2.Validation on Method of Microbial Limit Test of Povidone Iodine Solution

OBJECTIVE:To establish a microbial limit test method for povidone iodine solution.METHODS:Validation on methodology of the microbial limit test of povidone iodine solution was performed using plating method(Ⅰ),culture agent dilution method(Ⅱ),membrane-filter procedure method(Ⅲ)and sodium thiosulfate neutralization in combination with membrane-filter procedure method(Ⅳ).RESULTS:The recovery rates of all of the test organisms in method Ⅰ and method Ⅱ approached zero,less than 70% in method Ⅲ but above 85% in method Ⅳ.The growth of the control bacteria was abnormal in method Ⅱand method Ⅲ,but normal in method Ⅳ.CONCLUSION:It is advisable to adopt the method Ⅳ for microbial limit test of povidone iodine solution.
3.Clinical effect of self made Jianpi Yiqi Xiaoyu decoction combined with FMC chemotherapy for treatment of elderly patients with moderate to advanced gastric cancer: results of a randomized controlled study

Objective To discuss the clinical effect of self made Jianpi Yiqi Xiaoyu decoction combined with FMC chemotherapy for treatment of elderly patients with moderate to advanced gastric cancer and its impact on their quality of life.Methods One hundred and two elderly patients with moderate to advanced gastric cancer accompanied by the Chinese medicine dialectical syndrome belonging to deficiency of spleen and stomach qi and blood stasis admitted to Zhengzhou Traditional Chinese Medicine Hospital from January 2013 to January 2016 were enrolled,they were divided into observation group and control group according to random number table method and 2 patients who missed the follow-up in each group were excluded,resulting in 49 patients in each group;the patients in the control group were treated with FMC chemotherapy [including mitomycin 15 mg/m2,cisplatin 60 mg/m2 and fluorouracil 1000 mg/m2,slow intravenous drip (drop) after dilution with 60 mL 0.9% sodium chloride injection];the patients in observation group were treated with FMC chemotherapy combined with Jianpi Yiqi Xiaoyu decoction (consisting of astragali 30 g,codonopsis pilosula 20 g,atractylodes macrocephala 15 g,Chinese yam 15 g,hoelen 15 g,ligustici 10 g,moutan cortex 10 g,aurantii 10 g,rehmannia dride rhizome 10 g,caulis dendrobii 10 g,500 mL water was added to make a decoction,boiled and concentrated to 200 mL decoction,100 mL taken in the morning and another 100 mL in the evening orally in one day).The clinical efficacy,the change of quality of life and incidence of adverse reactions were compared between the two groups.Results The proportion of patients whose quality of life improved in the observation group was significantly higher than that in the control group [55.10% (27/49) vs.16.33% (8/49),P < 0.05].The total effective rate was obviously higher in the observation group than that in the control group [51.02% (25/49) vs.22.45% (11/49),P < 0.05];the incidence of reduction of hemoglobin [30.61% (15/49) vs.63.27% (31/49)],aleucocytosis [28.57% (14/49) vs.67.35% (33/49)],thrombocytopenia [28.57% (14/49) vs.69.39% (34/49)],nausea and vomiting [44.90% (22/49) vs.71.43% (35/49)],diarrhea [32.65% (16/49) vs.63.27% (31/49)] and neurotoxicity [34.69% (17/49) vs.55.10% (27/49)] were significantly lower in observation group than those in the control group (all P < 0.05).Conclusions Self made Jianpi Yiqi Xiaoyu decoction combined with FMC chemotherapy can significantly improve the clinical efficacy of theelderly patients with moderate to advanced gastric cancer.It can improve the patients' quality of life and reduce the incidence of adverse reaction of chemotherapy.
4.The application of high frequency colour ultrasound in children with internal abdominal hernia

Objective To summarize the characteristics of pediatric abdominal hernia ultrasonic image and review the misdiagnosis of cases,to explore the values of high frequency color doppler ultrasound in the diagnosis of internal abdominal hernia. Methods Retrospective analysis the cases which diagnosed of internal hernia by abdomen colour ultrasound in January 2012 to December 2015 at our medical center,as well as the patients had surgery in our medical center for surgical treatment of 35 children (part results with pathological diagnosis), Retrospectively analyzed the characteristics of preoperative ultrasound image,and compare the results of abdominal operation results,and the pathological diagnosis. Results 30 cases confirmed by surgery for internal hernias. Conclusion ultrasound have higher value to exceed the diagnosis of internal hernia in childen,which most of the following intestinal obstruction in the diagnosis.
5.Effect of Incremental Exercise on the of Maximum Force of Femur and Lumbar Vertebrae in SD Rats

Objective The aim of the study is to investigate whether the progressive incremental exercise can induce the change in bone maximum force.Methods One hundred and seventy healthy SD rats,aged 3 months,were used as samples after adaptive exercise for one week,and then they were divided to a basic control group(C_0),8 control groups and 8 exercise groups according to the training duration(2W,4W,6W,8W,9W,11W,13W and 15W).Based on Bedford and our pre-study.the longest training duration of the rats was 15 weeks.The maximum force(F) of femoral bone and the 2nd lumbar vertebrae(L_2)were tested by three-point bending test and compressive test separately.Results It was shown that there was no pronounced difference in F of femoral bone between con trol groups and exercise groups.But significant differences in F of L_2 were found between the control groups and exercise groups(P<0.001).Conclusions Changes in different degree were found in the F of femoral bone and L_2 during the process of growth.The differential effects of incremental exercise on F were caused by the difference in training intensities and in the bone types.
6.Expression of myocardial NF-?B,iNOS and COX-2 in diabetic rats

