1.Interaction between regulatory proteins related to the activation of eNOS and CYP4A/20-HETE

Aim To investigate the interaction between regulatory proteins related to the activation of eNOS and CYP4A/20-HETE.Methods The endothelia of pulmonary artery of new born bovine were cultured and divided into four groups: controlling group: ethanol solvent as vehicle (the volume is the same with 1?10~ -6 mol?L~ -1 20-HETE) was added to culture dish and incubated for 10 min; 20-HETE 5 min treatment group:1?10~ -6 mol?L~ -1 20-HETE was added to culture dish and incubated for 5 min; 20-HETE 10 min treatment group:1?10~ -6 mol?L~ -1 20-HETE was added to culture dish and incubated for 10 min; VEGF 10 min treatment group: 1?10~ -8 mol?L~ -1 VEGF was added to culture dish and incubated for 10 min. CYP4A,eNOS and proteins related to the activation of eNOS were measured by Immunoprecipitating and immunobloting.Results Expressions of eNOS and phospho-eNOS(Ser1177)in PAECs were upregulated after the cells were treated with 20-HETE, and similar phenomena were observed when 20-HETE was replaced by VEGF; 20-HETE increased the binding of Hsp、Akt and eNOS.Conclusions ①20-HETE phosphorylates eNOS at Ser1177 site which activates the eNOS to catalyze L-aginine to produce nitric oxide.②eNOS binds with Hsp90 and Akt in endothelial cell of pulmonary artery and 20-HETE increase the binding. ③eNOS binds with CYP4A in endothelial cell of pulmonary artery.
2.Research progress in the mandibular molar implant restoration with frontal resorption

Mandibular molar loss, which is in a high ratio, is found accompanying with alveolar bone resorption frequently while being restored in clinic. Therefore, due to the existence of mandibular neural canal, the success ratio of implant denture in this area can be directly affected by the height reduction of alveolar bone. In clinic, there are some achievements obtained to improve the survival rate of implant denture, such as increasing the implant support area, enhancing alveolar bone height through surgery, optimizing the design of the superior part of implant. This article summarizes the research progress on the mandibular molar implant denture with alveolar bone absorbed and aims to search a selective design of mandibular molar implant denture under the high absorption of alveolar bone.
3.Correlation of apolipoprotein H exon-8 polymorphism with coronary atherosclerotic heart disease and lipometabolism

BACKGROUND: Human apolipoprotein H which characterizes by polymorphism is related to metabolism of triacylglycerol (TG) and function of platelet; therefore, it is suspected that it is possibly related to coronary atherosclerotic heart disease (CAHD).OBJECTIVE: To investigate the correlation among apolipoprotein H(ApoH) exon-8 polymorphism with CAHD and its effect on lipometabolism.DESIGN: Case-controlled observation.SETTING: Department of Cardiology, Haici Medical Group, Qingdao,Shangdong. PARTICIPANTS: 110 CAHD patients selected from Qingdao Haici Medical Group were regarded as CAHD group, and other 100 healthy subjects were regarded as control group.METHODS: 2 mL blood was collected from peripheral vein of all cases. Polymerase chain reaction and technique of restriction fragment length polymorphism were used to determine the genotype of ApoH exon-8; meanwhile, lipids were measured with oxidase technique.MAIN OUTCOME MEASURES: ① polymorphism of ApoH exon-8 genotype; ② gene frequency of ApoH exon-8 (Try316Ser); ③ comparisons of lipid level of cases with various genetypes.RESULTS: All 210 cases were involved in the final analysis. ① G1025C (Try316Ser) existed in ApoH exon-8, including types of GG and GC, butnot CC type. ② The frequency of GC in the CAHD group was 25.5%, and the frequency of C allele was 0.13; they were significantly higher than those in control group (10%, 0.05, P <0.05). ③ TG level in genotype GC of CAHD group was significantly higher than that both in GG genotype and in any genotype of control group [(1.38±0.24), (1.16±0.10),(1.09±0.78), (1.12±0.76) mmol/L, P < 0.05]. Level of low-density lipoprotein cholesterol (C-LDL) was higher in CAHD group than that in control group (P<0.05).CONCLUSION: The polymorphism of ApoH exon-8 is closely related to CAHD and TG level.
4.Mandibular first molar implant finite element models established by reverse engineering method

