1.Antitumor efficacy of naked DNA expressed murine IL-12 in a murine hepatocellular carcinoma model

Purpose: To investigate the antitumor efficacy of intra-tumoral administration of plasmid DNA expressing mIL-12 in murine H22 liver tumor models grafted subcutaneously. Methods: Plasmid encoding mIL12 was constructed and examined the expression of cytokine in the eukaryotic cell through enzyme-linked immunosorbent assay (ELISA). The proliferation assay of T lymphoblasts was performed for measuring the biological activity of expressed mIL12. After intra-tumoral administration of plasmid DNA, mean diameters of the tumor mass and survival time were measured in each murine models group. Lactic dehydrogenase ( LDH) assay was performed to examine whether or not treatment with different plasmid DNA could induce systemic cytolytic activity of lymphocytes against parental H22 cells. Histopathological analysis was operated after administration of plasmid DNA vectors in each murine model group. Results: Growth of liver tumor was significantly inhibited( F =4. 10, P =0. 03), and activity of CTL against H22 was enhanced in mIL12 gene therapy group as compared with the control group. In the focal treated with pDC511mIL12 plasmid DNA, inflammatory cell infiltration was more extensive and necrosis was more definite than control group at 1 month after DMA injection. Conclusions: Intra-tumoral administration of plasmid DNA encoding interleukin-12 could inhibit the growth of H22 liver tumor and induce the host antitumor immune response efficiently in the murine model.
2.Study of regional left ventricular longitudinal function in fetuses with gestational diabetes mellitus by velocity vector imaging

Objective To investigate the clinical value of velocity vector imaging in the assessment of regional left ventricular longitudinal myocardial function in fetuses with gestational diabetes mellitus (GDM). Methods Digital dynamic four-chamber views of 98 fetuses with GDM and 135 normal fetuses were collected and analyzed using velocity vector imaging. The regional tissue velocity, strain and strain rate of the interventricular septum and left lateral wall were measured in systole and diastole. Results Two hundred and seven of 233 cases were analyzed successfully. Systolic velocity(Vs) and diastolic velocity(Vd)were age-dependent, Vs and Vd were both gradually decreased from the basal segment to the apical segment in the left ventricle( P <0. 05). The Va after 32+0 weeks and Vd after 28+0 weeks and systolic strain(S),systolic strain rate(SRs) and diastolic strain rate(SRd) in left ventricle of fetuses with GDM after 24+0weeks were obviously lower than the corresponding parameters of normal fetuses ( P <0. 05). Conclusions Velocity vector imaging can evaluate the fetal regional left ventricular longitudinal myocardial function.
3.Quantitative assessment of myocardial segmental systolic function in patients with coronary disease by velocity vector imaging

Objective To investigate the clinical significance of velocity vector imaging(VVI)in evaluating the left ventricular(LV)segmental longitudinal systolic function in patients with coronary artery disease(CAD).Methods In 25 patients with myocardial ischemia,28 patients with myocardial infarction,26 patients with coronary lumen stenosis<50%,according to coronary arteriography and electrocardiogram,the myocardial segments of LV were divided into 4 groups:ischemic segments group,infarcted segments group,non-ischemic segments group and normal segments group.Twenty-eight healthy subjects were selected as control group.Dynamic imaging of all subjects were collected,the systolic peak strain(Smax)and strain rate(SRmax),the time to peak strain(PTs)and the time to peak strain rate(PTsr)were measured respectively.Results Smax and SRmax of the ischemic segments and infracted segments were significantly lower than those of the control group respectively,PTs and PTsr of the ischemic segments and infracted segments were significantly longer than those of the control group respectively.Smax and SRmax of infarcted segments were significantly lower than those of the ischemic segments,there were no differences of PTs and PTsr between ischemic segments and infracted segments.Smax and SRmax cutoff of -14.08%,-0.83 s-1 for detecting ischemic segments and cutoff of -6.65%,-0.38 s-1 for detecting infracted segments,respectively,gave an optimal sensitivity and specificity.Conclusions VVI is a kind of novel noninvasive-tool to quantitatively assess LV regional systolic function in CAD patients.It is competent to differentiate between the ischemic segments and infarcted segments.
4.Clinical application of percutaneous implantation of ~(125)I seeds for treating malignant tumor

Objective To investigate the therapeutic mechanism of ~(125)I seeds based on physical properties, the significance of therapeutic planning system (TPS), the therapeutic means and the clinical efficacy. Methods Eighteen cases with advanced carcinoma were treated through percutaneous implantation of 125 I for interstitial radiotherapy. Results No serious complications occurred in the patients after radiotherapy associated with apparent improved living quality. Two months after therapy, the tumors had shrunk in different degrees with conspicuous decrease in 12, of which 5 tumors disappeared. Conclusions Short-term efficacy demonstrates that intra-tissue radiotherapy of ~(125)I had significant efficacy for advanced tumors.
