1.Effect of Square Physical Fitness Exercise on Patients with Coronary Heart Disease

ObjectiveTo observe the effect of square physical fitness exercise on the indexes of disease conditions, biology and blood lipids in patients with coronary heart disease (CHD).Methods125 CHD patients were divided into the exercise group (77 cases, taking part in the square physical fitness exercise) and control group (48 cases, not taking part in the square physical fitness exercise). The indexes of biology, disease conditions and blood lipids of all patients in two groups were tested.ResultsAfter exercise, the body mass, body mass index, waist circumference, numbers of ischemia attacks and duration of ischemia attack in patients of the exercise group were obviously lower than those before exercise ( P<0.05~0.01) and also lower than that of the control group ( P<0.05); the average levels of total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterol (LDL-C) in serum decreased and high density lipoprotein cholesterol (HDL-C) increased significantly compared with that before exercise ( P<0.05~0.01), TG, LDL-C were also lower than that of the control group ( P<0.05). In the control group, only indexes of numbers of ischemia attacks, duration of ischemia attack and TG were lower than that in the first test ( P<0.05).ConclusionSquare physical fitness exercise can improve the indexes of disease conditions, biology and blood lipids.
2.Effects of pain on fatigue in patients with rheumatoid arthritis: multiple mediating effects of negative emotions and insomnia

Objective:To understand the relationship between pain, negative emotions (anxiety and depression), insomnia and fatigue in patients with rheumatoid arthritis, and explore the multiple mediating effects of negative emotions and insomnia on pain and fatigue.Methods:A total of 240 patients with rheumatoid arthritis were selected from Northern Theater General Hospital from October 2018 to January 2019. The general questionnaire, Bristol Rheumatoid Arthritis Fatigue Multi-Dimensional Questionnaire,Visual Analogue Scale,The Hospital Anxiety and Depression Scale and the Asens Insomnia Scale were used to investigate the demographic data, fatigue, pain, anxiety, depression, and insomnia, respectively. SPSS 20.0 was used for correlation analysis, and Mplus 7.0 was used for mediation analysis.Results:The fatigue score of patients with rheumatoid arthritis was (28.88±14.89), which was moderate. The score of Visual Analogue Scale was 4(2-7), the score of insomnia was 7(3-11), the score of anxiety was 5(3-8), the score of depression was 7(4-11). There was a positive correlation between fatigue and pain, negative emotion and insomnia ( r values were 0.432-0.950, P<0.01). Negative emotions and insomnia had a partial mediating effect between pain and fatigue, with mediating effects accounting for 56.44% of the total effect. Conclusions:The fatigue of patients with rheumatoid arthritis is moderate; fatigue is associated with pain, negative emotions, and insomnia; negative emotions and insomnia have multiple mediating effects between pain and fatigue.
3.Detection and analysis of platelet-derived growth factor-c and vascular endothelial growth factor in ocular aqueous humor with neovascular glaucoma

Background Neovascular glaucoma (NVG) is a serious eye disease with a variety of causes.It results from the secretion of growth factors by hypoxic retinal tissue,such as vascular endothelial growth factor (VEGF),but anti-VEGF treatment alone dose not completely inhibit neovascularization.Our previous study found that platelet-derived growth factor-c (PDGF-C) is also an important factor in pathological angiogenesis.However,the role of PDGF-C in the development of NVG remains unknown.Objective This study was to quantitatively detect the levels of VEGF and PDGF-C in the aqueous humor of patients with NVG and provide a basis for the anti-angiogenesis therapy.Methods A prospective cohort study was carried out.Sixty-two eyes of 62 patients with advanced NVG and 11 control subjects with age related cataract were included in Xijing Hospital of the Fourth Military Medical University from January 2014 to August 2015.Ten of 62 NVG eyes received retinal photocoagulation and/or cryotherapy,and 16 eyes resulted from central retinal vein occlusion,20 eyes from diabetic retinopathy,5 eyes from postopeartion of retinal detachment,4 eyes from Eales disease and 7 eyes from unknown cause.Iris neovascularization of grade Ⅱ was found in 13 eyes,grade Ⅲ in 29 eyes and grade Ⅳ in 10 eyes.Aqueous humor specimen was collected for 0.1-0.2 ml during the surgery,and the contents of VEGF and PDGF-C in the aqueous humor were detected by ELISA assay.Written informed consent was obtained from each subject prior to any medical procedure.Results The VEGF and PDGF-C concentrations in the aqueous humor were (1 138.17±69.31) ng/L and (29.80±1.64) ng/L in the NVG group,respectively,which were significantly higher than (679.54±49.81) ng/L and (18.60±1.85)ng/L in the control subjects (t=20.95,20.49,both at P<0.01).The VEGF and PDGF-C concentrations in aqueous humor were (1 095.99±52.71) ng/L and (28.55±0.94) ng/L in the retinal photocoagulation group,showing significantly decreases in comparison with (1 146.28±69.57) ng/L and (30.04± 1.64) ng/L in the untreatment group (t =-2.160,P =0.034;t =-2.760,P =0.008).The VEGF content in aqueous humor was positively correlated with PDGF-C content in the patients with NVG (r=0.346,P=0.006).However,the aqueous levels of VEGF and PDGF-C were not significantly different among various primary disease groups and different iris neovascularitation gradings (all at P>0.05).Conclusions Both VEGF and PDGF-C contents in the aqueous humour are considerably elevated in NVG patients.Retinal photocoagulation and/or cryotherapy can inhibit the release of VEGF and PDGF-C in the aqueous humor.PDGF-C may be a target for the treatment of NVG.
4.Progression in the basic research on neovascular glaucoma

