1.Retrospective analysis on pathological features of 1561 patients with colorectal cancers in fifteen years

Objective To retrospectively study pathological characteristics of 1561 patients with colorectal cancer in Nanchang area.Methods Analysis was made regarding the results of colonoscopy and pathological examination in patients from 1990 to 2004.Results Totally 21 853 patients received colonoscopy during the period,and 1561 patients were diagnosed as colorectal cancer.The overall diag- nostic yield was 7.1%.The average age diagnosed was 52 years old.Ratio of male to female was 1.4: 1.The lesion located in rectum accounted for 43.4%.The percentage of cancer in left colon was higher than that in right side (30.5% vs 26.1%,P<0.05).The proportion (57.5%) of patients with rectal can-cer under 30 years old was the highest in all patients with rectal cancer.Cancer located in sigmoid colon and left side of the colon increased with age.Well and moderately differentiated adenocarcinoma was observed in 83.6% of the patients.Comparing the clinical characteristics of every five years,increasing diagnostic yield was noted from 1990 to 1999,hut the decreased trend was found in last five years.The propor- tions of rectal cancer was also increased in the last fifteen years,and overall proportions of cancer located in left side of colon was decreased.Conclusions The overall diagnostic yield of colorectal cancer in Nanchang area was lower than other province,especially rectal cancer.The proportion of cancer of left side of colon was increased with age.The well and moderately differentiated adenocarcinoma was found increasing in the last fifteen years, while the poorly differentiated adenocarcinoma was decreased.
2.Molecular mechanism of Golgi protein 73 in inflammation

Objective To study the effect of Golgi protein 73(GP73) on inflammation, and to reveal the effect of GP73 on tumorigenesis and metastasis.Methods The transcriptional activity of NF-κB and the expression of IL-1β, IL-6 and TNF-αwith GP73 overexpression or knockdown were detected to illuminate the role of GP73 in inflammation.According to the TCGA database, the correlation between the transcriptional activity of GP73 and the expression of NF-κB, IL-1β, IL-6 and TNF-αwas analyzed to determine the role of GP73 in tumor inflammation.Results Correlative analysis showed that there was a positive correlation between the expression of GP73 with NF-κB, IL-1β, IL-6 and TNF-α.The transcriptional activity of NF-κB was upregulated by GP73 overexpression, but downregulated by GP73 knockdown.The expression of IL-1β, IL-6 and TNF-αwas upregulated by GP73 overexpression.Ammonium pyrrolidinedithiocarbamate ( PDTC ) was in-volved in inflammation reaction induced by GP73.Conclusion GP73 is possibly involved in inflammation and promotes tu-morigenesis and metastasis.
3.Construction of H22 GP73 knockout gene stable strain using CRISPR/Cas9 gene editing system and identification of functions

Objective To knock out the GP73 gene in H22 cells originating in mice using CRISPR/Cas9 gene editing system and construct H22 GP73 gene knockout stable strain for identification of its functions .Methods Two pairs of sgRNAs that could specifically identify the upstream and downstream of GP 73 gene first promoter were designed before a recombinant eukaryotic expressional plasmid was constructed using pX 459 .After enzyme digestion and sequencing , two pairs of recombinant plasmids were co-transfected into H22 cells before puromycin was used to screen positive cells and monoclonal cells which stably knocked out GP 73 gene were developed .The knockout effect was measured by Western blotting.Cell Titer 96? AQueous One Solution Assay was used to detect the effect on cell reproductive capacity when the GP73 was knocked out .The transferability was detected through wound healing test .Results The result of Western blotting suggested that GP73 protein was undetected in the construction of H22 GP73 knockout gene stable strain after transfection.The transfer and reproduction slowed down .Conclusion H22 GP73 gene knockout stable strain can be successfully built using CRISPR/Cas9 gene editing system ,thus facilitating studies on the function of GP 73 in hepatocarcinogenesis .
4.Amniotic fluid chromosome detection for 95 NIPT high-risk pregnant women

Objective To analysis of the detection result of amniotic fluid chromosome which in NIPT high-risk pregnant women.Methods Amniotic fluid cells via amniotic cavity puncture were cultured and analyzed,the chromosome karyotypes were observed.Results The highest positive predictive value of NIPT was for trisomy 21(85.00%),then trisomy 18(75.00%),sex chromosome abnormalities(68.00%),other chromosome abnormalities(41.67%),trisomy 13 (25.00%).Conclusion The highest accuracy of NIPT was shown in detection of Down''s syndrome by NIPT.NIPT was screening test which is effective and noninvasive in prenatal diagnosis.Amniotic fluid Chromosomal karyotype analysis was the gold standard in the diagnosis of fetal chromosomal disease.
5.BPOZ inhibits proliferation of breast cancer cell lines

