1.Selectin and Acute Pancreatitis

Objective To study the function of selectin in the pathogenesis and advancement of acute pancreatitis(AP), so as to guide further investigation and clinical treatment. Methods Correlative articles in recent years were reviewed. Results Selectins act as an indicator of the activation of endothelium. Their expression changes markedly during AP and is closely related to cytokines, oxygen free radicals and complements. Conclusion Selectin is a component which is engaged in the pathology of AP, the level of selectin is useful in guiding clinical observation and treatment.
2.Immune Response to Respiratory Syncytial Virus Infection in Ovalbumin Sensitized Mice

Objective To observe the lung inflammation and the expression of T - helper cell related cytokines in respiratory syncytial virus(RSV) infected Balb/c mice pre - sensitized with ovalbumin(OVA). Methods Mice were randomly divided into two series: nonsen-sitized and sensitized animal model with nebulized OVA once a day for 10 days. Each of the series was divided again into two groups: control group,RSV infected group with nasal draping plus nebulizing. All the mice were sacrificed 5 days later of infection. Viral isolation of lung organization was performed in each group. Pulmonary pathological detection and the levels of interleukin - 4(IL-4), interferon-? (IFN- ?) mRNA were evaluated by RT- PCR. Results The RSV were found only in RSV infected groups. The Balb/c mice developed typical interstitial pneumonia after RSV infection. When the mice pre- sensitized were infected with RSV, the pulmonary inflammation, lymphocyte and eosinophils infiltration and cell - collar peribronchiles were more severe in lung organization and bronchiole than those in the single RSV infection group. In the lung organization of control group, there were no mRNA expression of IL - 4 and IFN - ?. The mRNA of IFN -? was expressed in the lung organization of RSV infected groups while had not showed mRNA of IL- 4. In the OVA+ RSV group the mRNA expression of IL - 4 was obvious, while the expresses of IFN -? scarcely. Conclusions Pulmonary inflammation is more severe in the infection after OVA sensitization. RSV infection alone results in a Th1 - like cytokine response, while the infection after OVA sensitization results in Th2 - like response.
3.27-O-(E)-p-coumaric acyl ursolic acid via JNK/SAPK signal pathway regulates apoptosis of human breast cancer MDA-MB-231 cell line.

27-O-(E)-p-coumaric acyl ursolic acid( DY-17) from Ilex latifolia is a compound of the monomer. To investigate the DY-17 inducing apoptosis in the human breast cancer cell line, the MDA-MB-231 cells were used as research object in this experiment. The proliferation activity of the MDA-MB-231 cells stimulated with the different concentrations of DY-17 (20, 40 µmol · L(-1)) was detected at different time( 12, 24, 36, 48, 60,72 h) . We surveyed the DY-17 inducing apoptosis of the MDA-MB-231 cells with the fluorescent staining technology. The rate of MDA-MB-231 cells apoptosis and necrosis was determined by flow cell cytometry (FCC). Moreover, expression of JNK, phosphorylated JNK, Bax, PARP shear and caspase-3 shear related to JNK/SAPK pathways were investigated in every group ( control group, EGF group, EGF + DY-17 40 µmol · L(1) group and EGF + SP600125 group) with Western blot. The MTT results showed that, in the presence of DY-17, the proliferation activity of MDA-MB-231 cells decreased in a dose-dependent and time-dependent manner. The apoptosis and necrosis rates of MDA-MB-231 cells with DY-17(20, 40 µmol · L(-1)) groups was respectively 31.86%, 49.91% by flow cytometry and significantly increased compared with control group under Fluores- cence microscopy. Up-regulation of the JNK phosphorylation protein expression was observed in EGF group compared with control group. In addition, markedly decreased the expression of JNK phosphorylation protein were also surveyed in EGF + DY-17 40 µmol · L(-1) group compared with EGF group. The expression of Bax, shear PARP and shear caspase-3 protein in EGF + DY-17 40 µmol · L(-1) group were significantly increased in comparison with EGF group. The results showed DY-17 induced apoptosis of human breast cancer MDA-MB-231 cell line related to down-regulating JNK/SAPK signal pathways.

4.An optimal method for cryopreservation of microamount round spermatids of the mouse.

