1.Study on dehydroepiandrosteron permeation from Transdermal Therapeutic Systems

To study in vitro usability of Transdermal Therapeutic Systems (TTS) dehydroepiandrosteron (DHEA) on pig's ear skin. Results showed that: there was correlation about DHEA permeation between human and pig's ear skin. DHEA permeation speed was linear proportional with square root of time and the DHEA content in TTS. DHEA permeation speed via human skin had linear correlation with permeation speed via pig's ear skin with angular co-efficient 0.74; this showed that pig's ear skin can be used for exploratory research on establishing formulation of transdermal drugs. Pig skins are available material and easy to collect with large amount. However, this result also showed that permeation ability of human skin is lower than pig’s ear skin

2.The functions of GPⅠa/Ⅱa and GPⅣin platelet adhesion to collagen

Objective To study the functions of GPⅠa/Ⅱa and GPⅣduring the course of platelet adhesion to collagen. Methods Reaction system which contains anti-GPⅠa/Ⅱa antibody ,anti-GPⅣantibody or no blocking antibody was analyzed by flow cytomety. PE labeled anti-CD42b antibody was used to select platelet and fluorescence intension of fluorescein isothiocyanate ( FITC) was detected during the course of platelet adhesion to FITC-labeled collagen. Results The antibody against GPⅠa/Ⅱa inhibited platelet adhesion to collagen, especially to activated platelet(P 0. 05 ). Conclusion GPⅠa/Ⅱa plays an important roles in platelet adhesion to collagen. Blocking GPⅠa/Ⅱa may decrease the adhesion. GPⅣmay be helpful for acceleration of platelet adhesion to collagen at early stage.
3.Function of GPⅠa/Ⅱa in the platelet-collagen interactions

Objective To investigate the function of GPⅠa/Ⅱa in the platelet-collage adhesion and the platelet granules release induced by collagen. Methods With the flow cytometry, in either presence or absence of the anti-GPⅠa/Ⅱa antibody: (1) PE-labeled anti-CD42b antibody was used for selecting platelet and FITC fluorescence intension was detected during the course of platelet adhered to FITC-labeled collagen. (2) PerCP-labeled anti-CD61 antibody was used for selecting platelet and detecting CD62P with PE-labeled CD62P antibody during platelet granule release induced by collagen. Results Anti-GPⅠa/Ⅱa antibody inhibited the platelet-collagen adhesion, especially the activated platelet-collagen adhesion (P0.05). Conclusion GPⅠa/Ⅱa plays an important role in platelet-collagen adhesion. Blocking GPⅠa/Ⅱa can decrease the platelet-collagen adhesion, but has no effects on platelet granule release reaction.

5.Quantitative Determination of Ferulic Acid Content in Chrysanthemum Morifolium cv. ( Chuju) Continuous Cropping Soil Using Near Infrared Spectroscopy

A near infrared spectroscopy ( NIRS) method was used for rapid quality evaluation of ferulic acid content in chrysanthemum morifolium cv. ( Chuju) continuous cropping soil. Standard leverage, studentized residual and Mahalanobis distance were calculated to eliminate abnormal samples. After the initial near infrared spectrum was treated by two second derivative and Norris smoothing filter noise, 6000-4000 cm-1 wave number range and 7 factors were chosen for partial least squares ( PLS) calibration model. The results showed that good correlation was presented between the calibration set/validation set and the values determined by high performance liquid chromatography, and the calibration correlation coefficient ( Rc ) and validation correlation coefficient ( Rcv ) were 0. 9914 and 0. 9935, respectively. Root mean square error of calibration (RMSEC), root mean square error of validation (RMSEP) and root mean square error of cross-validation (RMSECV) were 0. 484, 0. 539 and 0. 615, respectively. This method was accurate, reliable, simple, rapid and nondestructive, and could be applied to the fast analysis for ferulic acid in continuous cropping soil.
6.ICOSL could upregulate immune response in Graves′s disease animal models

