International Journal of Pediatrics 2015;(4):461-463,467
Objective To analyze the change of cysteinyl leukotriene ( Cys-LTs) levels and 8-Isopros-tane (8-iso-PG) levels in the exhaled breath condensate (EBC) of asthmatic children from acute exacerbation to clinical remission, and investigate the role of the detection of Cys-LTs and 8-iso-PG in EBC in its severity and pathogenesis , and explore the relationship between the Cys-LTs and 8-iso-PG through measuring Cys-LTs levels and 8-iso-PG levels in the EBC of asthmatic children. Methods The outpatient or inpatient asthmatic children of the pediatrics and a group of healthy children were studied. All subjects′ EBC were collected by the R-Tube produced by American Respiratory Research. The concentration of Cys-LTs and 8-iso-PG in EBC were measured by enzyme-linked immunosorbent assay (ELISA), and compared among children in asthmatic exacerbation, asthmatic remission, and healthy condition. The relevance of their change would be explored at the same time. Results (1) Cys-LTs levels in EBC were higher in asthma exacerbation, compared to healthy controls (P<0. 05), while no significant difference were found between asthmatic remission and asthmatic exacerbation or healthy controls ( P>0. 05 ) . ( 2 ) 8-iso-PG levels was higher in asthmatic exacerbation compared to asth-matic remission ( P<0. 05 ) . Moreover, the 8-iso-PG levels were significantly higher in asthmatic remission than in healthy controls (P<0. 05). (3) Through the relevance analysis of the Cys-LTs and 8-iso-PG levels in EBC among the three groups, Cys-LTs levels in EBC of asthmatic exacerbation significantly were correlated with 8-iso-PG levels (n1 =35, r1 =0. 61, P<0. 05), while there was no significant correlation between 8-iso-PG levels and Cys-LTs levels in asthmatic remission. Conclusion The increase of 8-iso-PG levels in EBC of bronchial asthmatic patients correlates with the disease and its control. Therefore, 8-iso-PG can be an objective indicator for asthmatic diagnosis and healing efficacy. Cy-LTs levels increase in the EBC of bronchial asthmatic according to disease severity. The two levels correlate during asthmatic exacerbation, indicating that a link be-tween airway oxidative stress and inflammation among asthmatics.

Methods Eighty SPF male Sprague-Dawley rats, aged 8-10 weeks, weighing 220-250 g, were di-vided into 8 groups ( n=10 each) using a random number table method: control group ( group C) , sham operation group (group S), lidocaine group ( group L), dimethyl sulfoxide ( DMSO) group, low-dose curcumin group ( group CL ) , high-dose curcumin group ( group CH ) , curcumin plus ERK inhibitor PD98059 group (group P) and curcumin plus Akt inhibitor MK-2206 group (group M). The model of nerve injury was established by injecting 2% lidocaine 10μl via the catheter in anesthetized rats in the other six groups except for C and S groups. Drugs were injected through a microcatheter for 14 consecutive days starting from 3rd day after IT catheterization as follows: DMSO 10μl, curcumin 100μg∕10μl and curcu-min 500 μg∕10 μl were injected once a day in D, CL and CH groups, respectively; curcumin 500 μg∕10μl was injected once a day, and D9805910μg and MK-220612μg ( in 5μl DMSO) were intraperitoneal-ly injected once a week in P and M groups, respectively. The mechanical paw withdrawal threshold ( MWT) and thermal paw withdrawal threshold ( TWL) on the operated side were measured on 1 day before IT catheterization, before administration on 3rd day after IT catheterization, and on 14th day after adminis-tration. The spinal cord was removed on 1st day after the end of administration for determination of the ex-pression of ERK1∕2, phosphorylated ERK1∕2 (p-ERK1∕2), Akt, phosphorylated Akt (p-Akt), c-fos, Bcl-2 and Bax ( by Western blot) and expression of ERK1∕2 and Akt mRNA ( by real-time polymerase chain reaction) . Results Compared with group C, the MWT was significantly decreased, the TWL was short-ened, the expression of ERK1∕2 protein and mRNA, p-ERK1∕2, c-fos, Akt protein and mRNA, p-Akt and Bcl-2 was down-regulated, and the expression of Bax was up-regulated in group L (P<0. 05). Com-pared with group L, the MWT was significantly increased, and the TWL was prolonged in CL, CH, P and M groups, the expression of p-ERK1∕2, c-fos, p-Akt and Bcl-2 was up-regulated, and the expression of Bax was down-regulated in CL and CH groups, the expression of p-Akt and Bcl-2 was up-regulated, and the expression of Bax was down-regulated in group P , and the the expression of p-ERK1∕2 and c-fos was up-regulated in group M ( P<0. 05) . Compared with group CH, the MWT was significantly decreased, and the TWL was shortened in P and M groups, the expression of p-ERK1∕2 and c-fos was down-regulated in group P, and the expression of p-Akt and Bcl-2 was down-regulated, and the expression of Bax was up-reg-ulated in group M ( P<0. 05) . Conclusion Curcumin can reduce lidocaine-induced nerve injury, and the mechanism may be partly related to enhancing the activity of Akt and ERK1∕2 signaling pathway in rats.

