1.Effects of TAE and TACE on HIF-1?,MVD and PCNA in rabbit liver carcinoma

Objective To investigate expression and clinical significance of HIF-1?、MVD and PCNA after TAE or TACE for rabbit liver carcinoma,and to identify the mechanism of hepatic carcinoma cell proliferation after the operation.Methods Thirty rabbits were implanted with liver VX2 tumors for 1 week.Then they were randomly divided into 3 groups.Control group had hepatic arterial infusion of 0.5 to 0.8 mL saline.TAE group was embolized with 0.5 to 0.8 ml UFLP.TACE group was embolized with 0.5 to 0.8 ml UFLP mixed with 2 mg THP.Two weeks after treatment,HIF-1?、MVD and PCNA expression was detected.Results Growth of the liver tumor was found in the 3 groups,and there were 15,11 and 3 new tumors found in the control group,TAE group and TACE group respectively.The expressions of HIF-1?and PCNA in control group were higher than those in TACE group,but significantly lower than those in TAE group(P

BACKGROUND: Buqi tongluo prescription, which is characterized by invigorating qi and promoting blood circulation, dredging collaterals and dispersing stagnation, promoting tissue regeneration, and promoting blood circulation to remove obstruction in the collateral, can promote blood circulation at local injured part and ameliorate nutrien of nerve so as to accelerate axonal regeneration of peripheral nerve and functional recovery of nerve conduction.OBJECTIVE: To observe the effect of buqi tongluo prescription of various agents on recovery of nerve function after peripheral nerve injury and compare the results with the combination of vitamin B1 and B6.DESIGN: Randomized controlled animal study.SETTING: Department of Orthopaedics of the First Hospital Affiliated to Guangzhou University of Traditional Chinese Medicine.MATERIALS: A total of 48 Wistar rats of confined clean grade were selected in this study. After modeling on right sciatic nerve, rats were randomly divided into buqi tongluo capsule group, buqi tongluo parenteral solution group, vitamin group and blank group with 12 in each group.METHODS: All rats were cut their right sciatic nerve laterally, and then anastomosis in situ was used immediately to establish models of peripheral nerve injury. Administration began on the third day after operation once a day. ① Rats in buqi tongluo capsule group were perfused with powder of buqi tongluo capsule (including huangqi, renshen, danggui, chuanxiong and danshen which was diluted with saline) of 0.9 g/(kg·d) raw drugs (ig). ② Rats in buqi tongluo parenteral solution group were injected with 0.9 g/(kg·d) buqi tongluo parenteral solution (ip). ③ Rats in vitamin group were perfused with 15 mg/(kg·d) suspension (1.5 g/L) of vitamin B1 and B6.④ Rats in blank group were perfused with 0.01 mL/g saline.MAIN OUTCOME MEASURES: Four rats from each group were selected for 4, 8 and 12 week time points after administration to record bilateral nerve conduction velocity. Recovery rate was regarded as the ration between nerve conduction velocity at the injured side and nerve conduction velocity at the normal side.RESULTS: A total of 48 rats were involved in the final analysis. ① At each time point, nerve conduction velocity in two buqi tongluo prescription groups were faster than that in vitamin group and blank group (P < 0.05, 0.01), and that in vitamin group was faster than that in blank group (P < 0.05). ②At each time point, recovery rate of nerve conduction velocity in two buqi tongluo prescription groups was higher than that in vitamin group and blank group (P < 0.05, 0.01), and that in vitamin group was higher than that in blank group (P < 0.05).CONCLUSION: Both buqi tongluo capsule and buqi tongluo parenteral solution can promote recovery of nerve function after peripheral nerve injury, and the effect is superior to that of vitamin B1 and B6.
4.Effects of adenoviral vector mediated rat AT2R gene transfer on intima proliferation after rat carotid artery injury

