2.The study of lung adenocarcinoma serum biomarkers selected by surface-enhanced laser desorption ionization spectrometry

Background and Objective:Lung adenocarcinoma is,at present,the most common malignancy in the world and its overall 5-year survival rate is disappointing.Because most of the patients are diagnosed at advanced stage,early diagnosis may improve the prognosis of patients with lung adenocarcinoma.Surface-Enhanced Laser Desorption and Ionization Mass Spectrometry(SELDI) is one of the currently used techniques to identify biomarkers for cancers.This study explored the application of serum SELDI proteomic patterns to distinguish lung adenocarcinoma patients from healthy individuals.Methods:Serum samples from 71 lung adenocarcinoma patients,71 healthy volunteers with matched gender,age and history of smoking were analyzed using WCX2 ProteinChip to select potential biomarkers.Proteomic spectra were generated by mass spectrometry.Results:Five highly expressed potential biomarkers were identified with the relative molecular weights of 4047.79?1.60,4203.99?1.91,4959.81?2.13,5329.30?2.55 and 7760.12?4.11.The sensitivity for diagnosing lung adenocarcinoma was 90.14%,78.87%,50.70%,57.75%,73.24%;and specificity was 97.18%,92.96%,70.42%,76.06%,94.37%,when the critical point was made 1.5.Conclusions:SELDI-TOF-MS ProteinChip technology is a quick,easy,convenient,and highoutput analyzing method that it is capable of selecting several relatively specific,potential biomarkers from the serum of lung adenocarcinoma patients and may have clinic value.
3.Application of three-level storehouse system in ward drug management

Objective To explore the effect of the three level storehouse system on ward drug management.Methods The three level storehouse system was established in our hospital,which includes drug demanding quantity of ward,delivering quantity of the drug storehouse,and management supervision of pharmacy.Result After implementing three level storehouse system, the incidence of medicine expiry,improper storage,drug reception,confused marking ,improper placement and incorrect drug administration were decreased(P<0.05).Conclusion The three level storehouse system could improve the quality of hospital drug management as well as ensure clinical medication safety.
4.Clinical experience of laparoscopic resection of adrenal tumors

Objective To investigate the laparoscopie resection of the adrenal tumor effects and clinical experience. Methods Among the 24 cases of Laparoscopic adrenalectomy tumors, there were 8 cases of adrenal adenoma, 7 cases of pheochromoeytoma and 9 eases primary aldosteronism. Results One ease was transit to open operation, the other 23 were successful,each surgerical time was 40~130 min with an average of 98 rain. Hemorrhage was 50~300 mL, 140 ml average, no blood transfusion. It was 1~2 d after sur- gery to remove drainage tubes, and could take some activities and have fresh flow in the second day. The pa- tients didn't appear complications and discharged after 5~7 d. Follow-up of 6~24 months, there was no recurrence and long-term complications. Conclusion Laparoscopic adrenalectomy has the advantages of minor trauma surgery, less bleeding, good demonstrating, convenient operation, fewer complications and quick recovery.
5.Lycopene protects primary mouse cerebrocortical neurons against t-BHP-induced damage in vitro

AIM:To investigate the protective effect of lycopene on primary mouse cerebrocortical neurons ex -posed to tert-butyl hydroperoxide ( t-BHP) and its mechanisms of in vitro.METHODS:Primary cerebrocortical neurons of newborn C57 mice were extracted and divided into normal group , t-BHP group, lycopene +t-BHP group and lycopene group.The neuronal damage was induced by t-BHP exposure for 24 h, and the cell viability was examined by MTT assay . ROS content was measured by flow cytometry , and the protein levels of Bax , Bcl-2, caspase-3, cleaved caspase-3 and cyto-chrome C were examined by Western blot .RESULTS:The primary mouse cortical neurons expressed MAP-2 protein.Ly-copene at concentration of 4μmol/L reversed the decrease in cell viability .Flow cytometry revealed that lycopene treatment attenuated ROS content under the condition of t-BHP exposure.In addition, the protein level of Bcl-2 was increased, and the expression of Bax , cleaved caspase-3 and cytochrome-C was suppressed in lycopene +t-BHP group.CONCLUSION:The protective effect of lycopene on cortical neurons with t-BHP-induced injury may be involved in the mechanism of neuro-nal antioxidative response by down-regulating caspase-3 and Bax/Bcl-2 through the mitochondrial apoptotic pathway .
6.Tissue culture and plant regeneration of Rhodiola henryi.

OBJECTIVETo study the tissue culture and plant regeneration technologies and optimizing propagation system in vitro of Rhodiola henryi.
METHODOrthogonal experiment designs were used in the study of Rh. henryi callus induction, shoot formation and rooting, and the data were analyzed by range analysis and variance analysis.
RESULTThe optimal media to induce multiple callus from leaves were MS supplemented with 2,4-D 1.5 mg x L(-1) and 6-BA 0.5 mg x L the effect of the three factors was in sequence of explants > 2,4-D > 6-BA; The optimal media to induce multiple buds from stems were MS supplemented with 6-BA 1.5 mg x L\/1-1 NAA >6-BA; Plantlets were rooted on 1/2MS supplemented with IBA 1.0 mg x L-1, and rooting rate reached to 90% or more and transplant survival rate of plantlet reached 98% or more.
CONCLUSIONAn efficient system for tissue culture and plant regeneration of Rh. henryi was initially established.

7.Study on the relationship between glutathione S transferase mu gene deletion and leukemia in workers exposed to benzene.

