1.Expressed Sequence Tags(ESTs) Analysis of Angiostrongylus cantonensis

A total of 1 277 ESTs of Angiostrongylus cantonensis were downloaded from GenBank and analyzed with BlastX.SignalP V3.0 analysis was applied to predict potential putative antigen or allergen relative proteins with N-terminal secreted signal peptides or signal anchors.BlastX analysis showed that there were 614 ESTs scored more than 100, of which 14 were identical with A.cantonensis, 60 ESTs did not match any proteins in the databases.The identified 614 ESTs could be grouped into 10 categories, 80 ESTs expressed 22 antigen or allergen relative proteins, in which 12 had N-terminal secreted signal peptides and 3 had signal anchors.
2.Sequence anlysis of rDNA of Anisakid nematodes with zoonotic potential from Taiwan strait

To analyze the rDNA sequence of the Anisakid nematodes with zoonotic potential from Taiwan straits, Anisakid nematodes were obtained from the gut of marine fish and identified chiefly based on the morphological characteristics. The genomic DNA of nematodes were extracted, and the sequence of rDNA ITS were amplified by PCR using universal primers, then cloned and sequenced bidirectionally. Sequences analysis was conducted by blasting database and software DNAMAN. Results of the blasting database showed that no match could be demonstrated in 3 of 6 rDNA sequences obtained from 6 different nematode species;the new sequences were submitted to the database and the accession numbers in GenBank were obtained. The relationship between adult Contracaecum muraenesoxi and its putative third stage larva was validated by molecular evidences. Sequences relationship based on the inner similarity of the internal transcribed spacer sequences was constructed. It was found that the phylogenetic informative loci were mainly gathered in the second internal transcribed spacer(ITS2) for these anisakid nematodes because of the ITS2 has higher transition ratio than that of the ITS1. It is evident that the sequence in this region could provide valuable information for the molecular distinction of anisakid nematodes with zoonotic potential from Taiwan straits.
3.Influence of renal sympathetic denervation on cardiac function of dogs with heart failure

Objective:To study influence of renal sympathetic denervation (RDN)on cardiac function of dogs with heart failure (HF).Methods:A total of 40 dogs were randomly and equally divided into RDN group [received bilat- eral renal artery radiofrequency ablation (RFA)]and model group (only received femoral puncture).Pacemaker was implanted in every dog,and dog HF model was established using rapid right ventricular pacing.Cardiac and re-nal function indexes,BNP and sympathetic activity index levels were observed and compared between two groups be- fore RFA/sham operation,instant and four weeks after model establishment.Results:After operation four weeks, compared with model group,there were significant reductions in levels of epinephrine (E)[(362.69±42.54)ng/ml vs.(290.36±42.32)ng/ml],renin (R)[(305.46± 39.68)ng/ml vs.(230.04±32.80)ng/ml],aldosterone (AD)[(408.00±38.56)ng/ml vs.(246.00± 48.37)ng/ml],angiotensin Ⅱ (ATⅡ)[(280.00±48.08)pg/ml vs.(172.00±25.04)pg/ml]and norepinephrine (NE)[(425.65±50.54)ng/ml vs.(316.76±46.29)ng/ml]in RDN group (P<0.05 all);there were significant reductions in HR,respiratory rate (RR)and BNP level in RDN group,P<0.05 all;there were significant rise in SBP,LVEF,CO,CI,left ventricular pressure maximal rising rate (+dp/dtmax),left ventricular pressure maximal dropping rate (-dp/dtmax)and left ventricular end-systolic pressure (LVESP),and significant reductions in left ventricular end-systolic dimension (LVESd),left ventricular end-diastolic dimension (LVEDd)and left ventricular end-diastolic pressure (LVEDP)in RDN group,P<0.05 all.Conclusion:RDN can decrease renal sympathetic activity,improve heart function,inhibit myocardial remode- ling,its therapeutic effect is significant
4.Influence of long-term exercise training on carotid intima-media thickness in patients with mild-moderate hypertension

Objective: To observe influence of long-term exercise training on carotid intima-media thickness (IMT) in patients with mild-moderate hypertension. Methods: A total of 92 patients with essential mild-moderate hypertension were randomly divided into routine treatment group (n=46) and exercise group (n=46, received exercise training based on routine treatment) according to number table. All patients were followed up for one year, and the blood pressure, carotid artery diameter and carotid IMT were measured, compared and analyzed in two groups before and after treatment. Results: There was no significant difference in systolic and diastolic blood pressure before treatment between two groups (P>0.05). Compared with baseline value, there were significant decrease in systolic blood pressure [(176.66±l1.78)mmHg vs. (130.89±13.01)mmHg], diastolic blood pressure [(101.43±6.41)mmHg vs. (81.71±8.45)mmHg], carotid artery diameter [(6.62±0.97)mm vs. (6.22±1.01)mm] and carotid IMT [(0.98±0.12)mm vs. (0.84±0.11)mm] in exercise group after one-year training (P<0.05 all), and they were all significantly lower than those of routine treatment group (P<0.05 all ). Conclusion: Long-term exercise training can effectively control blood pressure, decrease carotid artery diameter and carotid intima-media thickness.
5.A preliminary study on the mechanism of lung toxicity of sub-chronic exposure to sodium fluoride in rats

Objective :
To study the relationship between different doses of subchronic sodium fluoride exposure and lung histopathological changes in rats,and to explore the effect and related mechanism of sodium fluoride induced lung tissue injury in rats.
Methods:
Thirty-two Wistar male rats were randomly divided into control,low,medium and high dose groups,each group included 8 rats,intragastric administration of sodium fluoride toxin solution 0, 12,24,and 48 mg / (kg · d) .The experiment lasted for 16 weeks.The incidence of dental fluorosis and weight in each group of rats was recorded,the lung coefficient was calculated,the pathological changes of lung tissue was observed by HE staining,enzyme-linked immunosorbent assay(ELISA) was used to detect the content of superoxide dismutase (SOD) and malondialdehyde (MDA) levels in rats serum,apoptosis level of lung cells was assessed by TUNEL staining,and the Caspase-3 protein expression levels in lung tissues were determined by immunohistochemistry.
Results:
At the end of exposure,the upper and lower incisors of rats in different dose groups showed different degrees of dental fluorosis.The increase in body weight of the rats in the high-dose group was lower than that in the other three groups (P<0. 05) .The coefficient of lung organs and the content of MDA in serum of the rats in the high-dose group were higher than those in the control group (P<0. 05) .The rats in the low,medium and high dose groups had different degrees of pulmonary interstitial inflammatory infiltration and alveolar morphological changes.Compared with the control group,the lung cell apoptosis rate and Caspase-3 protein expression of the rats in the low ,middle and high groups significantly increased ,and the differences were statistically significant ( P < 0. 05) .
Conclusion
Subchronic exposure to sodium fluoride can cause the lung tissue damage in rats ,and the mechanism may be related to the apoptosis induced by oxidative stress.