OBJECTIVETo search for an optimal protocol and freezing conditions for the cryopreservation of microamount round spermatids of the mouse.
METHODSWe compared the survival rates of frozen-thawed microamount round spermatids of the mouse achieved by vitrification or standard slow freezing with different concentrations of glycerol (5, 7, or 9%) and different lengths of equilibrium time (0, 15, 30, 45, or 60 min).
RESULTSUnder the conditions of 7% glycerol and 30 min equilibrium, both vitrification and standard slow freezing achieved high survival rates of spermatids, and the former obtained an even higher rate than the latter ([72.9 ± 15.4]% vs [58.2 ± 17.7]%, P < 0.05).
CONCLUSIONA high rate of frozen-thawed microamount round spermatids of the mouse can be achieved by vitrification under the conditions of 7% glycerol and 30 min equilibrium.

5.Standardize and pragmatism- the key to promote the healthy development of obesity and metabolic surgery in China.

After more than half a century of development, obesity and metabolic surgery in western countries has become a complete surgical specialties, but the introduction of obesity and metabolic surgery was only 10 years old in China. The surgical treatment of obesity and related metabolic diseases, especially obesity of type 2 diabetes has been recognized and acknowledgd by the majority of surgical colleagues and patients as a new direction of development. The current trend of obesity and metabolic surgery is favorable, but there are many problems in the development of obesity and metabolic surgery in China, for example, the surgery nomenclature is not standardized, surgical indications are loosened, the surgical procedure is modified arbitrarily, the evaluation criteria is confusing, the postoperative follow-up protocol is not unified, etc. These problems require the majority of surgical colleagues to work together to make the standards scientifically a nd objectively in accordance with the actual situatioin of our country, so as to promote the healthy development of obesity and metabolic surgery in China.

8.Case report: Leiomyoma of the anterior abdominal wall

Leiomyomas are benign soft tissue swellings of smooth
muscle origin, most commonly found in the uterus. Extra
uterine leiomyomas presenting as an abdominal mass is
often a diagnostic challenge as such occurrence is rare. We
present a rare case of primary abdominal wall leiomyoma,
and highlight the importance of laparoscopic approach in
the diagnosis and treatment of such tumour.

9."Application of ""Flipped Classroom"" and Experiment Project Modular Design in the Instrumental Analytical Experiment Teaching"

OBJECTIVE:To improve the effectiveness of experimental teaching and independent hands-on ability of the students.METHODS:Combined with teaching characteristics of newly established universities,the characteristics of instrumental analytical experiment teaching and the characteristics of pharmaceutical students,flipped classroom and project modular design were applied to instrumental analytical experiment teaching together.The instrumental analytical experiment teaching was expounded and integrated in respects of curriculum content design,teaching implementation and curriculum assessment.RESULTS&CONCLUSIONS:The students achieve the knowledge learning after classroom,and complete the knowledge absorption and mastering in class with the help of video and audio information.It also improved independent learning ability of the students,strengthened the ability of interactive learning and cooperative leafing between teachers and students and among students.It is feasible to apply the flipped classroom and the experimental project modular design in the instrument analytical chemistry experiment teaching reform.This method also can provide some references for the reform of instrument analytical chemistry experiment teaching.
10.EGF and SCF promote the proliferation and differentiation of mouse spermatogenic cells in vitro.

OBJECTIVETo study the promoting effects of the epidermal growth factor (EGF) and stem cell factor (SCF) on the proliferation and differentiation of spermatogenic cells in mice.
METHODSWe cocultured in vitro spermatogenic cells of male mice aged 7 - 8 days in the medium with EGF and/or SCF at the concentrations of 5, 10, 20, 40 and 100 ng/ml, respectively. Then we observed the survival rate and morphological changes of the spermatogenic cells, detected the expressions of the pachytene-specific phosphoprotein gene (P19) and haploid sperm cell-specific transition protein gene (TP1), and analyzed the ploidy of the cells.
RESULTSAfter cocultured with EGF or SCF for 2 - 4 days, the spermatogenic cells began to proliferate in masses or chains in all concentration groups, most obviously in the 20 ng/ml EGF and 40 ng/ml SCF groups. At 7 days, both the number and survival rate of spermatogenic cells were significantly higher in the 20 ng/ml EGF and 40 ng/ml SCF groups than in the others (P < 0.05), and meanwhile, the P19/TP1 ratio was obviously decreased and the rate of haploid sperm markedly increased in the 40 ng/ml SCF group (P < 0.05). Combination of EGF and SCF remarkably promoted the proliferation of the spermatogenic cells (P < 0.05).
CONCLUSIONBoth EGF and SCF could increase the number and survival rate of spermatogenic cells. SCF could promote the formation of haploid sperm, and the combination of the two cytokines could enhance the effect on the proliferation of spermatogenic cells.