OBJECTIVETo study the relationship between glutathione S transferase M1(GST mu) gene deletion and leukemia in workers exposed to benzene.
METHODSA matched population-based case-control survey with multivariate Logistic regression analysis was conducted in this study.
RESULTSIn the population of 34 patients and their matched controls, the absence of the GST mu genotype conferred odds ratio of 3.6. It suggested that GST mu was an important determinant of heterogeneity in individual susceptibility to leukemia associated with exposure to benzene. The single-variance analysis indicated that these markedly significant factors were GST mu gene deletion, GST mu isoenzyme activity, duration of exposure, GST isoenzyme activity, smoking quantity and average concentration of benzene in workshop air. The multivariate analysis indicated that these markedly significant factors were GST mu gene deletion, duration of exposure to benzene and GST mu isoenzyme activity.
CONCLUSIONGST mu gene deletion may be associated with increased risk of leukemia in workers exposed to benzene and is one of genetically determined factors.

8.Knowledge,attitude and practice related with AIDS of floating population in Hongkou District,Shanghai

Objective To evaluate the knowledge,attitude and practice(KAP)related with AIDS of local floating population.Methods A total of 1 942 floating persons were sampled by multistage cluster sampling and interviewed with questionnaire in the agricultural markets,construction sites,and their habitats within 3 communities in Hongkou District.Results 82.9% of the interviewees were 20 ~49 years old.The average score was 34.77 ±3.52 (maximum score is 44)for knowledge,12.11 ±2.32(maximum score is 19)for attitude,5.50 ±0.95(maximum score is 7)for practice,respectively.The scores increased with educational level significantly.The scores of knowledge were 33.26 ±3.54,34.63 ±3.23,36.56 ±3.39 among the subjects with educational levels of primary school and below,junior high school,senior high school and above.The scores of attitude were 13.77 ±2.27,14.79 ±2.39,15.62 ±2.38,respectively.And the score of practice was 5.40 ±0.90,5.51 ±0.93,5.58 ±1.03,respectively.Conclusion In the present,the KAP relating AIDS of local floating population is poor in Hongkou District.A variety of intervention methods and operational patterns should be developed for the floating population with different educational levels and jobs.
9.Construction and functional analysis of a common gene targeting vector with double-selection markers.

Homologous recombination is an important technique that is used to modify mammalian genome. Here, we constructed an efficient common gene targeting vector based on the plasmid pBS246. The vector consisted two positive selection markers, neomycin resistance gene (neo) and enhanced green fluorescent protein gene (EGFP) flanked by locus of X-over P1 (LoxP) sites. Two synthesized multiple cloning sequences MCS-1 and MCS-2 that contain several "8 bp cutter" enzyme sites were placed in outside of LoxP sites. Additionally, a negative selection marker HSV-tk (herpes simplex virus thymidine kinase) gene was located adjacent to MCS-1 site. The constructed vector was named pGT-V1, and its functions were characterized in C2C12 cells. The vector had the following unique features: 1) EGFP was used to monitor instantly the transfection rate that was essential for increasing the efficiency of gene knockout (KO); 2) The EGFP marker located between two LoxP sites was able to be removed from KO positive cells to avoid the potential damage of selection markers to the recipient cells. The process could be monitored visually and the positive cells without selecting markers (the loss of green fluorescent cells) could be sorted out by either flow cytometry or immunomagnetic beads; 3) "8 bp cutter" restriction sites were embedded in MCS sequences, which then enhanced the versatility of this vector. In summary, the constructed plasmid optimized the vector of gene targeting and provided a new technique means for the transgenic animal research.

10.iTRAQ-based Proteomic Analysis of Serum Differential Proteins from Henoch-Schönlein Purpura Patients Before and After Qishun Baolier Treatment

ObjectiveHenoch-Schönlein purpura(HSP) is one of the dominant diseases in Mongolian medicine. Qishun Baolier(QSBLE), as the main prescription for the treatment of HSP, has significant clinical effect, but its mechanism is not yet clear. Baed on this, this study is intended to screen the differentially expressed proteins before and after treatment, and preliminarily explore the molecular mechanism of QSBLE in the treatment of HSP. MethodTaking oneself as the control, 30 HSP patients aged 6-45 years were collected, and QSBLE was taken orally at 12:00 and 24:00, respectively. The dose was adjusted according to age and the course of treatment was one week. The distribution of proteinuria, hematuria and skin purpura of all patients were determined before and after treatment. The serum samples of 10 patients with clinically significant remission after QSBLE treatment were randomly selected for proteomics. Isobaric tags for relative and absolute quantification(iTRAQ) combined with liquid chromatography tandem mass spectrometry(LC-MS/MS) was used to analyze the proteins in serum of HSP patients before and after treatment, and differential proteins were analyzed bioinformatically and the protein-protein interaction(PPI) networks were constructed. ResultA total of 378 proteins were identified from serum, including 18 differentially expressed proteins, of which 15 proteins were up-regulated and 3 proteins were down regulated. Bioinformatics showed that the differential proteins were mainly involved in biological processes such as immune response, immunoglobulin production, phagocytosis, adaptive immune response before and after treatment. Biological processes, pathways and proteins were used to construct the PPI network, the proteins represented by immunoglobulin heavy constant γ1(IGHG1), immunoglobulin λ-chain 7-43(IGLV7-43), gelsolin(GSN) and 60 kDa heat shock protein(HSPD1) were involved in biological processes and related pathways such as adaptive immune response, immunoglobulin production, leukocyte-mediated immunity, regulation of stress response, regulation of immune system processes, regulation of trauma response, and these proteins were at the center of the PPI network. ConclusionQSBLE may play a role in the treatment of HSP by regulating the expression of IGHG1, IGLV7-43, GSN, HSPD1 and other key proteins to affect immune-related biological processes.
