Objective:To study the pharmacokinetic paramet er s of tinidazole in saliva. Methods:Tinidazole was separated on U ltrasphere ODS (C 18 column, 5 ?m,4.6 mm?25 cm)column with a mobile phas e of MeOH-2.33?10 -3 mol/L acetic acid 32∶68(v/v). The sample s were quantified with an ultraviolet detector operated at 310 nm. Resul ts:The saliva T max of the tinidazole tablets was (1.64?0.94) h,C max (69.23?11.39) ?g/ml,K e 0.059 6?0.013 2,T 1/2 (11.952?2.374) h,Auc (83.42?11.49) ?g?h/ml,Cl r 60.64? 8.56 and V r (2 704.45?601.42) ml respectively.Conclus ion:The method can be used for clinical monitoring of tinidazole in sali va.
2.Corelation of PI3-K Phosphorylated Products and Th2 Cytokine in Patients with Active Lupus Nephritis

【Objective】To observe the expression of PI3-K phosphorylated products and elucidate the correlation between PI3-K phosphorylated products and Th2 cytokine in peripheral blood mononuclear cell (PBMC).【Methods】14 patients with active lupus nephritis and 12 controls were selected,PI3-K phosphorylated products were detected by immunoprecipitation and Western blotting,RT-PCR was used to observe interleukin-6 mRNA and interleukin-10 mRNA expression.【Results】In either spontaneous condition or stimulated by anti-CD3 antibody,the expression of PI3-K phosphorylated products in patients with active lupus nephritis were higher than those of the controls(1.14±0.23 vs 0.46±(0.12,P=0.023;2.09±0.63 vs 0.65±0.14,P=0.016).The expression of PI3-K phosphorylated products in active lupus nephritis showed a positive correlation with interleukin-6 mRNA and interleukin-10 mRNA (r=0.652,P=0.008;r=0.718,P=0.007).PY294002,one of specific inhibitor of PI3-K,inhibited significantly the expression of interleukin-6 mRNA(2.32±0.51 vs 0.57±0.15,P=0.009) and interleukin-10 mRNA (1.71±0.33 vs 0.67±0.11,P=0.006) in stimulated PBMC in active lupus nephritis.【Conclusion】PI3-K can involve in the pathogenesis of lupus nephritis by inducing the overexpression of interleukin-6 and interleukin-10.
3. Occupational characteristics of 318 cases diagnosed as occupational asthma

Objective:
To summarize the clinical characteristics of occupational asthma and provide a basis for revising the diagnosis of occupational asthma in China.
Methods:
Taking "occupational asthma" , "occupational bronchial asthma" and "bronchial provocation test at work site" as key words, the case reports of occupational asthma in China were retrieved. The general data, latent period, allergen, clinical manifestation, and diagnostic methods were analyzed.
Results:
A total 318 cases from 14 published literatures were reported.The incidence of male and female is basically similar. The average age of onset is 38 years, and the latent period is from 2 months to 19 years. The top 3 allergens were isocyanates, penicillin and cephalosporins, formaldehyde. During the diagnosis process, 48.8% of the patients were diagnosed by bronchial provocation test, and 19 cases were diagnosed according to the specific IgE antibody. Only 1 case was diagnosed according to the specific skin test.
Conclusion
The etiological diagnosis of occupational asthma is not easy. Nearly half of the patients in our country have been diagnosed as occupational asthma by bronchial provocation test at work site. How to identify the relationship between occupational exposure and the occurrence of asthma is one of the main problems we need to solve.
4.Study of Apoptosis and Gene Expression Pattern Related to Human Degenerative Intervertebral Disc Tissue

