Objective To evaluate the heterozygous ambiguity resolution primers (HARPs) method in resolving ambiguous genotyping results of human leukocyte antigen (HLA) genes in Chinese Hart population, and choose some appropriate HARPs primers. Methods HLA-A, HLA-B and HLA-DRB1 genes of 416 southern Chinese Han individuals were genotyped by sequence-based-typing(SBT) method and then the ambiguous genotyping samples were sequenced again by HARPs primers provided by American Atria company. Results The percentage of ambiguous genotyping samples resolved by HARPs for HLA-A, HLA-B and HLA-DRBI locus was 86.3% (132/153), 73.9% (130/176) and 38.1% (85/223) respectively. Among them, 48.5% (64/132)HLA-A, 80.0% (104/130)HLA-B and all HLA-DRB1(85/85)samples only need one primer, 47.7 % (63/132)HLA-A and 20.0% (26/130)HLA-B samples need two primers. Three to six different HARPs primers can resolve more than 90% ambiguities. Conclusion HARPs is a convenient method and could be a routine method to resolve ambiguities for HLA-A, HLA-B and HLA-DRB1 genes genotyped by SBT in Chinese Han population.

Objective To investigate the expressions of erythropoietin(EPO)and its receptor(EPO-R)in the spinal cord after acute traumatic injury in adult rats. Methods Sixty-nine Wistar rats were randomly divided into three groups:normal control group(n=5),spinal cord injury group(n=32),and sham operation group(n= 32).The injury group and sham operation group were further randomly divided into eight subgroups respectively (n=4)(1h,6h,12h,24h,3d,7d,14d,28 dafter operation). The expressions of EPO and EPO-R at different time points were detected by RT-PCR,Western blot and immunohistochemical staining. Results EPO was nut detected at any time point in the normal control grnup, spinal cord injury group or sham operation group. The EPO-R expression was not found in the normal control group or sham operation group.RT-PCR and Western blot analyses revealed EPO-R mRNA and protein expressions in the injury group as early as 6 h after injury. The EPO-R mRNA and protein expressions sharply increased at 12 h,peaked at 24 h to 7 d,and gradually declined after 7 d. They were still higher than those in the control rats 28 d after injury. The EPO-R immunoreactivity was chiefly found in neurons, oligodendrocytes, vascular endothelial and ependymal cells. Conclusion The EPO-R expression can be up-regulated obviously in the injured spinal cord, which provides a molecular basis for the nerooprotection of exogenous EPO.

OBJECTIVETo investigate the effects of mannan-binding lectin (MBL) on the functions of human polymorphonuclear cells (PMNs).
METHODSELISA and Dot blot were performed to examine the binding between MBL and the microorganisms. Flow cytometry and fluorescence microscopy were employed to analyze the phagocytosis of FITC-labeled microorganisms by the PMNs. Real-time quantitative PCR was used to detect the expression levels of IL-1β, TNF-α and CD11b mRNA in the PMNs, and ELISA used to detect the levels of TNF-α and IL-6 in the supernatants of PMN culture. Nitro-blue tetrazolium reduction assay was used to estimate the levels of superoxide production.
RESULTSMBL bound to the microorganisms in a dose-dependent manner. MBL had no significant effect on phagocytosis of C. albicans and E.coli by the PMNs in the absence of human serum, but in presence of mixed MBL-deficient human sera, MBL promoted the phagocytosis of C. albicans, which could be blocked by mannan. Mannan treatment increased the expressions of IL-1β, TNF-α, IL-6 and CD11b and enhanced superoxide production in the PMNs.
CONCLUSIONMBL can promote phagocytosis of microorganisms by PMNs and increase the expressions of proinflammatory cytokines from PMNs in a complement lectin pathway-dependent manner.

ObjectiveTo analyze the coping style and its relationship with anxiety status among middle school students aged 13 to 18 in Anhui province during the COVID-19 epidemic. MethodsFrom February 13 to 19, 2020, a cross-sectional survey was conducted among middle school students in Anhui province by using convenience sampling method and network questionnaire. The assessment tools included the Coping Style Scale for Middle School Students (CSSMSS) and the Screen for Child Anxiety Related Emotional Disorders (SCARED). ResultsThe results of the CSSMSS evaluation showed that female students scored higher in emotion abreaction than male students [(8.27±2.98) vs. (7.84±2.91)]. The scores of tolerance [(9.74±2.73) vs. (9.11±2.60)], escape [(7.82±2.79) vs. (7.26±2.44)], emotion abreaction [(8.48±2.97) vs. (7.91±2.93)] and fantasy/denial [(9.79±3.56) vs. (9.26±3.47)] of senior high school students were higher than those of junior high school students, and the score of problem solving [(19.38±4.07) vs. (20.33±4.54)] was lower than that of junior high school students, with statistical significance (P<0.05 or 0.01). Correlation analysis results showed that the scores of tolerance, escape, emotion abreaction and fantasy/deny in CSSMSS were positively correlated with the scores of SCARED of middle school students in Anhui province (r=0.348, 0.287, 0.390, 0.501, P<0.01). ConclusionDuring COVID-19 epidemic, students of different genders and grades in Anhui province have different coping styles, and some coping styles may induce anxiety status.
