2.Identification of a Pair of Toxin-antitoxin (TA) Gene in the Chromosome of Cyanobacteria Synechocystis sp. PCC6803

Chromosomally encoded toxin–antitoxin (TA) systems are thought to result in growth arrest and eventual cell death upon exposure to environmental stress in E. coli. In the chromosome of cyanobacteria Synechocystis sp. PCC6803, the genetic organization of a 360 bp open reading frame (ORF), slr0664, and another small ORF of 256 bp, ssr1114, is similar to that of TA system. The predicted protein encoded by slr0664 is homologous to RelE, but neither homologue of ssr1114 nor ssr1114-encoding protein was found in TA system. To see whether slr0664 encodes a toxin protein, ssr1114 encodes an antitoxin, an expressing plasmid containing promoter Plac and PBAD, was constructed. In this construct, Both slr0664 and ssr1114 were controlled by Plac and PBAD, respectively. Expression of slr0664 in Escherichia coli results in the inhi-bition of bacterial growth, the expression of ssr1114 neutralize the toxicity of slr0664 expression. These re-sults show that slr0664 is toxin gene and ssr1114 is antitoxin gene, both ssr1114 and slr0664 constitute achromosomal TA system in Synechocystis sp. PCC6803.
3.Effects of FKBP12.6 gene transfection on the contractility of ventricular myocytes of rats with heart failure

Objective To investigate the effects of FKBP12.6 gene transfection on the contractility of ventricular myocytes of rats with heart failure.Methods Rat model of heart failure was established by a surgical operation of abdominal aortic stenosis.The enzymatic hydrolysis was employed to isolate the rats' ventricular myocytes.Mediated by adenovirus the FKBP12.6 gene was transfected into ventricular myocytes.Ad-GFP infected cardiac myocytes from heart failure rats and normal rats were used as control.Western blotting and reverse transcriptase-polymerase chain reaction(RT-PCR) were used to detect the specific overexpression of FKBP12.6.Transfected ventricular myocytes were loaded with the membrane-permeant Ca2+ dye X-rhod-1-AM.Under the condition of field stimulation,the laser confocal microscope in line-scan and plane-scan mode was used to detect the Ca2+ transients and myocytes contraction.Results The mRNA and protein levels of FKBP12.6 in the myocytes of rats with heart failure were significantly lower than that in normal myocytes(0.47?0.08 vs 0.96?0.12,0.25?0.04 vs 0.48?0.07,P
4. Effects of microgravity environment on intracellular signal transduction pathways

Microgravity environment is a stress and extracellular signal that affects cellular morphology and function through signal transduction system, thus leading to certain biological effect. At present, many signaling pathways have been reported to be involved in the regulation of cell function under microgravity environment, such as NF-κB signaling pathway, Notch signaling pathway, MAPK signaling pathway, HSP signaling pathway and so on, and these reports have laid a foundation for the molecular studies of cytolergy under outer space environment. The recent progress in the researches on intracellular signaling pathways affected by microgravity is herewith reviewed in present paper in the hope of providing references for understanding the cell activity in space environment, and to find the ways to alleviate the harmful effects caused by the microgravity environment.
5.Effect of swallowing training combined with acupuncture on dysphagia in nasopharyngeal carcinoma after radiotherapy

ObjectiveTo evaluated the effect of swallowing training and acupuncture on dysphagia in nasopharyngeal carcinoma after radiotherapy.MethodsThirty-eight post-radiotherapeutic nasopharyngeal carcinoma patients were divided into 2 groups: comprehensive group (n=20) were treated with swallowing training and acupuncture therapy, acupuncture group (n=18) were treated with acupuncture only. The patients were assessed with water-swallow test and videofluoroscopy (VFS) before and after treatment.ResultsBoth groups improved their score of water-swallow test after treatment (P<0.01), but the scores of comprehensive group were better than that of acupuncture group(P<0.05).Under the VFS, the cure rate was 70.0% or 38.9% respectively(P=0.089). ConclusionSwallowing training can improve the swallow ability of bulbar palsy patients.