Objective To study the effect of rat angiotensin Ⅱ receptor (AT2R) gene transfer mediated by adenoviral vector on neointimal proliferation after rat carotid artery injury. Methods AT2R gene was transducted into the carotid artery by adenoviral vector with GFP after carotid balloon injury. Restenosis models were established in Wistar rats. Immunostaining was performed to examin the expression of AT2R in local arteries. Neointima/media area ratio at day 21 after gene transfer was measured by morphometric analysis. Results The transduction rate of AT2R gene in rat carotid arteries was 40% and the gene expressed constantly in the vessel walls. Neointima/media area ratio of the gene transduction group was significantly reduced (48%) compared with the control group at day 21 after gene transfer. Conclusion The results suggest the possibility that AT2R gene transfer can inhibit proliferation of arterial intima after balloon injury and it is a potential therapeutic approach to prevent neointimal hyperplasia.
5.The immune function injury and its mechanism in drug abuser

Objective To explore the immune function injury and its mechanism in drug abuser.Methods The immune function changes in 50 drug abusers were compared with normal healthy populations by detection of the indexes of subgroups of Th cells, transformation rate of lymphocytes,IgA,IgM,IgG,IgE,compliment C3,C4,IL 1, IL 2,IL 6 TNF and NO.Result In peripheral blood the percentage of Th1 cell, transformation rate of lymphocyte, IgA, IgM, IgG, IgE content, compliment C4,C4, IL 1, IL 2,IL 6,and TNF levels were significantly lower than normal(P< 0.01).The value of Th1/Th2 was lower than normal as well(P< 0.05).NO content was significantly higher than normal(P< 0.001).Conclusion The mechanism of immune function injury in drug abuser might be correlative to direct injury of drugs and their inhibition effect on the thymus hypothalamus hypophysis adrenal axis.

Objective To construct the recombinant adenovirus of Angiotensin Ⅱ type 2 receptor (AT2R), and study the effect of AT2R on prevention of neointimal hyperplasia in rat carotid arteries after balloon angioplasty. Methods The recombinant adenovirus of AT2R with green fluorescent protein was constructed by using the method of homogenous recombination in bacteria. AT2R gene was transduced into rat carotid arteries with pAdCMV/AT2R after the reproduction of rat carotid balloon injury restenosis model. The expression and the transfection efficiency of AT2R gene were evaluated by RT-PCR, immunofluorescence staining, and confocal microscope. The intimal/medial area ratio was measured by digital analysis system. Results The titre of purified recombinant adenovirus was 1.5?10 12pfu/ml, and it was proved by PCR. pAdCMV/AT2R transfection efficiency in carotid artery was about 40% at day14, and localized to the neointima, as well as to the media and the adventitia. pAdCMV/AT2R delivered into injured rat carotid arteries significantly up-regulated AT2RmRNA and protein expression in neointima from day 7 to 21 after injury. Objective: To construct recombinant adenovirus vector of Antisense BTEB2 cDNA and study the effect of antisense BTEB2 on proliferation of vascular smooth muscle cells and neointimal hyperplasia.Methods:BTEB2 cDNA was prepared by RT PCR technique and was subcloned reversedly into shuttle plasmid pDC315 to constructed recombinant shuttle plasmid pDC315AS BTEB2.Then the recombinant shuttle plasmid and adenovirus genomic plasmid pBHGlox?E1,3Cre were cotransfected into 293 cell to obtain recombinant adenovirus. The PCR technique was used to detect target gene fragment and adenovirus genomic characteristic fragment. After the recombinant adenovirus infected vascular smooth muscle cells, antisense RNA expression of BTEB2 was detected by RT PCR. Results:There was recombinant adenovirus containing BTEB2 cDNA in the lysate of cotransfected 293 cells.The recombinant adenovirus infected 293 cells and replicated in 293 cells. The expression of BTEB2 antisense RNA was very obvious in vascular smooth muscle cells after infected by recombinant adenovirus. Conclusion:The construction of recombinant adenovirus vector of Antisense BTEB2 has been achieved, and Antisense RNA of BTEB2 can express in vascular smooth muscle cells. This study has paved the way for furthur research.
