2.DNA super-barcoding of several medicinal species in Gentiana from Yunnan province.

Gentiana is an important but complicated group in Gentianaceae. The genus covers numerous medicinal plants which are difficult to be identified. In the present study, several medicinal species in Gentiana from Yunnan province, including G. rigescens, G.rhodantha, and G. delavayi, were sequenced using the Illumina HiSeq 2500 system. Three complete chloroplast genome sequences were obtained after assembly and annotation. According to several published genome sequences of G. crassicaulis, the DNA super-barcoding of species in Gentiana was preliminarily carried out. The results revealed that chloroplast genomes of the three species were conservative with short lengths(146 944, 148 992, and 148 796 bp, respectively). The genomes encoded 114 genes, including 78 protein-coding genes, 30 tRNA genes, 4 rRNA genes, and 2 pseudogenes. Furthermore, these medicinal species in Yunnan province were identified using DNA super-barcoding based on chloroplast genomes. The results showed that the Gentiana species could be gathered into monophyletic branches with a high support value(100%). It indicated that DNA super-barcoding possessed obvious advantages in discriminating species in complicated genera. This study is expected to provide a scientific basis for the identification, utilization, and conservation of Gentiana species.

5.The diagnosis and surgical treatment of primary gastric lymphoma

Objeetive To probe the clinical diagnosis and surgical treatment of primary gastric lymphoma (PGL). Methods Clinical data of 23 PGL patients identified by postoperative pathology were analyzed retrospectively. Sixteen patients underwent subtotal gastrectomy,3 patients underwent total gastrectomy,2 patients underwent palliative resection and 2 patients underwent exploratory laparotomy only. Postoperatively 21 patients received adjuvant treatment(chemotherapy and/or radiotherapy).The variables analyzed were type of surgery, histological type in accordance with Kiel's classification, Involvement of lymph nodes. Ann Arbor stage classification. Results The overall 5-year survival rate of these patients Was 80%,that of low grade malignancy patients was 90%,and of high grade malignancy was 40%.Thirteen patients were classified as stage Ⅰ E and seven as stageⅡE and three as stageⅢor Ⅳ,the 5-year survival rate was 90%,67%and 0,respectively.Nineteen patients underwent radical resection with 5-year survival rate of 92.3%.No patients undergoing palliative resection have survived more than 5 years. The prognosis of PGL with low grade malignancy and that of early stages(IE and Ⅱ E)and those undergoing radical excision was better than those with high grade malignancy, that of advanced stage(Ⅲand Ⅳ),and that undergoing palliative resection. Conclusions Preoperative diagnosis relies mainly on gastroscopy with biopsy and CT scan. For patients with early stage disease,radical resection combined with adjuvant therapy is the key factor in improving the prognosis. Chemotherapy or/and radiotherapy is useful management for patients with advanced stage disease.

7.Study on the Mechanism of Periplaneta americana Extract Reversing Multi-drug Resistance of Human Hepato- cellular Carcinoma HepG 2/ADM Cells

OBJECTIVE:To study the mechanism of Periplaneta americana extract degreasing cream and CⅡ-3(shorted for “degreasing cream ”and“CⅡ-3”)reversing the multi-drug resistance of human HepG 2/ADM cells. METHODS :MTT assay was used to investigate the toxicity effects of different concentrations of sorafenib (positive control ),degreasing cream and C Ⅱ-3 on HepG2/ADM cells ,then IC 20 was calculated. The experiment was divided into sensitivity drug ,drug-resistance group ,sorafenib group,degreasing cream group and C Ⅱ-3 group. HepG 2 cells were included in sensitivity group ,and HepG 2/ADM cells were included in the latter 4 groups. Sensitivity group and drug-resistance group were treated with routine medium ,and other 3 groups were treated with relevant medicine (IC20 as drug concentration ). The content of ADM in HepG 2/ADM cells was determined by Laser scanning confocal microscopy. The expression of apoptosis-related protein as Bcl- 2 and Cleaved-Caspase- 9 p37 were detected by Western blotting assay. RT-qPCR and immunocytochemistry were adopted to detect mRNA and protein expressions that related to multidrug resistance [P-gp (expression produce of MDR1 gene),LRP,BCRP] and that related to enzyme-mediated multidrug resistance pathway (GST-π and Topo Ⅱ). RESULTS :The IC 20 of degreasing cream ,CⅡ-3 and sorafenib were (2.40±0.16), (200.44±27.52),(18.00±1.82)μg/mL,respectively. Compared with sensitivity group ,the protein expressions of Bcl- 2,P-gp, LRP,BCRP and Topo Ⅱ,the mRNA expressions of MDR 1, LRP,BCRP and GST-π were increased significantly in drug resistance group (P<0.05 or P<0.01). Compared with @qq.com drug-resistance group ,the mRNA and protein expression of MDR1 mRNA and LRP ,BCRP,GST-π were significantly decreased in degreasing cream group and C Ⅱ-3 group(P< 0.05 or P<0.01);the protein expression of Bcl- 2 and the mRNA expression of Topo Ⅱ were significantly decreased (P<0.01), while the protein expression level of Cleaved-Caspase- 9 p37 was significantly increased in C Ⅱ -3 group (P<0.05). CONCLUSIONS:Degreasing cream and C Ⅱ-3 can reverse multidrug resistance of HepG 2/ADM cells by reducing drug efflux , promoting cell apoptosis ,reducing the mRNA and protein expression of multi-drug resistance gene as well as gene in enzyme-mediated multi-drug resistance pathway. The effect of C Ⅱ-3 is better than that of degreasing cream.
8.Clinical analysis of 24 patients with POEMS syndrome