ObjectiveTo investigate the relationship between the degenerative mechanisms of lumbar intervertebral disc (LID) and apoptosis.MethodsThe total RNAs were isolated from human LID tissues. Both the mRNAs from the degeneration and normal LID were reversely transcribed to the cDNAs. The cDNAs were labeled with the incorporations of fluorescent dUTP, for preparing the hybridization probes. The mixed probes were then hybridized to the cDNA microarray. After high-stringent washing, the cDNA microarray was scanned for the fluorescent signals and analyzed by computer image analysis. The apoptotic status and the expression of Bcl-2 and Bax in 12 cases of degenerative LID and 10 cases of normal LID were detected with TdT-mediated dUTP-biotin nick end labeling (TUNEL) and immunohistochemistry methods.ResultsAmong the 4096 targets, there were 10 genes related to apoptosis. The expression related to Bax protein gene was up-regulated and it was down-regulated for Bcl-2 protein. In group of normal LID, the average apoptotic index (AI) was (24.897±3.620); percentage of Bcl-2 positive cells was (31.440±4.150)%; percentage of Bax positive cells was (29.372±2.588)%, average optical density (OD) values of positive particles were (0.183± 0.010 ), ( 0.203 ±0.012) and (0.169±0.005) respectively. In group of degenerative LID, the average AI was (49.232±3.440); percentage of Bcl-2 positive cells was (18.239±2.470)%; percentage of Bax positive cells was (52.349±3.764)%; average OD values of positive particles were (0.152±0.003), (0.310±0.008) and (0.262±0.014) respectively. There were significantly differences in AI and expressions of Bcl-2 and Bax proteins between normal LID and degenerative LID (P<0.05).ConclusionCell apoptosis plays an important role in the process of LID degeneration. Both Bcl-2 and Bax take part in the occurrence and progression of LID.
6. Effect of nerve growth factor on chronic peripheral neuropathy in rats induced by 1-bromopropane

Objective:
To observe the effect of nerve growth factor (NGF) and Mecobalamin on chronic peripheral neuropathy in rats induced by 1-bromopropane.
Methods:
36 male SD rats were exposed to 1-bromopropane vapor at concentrations of 4 000 mg/m3, 6 hours per day, 5 days per week for 12 weeks. The rats were randomed divided into 4 groups, and treated by Mecobalamin for 300 μg/kg qd, NGF for 40 μg/kg qd, Mecobalamin+NGF with the dose as mentioned above, respecively. The control group were fed in normal condition. The changes of Sciatic nerve conduction velocity (NCV) , electromyography (EMG) and pathology were observed 30 days later.
Results:
The nerve conduction velocity were decreased in all the rats. Compared with the control group, the motor nerve conduction velocity (MCV) was improved in group Mecobalamin and group Mecobalamin+NGF, The difference was statistically significant, as the sensory nerve conduction velocity (SCV) was improved only in group Mecobalamin+NGF. Sciatic nerve biopsy observed by electron microscope showed that myelinated nerve fibers were obvious swelling, lamellar separation, partial myelin vacuolization, and axonal degeneration. After treatment with exogenous nerve growth factor, the number and severity of damaged nerve fibers were restored.
Conclusion
Exogenous nerve growth factor contributes to the recovery of peripheral nerve damage induced by 1-bromopropane.
7. Comparison of whole blood indium and the serum indium of the population exposed to the indium

Objective:
The method was established for the detection of whole blood indium and serum indium. By comparing the results of two samples, it is possible to explore the significance of whole blood indium and serum indium in the population exposed to indium compounds.
Methods:
According to GBZ/T 295-2017 and GBZ 294-2017, the whole blood and serum samples were diluted 20 times by 0.5% nitric acid solution (including 0.05% Triton X-100) . Under the standard mode of inductively coupled plasma mass spectrometry (ICP-MS) , whole blood indium and serum indium of indirect exposure group, low exposure group and high exposure group in an indium mine were detected with 20 μg/L rhodium standard solution as internal standard.

8.Clinical value of seminal paraoxonase-1 activity evaluation in the diagnosis of male infertility.

