1.The clinical significance of K-ras mutation detected in plasma cell-free DNA from patients with colorectal carcinoma

Objective To evaluate the clinical significance of K-ras mutation in plasma cell-free DNA from patients with colorec-tal carcinoma .Methods Plasma cell-free DNA in 212 patients with colorectal carcinoma were extracted and identified by Globin gene .K-ras mutation was amplified by RE-PCR and compared with results of pathological biopsy samples DNA .Results Plasma cell-free DNA were successfully obtained from 212 cases .But no plasma cell-free DNA were found in normal control .Out of 212 ca-ses ,34 cases showed K-ras mutation(16 .00% )in cell-free DNA ,and 33 cases showed positive in pathological biopsy samples DNA (15 .57% ) .It was found two kinds of K-ras mutation on code 12-GGT to GAT ;GGT to GTT .K-ras mutation was not related to tumors′position(χ2 =3 .34 ,P=0 .61);to differentiation degree(χ2 =2 .38 ,P=0 .29);to Dukes staging(χ2 = 2 .84 ,P=0 .38);to lymphoid nodes′metastasis(χ2 =0 .98 ,P=0 .37);to age(χ2 =1 .86 ,P=0 .39) .Conclusion Tumor-derived DNA can be detected in plasma of underlying cancer patients .For colorectal carcinoma patients ,detecting K-ras mutation of plasma cell-free DNA have the same clinical significance as biopsy samples .Moreover ,it′s more convenient and non-invasive .

BACKGROUND: Chronic myeloid leukemia (CML) is characterized by the clonal expansion of hematopoietic stem cells. Without effective treat ment, individuals in the indolent, chronic phase (CP) of CML will undergo blast crisis (BC), the prognosis for which is poor. Therefore, it is important to clarify the mechanism underlying CML from a whole-genome perspec tive. OBJECTIVE: To investigate the gene expression profile of bone marrow mononuclear cells from CML with Applied Biosystems Expression Array System.DESIGN: Observation and controlled analysis.SETTING: Institute of Gene Engineering, Southern Medical University PARTICIPANTS: Samples of two cases of bone marrow (a chronic myeloid leukemia patient and a healthy person).METHODS: This experiment was conducted at the Institute of Gene Engineering, Southern Medical University from October 2004 to September 2005.The total RNAs were extracted and purified from bone marrow mononuclear cells derived from a CML patient and a healthy person. mRNAs were purified using an oligo (dT)-cellulose mRNA purification kits and labeled using reverse transcription, in vitro transcription (RT-IVT), then hybridized with microarray. Gene expression differentiation of the bone marrow mononuclear cells were examined by ABI 1700 Chemiluminescent Microarray Analyzer. Reproducibility of microarray results was assessed by comparing data sets obtained from the same sample and analyzed by two different arrays.MAIN OUTCOME MEASURES: ①Assessment of quality of total RNA and labled cRNA. ②Reproducibility of microarray. ③ Hybridization of array.④Results of semi-quantitative reverse transcription-polymerase chain reaction RESULTS: ①Using statistical data analysis tools, we identified 6 706 genes that were up- or down-regulated in CML patient compared with the healthy person. In these genes, we found that 17 genes were up-regulated while 51 genes were down-regulated among 68 genes closely related to CML. ②most differentially expressed genes in C/EBPalpha mediated path way and CD40L signaling pathway had reduced expression. ③Good repro ducibility of microarray was confirmed by analysis of correlation and detection concordance in technical replicates. The correlation coefficient of the detectable probe in technical replicates was 0.991 for the CML patient and 0.988 for the healthy person. ④The results of semi-quantitative RT-PCR experiments supported the reliability of our microarray analysis.CONCLUSION: By comparing expression patterns of CML with those of the healthy person, we identified a large number of genes that, were up- or down-regulated in CML patients. These data should provide useful information for finding candidate genes whose products might serve as molecular targets for treatment of CML patients.
4.National external quality assessment for HLA low-resolution molecular typing: 2005 -2008 activities

Objective National external quality assessment (EQA) results from 2005 to 2008 for HLA class Ⅰ (A, B) and class Ⅱ (DRB1) low-resolution DNA typing were summarized with the goal of exploring strategies to assure and improve HLA DNA typing performance in clinical testing. Methods HLA allele results from the four consecutive years EQA events were analysed. Different kinds of errors were described and classified, and the possible causes were discussed. Results Participant laboratories were increasing in the four consecutive years with the number of 22, 28, 47 and 61 in 2005, 2006, 2007 and 2008,respectively. 2 844 HLA DNA typings were returned from the participants during the 4 years EQA surveys, and overall 30 errors (1.05%) were identified. These 30 errors were classified into two major types of errors including 25 technical genotyping errors and 5 human errors. The proportion of laboratory participants which made mistakes was 13. 6%, 10. 7%, 10. 6% and 16.4% in 2005, 2006, 2007 and 2008, respectively. Conclusions Above 10% of participant laboratories exhibited errors in the four consecutive years HLA molecular typing EQA surveys. Relevant important attentions should be greatly paid to clinical HLA molecular typing test.
5.Antibacterial and mechanical properties of ceramic orthodontic brackets with nano silver hydroxyapatite coating