AIM:To investigate the expression of nuclear factor-?B(NF-?B),inducable nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2)in diabetic myocardium.METHODS:Thirty SD rats were randomly divided into control and experimental groups.Diabetes was induced by a single intraperitoneal injection of STZ.The weight,blood glucose level and heart weight index(HWI)were measured 24 weeks after injection.The myocardial NF-?B,iNOS and COX-2 were stained at the same time.Furthermore,NF-?B activation in myocardium was investigated by electrophoretic mobility shift assay(EMSA).RESULTS:(1)Compared to the normal rats,the NF-?B-positive cells in the myocardium in diabetic rats significantly increased.(2)NF-?B activation in myocardium by EMSA was significantly higher in the diabetic rats than that in the normal rats.(3)The iNOS was not expressed in normal rat myocardium and was significantly expressed in the diabetic rat myocardium.(4)The COX-2 was rarely expressed in normal rat myocardium and was significantly expressed in the diabetic rat myocardium.CONCLUSION:The expressions of NF-?B,iNOS and COX-2 are significantly enhanced in the diabetic myocardium.

Sirt3 is a kind of nicotinamide adenine dinucleotide (NAD+)-dependent deacetylases.Sirt3 is localized in mitochondria and has been involved in a wide range of mitochondrial biological functions, such as nutrient oxidation、ATP generation, reactive oxygen species (ROS) detoxification and mitochondrial homeostasis.Sirt3 plays an important role in the occurrence and development of cardiovascular diseases.Increased expression of Sirt3 gene has been associated with extended lifespan of humans.
8.Study on the correlation of SIRT3 with metabolic syndrome and myocardial hypertrophy

[Summary] SIRT3 is a member of the silent information regulator 2 ( Sir2) family, located in the inner mitochondrial membrane, with a strong effect of deacetylation. SIRT3 not only modulates energy metabolism, cell apoptosis, tumor growth, anti-aging etc. , but also plays an important role in the field of cardiovascular diseases. In this review, we summarize recent findings related to SIRT3 with metabolic syndrome and cardiac hypertrophy, to provide a theoretical basis for the further study on the potential role of SIRT3 in cardiovascular diseases.
9. Expresssion of miRNA-1 in esophageal squamous cell carcinoma tissues and its mechanism

Objective: To investigate the expression of microRNA-1 (miRNA-1) in human esophageal squamous cell carcinoma (ESCC) tissues and its possible mechanism. Methods: The expression levels of miRNA-1 and LIM and SH3 domain protein 1 (LASP1) mRNA and LASP1 protein in 55 cases of ESCC and its adjacent para-cancerous tissues were detected by real-time fluorescent quantitative PCR and Western blotting, respectively. The potential target gene of miRNA-1 was predicted by online bioinformatics software. The target gene of miRNA-1 after Eca109 cells co-transfection with recombination dual luciferase reporter vector psiCHECK-2-LASP1 containing 3'-untranslated region (3'-UTR) with miRNA-1 binding site of LASP1 gene and miRNA-1 mimic was verified by dual-luciferase reporter assay system. The expressions of miRNA-1 and LASP1 protein in Eca109 cells after transfection with miRNA-1 mimic or miRNA-1 inhibitor were detected by real-time fluorescent quantitative-PCR and Western blotting, respectively. Results: The expression level of miRNA-1 in ESCC tissues was lower than that in adjacent para-cancerous tissues (P < 0.01), and the expression levels of LASP1 mRNA and protein were opposite (all P < 0.05). The expressions of miRNA-1 and LASP1 were associated with lymph metastasis, TNM staging and histological grade (all P < 0.05). The miRNA-1 expression in ESCC tissues was positively associated with LASP1 (r = +0.45, P < 0.05). LASP1 was target gene of miRNA-1. miRNA-1 could directly target the LASP1 3'-UTR. The expression level of miRNA-1 was up-regulated in Eca109 cells after transfection with miRNA-1 mimic (P < 0.01), and the expression level of LASP1 protein was down-regulated (P < 0.05). The expression level of miRNA-1 was down-regulated in Eca109 cells after transfection with miRNA-1 inhibitor (P < 0.05), and the expression level of LASP1 protein was up-regulated (P < 0.05). Conclusion: The expression level of miRNA-1 is low in ESCC tissues, and its mechanism may be associated with regulation of target gene LASP1.
10.Effect of melatonin on bioactivity of fibroblasts from human hypertrophic scar

Objective To investigate the effect and mechanism of melatonin on bioactivity of fibroblasts from human hypertrophic scar.Methods Fibroblasts from hypertrophic scar were cultured and incubated with melatonin,melatonin and Luzindole,and Luzindole,respectively,for 24 h,and the media as control.XTT/PMS assay was used to measure the proliferation of fibroblasts,ELISA assay to detect the TGF-β1 production of fibroblasts,and the expression of cell a-SMA,collagen Ⅰ,collagen Ⅲ mRNA were determined with real-time PCR method.Results Compared with the control,melatonin at the concentration of 10-5mmol/L,10-3mmol/L,and 1 mmol/L could inhibit the proliferation of fibroblasts in a does-dependent manner (P<0.05); melatonin at the concentration of 10-3 mmol/L could significantly decrease the TGF-β1 production and expression of a-SMA mRNA and collagen Ⅰ mRNA in fibroblasts from human hypertrophic scar (P<0.05) ; the effect of melatonin on fibroblast was significantly blocked by Luzindole (P<0.05),but melatonin could not inhibit collagen Ⅲ mRNA expression (P>0.05).Conclusions Melatonin can significantly regulate the biological activity of fibroblasts from human hypertrophic scar through a receptor pathway.