BACKGROUND: Reverse engineering modeling has been widely used in previous studies. However, there are a few studies on implantation modeling, especially in aspects of mandibular molar implantation. The mandibular molar has high miss rate in dentition. Thus, to establish three-dimensional finite element models of a jaw bones with mandibular molar implant denture using reverse engineering shows a great parameter value for research of mandibular molar implantation repair. OBJECTIVE: To establish a three-dimensional finite element model of a mandibular molar with dental implant inserted in mandibular molar area using reverse engineering, and to provide mathematical model basis for biomechanics researches of stomatology.METHODS: Establish three-dimensional model, divide mesh and calculate by Solidworks 2007 through scanning mandible by screw jaw using helix CT, obtaining data from images, separating data, rebuilding three-dimension using Mimics software,optimizing the design of mesh by Magics software, and dealing with solid model by Pro/E preprocessor.RESULTS AND CONCLUSION: A three-dimensional finite element model of mandibular first molar implant with regular depth was established, which has high geometrical similarity, good properties of mechanics and mesh, and can be easily modified. This can provide excellent model support for studying the stress analysis of different designs on restoring mandibular first molar implant.
5.Recombinant PML adenovirus suppresses human gallbladder cancer cell growth by inducing apoptosis

Objective To explore the molecular mechanism by which promyelocytric leuxemia (PML) suppresses human gallbladder cancer cell line (GBC-SD) growth.Methods GBC-SD cells were infected by green fluorescent protein recombinant adenovirus (Ad-GFP), GFP-positive cells were examined by microscopy.Cultured gallbladder carcinoma cells (GBC-SD) were infected with Ad-PML or Ad-control.Cell death was detected by DNA laddering and TUNNEL analysis.Cell cycle was analyzed by flow cytometer.Results An infection efficiency of 100% can be achieved at a concentration of 100 multiplicity of infection (MOI).The growth rate of the Ad-PML-infected GBC-SD cells was significantly inhibited.DNA laddering was detected at 72 h post-infection.The amount of apoptotic cells significantly increased in the Ad-PML-infected GBC-SD cells without evident alterations in cell cycle distribution.Conclusion PML suppresses growth of human gallbladder cancer cell line GBC-SD by inducing apoptosis.
6.MECP 2 duplication syndrome:a pedigree report and literature review

Objective To explore the clinical features and the gene mutations in MECP 2 duplication syndrome. Methods The clinical data of a child with developmental retardation and hypophrenia accompanied with respiratory tract infection was analyzed retrospectively. Microarray analysis technique was used to detect the genes in the patient and his family. The pertinent literature was reviewed. Results A 1-year and 7-month old boy was found to have hypotonia, developmental delay, and recurrent respiratory tract infections after birth. Microarray analysis showed a duplication of 441.88kb in Xq28 area and diagnosis of MECP2 duplication syndrome was confirmed. His grandmother, mother, and two aunts were found duplication of 441.73-441.88kb in Xq28 area, all of whom were MECP2’s female carrier. Conclusions The improvement of chromosome chip technology inspection is helpful to the early diagnosis of MECP2 duplication syndrome.
7.The effects of hyperbaric oxygen therapy on hypoxic-ischemic brain damage in newborn rats

Objective To track the effects of hyperbaric oxygen (HBO) therapy on neonatal rats with hypoxic-ischernic brain damage (HIBD).MethodsA total of 126 healthy,seven-day-old SD rats were used to model the HIBD and then randomly divided into seven groups of 18:a sham group,a hypoxic-ischemic (HI) group,a 6 h HBO group,a 24 h HBO group,a 48 h HBO group,a 72 h HBO group and a 1 week HBO group.One hour of HBO treatment at 2 atmospheres absolute pressure was administered once daily for a week to the rats in the latter 5groups,starting at 6,24,48,72 hours and 1 week post the HIBD modeling,while no HBO was administered to the sham and HI groups.The effects were assessed in terms of histological changes ( the neuron density and the percentage of neuron apoptosis in the CA1 region of the hippocampus) and nitric oxide (NO),malondialdehyde (MDA) and superoxygen dismustase (SOD) concentrations after 15 days.ResultsIn the 6 h HBO,24 h HBO,48 h HBO and 72 h HBO groups,average neuron density in the CA1 region was significantly higher than in the HI group.But in the 1 week HBO group the average density was not significantly different than in the HI group.In the 6 h HBO,24 h HBO,48 h HBO and 72 h HBO groups the average percentage of neuron cell apoptosis in the CA1 area of the hippocampus was significantly lower than in the HI group,but the 1 week HBO group again showed no significant difference.There were significant differences in average NO,MDA or SOD levels among the 6 h HBO,24 h HBO,48 h HBO,72 h HBO and HI groups,but the 1 week HBO group showed no significantly higher average levels than the HI group.ConclusionsThe optimal therapeutic window for using HBO to protect against HIBD is within the first week.The best effect can be obtained in the first 6 hours,but after 1 week HBO no longer has a significant effect.The earlier the HBO is administered,the better the effect obtained.
8.Relationship between β-fibrinogen gene - 455G/A,- 148C/T polymorphisms and atrial fibrillation with cerebral infarction