ObjectiveTo analyze the characters of POEMS syndrome, raise physician awareness of diagnosis and therapy,and explore the optimal treatment strategies.MethodsThe clinical features,laboratory examination and therapy of 24 patients with POEMS syndrome were analyzed,and relative literatures were reviewed. ResultsA strong predominance of male over female was found, 18 vs 6. All patients were over 40 years old,with a mean age 56.5 years old,indicating a common adult involvement.All presented with polyneuropathy,which was also the most common complaints when admitted,which reminded neurologists of underlying possibility of a rare plasma cell disorder.Organomegaly was found,including 19 cases with hepatomegaly,17 patients with splenomegaly.Endocrinal abnormalities were also found in 15 cases.18 patients were M protein positive. Skin pigmentation was recognized in 21 cases. Melphalan in combination with prednisone was applied and 100 % response was observed. One of the patients received peripheral blood stem cell transplantation(PBSCT)2 years ago and a durable response was observed with a continuous complete remission of polyneuropathy and the absence of M protein following 2 years post PBSCT.ConclusionPOEMS syndrome is a rare multisystem disorder, the combination of symptoms and signs is highly complex,which is easy to misdiagnose or missed diagnose.A polyneuropathy with either organomegaly,endocrinal abnormalities, skin disorder or serositis is required a further investigation of clonal M protein and bone marrow.Melphalan and prednisone probable are the optimal regimens.PBSCT provides a new choice for therapy and research of POEMS syndrome.
9.Effect of Niaochangshu Capsule on Urodynamics of Patients with Post-menopause Urethral Syndrome

Objective To investigate the effect of Niaochangshu capsule on urodynamics in bladder filling period of patients with post-menopause urethral syndrome of renal deficiency and blood stasis type.Methods A total of 36 patients with post-menopause urethral syndrome of renal deficiency and blood stasis type were randomly assigned to two groups:the trial group(24 patients were treated with Niaochangshu capsule) and the control group(12 patients were treated with Nilestriol).All patients were treated for 8 weeks.The changes of the urodynamic indexes in bladder filling period(the capacity of the bladder,the compliance of the bladder and the sensitivity of the bladder) were compared between the two groups.Results After treatment,the urodynamic index,including the maximum cystic capacity,the volume at the first desire to void and the cystic compliance were evidently improved in both of the trial group and the control group(P0.05).Conclusion Niaochangshu capsule is effective in treating the patients with post-menopause urethral syndrome of renal deficiency and blood stasis type.It can increase the cystic capacity and compliance,step down the sensitivity of the bladder.Its effect is similar to Nilestriol.
10. Quality evaluation and quality equivalence of Gentiana rigescens Franch. from different populations

OBJECTIVE: To establish evaluation methods of the quality and quality equivalence of Gentiana rigescens Franch. from different populations. METHODS: The contents of the active ingredients, gentiopicroside, swertiamarin and sweroside in the roots of Gentiana rigescens Franch., were determined by HPLC, and the HPLC fingerprint was established. The quality and quality e-quivalence were evaluated by One-way ANOVA, cluster analysis and principal component analysis (PCA). RESULTS: The overall quality of Gentiana rigescens Franch. from different populations in Dali Yunnan was better. The content of gentiopicroside in the sample from Weishan population was lower than those in the other populations, while it was more than twice the national standard. There was significant difference (P<0.01) in the contents of gentiopicroside, swertiamarin, and sweroside in the roots of Gentiana rigescens Franch. from different populations. The quality equivalence of Gentiana rigescens Franch. from different populations was analyzed, and 11 common peaks in HPLC fingerprint were selected to evaluate the similarities of the crude drugs. The similarities of the characteristic peaks were all above 0.988. The three characteristic peaks of gentiopicroside, swertiamarin and sweroside which were related to the activity of Gentiana rigescens Franch were identified, and the qualities of different populations were basically equivalent. The total contents of gentiopicroside, swertiamarin and sweroside of Gentiana rigescens Franch. from different populations were similar as shown by cluster analysis and PCA of HPLC fingerprint. CONCLUSION: Jianchuan and Heqing polulations are quality germplasm populations according to the contents of effective components and HPLC fingerprint. The evaluation methods of quality and quality equivalence of Gentiana rigescens Franch. from different populations are established, which would provide the theoretical basis for further filtering of quality germplasm resources of Gentiana rigescens Franch. from wild populations.
