1.Extraction of Total Alkaloids from Yaotong Granules

Objective:To study the optimum extraction conditions for Yaotong granules. Methods:An orthogonal test was adopted to optimize the percolation extraction process of total alkaloids by orthogonal experiment with the yield of dry extract and total extraction rate of tetrandrine and fangchinoline as the indices. Results:The optimal extraction conditions were as follows:the medicinal materials were soaked with crude powder in 60% ethanol for 48h,then percolated at the rate of 2 ml·min-1 ·kg-1 . Conclusion:The extraction technology for Yaotong granules is simple and rapid with stable results,which can provide theoretical basis for the production of Yaotong granules.
2.Expression of BRCA1 and BRCA2 in sporadic breast cancer in women of Jiaodong peninsula

Objective To study the expression and clinical significance of BRCA1 and BRCA2 in sporadic breast cancer in women of Jiaodong peninsula. Methods Immunohistochemistry and tissue array were used to detect the expression of BRCA1 and BRCA2 in 100 cases of sporadic breast cancer and 30 cases of benign breast tumor in women of Jiaodong peninsula. Results ① The expression rate of BRCA1 and BRCA2 was 49% (49/100) and 50% (50/100) in breast cancer, 80% (24/30) and 83.33% (25/30) in benign breast lesions respectively. The expression rate of BRCA1 and BRCA2 in breast cancer was lower than that in benign breast lesions (P<0.05). ② The expression of BRCA1 and BRCA2 was uncorrelated with factors such as tumor size, lymphatic metastases, age and menopause or not(P>0.05). ③ There was no dependency between the expression of BRCA1 and BRCA2 (P> 0. 05). Conclusions The expression rate of BRCA1 and BRCA2 in breast carcinoma in women of Jiaodong peninsula was lower than that in benign breast lesions, suggesting that the expression of BRCA1 and BRCA2 was related to the occurrence of sporadic breast carcinoma in women of Jiaodong peninsula. However, the role of BRCA1 and BRCA2 in the genesis and development of the breast carcinoma is independent.
3.Applying vector analysis to evaluate influence of ocular residual astigmatism on astigmatism correction by FS-LASIK

Objective To investigate the influence of ocular residual astigmatism (ORA) on the correction of astigmatism by FS-LASIK with vector analysis.Methods The records of 182 patients who had accept FS-LASIK between January,2016 and April,2016 were retrosepectively reviewed.The patients whose ORA ≥ refractive cylinder were assigned to high ocular residual astigmatism group (HORA group),ORA < refractive cylinder were assigned to low ocular residual astigmatism group (LORA group).All of the patients were followed 6 months or more.The visual acuity,error ratio and correction ratio were compared between HORA group and LORA group.Results The preoperative ORA of all patients was (0.61 ± 0.27) D,in which > 0.75 D were 58 cases (31.9%),and the HORA group was more than the LORA group (P < 0.05).At postoperative 6 months,there was no statistically significant difference in vision acuity between the HORA group (1.06 ± 0.15) and LORA group (1.08 ± 0.15) (t =0.97,P =0.35).There was statistically significant difference in the error ratio between the HORA group (58.11 ± 63.23) % and LORA group (26.12 ± 35.37) % (t =3.43,P < 0.05).There was statistically significant difference in the correction ratio between the HORA group (146.45 ± 86.63) % and LORA group (122.56 ± 36.31) % (t =2.81,P < 0.05).Conclusion The error ratio and correction ratio of astigmatic correction by FSLASIK is significantly higher in eyes with high ORA than in eyes with low ORA.Vector analysis should been carried out before the FS-LASIK.
4.Investigation in anxiety and depression of patients with chronic prostatitis and psychological nursing countermeasures

Objective To provide therapeutic strategy by investigating the anxiety and depression status as well as the correlated impact factors in patients with chronic prostatitis. Methods The self-rating anxiety scale (SAS)and self-rating depression scale (SDS)were employed to evaluate psychological response of 360 patients with chronic prostatitis. Results Both the average scores of SAS and SDS in patients with chronic prostatitis were higher than normal levels of Chinese population. The impact factors for anxiety included income status, educational level and disease course. Conclusions The mental health status of chronic prostatitis patients is poor and certain factors affect the occurrence of anxiety and depression. Therefore, it is necessary to treat the patients with supportive psychotherapeutic intervention and with combined approaches.
5.A study on the factors influencing the prevalence of echinococciasis in Gansu Province