Objective To investigate the relationship between β-fibrinogen (β-Fg) gene -455G/A,- 148C/T polymorphisms and atrial fibrillation (AF) with cerebral infarction.Methods Polymerase chain reaction-restriction fragment length polymorphism was used to detect the genotypes of -455G/A and -148C/T in 97 patients with AF with cerebral infarction (AF with cerebral infarction group),80 patients with AF alone (AF group),and 98 health subjects (healthy control group).Results The A allele frequencies of -455G/A in the AF with cerebral infarction group and the AF group were 0.304 and 0.344 respectively.They were significantly higher than 0.179 in the healthy control group (all P<0.05).The T allele frequencies of - 148C/T in the AF with cerebral infarction group and the AF group were 0.348and 0.369,respectively.They were significant higher than 0.240 in the healthy control group (all P<0.05).Conclusions The A allele of -455G/A and the T allele of -148C/T were associated with AF with cerebral infarction and AF.
9.Effect of Transcutaneous Electric Nerve Stimulation on Flaccid Cerebral Palsy

Objective To observe the therapeutic effect of transcutaneous electric nerve stimulation on children with flaccid cerebral palsy. Methods 40 children with flaccid cerebral palsy were divided into 2 groups: treatment group (n=20) and control group (n=20). 2 groups were given conventional rehabilitation training for 30 d, while the treatment group added transcutaneous electric nerve stimulation on quadriceps femoris for 30 d. Quadriceps femoris was assessed by Modified Lovett classification, gross motor function was assessed by Gross Motor Function Measure (GMFM), and integrated electromyogram (iEMG) and root mean square (RMS) were recorded. Results There was significant improvement in 2 groups in Modified Lovett classification, GMFM, and iEMG and RMS (P<0.05) while the treatment group was better than the control group (P<0.05). Conclusion Transcutaneous electric nerve stimulation can enhance the muscle tone and strength of quadriceps femoris to improve gross motor function for children with flaccid cerebral palsy.
10.Relationship of age with carotid artery plaques and luminal stenosis

Objective To study the relationship of age with carotid artery plaques and luminal stenosis.Methods One hundred and sixty-five patients who underwent carotid artery ultrasonogra phy in our hospital were divided into ischemic stroke (IS) group (n=100) and IS-free group (n=65) according to their clinical symptoms and ultrasonographic findings.Risk factors for their IS were analyzed.The patients were further divided into ≤59 years old group (n=36),60-79 years old group (n=98) and ≥80 years old group (n=31).Their carotid artery stenosis (CAS) and plaques were analyzed.Results The age was significantly older,the incidence of hypertension,CAS and vulnerable plaques was significantly higher in IS group than in IS-free group (70.6±11.2 years vs 65.1±12.8 years,P=0.004;79.0% vs 60.0%,P=0.008;40.0% vs 24.6%,P=0.041;20.0% vs 6.2%,P=0.014).The detection rate of stable and vulnerable plaques was significantly higher in 60-79 years old group and ≥80 years old group than in ≤59 years old group (P<0.01).Multivariate logistic regression analysis showed that age was a risk factor for IS,CAS and vulnerablelaques(P=0.000,P=0.023).Conclusion Age is a risk factor for IS.The number of carotid artery vulnerable plaques is greater and the incidence of moderate and severe carotid artery luminal stenosis is higher in IS patients than IS-free patients.The number of carotid artery vulnerable plaques is greater in ≥60 years old patients than in ≤59 years old patients.The incidence of CAS is the highest in ≥80 years old patients who therefore have a higher risk of IS.
