1.Relationship between range of operative and postoperative ovary function in 55 endometriosis

ObjectiveTo study the relationship between the range of operations and the incidence of climacteric syndrom,the time of episode, degree and estrogen level in endometriosis after operations. MethodsThesexual hormone level was measured and climacteric syndrome was followed up after operation in 55 endometriosis. ResultsThe incidence of climacteric syndrom in oophorectomy was more frequent and earlier than partial oophorectomy. Meanwhile, the syndrome was much severe and the level of E2 was lower in the former group than those in the later one. ConclusionSo we should retain the ovary tissue and blood supply as much as possible. 6 months postoperation is suggested to be suitable to begin hormone replacement therapy. The dosage should be adviced to remain the blood hormone level just like one ovary does.

3.Clinical and experimental research of Jingshichuan Capsule on curing cholelithiasis

AIM: To observe the curative effect of Jingshichuan Capsule and to try to explore the therapeutic mechanism. METHODS: Medicines were taken orally. During experimental research an animal model of cholelithiasis was established through feeding guinea pig with food which placed a premium on shaping of cholith. The guinea pigs were divided into the pathologic group, the Jingshichuan group and the control group. The animal was executed after sixties days, the gallbladder was taken out by operation to inspect the shaping of cholith; the blood and the bile was collected to be measured; and parts of the tissue of liver and gallbladder were left for pathologic examination. RESULTS: The rate of clinic cure and the rate of effect of Jingshichuan group were separately 40.0% and 96.9%, which was significantly higher than the control group ( P
4.Obtainment and Characterization of The Evolved Enzymes From Arabidopsis thaliana Glutathione S-Transferase Zeta Class

The Arabidopsis thaliana glutathione S-transferases zeta class (AtGSTZ) is a multi-functional enzyme, which plays important role in cellular metabolism and environmental purification. Error-prone PCR and cycles of DNA shuffling were used to construct a mutagenesis library of AtGSTZ. The screening of the resultant libraries was carried out by a pH indicator dye-based colorimetric assay. Nine mutants which enhanced the dichloroacetic acid dechlorination activity were obtained. Among them, NN23 contained 25 amino acid substitutions with the activity improving 120%, whereas NN20 contained 24 amino acid substitutions with the activity improving 102%. EC1 contained 2 amino acid substitutions with the activity improving 47%. The rest 6 mutants contained one amino acid substitution with their activity increasing from 9% to 60%. The enzymatic characterization showed that all the evolved enzymes increased their catalytic efficiencies towards dichloroacetic acid and binding affinity towards glutathione whereas some of them increased the renaturability. However there is no obvious change in their thermostability. Based on these data, functional residues related to catalysis and refolding of AtGSTZ were discussed.
5.Functional change of globus pallidus and Parkinson's disease

As an important nucleus of basal ganglia, globus pallidus was concerned to involve in the neural mechanism of Parkins's disease (PD). Previous studies of PD patients and animal model indicate that neurotransmitters and receptors are changed in globus pallidus neurons, the balance between inhibition and excitation is broken in the output nuclei of basal ganglia, and globus pallidus neurons display bursting and oscillatory discharge, the synchronous tendency of neighboring neurons enhances. These neurophysiological changes are associated with PD symptoms, such as bradykinesia, rigidity and tremor, which reflects that globus pallidus plays an important role in the process of PD. In the present paper, the recent advances in pathophysiology in globus pallidus, its clinical application in treatment of Parkinson's disease, and the study direction in the future are reviewed.
6.Upregulation of B7 molecules expression by calcium ionorphore in K562 cells in vitro

Objective:To investigate the effect of calcium ionorphore on B7 costimulatory molecules of chronic myeloid leukemia cells line K562.Methods:Well growing K562 cells were cultured in the medium containing calcium ionorphore(375 ng/ml),with K562 cells without CI treatment as control.The cells′ viability and number were calculated by Trypan Blue exclusion at 0,48 and 96 h.Before and after 96 h of cultured,B7-1 and B7-2 expression was assayed by flow cytometry.The proliferation of allogeneic human T cells was measured by MTT colorimetry.Results:B7 costimulatory molecules were abcent or lowly expressed on K562 cells.After 96 h of CI treatment,B7 costimulatory molecules of K562 cells were markedly upregulated and marked activation of allogeneic T cells occurred.No notable morphological change was found during the culture.K562 cells cultured in medium with CI grow slowly than that without CI.Conclusion:B7 costimulatory molecules expression on chronic myeloid leukemia cells line K562 surface was defective.These costimulatory molecules on K562 cells can be upregulated by calcium ionorphore.Calcium ionorphore may inhibit the growth of K562 cells.
7.Establishment of model with exogenous human CYP2E1 expression and inducement of substrates

AIM: To investigate the effect of ethanol and chemical carcinogen N,N'-dinitrosopiperazine(DNP) on the exogenous expression of CYP2E1.METHODS: Exogenous hCYP2E1 was introduced into NIH3T3 mediated by lipofectamine. Then the integration of exogenous gene was showed by Southemrn blot. After treated with different concentration of ethanol and DNP,RT-PCR and Western blotting were used to analyze the expression change of hCYP2E1 in NIH 3T3. RESULTS: Two cell clones with integration and stable expression of exogenous hCYP2E1 were obtained. The RT-PCR and Western blotting showed that human CYP2E1 mRNA and protein expression was enhanced with increase in ethanol and DNP concentration. CONCLUSION: Exogenous expression of hCYP2E1 was steadily induced by ethanol and DNP. The mechanism may be due to the activation of its transcription. The DNP carcinogenesis might be related to its in situ activation by CYP2E1.

(TNF ?) gene polymorphism and ankylosing spondylitis (AS).Methods Genomic DNA from 98 Chinese AS patients and 70 ethnically matched controls were typed for TNF(308) polymorphism by allele specific polymerase chain reaction (AS PCR).Results The TNF genotypes in AS patients were respectively TNF1 homozygote 37%,TNF2 homozygote 10% and TNF1 and TNF2 heterozygote 53%.While TNF genotypes in controls group were respectively TNF1 homozygote 67%,TNF2 homozygote 3% and TNF1 and TNF2 heterozygote 30%.Significant difference was found in the distribution of TNF 308 genotype between both groups ( ? 2=15 73, P
10.High throughput screening atrazine chlorohydrolase mutants with enhanced activity through Haematococcus pluvialis expression system.

Developing a high-throughput screening method is of great importance for directed evolution of atrazine chlorohydrolase. A mutagenesis library of atzA from Pseudomonas sp. ADP and Arthrobacter sp. AD1 was constructed using error-prone PCR and DNA shuffling. Candidate mutants were screened through Haematococcus pluvialis expression system, using atrazine as selection pressure. Sequence analysis showed that mutations in the obtained 12 mutants with enhanced activity were all point-substitutions and scattered throughout the gene. Enzymatic activity analysis showed that the mutants all had higher activities than that of the wild type. The activities were 1.8-3.6 fold of the wild-type enzyme when cultured in BBM medium with 1 mg/L atrazine, whereas 1.8-2.6 fold with 2 mg/L atrazine. These results indicated that Haematococcus pluvialis expression system is an ideal high throughput screening system for directed evolution of atrazine chlorohydrolase.
