2.Potential role of Wnt signaling pathway in epidermal cell dedifferentiation under traumatic microenvironment

Objective To investigate the inductive role of traumatic microenvironment in dedifferentiation of epidermal cells and explore the potential role of Wnt signaling pathway in this biological process. Methods The sheets of human foreskin were digested overnight after removal of adipose tissue, and then the epidermis was separated from the dermis. The separated epidermis sheets were repeatedly adhered to type Ⅳ collagen and flushed to remove the epidermal stem cells. The obtained epidermis sheets were transplanted onto the full-thickness skin wounds on the back of BALB/c nude mice, five days after which the cell lineage was evaluated by immunohistochemistry and the expressions of Wnts and downstream components in the grafted epidermal sheets examined by RT-PCR and Western blot. Results The cells in the basal layer of full-thickness epidermal sheets were positive for CK19 and β1 integrin and negative for CK10. While the cells in uhrathin epidermal sheets treated with type Ⅳ collagen were fully positive for CK10. Five days after transplantation of the ultrathin epidermal sheets, cells negative for CK10 but positive for CK19 and β1 integrin emerged at the wound-neighboring side of the skin grafts. At the same time, the expressions of Wnt-10b, Wnt-4 and Wnt-7a mRNA were increased by about 3.1-fold, 2.2-fold and 1.4-fold independently after transplantation. Furthermore, the expressions of β-catenin and β-catenin target genes (cyclin D1 and c-myc) were elevated by about 3-fold, 1.5-fold and 2-fold respectively in the grafted epidermal sheets (P < 0.01). Conclusion Traumatic microenvironment can induce epidermal cell dedifferentiation, when the Wnt/β -catenin signaling pathway may play an important role.
3.Abnormal visual evoked potentials in patients with Parkinson’s disease

Objective To investigate the changes of visual evoked potentials (VEP) in patients with Parkinson’s disease (PD). Methods Pattern reversal checkerboard VEP and the unified Parkinson’s disease rating scale (UPDRS) were evaluated in 69 patients with PD and 61 healthy controls. The 69 patients and 61 healthy controls were divided into an aged group (≥60 years old) and a non aged group (0.05). Conclusions VEP reflected objectively the patients’ electrophysical changes of the visual pathway due to dopamine changes and it may be helpful in PD diagnosis.
4.Effects of exogenous adrenomedullin on expressions of IL-4 and IFN-γ in the early period of mechanical renal trauma of the rats

Objective To observe the effects of exogenous adrenomedullin(ADM)on expressions of IL-4 and IFN-γ following mechanical renal trauma in the rats. Methods A total of 104 healthy adult plain grade Wistar rats were,randomly divided into four groups,ie,normal control group(eight rats),simple trauma group(32 rats),prevention group(32 rats,injected with ADM before trauma)and treatment group(32 rats,injected with ADM after trauma).The experimental model of rat with mechanical renal trauma was prepared by striking the ridge of the left rib area with free dropping forrous hammer in all groups except for the control group.Ten minutes before and after mechanical renal trauma,ADM ly.All rats were sacrificed by draining out all the blood in their hearts at 1,6,12 and 24 hours after mechanical renal trauma.Then,the renal tissues were removed and fixed with 10% formalin for observing the positive expressions of IL-4 and IFN-γ by means of SABC staining. Results Compared with the simple trauma group,the positive expression of IL-4 in the prevention group Wag observed at 1 hour,gradually increased at 6 and 12 hours and reached the peak at 24 hours after mechanical renal trauma,with statistical difference(P<0.05).The positive expression of IL-4 in the treatment group WaS increased at one hour and reached the peak at 6 hours after mechanical renal trauma,with statistical difference(P<0.05).Compared with the simple trauma group,the IFN-γ expression in the prevention group was,increased at 6 and 24 hours but decreased at 12 hour,while that in the treatment group was decreased significantly at 1,12 and 24 hours.Compared with the normal control group,the IFN-γ expression in the treatment group was significantly decreased at 1 and 12 hours.The IFN-γ expression in the treatment group was lower than that in the prevention group at every time points,with statistical difference(P<0.05). Conclusions Preventive and therapeutic administration of exogenous ADM can enhance the expression of IL-4.IL-4 and IFN-γ play an antagonistic role in repair ofthe renal injury.The primary role of exogenous ADM is the dynamic regulation of IFN-γ expression.
5.Quercetin's protective effect against oxidative stress in and impact on biological activity of B10BR, an immortal mouse melanocyte cell line

Objective To investigate quercetin's protective effect against oxidative stress in and impact on the biological activity of mouse B10BR melanocytes. Methods B10BR cells were cultured and treated with different concentrations of quercetin followed by additional culture. Then, cell viability was measured by using MTT assay, hydrogen peroxide-induced cell apoptosis by flow cytometry, and cell morphological changes by microscopy. The tyrosinase activity in and melanin synthesis by B10BR cells were measured by dopa oxidation assay and sodium hydroxide (NaOH)-lysis method, respectively. Results After treatment with quercetin of 33.33 μmol/L for 24 hours, the survival rate of B10BR cells reached (94.22 ± 3.36)%, tyrosinase activity (107.15 ± 10.96)%, and melanin content (111.85 ± 9.49)%. A significant difference was observed in tyrosinase activity and melanin content between hydrogen peroxide-induced and 33.33 μmol/L quercetin-treated B10BR cells and those only induced by hydrogen peroxide (both P < 0.01). Flow cytometry revealed that quercetin inhibited hydrogen peroxide-induced apoptosis in melanocytes. Conclusion The protective effect of quercetin against hydrogen peroxide-induced apoptosis in melanocytes may provide a new idea for the treatment of vitiligo.
6.Isolation, culture and phenotypic investigation of dedifferentiation-derived epidermal stem cells

