1.Observation of estrous cycle in sexual maturation and ovariectomized fe-male ICR mice

AIM:To observe the characteristics of estrous cycle of female ICR mice and ovariectomized mice that had reached sexual maturity stage .METHODS:ICR mice were randomly divided into 3 groups, normal group, sham operation group and ovariectomized group .After ovariectomy and sham operating , at the 8th day, the mice were taken vagi-nal smears twice daily (09:00 and 16:00) for 12 consecutive days.The feature of vulva in the mice and vaginal smear cy-tology, occurrence rate of regular estrous cycle , duration of estrous cycle and each stage duration of estrous cycle were re -corded and observed .RESULTS:The characteristics of estrous cycle in normal group and sham operation group were simi-lar.Nearly 85%of the ICR mice in sexual maturity stage showed regular estrous cycle .The majority manifested 6-day cy-cle (83%), and only a small number exhibited 5-day cycle (17%).Compared with 5-day group, the duration of oestrus was significantly increased in 6-day group (P<0.05).The number and kind of the cells in vaginal smear were a little dif-ferent between 5-day and 6-day groups.The ovariectomized mice which showed irregular estrous cycle (76%) were charac-terized by continuous diestrus .CONCLUSION:Female ICR mice have the strain characteristics of estrus cycle .
2.Investigation research in value orientation during occupational choice and occupational development of nursing students with different background

Objective To understand the characteristic of value orientation during occupational choice and occupational development of nursing students with different background. Methods The study was non-experimental and descriptive with questionnaires. Results The correlation coefficients on 15 work value scales in different background students ranged from 0.561 to 0.940, P value ranged from 0.01 to 0.05. And the coefficients were significant. But there were significant differences on some scales in 15 work value of students who were different in grade, sex, family or areas and so on. Conclusions Nursing stu-dents with different background place emphasis on the same work value and ignore the same others because of the same social-cultural background. But there are some differences in recognition degree of some work val-ue in nursing students with different background.
3.Preparation and identification of Env-pseudotyped subtype B HIV-1

Objective To prepare the Env-pseudotyped subtype B HIV-1 with enhanced green fluorescent protein ( EG-FP) gene,explore HIV-1 infection mechanisms and develop feasible methods of identification .Methods The Env-pseudo-typed viruses were packaged in HEK293T cells by cotransfection, and the reporter gene and P24 protein were detected by PCR, Western blot and ELISA .Reporter gene amplification , viral titration assay and a single round of infection assay were performed after the env-pseudotyped viruses infected HIV-1 permissive cell .Results and Conclusion A generation and identification method of the pseudotyped HIV-1 was established . The Env-pseudotyped subtype B HIV-1 has been prepared, which is able to infect SupT1 and TZM-bl cells through infection assay .
4.Influence of narrative therapy on the family environment and values in adolescents with emotional disorder

Objective To explore the effect of narrative therapy on family environment and values in adolescents with emotional disorder.Methods 35 adolescents with emotional disorder and their parents from Affiliated Brain Hospital,Nanjing Medical University were enrolled and treated by narrative therapy.Family environment was assessed using Family Environment Scale-Chinese Version (FES-CV)and was compared between pre-and posttherapy.Results 28 adolescents with emotional disorder and their parents completed therapy.The cohesion,independence,intellectual orientation,active-recreation orientation and organization in FES-CV showed higher scores at post-therapy than at pre-therapy ((6.89 ± 2.56) vs.(4.21 ± 3.21) ; (5.54 ± 1.35) vs.(4.68 ± 1.51) ;(4.75±2.07)vs.(3.50±2.12);(4.07±2.65)vs.(2.50±2.32);(3.68±1.77)vs.(2.50±1.62)respectively).The contradict in FES-CV showed lower scores at post-therapy than at pre-therapy ((2.75 ± 2.44) vs (5.25 ± 2.50)).The values of adolescents with emotional disorder and their parents had a positive change after narrative therapy.Conclusion Narrative therapy is effective on the positive change of FES-CV and values of adolescents with emotional disorder and their parents.
5.Effect of hemoperfusion on micro-inflammation in patients with maintenance hemodialysis