Objective To investigate the effect of BTB/POZ ( broad complex, tramtrack and bric a brac/poxviruses and zinc finger) on proliferation of breast cancer cell lines.Methods The eukaryotic expression plasmid pCMV-HA-BPOZ was constructed by cloning from cDNA of human genome.Western blotting was used to detect the expression of pCMV-HA-BPOZ.Cells growth assay was used to detect the effect of BPOZ on proliferation and colony formation assay was used to detect the effect of BPOZ on cell growth ability.Results Western blotting showed that HA-BPOZ was efficiently expressed in cells.Moreover, the growth ability and proliferation of cells were significantly inhibited in BPOZ overexpressed cells compared with the control cells (both P <0.05).Conclusionp CMV-HA-BPOZ plasmid is constructed.BPOZ can restrain breast cancer cell lines MCF7 and ZR-75-1 cells from proliferating and growing.The results of our study can con-tribute to the study of functions of BPOZ in breast cancer.
6.Analysis of surveillance data of foodborne disease from a level a tertiary general hospital in Shandong province from 2019 to 2021

Objective Analyze the surveillance results of foodborne illness from a level A tertiary general hospital in Shandong Province from 2019 to 2021,to provide a realistic basis for the continuous development and improvement of foodborne disease surveillance.Methods The data were collected from a level A tertiary general hospital in Shandong Province from 2019 to 2021 in the food safety risk assessment business application platform of national health insurance information project of National Health Security Disease Control Information System,and stool/anal swab samples were collected as required for pathogen detec-tion.Results From 2019 to 2021,a total of 2 323 cases of foodborne diseases were reported and 682 samples were collected,with a sampling rate of 29.36%,and 134 positive cases were detected,with a detection rate of 19.65%.The difference between the detection rates of different years was statistically significant(P<0.05).The detection rate of norovirus was the highest(9.53%),followed by Vibrio parahaemolyticus(8.94%)and Salmonella(1.91%).The difference between the detection rates of different pathogens was statistically significant(P<0.05).The sex ratio of 2 323 cases was 1:1.28.Among the different age distributions,20-29 years old accounted for the highest proportion(29.19%),followed by 30-39 years old(27.21%)and<20 years old(11.71%).The highest number of cases was reported from June to September,the peak was reached from July to Au-gust,and the number of cases was gradually reduced in October and later.Among different occupational groups,housework and unemployed people accounted for the highest proportion(30.18%),followed by farmers(15.93%),students(13.65%)and commercial service workers(11.80%).The main symptoms of the digestive system were diarrhea in 2 093 cases(90.10%),abdominal pain in 1 741 cases(74.95%),nausea in 1 575 cases(67.8%)and vomiting in 1 401 cases(60.31%).The main diarrhea was watery stool in 1 954 cases(84.12%).The main systemic symptoms were fatigue in 454 cases(19.54%)and dehydration in 212 cases(9.13%).The main clinical diagnosis was acute gastroenteritis in 1 980 cases(85.23%)and infec-tious diarrhea in 199 cases(8.57%).The suspected exposed foods were mainly aquatic animals and their products(25.10%)and meat and meat products(19.37%).The main eating places of suspected exposed foods were families(57.34%),followed by restaurants(21.91%),other places(5.94%)and street food stalls(5.21%).Conclusion From 2019 to 2021,the pathogens of foodborne diseases detected were mainly norovirus and Vibrio parahaemolyticus,mainly in the age group of 20 to 39 years old,and the occupation distribution was mainly household and unemployed people,farmers,students and commercial serv-ice personnel,with digestive system symptoms such as nausea,vomiting,abdominal pain and diarrhea.The suspected exposed foods are mainly aquatic animals and their products and meat and meat products,and the main eating places are families,restau-rants and street food stalls.The surveillance and reporting of foodborne diseases should continue to be strengthened in the future.
7.Effect of melatonin on prefrontal cortex ischemia-induced cognitive impairment in rats and the receptor mechanism

