Objective To optimize the matrix formulation of the extract of Chuanxiong Rhizoma cataplasm. Methods The optimal proportion of matrix formulation was selected by uniform design and response surface method. The primary adhesion force, lasting adhesion force, peel strength and residuals on the anti-sticking paper were used as test parameters. The optimal ratios of NP-700, glycinate aluminum, glycerin, tartaric acid, the concentration of drug, and the volume of drug were acquired. Results The matrix formulation of the cataplasm was made with NP-700∶glycinate aluminum∶glycerin∶tartaric acid∶concentration of drug∶volume of drug=1.1∶0.08∶7∶0.04∶4∶5. Conclusion Uniform design combined with response surface method can be used for the optimization of the matrix formulation of cataplasm. The optimal matrix prescription has higher drug-loading, with good spreadability, moisture-retention capability and adhesive force.
4. Progress in the molecular relationship between zinc deficiency and esophageal cancer
Tumor 2017;37(11):1231-1237
Esophageal cancer is a common gastrointestinal malignancy and China has a high incidence of esophageal cancer. Zinc deficiency is a key risk factor for the occurrence and development of esophageal cancer and affects its progression through various ways, including changing the expression of coding genes and non-coding genes. In addition, the lack of zinc ions can also affect the stability of zinc finger protein (ZNF). The dysregulation of ZNF and its associated long non-coding RNAs (LncRNAs) play an important role in the progression of esophageal cancer. This paper summarizes the molecular relationship between zinc deficiency and esophageal cancer, hoping to provide new strategies and methods for the early diagnosis, targeted therapies and prognosis assessment of esophageal cancer.

OBJECTIVE:To investigate chemotherapeutic efficacy and safety of Paclitaxel combined with lobaplatin by inter-ventional embolization for cervical cancer. METHODS:Totally 68 cases of cervical cancer patients selected from our hospital dur-ing May 2010-Mar. 2014 were divided into control group and observation group according to therapy plan, with 34 cases in each group. Control group was given Paclitaxel liposome for injection 175 mg/m2 added into 5% Glucose injection 250 mL,ivgtt+Loba-platin for injection 80 mg/m2 added into 5% Glucose injection 250 mL,ivgtt. Observation group was given Paclitaxel liposome for injection 175 mg/m2+Lobaplatin for injection 80 mg/m2+5% Glucose injection 10 mL via percutaneous catheter after selecting uter-ine artery and tumor vessel by arterial catheterization arteriography, gelfoam embolization. Both groups were treated in the first day of every treatment course,21 d as a treatment course, for 3 courses. Clinical efficacies were observed in 2 groups as well as the levels of T lymphocyte subsets(CD4+,CD8+,CD4+/CD8+)before and after treatment. The occurrence of ADR during treatment,lo-cal recurrence and metastasis after 2 years, and survival situation were recorded. RESULTS:The total response rate of observatio group(85.29%)was significantly higher than that of control group(61.76%),with statistical significance(P<0.05). Before treat-ment,there was no statistical significance in the levels of CD4+,CD8+ or CD4+/CD8+ between 2 groups (P>0.05). After treat-ment,CD4+and CD4+/CD8+of 2 groups were increased significantly, while CD8+was decreased significantly;above indexes of ob-servation group were improved significantly compared to control group, with statistical significance (P<0.05). The survival time of observation group [(22.58±0.61)months] was significantly longer than that of control group [(17.61±1.32)months],and 2-year survival rate(70.59%)was significantly higher than control group(47.06%),with statistical significance(P<0.05). There were no statistical significance in incidence of disease control, incidence of total ADR and incidence of local recurrence and metastasis between 2 groups (P>0.05). CONCLUSIONS:Paclitaxel combined with lobaplatin by interventional embolization show definite chemotherapeutic efficacy for cervical cancer, and can improve the levels of T lymphocyte subset with good safety.

OBJECTIVETo observe the ability of triple helix-forming oligonucleotides (TFOs) modified with manganese porphyrin to combine with and cleave HBV DNA fractions.
METHODSTFO were modified with manganese porphyrin and acridines, and then reacted with the (32)P labeled HBV DNA fragments at 37 degrees C in vitro (pH 7.4). Electrophoretic mobility shift assays and DNase I footprinting tests were used to show the affinity and specificity of TFO to bind to target sequences. The ability of TFO to cleave HBV DNA fragments was tested by cleavage experiments.
RESULTSTFO modified with manganese porphyrin and acridine could bind to the target sequence in a sequence-dependent manner, with a Kd value of 3.5 x 10(-7) mol/L and a relative affinity of 0.008. In the presence of potassium monopersulfate (KHSO(5)), TFO modified with manganese porphyrin and acridine could cleave the target sequence where the triplex DNA was formed.
CONCLUSIONIn the presence of KHSO(5), TFO modified with manganese porphyrin and acridine could bind and cleave the target HBV-DNA in a sequence-dependent manner.