Neovascular glaucoma ( NVG ) is a series of blinding and intractable eye diseases, which caused by various intraocular or extraocular diseases. The iris and angle neovascularization and vascular membrane fiber contraction may eventually lead to progressive elevation of intraocular pressure and angle closure. Because of complex etiology, the treatment of NVG is intractable and inefficient. Based on the articles published in recent years, we reviewed the progressions of the relevant cytokines and animal models.
6.Determination of Emodin in Changtong Oral Liquid by HPLC

OBJECTIVE:To determine the content of emodin in Changtong oral liquid.METHODS:HPLC was used.The stationary phase was Nova-Pak C 18 column and the mobile phase was methanol-0.1%phosphoric acid solution(85∶15).The UV detection wavelength was254nm.RESULTS:There was a good linear relationship within the concentration range of0.48~2.4?g/ml of emodin in Changtong oral liquid(r=0.9998).The average recovery was99.73%(n=5),RSD=1.42%. CONCLUSION:This method is simple,fast and accurate.
7.Determination of Microelement Manganese in Changtong Oral Liquid by Atomic Absorption Spectrometry

OBJECTIVE:To establish atomic absorption spectrometry for the assaying of the microelement manganese in Changtong oral liquid.METHODS:The light source was hollow-cathode lamp with wavelength at 279.5nm,the electric current of the light was 3.0mA,the slit-width was 1.2nm,the air flow rate was 5.0L/min and the flow rate of acetylene was 1.0L/min.RESULTS:The calibration curves was in linear correlation with adsorption when the detection concentration range of manganese was 0.5~2.5?g/ml(r=0.9 997),the average recovery was 99.24%(RSD=2.06%,n=5).CONCLUSION:This method is simple,fast,accurate,sensitive and precise.
8.The effect of mutations in the upstream regulatory region of HPV-16 on the activity of virus early promoter

Background and purpose:The incidence of cervical cancer is rather high in Xinjiang, which is closely associated with the infection of human papilloma virus type 16 (HPV-16). The purpose of this study was to analyze the variants and function of HPV-16 upstream regulatory region (URR) in the tissues of cervical cancer biopsies from Xinjiang.Methods:The DNAs were extracted from the tissues of cervical epithelial atypical hyperplasia (CIN) and cervical cancer biopsies. HPV-16URR segments were ampliifed by PCR. Based on the sequence analysis of the URR, the representativeURR variants were selected and cloned into pGL3-Basic. The recombinant plasmids were transfected into Vero cell lines respetively. Luciferase activity of transfected cells was detected 48 h after transfection. Results:Fifty-ifve HPV-16URR DNA fragments were obtained through PCR, and 44 mutations were found from the URR fragments. 4 of these mutations, including nt7192(G→T) , nt7433(- →T), nt7435 (C→G) and nt7863 (A→-) occurred in all sequences. The mutation at nt7520 (G→A) occurred in 54URR sequences, and the 39 other mutations were present in different samples. Based on the location and frequency of the mutations in theURR fragments, 9URR variants were selected and cloned into pGL3-Basic. Then the luciferase activity of the cells transfected with pGL3-URR plasmids was detected respectively. Promoter activity ofURR mutants from cervical cancer are significantly higher than that ofURR mutants from CIN (P<0.01). Promoter activity ofURR fragments from some cervical cancer was signiifcantly higher than that of theURR fragments from SiHa and Caski cells.Conclusion:Multiple mutations occurred in HPV-16URR of cervical cancer patients from Xinjiang. The promoter activity and carcinogenicity of some URR mutants have been enhanced.
10.Effects of miR-30 a on cell cycle and dendritic cell-suppressive function of MSCs

Objective:Mesenchymal stem cells( MSCs) have self-renewal capacity and potential to differentiate into the cells.It was reported that the expression of miR-30a changed in some immune diseases.But it remains unclear the effect of miR-30a on the im-munoregulatory functions of MSCs.Here we studied the impact of miR-30a on the phenotype,cell viability,apoptosis,cell cycle and im-munoregulatory functions of MSCs.Methods: The mixed enzyme methods were used for the isolation of human umbilical cord MSCs.Flow cytometry(FCM)was used to investigate the effect of overexpressed miR-30a on the phenotype of MSCs.CCK-8 was used to examine the cell viability of miR-30a-overexpressed MSCs.Annexin V/PI was used for the detection of apoptosis of MSCs.Q-PCR and Western blot were used to investigate the effect of miR-30a on the expression of Cyclin E2( CCNE2).CCNE2 was one putative target of miR-30a predicted by Targetscan database.The effects of miR-30a-overexpressed MSCs on the maturation of dendritic cells(DCs)were determined.Results:Overexpression of miR-30a blocked the cell cycle of MSCs in the G0/G1 phase by inhibiting the expression of CCNE2,but did not affect the phenotype, cell viability and apoptosis of MSCs.When co-cultured with DCs, although MSCs down-regulated the expression of CD40 and CD86 on DCs,overexpression of miR-30a more significantly enhanced the suppressive impact of MSCs on the maturation of DCs.Conclusion: miR-30a affects the cell cycle of MSCs and enhances its immunosuppressive effect on DCs.