Objective:To evaluate the effect of melatonin on prefrontal cortex ischemia-induced cognitive impairment in rats and to investigate the receptor mechanism.Methods:Clean-grade adult male Sprague-Dawley rats, weighing 300 g, were selected, and a catheter was implanted into the prefrontal cortex.The experiment was performed in two parts.Experiment Ⅰ Twenty-four rats, in which catheters were successfully inserted into the prefrontal cortex, were assigned into 3 groups ( n=8 each) using a random number table method: control group (group C), model group (group M) and melatonin group (group ME). Normal saline 0.5 μl was injected into the prefrontal cortex in group C, 1 μmol/L endothelin 0.5 μl was microinjected into the prefrontal cortex in group M, and 1 μmol/L endothelin and 1 μmol/L melatonin 0.5 μl were injected into the prefrontal cortex in group ME.Experiment Ⅱ Forty-four rats, in which catheters were successfully inserted into the prefrontal cortex, were assigned into 4 groups ( n=11 each) using a random number table method: model group (group M), melatonin group (group ME), MT 1/2R antagonist luzindole + melatonin group (group L + ME) and MT 2R antagonist 4p-pdot + melatonin group (group P + ME). In group M, 1 μmol/l endothelin 0.5 μl was microinjected into the prefrontal cortex.In group ME, 1 μmol/L endothelin + 1 μmol/L melatonin 0.5 μl was injected into the prefrontal cortex.In group L + ME, 1 μmol/L endothelin + 1 μmol/L MT 1/2R antagonist + 1 μmol/L melatonin 0.5 μl was injected into the prefrontal cortex.In group P + ME, 1 μmol/L endothelin + 1 μmol/L MT 2R antagonist + 1 μmol/L melatonin 0.5 μl was injected into the prefrontal cortex.T-maze and the open field tests were performed at 1 week after administration. Results:Experiment Ⅰ There was no significant difference in the locomotor speed in open field test among C, M and ME groups ( P>0.05). The rate of correct selection in T-maze test was significantly lower in M and ME groups than in group C and higher in group ME than in group M( P<0.05). Experiment Ⅱ There was no significant difference in the locomotor speed in open field test among the four groups( P>0.05). Compared with group M, the rate of correct selection in open field test was significantly increased in ME and P+ ME groups ( P<0.05), and no significant change was found in group L+ ME ( P>0.05). Compared with group ME, the rate of correct selection in open field test was significantly decreased in group L+ ME ( P<0.05), and no significant change was found in group P+ ME( P>0.05). Conclusion:Melatonin can attenuate prefrontal cortex ischemia-induced cognitive impairment in the rats, and the mechanism is related to activation of MT 1R.
8.Genomic characteristics of an echovirus 20 strain (KM/EV20/2010) isolated in Kunming, Yunnan, China.

OBJECTIVETo identify the complete genome sequence of an echovirus 20 isolate (KM/EV20/2010) and understand its genetic variation and evolution characteristics.
METHODSSeven overlapping clones covering the whole viral genome (excluding the poly-A tail) were obtained by RT-PCR and sequenced, and their nucleotide and amino acid sequences were aligned with other echovirus 20 isolates. Phylogenetic and pairwise alignment analyses based on genome and complete VP1 regions were conducted with software Mega 4.1, RDP3 and SimPlot 3.5.1.
RESULTSThe genome of the echovirus strain was 7 395 nucleotides in length, and contained a 744-nt non-translated region (NTR) at the 5' end and a 96-nt NTR at the 3' end. The entire open reading frame contained 6 549 nt, encoding a 2 183-aa polyprotein. In the coding region, there was no nucleotide deletion or insertion. Based on the complete genome sequence alignments, the echovirus strain showed 80.1% nucleotide and 96.7% amino acid homology to echovirus 20 prototype JV-1 strain. The phylogenetic trees constructed on the genome and complete VP1 regions all indicated that the echovirus strain was not in one cluster with echovirus 20 prototype JV-1 strain, while had a closer relationship with echovirus 30 prototype Bastianni. Genotyping results from phylogenetic analysis showed that echovirus 20 has six genotypes. The strain used in this study belonged to genotype IV. The nucleotide divergence was 9.4%-21.7% among the 6 genotypes. The possible putative recombination was detected in its non-coding sequence of the echovirus 20 strain used in this study.
CONCLUSIONBased on the bioinformatics analysis. The echovirus 20 strains isolated in China could be divided into six genotypes, and the echovirus 20 in this study belonged to genotype IV.
