2.RELATIONSHIP BETWEEN THE ACTIVITY OF ALKALINE PHOSPHATES AND DEGENERATIVE CARDIAC VALVULAR CALCIFICATION

To explore the association between alkaline phosphatase (ALP) and degenerative cardiac valvular calcification, the activity of ALP and the grades 0~Ⅳ degenerative calcified valves were determined by biochemical and immunocytochemical methods. ALP positive substance was present in the matrix around the foci of calcification and in the cytoplasm of fibroblasts. The bioactivity of ALP was much higher in grades Ⅰ and Ⅱ calcified valves than that in grades 0 and Ⅲ~Ⅳ calcified valves. The results indicate that the level of ALP bioactivity is predominantly dependent on the process of extruding matrix vesicles from senescent cells in the valves.
3.Evaluation of Therapeutic Effect of Recombinant Human Granulocyte-macrophage Colony-stimulating Factor Combined with SD-Ag Unguent on Deep Second Degree Burn

Objective To evaluate the therapeutic effect of recombinant human granulocyte-macrophage colony stimulating factor ( rhGM-CSF) combined with sulfadiazine silver ( SD-Ag) unguent on deep second degree burn. Methods Eighty-nine patients with deep second-degree burn (burns areas<10%) were enrolled.These patients were divided into two groups at random (Group A and Group B).The patients in Group A were treated with SD-Ag unguent only, and those in Group B were treated with rhGM-CSF and SD-Ag unguent.The therapeutic effects and the adverse drug reaction were recorded. Results The healing time in Group A [(19.79±1.47) days] was obviously shorter than that in Group B [(15.76±1.63) days].The total wound healing rates in Group B [on day 9:(76.41±3.24)%, day 13:(95.01±1.43)%, day 16:(99.54±0.88)%, and day 21 (100.00±0.00)%] were higher than those of Group A [on day 9: (67.24±2.33)%, day 13: (75.54±1.11)%, day 16:(88.33±1.32)%, day 21:(99.14±1.95)%].During the treatment, there was no obvious adverse reaction was observed in the two groups. Conclusion The application of rhGM-CSF combined with SD-Ag unguent can not only accelerate the healing rates of deep second-degree burn, but also has curative effect with safety.
4.UHRF1 expression inhibition by RNA interference enhances the radiosensitivity of esophageal cancer cells

Objective To study the effect of UHRF1 expression inhibition by RNA interference on the radiosensitivity of esophageal cancer cell line TE-1 and its mechanism.Methods Short hairpin RNA (shRNA) targeting UHRF1 gene was introduced into TE-1 cells by lentivector-mediated transfer.The cells were divided into three groups:non-transfected group,negative control (NC)-shRNA-transfected group,and UHRF1-shRNA-transfected group.The mRNA and protein expression levels of UHRF1 in TE-1 cells were measured by RT-PCR and Western blot before and after transfection.After transfection and X-ray radiation,the radiosensitivity of TE-1 cells was evaluated by colony formation assay; the cell cycle and cell apoptosis were determined by flow cytometry; the γ-H2AX (as a marker of DNA damage) level was measured by Western blot.Results After transfection with UHRF1-shRNA,the mRNA and protein expression levels of UHRF1 were significantly decreased in TE-1 cells,as compared with those in the NC-shRNA-transfected group and non-transfected group (0.11 vs 0.96 and 0.98,F =124.21,P =0.000;0.10 vs 0.89 and 0.94,F =125.25,P =0.000).The UHRF1-shRNA-transfected group had sensitization enhancement ratios of 1.53 (D0 ratio) and 1.95 (Dq ratio).X-ray radiation could cause G2/M arrest and increase apoptotic rate and γ-H2AX expression in TE-1 cells.Compared with the two control groups,the UHRF1-shRNA-transfected group showed significantly less G2/M arrest (F =500.15,P =0.000),a significantly higher apoptotic rate (F =100.10,P =0.000),and significantly higher residual γ-H2AX expression (F =61.00,P =0.000) at 24 hours after X-ray radiation.Conclusions RNA interference can effectively inhibit the UHRF1 expression and enhance the radiosensitivity of TE-1 cells.The mechanism may be related to cell cycle regulation,cell apoptosis,and DNA damage repair.