Objective To investigate the factors influencing the prevalence of echinococciasis in Gansu Province.Methods With the method of stratified random sampling,all the administrative villages in pastoral areas,half pastoral areas,agriculture and urban areas in the 72 counties in Gansu Province were selected;according to the layers of the population of the county population proportion calculated,the layer investigation numbers were determined,and a total of 16 administrative villages were selected in each county.In the 16 administrative villages,more than 200 people of permanent residents were surveyed in each village (shortage was made up from a nearby village),at least 3 200 people were investigated in a county.By using the combined method of inquiry and field observation,natural factors,such as drinking water,farming and animal husbandry production data were surveyed.All data were analyzed using SPSS 19.0 statistical software.Unauy linear and multiple linear regression analysis of influencing factors and the relationship between the prevalence were analyzed.Results Unary linear regression analysis showed that longitudes,latitudes,altitudes,average annual temperature,annual rainfalls,secondary industries,animal husbandry towns,drinking water sources,the number of epidemic towns,epidemic villages,per capita net incomes in animal husbandry regions,the total number of livestock and the number of sheep were factors influencing the prevalence of echinococciasis (F =4.705,P <0.05).Latitude,altitude,animal husbandry towns,the number of epidemic towns,epidemic villages,per capita net incomes in animal husbandry regions,the total number of livestock and the number of sheep and the prevalence of echinococciasis was positively correlated (r =0.282,0.285,0.387,0.508,0.540,0.317,0.475,0.594,all P <0.05);longitude,average annual temperature,annual rainfall,secondary industries,drinking water sources and the prevalence of echinococciasis was negatively correlated (r =-0.311,-0.244,-0.244,-0.389,-0.311,all P <0.05).Multiple linear regression analysis showed that differences of interactions of annual rainfalls,drinking water sources,epidemic villages,per capita net incomes of herdsmen,secondary industries and the total number of livestock between groups were statistically significant (t =-1.822,-3.920,3.013,1.715,-1.609,3.264,all P <0.05).Conclusion The factors influencing the prevalence of echinococciasis in Gansu Province are correlated with annual rainfalls,drinking water sources,epidemic villages,per capita net incomes of herdsmen,secondary industries and the total number of livestock.
6.Direct acting antiviral agent for Chinese patients with chronic hepatitis C genotype 1b infection——a real world experience

Objective To evaluate the effectiveness and safety of direct-acting antiviral agents (DAA) treatment in Chinese chronic hepatitis C (CHC) patients with genotype (GT) 1b HCV infection in a real world setting .Methods The consecutive GT1b CHC Chinese patients treated with sofosbuvir (SOF) plus daclatasvir (DCV) (n=62) or SOF plus ledipasvir (LDV) (n=171) were enrolled from July 2014 to December 2016 at 302 Military Hospital of China .The treatment duration for all the patients was 12 weeks .All the clinical parameters were measured at baseline and then 4-weekly till 12 weeks after the end-of-treatment (EOT ).Baseline clinical characteristics ,treatment efficacy ,safety and tolerance were compared .Serum HCV RNA concentration was detected by means of COBAS TaqMan assay with a lower detection limit of 15 IU/mL ,and liver stiffness was measured using FibroScan?.Sustained virologic response (SVR) was defined as HCV RNA under the lower limit of quantification 12 weeks after EOT (SVR12).Students′t-test ,pearson χ2 test ,Spearman rank correlation analysis and Fisher exact test were used for comparison between groups when appropriate .Results Among 233 patients ,173 cases had baseline HCV RNA level ≥ 6 .0 lg IU/mL and 97 cases hade liver stiffness measurement (LSM )≥17.5 kPa.The baseline liver inflamation ,liver fibrosis ,and HCV RNA load of patients in the two groups were not significantly different (all P>0 .05).The HCV RNA of all the 233 patients was undetectable at the end of 12-week treatment ,while 2 patients relapsed after 12 weeks of EOT with the overall SVR12 of 99.1% .HCV RNA decline was significantly faster in patients with lower LSM than those with higher LSM (ρ=0 .233 ,P=0 .001) ,and SVR12 was higher in those with lower LSM .In terms of other clinical characteristics of SOF+DCV and SOF+LVD groups ,alanine transaminase declined from (68 .0 ± 60 .1) and (70 .1 ± 56 .1) U/L to (21 .1 ± 10 .9) U/L and (15 .3 ± 9 .5) U/L ,respectively ,total bilirubin declined from (21 .3 ± 17 .3) and (18 .2 ± 14 .0) μmol/L to (13 .2 ± 6 .7) and (10 .2 ± 4 .6) μmol/L , respectively ,AFP declined from 19 .6 (10 .6 ,62 .3) and 15 .0 (12 .0 ,25 .0) μg/L to 6 .5(4 .5 ,18 .7) and 7 .8(5 .3 ,15 .4) μg/L ,respectively ,LSM declined from 17 .6 (8 .9 ,25 .4) and 15 .7 (7 .8 ,23 .9) kPa to 13.9(6 .5 ,21 .4) and 9 .1(5 .6 ,19 .9) kPa ,respectively ,serum album elevated form (37 .5 ± 5 .8) and (38 .7 ± 5 .5) g/L to (41 .3 ± 4 .7) and (42 .8 ± 5 .1) g/L ,respectively ,platelet elevated from (120.9 ± 78 . 2)×109/L and (136 .6 ± 65 .8 )× 109/L to (139 .5 ± 71.8 )× 109/L and (149 .7 ± 71.4 )× 109/L , respectively .Reports of adverse events were low in both groups .Conclusions Both SOF + DCV and SOF/LDV therapy are highly effective with > 98% of SVR12 and reduce LSM value significantly with good safety for CHC GT1b Chinese patients .
7.Sampling study design and radiography protocol of a large-sample investigation on skeletal maturation in 3 to 18-year-old children in China

