1.Bone formation of alveolar augmentation with hydroxylapatite, bone morphogenetic protein and fibrin sealant composite

AIM: To investigate new bone formation of alveolar augmentation with hydroxylapatite(HA)+bone morphogenetic protein(BMP)+fibrin sealant FS(HA+BMP+FS) composite.METHODS:Bone morphogenetic protein was extracted from bovine cortical bone. Hydroxylapatite particles were combined with fibrin sealant and BMP, which is of high efficient osteoinduction, forming a new complex material. The alveolar augmentation was made with this composite. The samples were investigated 3, 6,12 and 24 weeks postoperatively, with gross observation, microscope, calculation of new bone formation, tetracycline fluorescent microscope, scanning electron microscope, backscattered electron image and X-ray energy analysis.RESULTS: 1. In experimental group, weave bone had been seen since 3 weeks postoperatively and more new bone 6 weeks postoperatively; remolded bones united and distributed to all spaces of HA particles 12~24 weeks postoperatively. In control group, no new bone appeared 3 weeks postoperatively, only some HA particles contacted base bone 6 weeks postoperatively, a little of new bone grew into HA spaces 12~24 weeks postoperatively. 2. New bone directly incorporated with HA. The quantity of calcium and phosphate are similar to HA and base bone. CONCLUSION:This composite in alveolar augmentation obviously promotes incorporation HA with base bone, speeds formation of new bone and adds amount of new bone, reconstructing a bony alveolar ridge.
2.Effect of radiation on multidrug resistance of tongue squamous cell carcinoma cell line Tca 8113

3.Experimental study on microcosmic changes of compressed premaxillary suture

Objective: To study the microcosmic and pathological changes of compressed premaxillary suture in rabbit models with the cleft in premaxilla.Method: 12 male New Zealand rabbits, at the age of 3 weeks were designed randomly into 3 groups with 4 in each. In group 1and 2 a retropress force of 2.24 and 1.12 N was applied on the maxilla of the animals respectively. The other group was the control. The experimental animals were killed and compressed premaxilla suture specimens were examined by microscopy and electron microscopy 3,6,9 and 12 weeks after operation respectively. Result:In group 1 disturbance of microstructure and degeneration of osteoblasts were observed 3 and 6 weeks after application of the compress force. In control group normal bone structure and development of maxilla were observed. The microcosmic changes in group 2 were similar to those in group 1, but slighter. The microcostructure in the compressed premaxilla tended to be normal 9 weeks after operation. Conclusion:Compress force on premaxillary suture may result in limited pathological changes of the bone during early developing stage.
4.Spatial and temporal features of proliferating vessels during mandibular distraction osteogenesis

Objective:To study the spatial and temporal features of p roliferating vessels during mandibular distraction osteogenesis. Methods :Bilateral mandibles of twelve adult goats were lengthened by 10 mm wit h a custom-made distractor at 1 mm/d, two animals were killed at each intervals . Two additional goats were used as normal control. SEM microvascular corrosion casting was used to examine the spatial and temporal features of proliferative v essels within distraction gap. Results:During distraction perio d, numerous vessels from bone marrow and periosteum were regenerated into distra ction gap and began to form vascular network. In the early phase of fixation per iod, the orientation of vein sinus was in accord with distraction orientation. A long with the time, more vessles were connected, regeneration of vessels vanishe d, and the vascular system within distraction gap became more mature.Con clusion:Angiogenesis during DO is closely related to the osteogenesis; the blood supply of new bone in distraction gap is from both bone marrow and per iosteum.
5.Stage-I functional reconstruction of traumatic deviated nose deformity with chronic rhino-sinusitis

OBJECTIVE To explore the feasibility, surgical method, surgical effect and postoperative complications of stage-I functional reconstruction of traumatic deviated nose deformityaccompanied with chronic rhino-sinusitis.METHODS 37 patients with traumatic deviated nose deformity accompanied with chronic rhino-sinusitis were selected in this study.Methods of stage-I reconstruction included endoscopic nasal septoplasty, functional endoscopic sinsus surgery(FESS), turbinectomy, rhinoplasty and so on.The patients were followed up for one month after operation.The shape of the nose was observed and the nasal bias was measured also.At 3 months after operation, the therapeutic effect about the chronic nasosinusitis was estimated by endoscopic check and patients' subjective symptom.The data obtained preoperatively and postoperatively were treated with statistical methods.RESULTS 37 patients with traumatic deviated nose deformity were selected in this study.The postoperative bias was 2.04 ?0.97 mm and preoperative bias was(6.64?2.21) mm.A t-test revealed the significant difference between pre-and postoperative bias(t=6.374, P