Objective To observe the effect of hemoperfusion on micro-inflammation in patients with maintenance hemodialysis (MHD).Methods Fifty MHD patients (MHD group) and 25 healthy volunteers (control group) were involved in this study.The MHD patients were divided into two groups by random digits table:hemoperfusion combined with hemodialysis group (HP + HD group,25 cases) and hemodialyais group (HD group,25 cases).The plasma levels of micro-inflammatory cytokines including high-sensitive C-reactive protein (hs-CRP),interleukin-6 (IL-6) and tumor necrosis faetor-α (TNF-α ) were measured before and after treatment.Results The plasma levels of hs-CRP,IL-6 and TNF-α in MHD group were significantly higher than those in control group [(6.72 ± 2.63) mg/L vs.(1.35 ± 0.92) mg/L,(348.83 ± 64.41) ng/L vs.(54.49 ±22.47) ng/L,(7.52 ± 3.17) ng/L vs.(2.53 ±0.88) ng/L](P<0.05).There was no significant difference in the plasma levels of hs-CRP,IL-6,TNF-α before treatment between HD group and HP+HD group (P>0.05),Compared with those in HD group,the plasma levels of hs-CRP,IL-6 and TNF-α after treatment in HP+HD group were significantly decreased [(4.78 ±2.49) mg/L vs.(6.89 ±2.69) mg/L,(260.54 ±56.72) ng/L vs.(357.14 ±56.37) ng/L,(5.36 ±2.41) ng/L vs.(7.49 ±2.87) ng/L] (P <0.05).Conclusion Hemoperfusion improves micro-inflammation in patients with MHD.
6.Ultrasound-guided multiple-spot sector stereotaxis in unipolar cooling cycle radiofrequency ablation of hepatic carcinomas

Objective To assess the effect of ultrasound-guided multiple-spot sector stereotaxis in unipolar cooling cycle radiofrequency ablation of hepatic carcinomas. Methods Ninety-six patients with 112 liver malignancies were treated with ultrasound-guided percutaneous radiofrequency ablation using multiple-spot sector stereotaxis method. Conventional multi-section ultrasound scanning was performed to determine the exact location of tumors. Contrast enhanced ultrasound (CEUS) was performed 15 minutes after treatment to determine whether the tumors were completely inactivated. Contrast enhanced CT (CECT) and CEUS were performed 1 month after treatment to evaluate the therapeutic effect. Results A total of 293 punctures were performed for 112 tumors, technical successful rate with a successful rate of puncture was 91.81% (269/293). CEUS showed no blood flow signals in the tumors 15 minutes after therapy. One month after therapy, CT showed the volume of tumors shrunk, no enhancement was found in 100 tumors (100/112, 89.29%), while partial reinforcement was noticed in 12 tumors (10.71%). Meanwhile, CEUS showed no enhancement in 90 tumors (90/103, 87.38%), when partial reinforcement was noticed in 13 tumors (13/103, 12.62%). Conclusion Ultrasound-guided multiple-spot sector stereotaxis unipolar cooling cycle radiofrequency ablation is an effective and reliable method to treat hepatic carcinomas.
7.Study on rejoin detection mode for unpaid blood donors with HBsAgor anti-HCV positive reactivity

Objective To investigate the detection mode for those unpaid blood donors screening as positive HBsAg+ and/or positive anti-HCV could be permitted to recall again for re-detection under the regulation condition in order to determine whether or not regain their qualifications of donating blood and rejoin the blood donation again.Methods The unpaid blood donors preliminari-ly screening as positive HBsAg and/or positive anti-HCV in Shenzhen from Otcober 2007 to December 2013 and conforming to the rules for recalling to re-detection formulated by our center were analyzed and researched for conducting the feasibility discussion on the rejoin mode of unpaid blood donors.Results A total of 415 759 case-times of blood donation were conducted during 2007 ~2013.Among them,2 506 cases(0.60%)and 1 357 cases(0.33%)were screened as positive HBsAg or positive anti-HCV,respec-tively.The recall process of rejoin re-detection was initiated in 59 positive HBsAg donors and 16 positive anti-HCV donors with many times of blood donation.But only 31 positive HBsAg donors and 9 positive anti-HCV donors successfully completed the detec-tion items of re-detection process.Among them 29 positive HBsAg donor regained the qualifications of donating blood and 2 cases were shielded for the blood donation qualification due to unqualification in the following detection.All of the 9 recalled donators with positive anti-HCV regained the blood donation qualification.Conclusion Under present detection mode,the detection tech-nique of blood screening is hard to avoid the occurrence of false positive results caused by the reagents,instruments and personnel operating.In order to protect the donation qualifications of the unpaid blood donators,a set of scientific,reasonable and practical re-detection mode for rejoin of the blood donators should be established for protecting the limited blood donation resource.
8.Preparation and evaluation of flutide-loaded PLGA nanoparticles modified with cell-penetrating peptides

