1.Effect of folic acid and vitamin B12 on the plasma homocysteine levels and neurological function in young and middle-aged acute ischemic stroke patients with hyperhomocysteinemia

Objective To investigate folic acid and vitamin B12 on the plasma homocysteine (Hcy) levels and neurological function in young and middle-aged acute ischemic stroke patients with hyperhomocysteinemia (HHcy).Methods Young and middle-aged acute ischemic stroke patients with HHcy were enrolled.They were randomly divided into either an intervention group or a control group.The patients with cerebral infarction in both groups were treated with conventional treatment.At the same time,the patients in the intervention group were treated with folic acid 5 mg and vitamin B12 25 μg,3 times a day for 4 weeks.Those in the control group were given placebo.The next day after admission and thereafter,the plasma Hcy levels were determined once every week.At the time of admission and after 4-week treatment,the National Institutes of Health Stroke Scale (NIHSS) was used to evaluate neurological deficits.Results A total of 78young and middle-aged acute lschemic stroke patients with HHcy were enrolled (n =39 in each group).The baseline plasma Hcy levels and the NIHSS scores in all patients had significant linear correlation (r =0.717;P<0.01).Four weeks after treatment,the plasma Hcy levels of the intervention group were significantly lower than those of the control group (15.07 ± 2.01μmol/L vs.21.29 ± 2.48 μmol/L; t =4.539; P <0.05),but there was no significant difference in the NIHSS scores (4.87 ± 2.17 vs.5.13 ± 1.67; t =0.585,P =0.560).Conclusions The baseline plasma Hcy level was positively correlated with the severity of stroke.Folic acid in combination with vitamin B12 could effectively decrease the plasma Hey level in young and middle-aged acute ischemic stroke patients with HHcy,but it had no significant effect on the improvement of neurological function.
2.Application value of multifunctional dynamic fiat panel X-ray detector in intravenous pyelography

Objective To investigate the application value of multifunctional dynamic flat panel X-ray detector in intravenous pyelography.Methods Total 540 patients with intravenous pyelography were divided into multifunctional dynamic flat panel X-ray detector group (300 cases) and conventional flat panel X-ray detector group (240 cases) by random digits table method.Image quality,radiation dosage and examination time were analyzed and compared between the two groups.Results There was no statistical difference between the two groups in image quality (P > 0.05).The examination time and radiation dosage was (25.5 ± 8.2) min and (12.5 ± 6.8) mAs respectively in multifunctional dynamic flat panel X-ray detector group and (39.2 ± 12.1) min and (23.6 ± 7.6) mAs in conventional flat panel X-ray detector group.The differences had statistical significance (P < 0.01).Conclusions Multifunctional dynamic flat panel X-ray detector in intravenous pyelography can provide lower radiation dose,shorter examination time and higher image quality.It has obvious advantages in urography.
3.Research on effect of midbrain VTA dopaminergic neurons in general anesthesia

Objective To investigate the effect of dopaminergic neurons of midbrain ventral tegmental area(VTA) in general anesthesia.Methods Forty adult healthy male SD rats were randomly divided into lesion group (n=20) and control group (n=20).The lesion group was given the bilateral infusion of specific dopaminergic neuron injury agent 6-OHDA in midbrain lateral VTA,while the control group received the same volume of normal saline at the same areas.The time of loss of righting reflex (LORR)loss and recovery of righting reflex(RORR)at postoperative 2 week were observed in each group.Results Compared with the control group,the LORR time in the lesion group was shortened and the RORR time was significantly prolonged under propofol-induced anesthesia (P<0.05).However,the LORR time under the isoflurane anesthesia had no statistically significant difference between the two groups(P>0.05),while the RORR time in the lesion group was increased(P<0.05).Conclusion Dopaminergic neurons in midbrain VTA might play different roles in the induction and recovery of different general anesthetics.
4.The study on implicit memory in patients with amnestic mild cognitive impairment

Objective To investigate the characteristics of memory impairment in patients with amnestic mild cognitive impairment (aMCI). Methods Thirty-five patients with aMCI and 35 healthy adults matched with age and education level were administered with a neuropsychological battery of tests including conception and perception implicit priming tasks (category exemplar, picture identification), as well as explicit memory tasks (immediate recall, delay recall, delay recognition ). Results Compared with healthy elders, patients with aMCI were impaired in the conception implicit priming task(t=-4.33, P<0.01), as well as in explicit memory (immediate recall, t=6.40, P<0.01;delay recall, t=9.29,P<0.01; delay recognition, t=7.65,P<0.01),but not in perception implicit priming task (t=-0.78, P>0.05).The conception implicit priming is positively correlated with verbal fluency (r=0.74,P<0.01). Conclusions The present results indicate that patients with aMCI are impaired in both explicit memory and conception implicit priming. The conception implicit priming impairment in aMCI may be related to their frontal lobe dysfunction.
5.Application of Tissue-explant Technique in Culturing Rat Submandibular Gland Cells