砄bjective: To seek for the method of reinnervation of denervated pectoralis major myoflap. Method:The model of pectoralis major myoflap transposition in 20 New Zealand rabbits was used. After the myoflaps were transferred (original nerve innervation,ON) and ON denervation followed by hypoglossal genioglossus muscle neuromuscular pedicle implantation (NMP) wre performed. The flaps were examined at the intervals of 4,8,12 and 16 weeks respectively after operation. Results:In NMP myoflaps contracted when the hypoglosal nerve was stimulated,some type Ⅱ fibers in the myoflaps transformed into type I, but the pattern of distribution of the muscle fibers displayed mosaic and was similar to that of genioglossus muscle. Conclusion:The denervated pectoralis major myoflap can be reinnervated by hypoglossal-genioglossus muscle neuromuscular pedicle transplantation.
7.Distraction osteogenesis for treatment of cleft palate——LSCM fluorescent labeling quantitive study

Objective:To study the new bone formation after distraction osteogenesis (DO) to repair cleft palate (CP) by laser scanning confocal microscope (LSCM) and quantitive image analysis. Methods: 20 cats were assigned randomly to 3 groups: Experimental group (15 cats), the CP bone defects in hard palate were corrected with DO repair at the rate of 0.4 mm twice a day, till the transport disc (TD) reached the opposite edge across the defect region. The tetracycline labeling (30 mg/kg) were intramuscularly injected 6 days before specimens retrieval of each 3 cats at 2, 4, 6, 8 and 12 weeks after the completion of distraction. The sections were observed and analyzed by LSCM and quantitive image analysis. The results were then compared with empty control (2 cats) and experimental control group (3 cats). Results: The data showed that active new bone formation were observed in 2 weeks and reached the summit in 4 weeks. The bone formation slowed down through 8 to 12 weeks but still significantly stronger than empty and experimental control groups in which no new bone growth were observed. Conclusion:The hard palate bone defect is eventually reconstructed by DO repair and the distraction gap was filled with active de nove intramembranous osteogenesis. A consecutive dynamic curve of fluorescent labeling deposition implies active bone formation and remodeling is revealed.
8.Treatment of orbital floor fracture with orbital floor mesh plate

Objective:To evaluate the effect of orbital floor mesh plate in the treatment of orbital floor fracture. Methods:27 patients with orbital margin fractures were treated by surgical operation with orbital floor mesh plates or medial wall plates. All patients were observed for 3 months after operation.Results:It was observed that all patients with enophthalmos were cured and the patients with nerve injury recovered satisfactorily.Conclusion:Orbital floor mesh plate is effective in the treatment of orbital floor fracture.
9.Study on quality standard for Yanqingsong Granule

AIM: To establish the quality standard for Yanqingsong Granule (Radix P uerariae, Rhizoma Chuanxiong, Radix Paeoniae Alba, Radix Angelicae Dahuricae, et c.). METHODS: Radix Puerariae; Rhizoma Chuanxio ng; Radix Paeoniae Alba; Radix Angelicae Dahuricae. in Yanqingsong Granule were identified by TLC, and the content of puerarin was determined by HPLC. RESULTS: Radix Puerariae; Rhizoma Chuanxio ng; Radix Paeoniae Alba; Radix Angelicae Dahuricae could be identified by TLC. P uerarin showed a good linear relationship at a ran ge of 0.1716~0.8580?g,r=0.9999. The average recovery was 97.49% (RSD= 1.77% and n=6). CONCLUSION: The methods are available with a reproducibility and ca n control the quality of this granule effectively.
10.The effect of thermo-radiotherapy on multidrug resistance in tongue squamous cell carcinoma cell line Tca 8113

Objective To study the effect of thermo-radiotherapy on multidrug resistance (MDR) and levels of intracellular adriamycin (ADM) in tongue squamous cell carcinoma cell line Tca 8113 and its MDR cell line Tca 8113/CBDEA. Methods Samples of the two cell lines were treated with thermo-radiotherapy (42℃ for 0.5 h and 2 Gy of radiation). Four and 24 hours later, the expression of the MDR relative proteins P-glycoprotein (P-gp),multidrug resistance associate protein 1 (MRP1) and glutathione s-tranferase-π (GST-π) were detected using immu-nohistochemistry. Intracellular ADM concentrations were measured using an HTS 7000 Plus bioassay reader. Re-sults No change in the expression of P-gp was observed in the Tca 8113/CBDEA and Tca 8113 cell lines after 4 or 24 hours. Expression of MRP1 was not significantly altered in the Tca 8113/CBDEA cell line, but there was a signifi-cant drop in the Tca 8113 cell line 24 hours post-thermo-radiotherapy. Expression of GST-π was not altered in either the Tca 8113/CBDEA or the Tca 8113 cell line at 4 hours post-thermo-radiotherapy, but there was a significant de-crease at 24 hours. At both 4 and 24 hours, drug tolerance had decreased and intracellular drug concentration had in-creased significantly in both cell lines. Conclusions Thermo-radiotherapy can enhance the effects of chemotherapy and suppress the expression of MDR factors induced by radiation. The combination of hyperthermia and radiotherapy does not induce MDR.