BACKGROUND:In the clinical orthodontics, ceramic brackets have deficiencies in the aspects of antibacterial and mechanical properties, which easily lead to the emergence of a variety of adverse events and influence the orthodontic effect. OBJECTIVE:To observe the antibacterial and mechanical properties of nano silver hydroxyapatite coating ceramic brackets. METHODS:The nano silver hydroxyapatite coating ceramic brackets were prepared. Scanning electron microscopy was used to observe the coating surface. Coating antibacterial experiment was conducted. Totaly 50 in vitro human maxilary premolars were randomly divided into two groups (n=25 per group): experimental and control groups. Premolars in the experimental group were bonded to nano silver coating hydroxyapatite ceramic brackets, and premolars in the control group were bonded to ordinary ceramic brackets. The shear strength was detected in these two groups.RESULTS AND CONCLUSION: The overall structure of nano silver hydroxyapatite coating was order, uniform and compact. Hydroxyapatite had a porous structure with a micro-nanometer aperture and there were a large number of nano-silver particles uniformly distributed. Quantitative antibacterial experiments showed that nano silver hydroxyapatite coating ceramic brackets had a strong inhibition to Escherichia coli and Staphylococcus albus, with an antibacterial rate of more than 95%. The shear strength in the experimental group was lower than that of the control group (P < 0.05). These results demonstrate that the nano silver hydroxyapatite coating ceramic brackets have good antibacterial and mechanical properties, which meet the requirement of mechanical change in the clinical orthodontics.

Objective To describe the MR appearancses of growth-plate of the developmental long bones,and to study the correlativity of MRI with the anatomy and histology findings.Methods 40 pigs with normal knees were examined by 1.0 T scanning system with various orientations and sequences,MR imagings were compared with the results of sectional anatomy and histology.Results According to MRI,we divided the evolution of growth plate into four developmental stages;the earlier forming stage of growth plate(aged the 80~95 days of pregnancy),the forming stage(aged the 95 days of pregnancy-2 months),the maintaining stage (aged 2 months-3.5 years),the closing stage(aged 3.5~4.5 years).The MRI manifestations of growth plate of knees reflected the findings of sectional anatomy perfectly and histology to a certain extent in each developmental stage.Conclusion MRI is very useful and helpful in dividing the developmental stages of growth-plate,guessing the foundation of anatomy and histology,diagnosing and differentiating all kinds of growth plate lesions.
9.Determination of Loganin in Tiaojing Zhuyun Granules by HPLC

Objective:To establish a determination method for loganin in Tiaojing Zhuyun granules by HPLC. Methods: HPLC was used with a SinoChrom ODS-BP (250 mm × 4. 6 mm,5μm) as the analytical column. The mobile phase was composed of acetoni-trile-0. 1% phosphoric acid solution(14∶86). The detection wavelength was set at 240 nm and the column temperature was 30℃. Re-sults:The calibration curve of loganin was in a good linearity over the range of 50. 40-1 008. 00 ng(r=0. 999 9). The average recov-ery was 99. 33%(RSD=1. 41%, n=6). Conclusion:The method is simple,feasible and reproducible,and can be used in the quality control of Tiaojing Zhuyun granules.
10.Intra-operative rectal washout can effectively prevent local tumor recurrence: a Meta-analysis

Objective To investigate the effect of rectal washout on prevention of local recurrence in patients with resectable rectal cancer.Methods Relevant studies published on PubMed,Embase database and Google between January 1980 and June 2013 were identified and reviewed.A meta-analysis was performed to compute the risk ratio (RR) and evaluate the effect of rectal washout on local recurrence in patients included in the reviewed studies.Results Eight studies were included in the meta-analysis,in which a total of 427 patients underwent rectal washout and 492 did not during rectal cancer resection.The patients had an overall local recurrence (LR) rate of 6.64 % (61/919).Rectal washout,no matter which type of washout solution was used,significantly reduced the risks of local recurrence (3.75 %,16/427 vs 9.15 %,45/492,RR =0.46,95 % CI =0.26-0.82,P =0.008).Patients with rectal washout showed a low LR rate,when compared with those in the three subgroups who underwent total mesorectal excision (TME) (5.18 %,10/193 vs 5.32 %,10/188).However,there was no statistically significant difference in the risk of LR among the three subgroups (RR =0.99,95 % CI =0.42-2.30,P =0.98; RR =0.44,95 % CI =0.18-1.04,P =0.06; RR =0.46,95 % CI =0.20-1.04,P =0.06,respectively).Conclusions The intra-operative rectal washout largely reduces local recurrence after anterior resection of rectal cancer.Rectal washout should be applied routinely during anterior resection.