Objective To explore the application of tissue-explant technique in culturing rat submandibular gland cell(RSGC)and the characteristics of RSGC.Methods RSGCs were cultured using tissue-explant technique.The cells were purified by enzymatic digestion and differential adhesion.The cell phenotype was identified by cytokeratin-8 immunohistochemical staining.Cellular morphology was observed and photographed under inverted microscope.The cell viability and growth were determined by a double-staining procedure using FDA-PI and MTT assay,respectively.Results Cytokeratin-8 was positive stained in the immunohistochemical staining.The cell viability was more than 95%.The cell growth curve showed that RSGCs were in logarithmic phase since day 5.Conclusion Tissue-explant technique is an easy way to purify plentiful RSGC with normal functions,and it can be used in further research of tissue engineering of submandibular gland.
6.Inhibiting Effect of Ultra-micro Powder Qiweibaizhusan on Bacteria in Vitro

Objective To observe the bacteriostatic effect of single herbs, traditional complex prescription and ultra-micro powder of Qiweibaizhusan. Methods The inhibiting zone and MIC of single herb and compound of Qiweibaizhusan on Escherichia coli, Staphylococcus aureus, Eubacterium aerofaciens, Pseudomonas aeruginosa, Salmonella sp, Saccharomyces cerevisiaes and Candida glabrata were measured by filter paper method. Results The growth of the tested bacteria and yeast except Staphylococcus aureus and Salmonella sp were inhibited by ginseng. The antibacterial effect of licorice was the best, and only Pseudomonas aeruginosa’s growth was not inhibited by licorice. The growth of Staphylococcus aureus and Eubacterium aerofaciens were inhibited by Agastache rugosa. The growth of Escherichia coli, Staphylococcus aureus, Eubacterium aerofaciens and Pseudomonas aeruginosa were inhibited by Poria cocos. Only Eubacterium aerofaciens’s growth was inhibited by Radix aucklandiae and fried Atractylodes macrocephala. The growth of all the bacteria and yeast were not inhibited by Radix puerariae. The growth of Staphylococcus aureus, Eubacterium aerofaciens and Salmonella sp were inhibited by the traditional decoction and ultra-micro powder of complex prescription of Qiweibaizhusan, and all the MIC of ultra-micro powder were smaller than the traditional decoction. Conclusion The main antibacterial component of Qiweibaizhusan was ginseng and licorice. The inhibiting effect of ultra-micro powder on bacteria was better than traditional decoction of Qiweibaizhusan in vitro.
7.Effect of lumbar puncture cerebrospinal fluid replacement on nuclear factor κB in cerebrospinal fluid and prognosis in patients with aneurismal subarachnoid hemorrhage after endovascular embolization

Objective To investigate the effect of early cerebrospinal fluid replacement on nuclear factor-κB (NF-κB) level and clinical outcomes in patients with aneurismal subarachnoid hemorrhage (aSAH) after endovascular embolization.Methods Patients with aSAH received aneurysm embolization were enrolled.They were divided into a cerebrospinal fluid replacement group and a non-cerebrospinal fluid replacement group according to the treatment scheme.All patients were treated with cerebral aneurysm coil embolization within 3 days after admission.The cerebrospinal fluid replacement group performed lumbar puncture cerebrospinal fluid replacement within 24 h after coil embolization,once every other day,20-30 ml of cerebrospinal fluid was replaced each time and 3 mg dexamethasone was injected intrathecally.The NF-κB levels in cerebrospinal fluid were detected at day 1,7 and 14 after the coil embolization.The primary outcome measures were the clinical outcomes determined by the modified Rankin scale (mRS) and the Glasgow outcome scale (GOS) at 3 months after onset.Good outcome was defined as mRS score 0-2 or GOS > 3.The secondary outcome measures included severe complications (hydrocephalus,cerebral vasospasm,cerebral infarction,and rebleeding) and death.Results A total of 81 patients with aSAH received aneurysm embolization were enrolled,including 42 in the cerebrospinal fluid replacement group and 39 in the non-cerebrospinal fluid replacement group.There was no significant differences in the baseline data between the cerebrospinal fluid replacement group and the non-cerebrospinal fluid replacement group (all P >0.05).The duration of neck stiffness in the cerebrospinal fluid replacement group was significantly shorter than that in the non-cerebrospinal fluid replacement group (11.3 ± 3.2 d vs.16.5 ± 3.5 d;t =6.985,P < 0.001).The cerebrospinal fluid NF-κB levels were progressively reduced at day 1,7 and 14 after coil embolization in the cerebrospinal fluid rephcement group and non-cerebrospinal fluid rephcement group (all P <0.05),but the ccerebrospinal fluid levels of NF-κB in the cerebrospinal fluid replacement group at each time point were significantly lower than those in the non-cerebrospinal fluid replacement group (all P < 0.01).The good outcome rates evaluated according to the mRS score (92.9% vs.56.4%;x2 =14.446,P < 0.001) and GOS score (97.6% vs.76.9%;x2 =8.004,P=0.005) in the cerebrospinal fluid replacement group at 3 months were significantly higher than those in the non-cerebrospinal fluid replacement group,and the incidence of cerebral vasospasm was significantly lower than that in the non-cerebrospinal fluid replacement group (14.3% vs.33.3%;x2 =4.086,P =0.043).Conelusiom Cerebrospinal fluid replacement therapy can reduce the incidence of cerebral vasospasm in patients with aSAH receiving aneurysm embolization and improve clinical outcomes.Its mechanism may be associated with the decrease of NF-κB level in cerebrospinal fluid.
8.Investigation on the use of new types of drugs among HIV/AIDS patients in Ningbo City