Objective To isolate the dedifferentiation-derived epidermal stem cells (DDESCs)to further investigate their phenotypic characteristics. Methods The sheets of human foreskin were digested overnight after removal of adipose tissues and then the epidermis was separated from the dermis.The epidermis sheets which eliminated basal stem cells by repeated adhesion to type Ⅳ collagen and flushing were transplanted onto the full-thickness skin wounds on the back of BALB/c nude mice. After five days, the sheets were collected and digested into single cells, after which the percentages of positive cells of CK10, CK19 and β1 integrin were detected by flow cytometric analysis. DDESCs were isolated by rapid adhesion to type Ⅳ collagen. The expressions of CK19, β1 integrin, Oct4 and Nanog in the cells were examined using immunofluorescence and quantitative real-time polymerase chain reaction (RTPCR). Results The percentages of positive cells of CK19 and 31 integrin were increased (P <0.01 )and those of CK10 in the transplanted sheets decreased ( P <0.01 ) five days after transplantation. Isolation of DDESCs by repeated adhesion to type Ⅳ collagen showed 4.56% adhering cells in the transplantation group within 10 minutes. The in vitro phenotypic assays showed that the expressions of CK19, β1 integrin, Oct4 and Nanog in DDESCs were similar to those of original epidermal stem cells ( P >0.05 ) but remarkably higher than those in the control group ( P < 0.01 ). Conclusion The phenotypic characteristics of DDESCs cultured in vitro are similar to those of epidermal stem cells, indicating a new approach for wound repair and regeneration.
7.Application of 3D-power Doppler ultrasound in diagnosis of breast carcinoma in elderly patients

Objective To explore the diagnosis value of 3D-power Doppler ultrasound in differentiating benign and malignant breast masses in elderly patients.Methods A retrospective study of 53 lesions in 45 patients was performed,and breast masses were divided into benign and malignant group by pathological diagnosis after operation.Display of cross,vertical and coronal plane of total 53 masses were automatically acquired by 3D-power Doppler examination before operation,and 3D imaging of masses were reconstructed by virtual organ computer-aided analysis.The vascular morphology inside and beside the breast neoplasm was observed by tomographic ultrasound imagine layer by layer in different sections.Vascular distribution characteristics were divided into penetrating,central,surrounding and few vessel type.Quantitative analysis of vascularization-flow index inside the breast neoplasm was measured by 3D-power Doppler histogram.Results The difference in vascular distribution characteristics by 3D-power Doppler ultrasound was statistically significant between 15 (28.3%) benign and 38 (71.7%) malignant lesions(x2=19.52,P<0.01).The case number of presence of penetrating vessels was 26(68.4%)in the malignantuniors.The sensitivity and specificity of penetrating vessel diagnosed malignant breast tumor were 68.4% and 86.7%.Vascular distribution characteristics of benign masses was mainly of surrounding vessel type(53.3%).Vascularization-flow index was higher in malignant group than in benign group [(1.32 ± 1.58) vs.(0.63 ± 0.53),t=2.187,P<0.05].Conclusions 3D-power Doppler ultrasound completely presents the vessel stereoscopical distribution and shows quantificationally blood perfusion of breast neoplasm.The 3D-power Doppler ultrasound is superior to conventional ultrasound for enhancing differential diagnosis ability between benign and malignant breast tumors.
8.Epidemiological situation of human parasitic diseases through three investi-gations in Jinhu County,Jiangsu Province

Objective To understand the epidemiological situation of human parasitic diseases in Jinhu County,so as to pro?vide the evidence for formulating further control strategy. Methods The investigated local residents were sampled by the ran?dom cluster sampling method in 1989,1999 and 2015. The infections of intestinal helminthes were detected by Kato?Katz tech?nique,the eggs of Enterubius vermicularis were examined by cellophane anal swab for children,the intestinal protozoa were ex?amined by the saline smear and iodine staining methods. Results The total rates of parasitic infections were 62.57%,9.32%and 0.49%in 1989,1999 and 2015,respectively. Compared to those in 1989 and in 1999,the infection rate in 2015 was de?creased by 99.22%and 94.74%,respectively. The numbers of detected parasite species were 14,10 and 4 in 1989,1999 and 2015,respectively. The intensities of infections were mainly mild in three investigations,and the intensities of all the infections in 2015 were mild. The species of infected parasites were mainly single,however,multiple infections were observed in 1989,in?cluding 4 parasite species(0.72%)and 3 parasite species(7.02%). Only in 1989,the difference between sexes was significant and the infection rate of the female was higher than that of the male(χ2=18.01,P<0.01). Conclusions The infection rates of human parasites are decreased gradually and stabilized at the low level in Jinhu County. However,the surveillance work still should be strengthened to consolidate the achievement.

10.Changes of intelligence and event-related potential P300 in the elderly patients with asymptomatic cerebral infarction

Objective To explore the characteristics of intellectual impairment and event -related potential(ERP) in the elderly patients with asymptomatic cerebral infarction (ACI). Methods By using WAIS-RC and sensory event-related potential, VIQ, PIQ, FIQ, the latency of all the waves of ERP and the amplitude of P300 were measured in 53 elderly patients with ACI and 50 healthy controls. The 53 ACI patients were divided into a single-infarct cerebral lesion (30 cases) and multi-infarct cerebral lesion (23 cases).