Objective:To report the sampling study design and radiography protocol of a large-sample investigation on skeletal maturation of 3 to 18-year-old children in China.Methods:Multi-stage stratified random sampling was employed in this study. Two provinces, municipalities, or autonomous regions were randomly selected from each of the seven regions of China, including Northeast China, Northwest China, North China, Central China, East China, Southwest China, and South China. Then one rural and one urban investigation site were randomly selected from each province, municipality, or autonomous region. In total 28 sites were included. Among those sites, four residential districts were randomly selected from each urban site, and four townships from each rural site. For each residential district or township, 1-4 kindergartens, primary schools, and middle schools were chosen. Random cluster sampling was used to extract 3-<6-year-old children in kindergartens, and 6-18-year-old children in primary schools and middle schools. The investigation on skeletal maturation was sampled proportionate to the sampling of the whole study. The estimated simple size was 780 for each site, and 21 840 for all 28 sites in total. There were six groups of 3-<6-year-old children classified at 0.5-year intervals, and 12 groups of 6-18-year-old children classified at 1-year intervals. Posteroanterior position radiography of the left hand and wrist was achieved for all subjects.Results:The study was performed from August 26, 2019 to October 16, 2021. In total, 20 444 children received posteroanterior position radiography of the left hand and wrist, including 10 196 males and 10 248 females, 9 711 urban and 10 733 rural, respectively. The 3-<6-year-old group included 1 611 (male 819, female 792) subjects, and the 6 to 18-year-old group included 18 833 (male 9 377, female 9 456) subjects.Conclusion:This nationwide investigation on skeletal maturation of 3 to 18-year-old children in seven regions of China was successfully preformed. The results of this study can provide an important reference for establishing the current evaluation criteria of bone age in Chinese children and adolescents.
8.Peptide Mass Mapping Analysis and Localization of Disulfide Bonds of Recombinant Human Granulocyte- macrophage Colony Stimulating Factor

OBJECTIVE To analyze the peptide mass mapping and localization of disulfide bonds of recombinant human granulocyte-macrophage colony stimulating factor(rhGM-CSF) by using mass spectrometry combined with ultra-performance liquid chromatography. METHODS The bulk of rhGM-CSF was denatured, reduced, alkylated and finally digested by the enzyme of trypsin and V8 respectively to analyse the peptide mass mapping. The bulk was denatured, alkylated and digested by V8 and V8 with trypsin respectively to identify the sites of disulfide bonds. RESULTS Fourteen matched peptides were found in the peptide mass mapping of rhGM-CSF digested by trypsin and eighteen matched peptides were found digested by V8. The fraction of coverage was both 100.0%. The site of disulfide bond Cys55-Cys97 was identified by digestion of V8, and the types of disulfide bonds Cys55-Cys97 and Cys89-Cys122 were identified by digestion of V8 with trypsin. CONCLUSION The peptide mass mapping analysis and localization of disulfide bonds using LC-MS/MS can be used for the quality control of recombination protein.