A high-altitude environment is characterized by low oxygen levels, low pressure, and low temperatures.Exposure to the plateau environment often causes damage to the body, leading to the occurrence of acute mountain sickness/chronic mountain sickness(AMS/CMS).Research indicates that acute or chronic exposure to the special environment can result in overall organ dysfunctions, such as those in the heart and gastrointestinal tract. The damage to the body caused by exposure to the plateau environment is closely related to acute and chronic hypoxia.Physiological maladjustment or disease is usually accompanied by changes in the structure of the gut microbiota. There have been reports on the correlations between the gut microbiota and bodily harm caused by high-altitude exposure. However, the specific types of bacteria involved and the mechanisms of action are still under investigation. This article reviews the intestinal tissue damage caused by low oxygen levels, immune activation, changes in microbial community structure, and differential metabolic products. The association and underlying mechanisms between bodily harm due to high-altitude exposure and the intestinal microbiota are also explored in hopes of stimulating new lines of thought related to the prevention and treatment of bodily harm caused by exposure to the plateau environment.

ObjectiveTo establish and evaluate a chronic obstructive pulmonary disease (COPD) model with lung-spleen qi deficiency. MethodA rat model mimicking COPD with lung-spleen qi deficiency was established by the combination of cigarette smoking and intratracheal instillation of lipopolysaccharide (LPS) along with gavage of Sennae Folium infusion. Forty male SPF-grade SD rats were randomly assigned to blank, model, and low- (L-FXY), medium- (M-FXY), and high-dose (H-FXY) Sennae Folium infusion groups. Other groups except the blank group were exposed to daily cigarette smoke, with LPS administrated via intratracheal instillation on the 1st and 14th days. On the 28th day of modeling, the L-FXY, M-FXY, and H-FXY groups were administrated with Sennae Folium infusion at 5, 10, and 20 g·kg-1, respectively, and at 4 ℃ for three weeks. The modeling lasted for 49 days. The general conditions (body mass, food intake, fecal water content, and anal temperature) and behaviors (grip strength test and tail suspension test) of rats in different groups were examined. The lung function, lung histopathology, D-xylose, amylase, and gastrin levels in the serum, interleukin(IL)-1β and IL-6 levels in the alveolar lavage fluid, levels of T-lymphocyte subsets (CD4+, CD8+, and CD4+/CD8+) in the peripheral blood, and thymus and spleen indices were measured. ResultTwo rats died in the H-FXY group. Compared with the blank group, both the M-FXY and H-FXY groups exhibited reduced body mass and food intake (P<0.01) and increased fecal water content (P<0.01). The anal temperature in the H-FXY group was lower than that in the blank group (P<0.01). The grip strength decreased in the modeling groups compared with the blank group (P<0.01), and the duration of immobility in the tail suspension test increased in the M-FXY and H-FXY groups (P<0.05, P<0.01). Compared with the blank group, the modeling groups showed reduced 0.3 second forced expiratory volume (FEV0.3), FEV0.3/forced vital capacity (FVC)(P<0.01), thickening of bronchial walls, proliferation of goblet cells, and the presence of emphysematous changes. In terms of gastrointestinal function, the M-FXY and H-FXY groups had lower levels of D-xylose, gastrin, and α-amylase than the blank group (P<0.01). Regarding the immune and inflammatory indices, the M-FXY and H-FXY groups showed lower thymus and spleen indices than the blank group (P<0.01). Compared with the blank group, the modeling groups presented lowered CD4+ level (P<0.01) and CD4+/CD8+ ratio (P<0.05, P<0.01) in the peripheral blood and elevated levels of IL-1β and IL-6 in the alveolar lavage fluid (P<0.01) than the blank group. ConclusionA model of COPD with lung-spleen Qi deficiency was established through the combination of daily cigarette smoke, intratracheal instillation with LPS, and gavage of Sennae Folium infusion. The comprehensive evaluation results suggested medium-dose (10 g·kg-1) Sennae Folium infusion for gavage during the modeling of COPD with lung-spleen Qi deficiency.