Objective:
To investigate the use of new types of drugs among HIV/AIDS patients in Ningbo City, Zhejiang Province, so as to provide insights into surveillance and interventions for use of new types of drugs among HIV/AIDS patients.
Methods:
The HIV/AIDS patients in Ningbo City were randomly sampled from the HIV/AIDS Prevention and Control Information System of Chinese Disease Prevention and Control Information System. Patients' demographics, use of new types of drugs and sexual behaviors were collected using questionnaire surveys. Patients' hair samples were collected, and new types of drugs were determined in hair using liquid chromatography-mass spectrometry (LC-MS). In addition, factors affecting the use of new types of drugs were identified using a multivariable logistic regression model.
Results:
A total of 254 HIV/AIDS patients were enrolled, including 214 men (84.25%), 31 cases aged under 25 years (12.20%), 66 cases aged 25 to 30 years (25.98%), and 157 cases aged 31 to 75 years (61.81%). There were 30 cases reported previous use of new drugs (11.81%), including 27 cases reported previous use of methamphetamine (90.00%). There were 48 cases tested positive of new types of drugs, with a positive rate of 18.90%, including 44 cases tested positive for methamphetamine (91.67%). Multivariable logistic regression analysis identified age (25 to 30 years, OR=6.926, 95%CI: 1.412-33.969), occupation (students/teachers/cadres/employees/retirees, OR=6.971, 95%CI: 2.123-22.889; housekeeping and housework servants/unemployed, OR=3.356, 95%CI: 1.289-8.739; business/public place servants, OR=2.447, 95%CI: 1.033-4.448) and syphilis infection during recent six months (OR=3.491, 95%CI: 1.664-7.326) as factors affecting the use of new types drugs among HIV/AIDS patients.
Conclusions
The use of new types of drugs exists in HIV/AIDS patients in Ningbo City. Age, occupation and syphilis infection were associated with the use of new types of drugs among HIV/AIDS patients.
9.Epidemiological investigation of an incident of suspected intentional transmission of AIDS

Abstract
On 18 May 2021, the Center for Disease Control and Prevention (CDC) of X District in P City, Z Province received a co-investigation of a suspected case of intentional HIV transmission from the public security branch, and conducted epidemiological investigations on Zhao and Wang (both males). Wang was confirmed HIV-positive in 2019. Zhao had unprotected sexual encounters several times with Wang in March 2021 without being informed of Wang's HIV infection. Zhao developed fever, sore throat and other symptoms of acute infection phase on 28 March, and were confirmed HIV positive by the CDC of P City on 11 May. Zhao did not have sex with anyone else before or after having sex with Wang. In addition, Zhao had no history of surgery, blood transfusions, drug use or any other history of HIV exposure. Laboratory tests conducted by the CDC of Z Province showed that the HIV nucleic acid sequences between the samples of Zhao and Wang had a high degree of homology. Combined with the epidemiological investigation, laboratory testing and the evidence from the public security branch, it was concluded that Wang intentionally transmitted HIV to Zhao through unprotected anal sex without disclosing his HIV infection status.
10.Optimization of attachment conditions for rabbit mesenchymal stem cells in cytodex 3 microcarrier culture systems.

Mesenchymal stem cells are of great value in tissue engineering and genetic engineering. To study the in vitro dynamic expansion of mesenchymal stem cells by microcarrier technology, we began the research with the investigation of attachment of rabbit mesenchymal stem cells on cytodex 3 microcarriers after inoculation. The result showed a poor attachment efficiency of 16.7% +/- 1.1% under general conditions, so the attachment efficiency must be increased through the optimization of inoculation conditions. Intermittent stirring, inoculation in 50% of the final culture volume and reduction of the fetal bovine serum concentration at inoculation all led to notable increases in attachment efficiency. Ratio of rMSCs attached in aMEM cultures was 39.8% higher than that of DMEM's on the average. When all these optimal conditions were adopted, the attachment efficiency (65.5%) was significantly higher than that (26.6%) under general conditions; accordingly the final growth extents of rMSCs were 4. 50 and 2. 01, respectively. Therefore, the optimization of attachment conditions promoted the expansion of rMSCs on microcarriers.