6.Analysis of CYP2C19 metabolism phenotypes distribution of 76 patients with clopidogrel treatment in Huangshi

Objective To investigate the distribution of CYP2C19 metabolism phenotype in patients with clopidogrel treatment in Huangshi.Methods The peripheral blood samples were obtained from 76 patients with clopidogrel treatment in Cardiovascular Department.CYP2C19 genotypes were determined by the gene chip,CYP2C19 metabolism phenotype were investigated and com-pared with the data of healthy Han Chinese from the published papers.Results There were three kinds of metabolizers about the metabolism phenotypes of CYP2C19,which were extensive metabolizers,intermediate metabolizers and poor metabolizers.In the 76 patients,the ratios of these metabolism phenotypes were 39.47%,44.74% and 15.79%,respectively.The result coincided with the healthy Han Chinese.Conclusion The clinical individualized medication of healthy Han Chinese could be refered to the clinical indi-vidualized medication of patients with clopidogrel treatment in Huangshi.
7.Study on the determination method of chromium picolinate in health food

For developing the determination method of chromium picolinate in health food by HPLC,extraction method and HPLC operation conditions such as columns,mobile phase had been optimized.Long term stability test of chromium picolinate had been done.Recovery of the standard spiked >93%,the RSD was 3.1,the lowest detection limit was 10.0 mg/kg,the result fits the demand of the analysis of health food.This method can be used in analysis of the content of chromium picolinate in types of tablet and capsule health food.2 years store experiment in room temperature showed that chromium picolinate was stable.
8.Research progress in bacterial resistance to antibiotics analyzed based on MALDI-TOF MS mass peak

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has become a widely used method in the field of microbial identification. Its obvious advantages including rapidity, great accuracy and high throughput attract many researchers to investigate its potential for usage in other microbiological fields. Currently, several studies have reported MALDI-TOF MS-based analysis of bacterial resistance to antibiotics, which can identify the specific heterogeneous spectrum peaks related to drug resistance through simple visual analysis of the spectrum or more complex analysis of the entire spectrum using informatics methods and statistical approaches. Therefore, MALDI-TOF MS has become a potential tool for detecting antibiotic resistance in bacteria. This review mainly summarized the progress in MALDI-TOF MS-based analysis of bacterial resistance to antibiotics.
9.Long-term efficacy of intensity-modulated radiotherapy with or without chemotherapy in treatment of esophageal carcinoma:an analysis of 349 patients

Objective To observe the efficacy and adverse reactions of intensity-modulated radiotherapy (IMRT) with or without chemotherapy in the treatment of esophageal carcinoma,and to explore the influencing factors for prognosis.Methods The short-term outcomes and acute adverse reactions in 349 patients with esophageal carcinoma who received IMRT with or without chemotherapy from 2006 to 2012 were retrospectively analyzed.The 1-,3-,and 5-year local control (LC) rates and overall survival (OS) rates were calculated with the Kaplan-Meier method.The influencing factors for survival were analyzed using the Cox regression model.Results The sample sizes at 3 and 5 years were 174 and 63,respectively.For all patients,the 1-,3-,and 5-year LC rates were 72.9%,61.2%,and 58.4%,respectively,and the 1-,3-,and 5-year OS rates were 66.5%,39.1%,and 24%,respectively.According to the results of subgroup analysis,the 1-,3-,and 5-year LC and OS rates in patients with a tumor volume of < 54.73 cm3 were significantly higher than those in patients with a tumor size of ≥54.73 cm3 (P =0.001 and 0.000).There were no significant differences in 1-,3-,and 5-year LC rates between patients with and without lymph node metastasis (P =?).However,the 1-,3-,and 5-year OS rates were significantly lower in patients with lymph node metastasis than in patients without lymph node metastasis (62.7% vs.83.1%;35.9% vs.53.3%;20.4% vs.38.3%;P =0.003).There were significant differences in the 1-,3-,and 5-year LC and OS rates between patients with complete response,partial response,and no response (P =0.000 and 0.000).The incidence rates of grade ≥ 2 acute radiation pneumonitis and grade ≥ 3 acute radiation esophagitis were 11.3% and 9.0%,respectively.The tumor volume,short-term outcome,and lymph node metastasis were the influencing factors for OS (P =0.038,0.000,and 0.008).Conclusions IMRT with or without chemotherapy is effective and safe in the treatment of esophageal carcinoma.The prognosis becomes poor along with increased tumor volume and regional lymph node metastasis.The evaluation of short-term outcomes is closely correlated with LC and OS.
10.Relationship between the genetic variants in SLCOI B1 gene and the response to repaglinide treatment in newly-diagnosed type 2 diabetic patients

Objective To investigate the effect of solute cartier organic anion transporter family, member 1B1 (SLCOIBI) gene variants on the response to therapy with repaglinide in type 2 diabetes. Methods 100 newly-diagnosed type 2 diabetic patients were treated with repaglinide during a course of 48 weeks. Anthropometrie parameters and indices related to glucose metabolism were measured periodically. Genotypes of SLCO1B1 D130N and V174A were detected by PCR-restriction fragment length polymorphism (RFLP) and sequencing respectively. Results Eighty-nine patients accomplished the 48-week follow-up visits. D130N variant in SLCO1B1 gene was associated with repaglinide treatment, DD genotype had better HbA1C lowering effect than N allele carrier [△HbA1C: (-2.29±0.23) % vs (-1.49±0.21)%, P<0.05]. No association was detected between D130N and the other effects of repaglinide on glucose metabolism related phenotypes. Conclusion D130N variant in SLCO1B1 gene is associated with the response to repaglinide treatment in patients with type 2 diabetes. DD homozygotes had a better effect than N allele carriers.