OBJECTIVETo investigate the changes in seminal paraoxonase-1 (PON-1) activity in infertile male patients and assess the clinical value of seminal PON-1 examination in the diagnosis of male infertility.
METHODSSeminal PON-1 activity was detected by spectrophotometric method in the semen samples from 270 infertile male patients and 50 health fertile males (control), and the semen parameters were analyzed using a computer-assisted semen analysis system.
RESULTSIn the male infertility group, seminal PON-1 activity was 1.22∓0.76 U/L in the patients with normal semen parameters and 0.64∓0.54 in the patients with abnormal semen parameters, both significantly lower than that of the control group (3.17∓0.89 U/L, P<0.01). In patients with asthenospermia, the declined sperm motility was associated with decreased seminal PON-1 activity, which showed significant differences between patients with mild, moderate, and severe asthenospermia. Seminal PON-1 activity was positively correlated with the percentage of sperm viability (P<0.01), but inversely with the percentage of morphologically abnormal sperm (P<0.01). According to ROC curves, the area of seminal PON-1 activity under the curve was 0.907, showing a statistical significance (P<0.01).
CONCLUSIONThe detection of seminal PON-1 activity can provide a laboratory evidence for the diagnosis of male infertility.

9. Study on the establishment of HMLD model and the expression of KL-6/TGF-beta in rat

Objective:
To establish an animal model of hard metal lung disease (HMLD) in rats, and to screen the indications for diagnosis of HMLD.
Methods:
The rats were randomly divided into 5 groups, each group included 8 rats: saline group, pure cobalt group, pure tungsten carbide group, silica group and hard metal (HM) group. 10 mg subjects were administered in each group by using the pulmonary endotracheal tube. After 8 week, the lung CT scan and lung tissue pathology were observed, the serum and bronchoalveolar lavage fluid (BALF) were collected for KL-6, TGF-beta1 and TGF-beta2.
Results:
The lung tissue structure of HM group was destroyed, a large number of nuclear giant cells and epithelial like cells appeared in the stroma, and uncommon CT scan images appeared in the lung. KL-6, TGF-beta1, TGF-beta2 expression in each group was not the same, the difference was statistically significant (
10.Mental health disparities in people living with human immunodeficiency virus: A cross-sectional study on physician-patient concordance and treatment regimens

Background::Human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS) can profoundly affect the mental health of the people living with HIV (PLWH), with higher rates of anxiety, depression, and sleep disturbances. The disparities in neuropsychological problems evaluated by physicians and self-assessed by patients are still unknown.Methods::A total of 5000 PLWH and 500 physicians from 167 hospitals were enrolled in this cross-sectional study from September 2022 to February 2023. 4-Item Patient Health Questionnaire (PHQ-4) was used for the evaluation of depressive issues and anxiety issues by PLWH. Each physician assessed 10 PLWH under their care for the presence of depressive or anxiety issues. The primary outcomes of this study are the concordance rates on the depressive issues and anxiety issues evaluation between physicians and PLWH. The Cohen’s kappa test was used to assess the agreement between physicians and PLWH.Results::The concordance rate for the evaluation of depressive issues is 73.84% (95% confidence interval [CI]: 72.60-75.04%), and it is significantly different from the expected rate of 80% ( P <0.001). Similarly, the concordance rate for the evaluation of anxiety issues is 71.74% (95% CI: 70.47-72.97%), which is significantly different from the expected rate of 80% as per the null hypothesis ( P <0.001). The overestimation rate by physicians on depressive issues is 12.20% (95% CI: 11.32-13.14%), and for anxiety issues is 12.76% (95% CI: 11.86-13.71%). The mismatch rate for depressive issues is 26.16% (95% CI: 24.96-27.40%), and for anxiety issues is 28.26% (95% CI: 27.02-29.53%). The underestimation rate by physicians on depressive issues is 13.96% (95% CI: 13.03-14.95%), and for anxiety issues is 15.50% (95% CI: 14.52-16.53%). For the treatment regiments, PLWH sustained on innovative treatment regimen (IR) related to a lower prevalence of depressive issues (odds ratio [OR] = 0.71, 95% CI: 0.59-0.87, P = 0.003) and a lower prevalence of anxiety issues (OR = 0.63, 95% CI: 0.52-0.76, P <0.001). PLWH switch from conventional treatment regimen (CR) to IR also related to a lower prevalence of depressive issues (OR = 0.79, 95% CI: 0.64-0.98) and a lower prevalence of anxiety issues (OR = 0.81, 95% CI: 0.67-0.99). Conclusion::Nearly one in three PLWH had their condition misjudged by their physicians. The findings underscore the need for improved communication and standardized assessment protocols in the care of PLWH, especially during the acute phase of HIV infection.
