1.Evaluation of regulational function of ingredients from hot herbs on TRPV1 channel based on 7900 PCR instrument

Aim To further study the molecular mecha-nism of the herbs with hot nature on the regulational action on TRPV1 channel based on the 7900 Real-time PCR instrument. Methods 7900 PCR instrument was applied to detect the intracellular flurescence of TRPV1 channel in the dorsal root ganglions ( DRG ) neurons and the effect on the TRPV1 ’ s thermo-sensational functions of the selected 11 ingredients from hot herbs was explored. Results TRPV1 channels could be ac-tivated by gradually elevated temperature; the activa-tion process could be blocked by the TRPV1 specific blocking agent capsaizepine. Most of the ingredients from hot-nature herbs had the potential to up-regualate TRPV1 channel function. Conclusions The estab-lished TRPV1 channel detection system based on PCR instrument is suitable for the analysis of regulational functions of drugs on the heat-activated TRPV1 chan-nel;the functions of hot herbs may be related to the up-regualtional effects of its active ingredients on the TRPV1 channel which will further up-regulate energy metabolism.
2.Relationship between the gene polymorphisms of transforming growth factor beta 1 and interleukin 1 beta and primary knee osteoarthritis in Xinjiang Han and Uygur populations

BACKGROUND:Transforming growth factor β1 (TGF-β1) and interleukin 1β (IL-1β) have been reported to play an important role in the occurrence and development of primary osteoarthritis. OBJECTIVE:To investigate the association between gene polymorphisms of TGF-β1 and IL-1β and primary knee osteoarthritis. METHODS: Han and Uyghur elderly populations in Xinjiang Uygur Autonomous Region, China were surveyed on the prevalence of osteoarthritis, followed by allotted to osteoarthritis and health groups according the symptoms and radiography. Genotyping TGF-β1-509C/T and -1348C/T and IL-1β-511C/T was performed to analyze the relationship between the gene polymorphisms of TGF-β1 and IL-1β and osteoarthritis. RESULTS AND CONCLUSION:Alleles T and C with genotypes CC, CT and TT were detected in both two groups. In the Uygur population, the genotype frequency of TGF-β1-509C/T and IL-1β-511C/T showed significant difference between osteoarthritis and health groups (P < 0.05). In the Han population, the genotype frequency of TGF-β1-1348C/T showed significant difference between two groups (P < 0.05). These results suggest that the Uygur individuals carrying TT genotype of TGF-β1-509C/T and IL-1β-511C/T, and Han people carrying the TT genotype of TGF-β -1348T are more susceptible to osteoarthritis.
3.Circular RNA FBXO11 regulates the proliferation and apoptosis of gastric cancer SNU-1 cells through targeting miR-376a-3p/SNRPB axis

[Abstract] Objective: To investigate the effect of circular RNA FBXO11 (circFBXO11) regulating the miR-376a-3p/SNRPB (small nuclear ribonucleoprotein polypeptides B gene) axis on the proliferation and apoptosis of gastric cancer SNU-1 cells. Methods: Cancer and para-cancerous tissue samples from 30 patients with gastric cancer who underwent surgical resection were surgically resected in the Department of Oncosurgery, the Affiliated Hospital of North China University of Science and Technology from January 2018 to January 2019 were collected. The positive expression rate of SNRPB protein in gastric cancer tissues was detected by Immunohistochemical staining. The expression levels of circFBXO11, miR-376a-3p and SNRPB mRNA in gastric cancer tissues, gastric cancer cell lines (SNU-1, AGS and HS-746T) and gastric mucosal cell line GES1 were detected by qPCR. Dual luciferase reporter gene assay was used to determine the relationship between circFBXO11 and miR-376a-3p as well as between miR-376a-3p and SNRPB. The si-NC, si-circFBXO11, miR-NC, miR-376a-3p, si-SNRPB, si-circFBXO11+anti-miR-NC, si-circFBXO11+anti-miR-376a-3p, si-circFBXO11+pcDNA-NC, si-circFBXO11+pcDNA-SNRPB were transfected into gastric cancer SNU-1 cells, respectively. CCK-8 assay, Flow cytometry and WB assay were used to detect cell proliferation activity, apoptosis rate and protein expressions of SNRPB, cyclin D1 and C-caspase-3, respectively. Results: Compared with para-cancerous tissues, the expression level of circFBXO11 and the positive rate of SNRPB protein in gastric cancer tissues were significantly increased (all P<0.01), while the expression of miR-376a-3p was significantly decreased (P<0.01). Compared with GES1 cells, the expressions of circFBXO11 and SNRPB were significantly increased, while the expression of miR-376a-3p was significantly decreased (all P<0.01) in gastric cancer cells. circFBXO11 negatively regulated miR-376a-3p expression, and miR-376a-3p negatively regulated SNRPB expression. After inhibiting the expression of circFBXO11 or over-expressing miR-376a-3p or suppressing the expression of SNRPB, the proliferation viability of SNU-1 cells was decreased, and the apoptosis rate was increased (P<0.01). Either inhibiting miR-376a-3p or over-expressing SNRPB could partially reverse the effect of circFBXO11 suppression on proliferation and apoptosis of SNU-1 cells (all P<0.01). Conclusion: circFBXO11 is highly expressed in gastric cancer tissues. Inhibiting circFBXO11 inhibits the proliferation and induces apoptosis of gastric cancer cells, and the mechanism is related to the regulation of miR-376A-3p/SNRPB pathway.
4.Plasmodium falciparum malaria with acute abdominal pain as the first symptom: a case report

Plasmodium falciparum malaria, caused by Plasmodium falciparum infection, is an Anopheles mosquito-transmitted infectious diseases, which predominantly occurs in tropical areas of Africa. P. falciparum malaria is characterized by complex and atypical clinical manifestations, and high likelihood of misdiagnosis and missing diagnosis, and may be life-threatening if treated untimely. This case report presents the diagnosis and treatment of a P. falciparum malaria case with acute abdominal pain as the first symptom.
