMK. Hinokitiol induced autophagy of J82 cells, increased LC3 expression and down-regulated P62 expression. 3-MA is able to rescue Tet-induced cell death. Conclusion:Hinokitiol can inhibit the proliferation of J82 cells and induce cell apoptosis via autophagy activation.
7.Effects of equol on colon cancer cell proliferation

Objective:To investigate the effect of equol on the proliferation of colom cancer cells and to explore the mechanisms.Methods: Colon cancer cells (DLD1,HCT15,COLO205,LOVO,SW480) were incubated, the cell proliferation was identified by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell proliferation assay.Reverse transcription PCR and Western blot were used to measure the mRNA and the protein expression of estrogen receptor and nuclear factor (erythroid-derived 2)-like 2 (Nrf2)in the colon cancer cells, respectively.Moreover, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell proliferation assay was used to investigate the effect of estrogen receptor(ER) inhibitor,ERα agonist, and estrogen receptor ERβagonist on the cell proliferation.Results: ERα was faintly expressed in the DLD-1 and HCT-15 cells.However, ERβ expression in DLD1, HCT15, COLO205, LOVO, and SW480 colon cancer cells.Different concentrations of equol (0, 0.5, 1, 5, 10 μmol/L) significantly inhibited the growth of HCT-15 cell with the expression of ERα and ERβ.More-over, different concentrations of equol (0, 0.5, 1, 5, 10 μmol/L) significantly inhibited the growth of LOVO, and SW480 cells with the ERβ expression in a dose-dependent manner as demonstrated with a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell proliferation assay.mRNA expressions of ERα and ERβ in HCT-15 were stimulated significantly.Western blotting proved that the protein expressions of ERα and ERβ increased with the increasing of equol dose.Moreover we found significant difference of Nrf2 protein expression in HCT-15 cell stimulated by different concentrationss of equol.After the similation of estrogen receptor inhibitor, ERα agonist, or ERβ agonist, we found that only dif-ferent concentrations of ERβ agonist(0, 1, 10, 100, 1 000, 10 000 nmol/L) significantly inhibited the growth of HCT-15, LOVO, and SW480 in adose-dependent manner.Estrogen receptor inhibitor and ERα agonistdid not present significant effect on the cell proliferation of HCT-15, LOVO, and SW480.Conclusion: Equol inhibited the colon cancer cell proliferation by its estrogenic activities and antioxidant activities.
8.The expression and significance of the multidrug resistance-related proteins P-gp, MRP, LRP and GST-? in lung cancer using tissue microarray

Background and purpose:Resistance to anticarcinogen is one of the key factors that affect the treatment efficiency in lung cancer. The purpose of this study was to investigate the clinical significance of the multidurg resistance-related proteins P-gp, multidrug resistance-related proteins(MRP),lung resistance associated protien(LRP) and GST-?by detecting their expression in lung cancer and to investigate the mechanism of resistance to anticarcinogen. Methods:S-P immunohistochemistry was used to examine the expression level of proteins P-gp, MRP, LRP and GST- ?in 226 samples of lung cancer and 23 samples of normal lung tissues. Results:The positive rates of P-gp, MRP, LRP and GST-? in lung cancers were 46.0%, 42.0%, 54.4%, 62.4% respectively. Significant difference existed between tumorous tissue and normal lung tissue (17.4%, 13.0%, 17.4%, 21.7%). The positive rates of P-gp, MRP, LRP and GST-? in poorly differentiated-type of NSCLC were 33.3%, 22.8%, 33.3%, 47.4%, compared with differentiated-type of NSCLC (59.7%, 58.1%, 73.6%, 79.1%) (P
9.Effects of oleanolic acid on proliferation of SGC-7901/CDDP in vitro and its potential mechanisms

Aim To investigate the effects of oleanolic acid on proliferation of SGC-7901/CDDP cells in vitro and its potential mechanisms.Methods Model of SGC-7901 cells resistant to cisplatin(SGC-7901/CDDP)was established in vitro.MTT assay was used to evaluate the viability of SGC-7901/CDDP cells in vitro.Real-time PCR was used to analyse the expression of Bcl-2 and Bax gene.Results MTT assay showed that oleanolic acid dramatically inhibited the proliferation of SGC-7901/CDDP cells in vitro,more than 62% SGC-7901/CDDP cells were inhibited when the cells were treated with 100 ?mol?L-1 oleanolic acid for 72 h;Real-time PCR showed that Bax(pro-apoptotic gene)was upregulated and Bcl-2(anti-apoptotic)was downregulated after being treated with oleanolic acid.Conclusions These results suggest that oleanolic acid inhibits proliferation of SGC-7901/CDDP cells in vitro,and its potential mechanisms may be through the upregulation of Bax and the downregulation of Bcl-2.