Objective To prepare and evaluate flutide-loaded PLGA nanoparticles modified with cell-penetrating peptide-TAT.Methods The sequence of TAT was synthesized with florenl methyoxycarbonyl amino acids .The purity and molecular weight of TAT were determined using RP-HPLC and MALDI-TOF-MS.PLGA was modified with the TAT peptide and then prepared into flutide-loaded nanoparticles ( TAT-PLGA NPs) with the double emulsion method .The physical and chemical properties were evaluated , including size distribution, Zeta potential, SEM of nanoparticles , loading ratio of drug content and release profiles of TAT-PLGA NPs in vitro.The cytotoxicity of TAT-PLGA NPs was evaluated by CCK-8 methods.Results The purity of synthesized TAT was 95.6%, and molecular weight was 1495.8.The mean diameter,Zeta potential, drug loading ratio of TAT-PLGA nanoparticals were (159.5 ±2.1) nm, -(1.87 ±0.6) mV, and (5.75 ±0.17)μg/mg, respectively.The nanoparticles observed by transmission electron microscopy (TEM) had a spherical shape and uniform size without aggregation .In vitro release test showed sustained release of flutide from TAT-PLGA nanoparticles .Cell proliferation assay revealed that the TAT-PLGA nanoparticles did not damage the cell growth in vitro and showed good compatibility.Conclusion TAT-PLGA nanoparticles are prepared successfully by double emulsion method,and have sustained-release effect and good compatibility in vitro.They have potential application prospect in prevention and treatment of influenza .
9.Effect of indomethacin on the migration of breast cancer cells in vitro

ObjectiveTo observe the effect of indomethacin on the migration of breast cancer cell line MCF-7 in vitro and investigate the mechanism involved.MethodsThe migration of MCF-7 cell line stimulated with or without indomethacin were tested using transwell plates consisting upper and lower chambers separated by Millipore polycarbonate membrance filters with 8 μm pore sizes; the levels of chemokine receptor 4(CXCR4),cyclooxygenase(COX-2),epidermal growth factor receptor(EGFR) and vascular endothelial growth factor(VEGF)expression in MCF-7 cell line were detected by flow cytometry,Real-time PCR and ELISA,respectively.Results Indomethacin decreased the migration ability of MCF-7 cell line significandy.CXCR4 membrane expression was significantly reduced in a time-dose dependent manner,and CXCR4,COX-2 and EGFR mRNA levels were significantly downregulated after indomethacin stimulation.However,exposure to indometahcin had no major effect on VEGF production of cells.ConclusionThe downregulation of CXCR4,COX-2 and EGFR expression might be the primary mechanism involved in the inhibitory effect of indomethacin on the migration of MCF-7 cell line.
10.A survey on the AIDS-related knowledge of dental undergraduate students in three universities

Objective To understand the present condition of preclinical dental undergraduate in-terns' AIDS-related knowledge. Methods The questionnaire was designed referring to domestic and foreign relevant questionnaires. The cluster sampling method was used to conduct the questionnaire survey among the undergraduates who started their clinical internship in July 2016 in three domestic colleges, using SPSS 14.0 statistical software to analyze it. Results In this survey, 144 questionnaires were distributed, and 144 ones were filled out and sent back. The survey results suggested that 97.22% (140) of interns held a posi-tive attitude towards aiding HIV-infected patients despite that 96.53% (139) of interns knew that dental medical staff was considered to be highly susceptible with HIV. In spite of the fact that most interns knew AIDS-related knowledge and protection measures, 40.28% (58) of interns were not clear about incubation period of AIDS, and the average awareness of children's AIDS and methods to disinfect HIV-infected dental instruments was less than half in the interns. Meanwhile it was found that 86.81% (125) of interns didn't perform well in terms of complying with the principles of confidentiality in AIDS prevention and treatment. Conclusion Dental clinical interns' attitudes towards AIDS-infected patients should be praised. Further-more, they should accept education about infection control to have a deeper understanding about AIDS-related clinical manifestations and legal knowledge in order to reduce the probability of nosocomial infec-tion of AIDS.