Objective:To study the pathological mechanism of the inducible co-stimulator molecular and ligand ( ICOS/ICOSL) in Graves disease animal.Methods:45 out-bred BALB/c mice were randomly divided into three groups with 15 rats in each group;using gene gun to deliver different plasmid injection.Group A was delivered with pCDNA3.0-mICOSL and pCDNA3.0-hTSHR, Group B with pCDNA3.0-hTSHR and null pCDNA3.0 with Group C for immunization as the control group.The concentration of serum free thyroxine immunization was deter mined with immunoassay and serum thyrotropin receptor antibody ( TRAb ) with ELISA, supernatant of IFN-γconcentration in mouse spleen cells was measured with radioimmunoassay,and hTSHR transected CHO cells were incubated to detect the concentration of cAMP to deter mine autoantibody TRAb activity.Results: After plasmid injection serum FT4 level in Group A (0.49±0.25) pg/ml ( q=6.571,P=0.023) was higher than that in Group C,the standard rate was higher than Group B and C (χ2=14.47,P=0.005).IFN-γconcentration of mice spleen cultured supernatant in Group A (1.88±0.41) pmol/L was significantly higher than the other two groups.The activity of autoantibody TRAb in Group A 188.3 (179.7-260.2) %was higher than that in the other two groups ( P=0.027 ) .Conclusion: Exogenous delivery of pCDNA3.0-mICOSL plasmid in GD mice could stimulate the spleen lymphocytes to secrete more IFN-γ,increase the activity of TRAb autoantibodies and might lead to upregulation of immune response in Graves animal model in vivo.

8.Effects of Cistanchis Glycosides on Learning and Memory Impairment in Mice

Objective To observe the effects of cistanchis glycosides on scopolamine -induced impariment of learning and memory and on sodium nitrite -induced impariment of learning and memo ry consolidation in mice.Methods Step down test was performed to reflect th e learning and memory of mice.Cistan chis glycosides were administered c onsecutive-ly for 30days.The mice were trained o n the 29th day and measurements were c arried out on the 30th day .Latent period and number of errors within 5min were noted.Scopolamine was given by intr aperitoneal injection 15min before train-ing,and sodium nitrite was administered subcutaneously right after trainin g.Results(1)Compared with model mice in-duced by scopolamine,cistanchis glycosides 200and 400mg /kg together with piracetam 600mg /k g considerably in-creased latent period and decreased error times.Cistanchis glycosides100mg /kg also increased latent time but decreased error times to some extent.(2)Compared with model mice induced by s odium nitrite,cistanchis glycosides 200and 400mg /kg increased latent time significantly but had no significant effect on the error times.Cistanchis glycosides 100mg /kg decreased latent time and error time s to some extent.Conclusion Cistanchis glycosides could obviou sly improve scopo-lamine -induced impariment of learn ing and memory and on sodium nitrite -induced impariment of learning and memory in mice.
9.Evaluation of the efficacy of cynoff 25 ULV to eradicate aedes species, dengue vectors in Vietnam, 2004

Background: Dengue fever remains a major public health problem in Vietnam. It was transmitted through two main Aedes species, namely Aedes aegypti and Aedes albopictus in which Aedes aegypti played the predominant role in transmitting the dengue viruses (accounted for 94% of the Aedes genus).Different groups of insecticides have been widely used in the community. As a consequence, it increased the resistance level of Aedes vectors to the insecticides. Therefore, a new synthesized insecticide was required for future control and prevention of dengue epidemics. \r\n', u'Objectives: To evaluate the efficacy of Cynoff 25 ULV in suppressing Aedes species in Vietnam. \r\n', u'Subjects and methods: The study was conducted in the entomology laboratory and in the fields of Ha Tay province. ULV spraying was implemented in Hiengiang commune, Thuongtin district, in comparison with a control area, Van Mo commune in Ha Dong town.\r\n', u'Results and conclusions: : Cynoff 25 ULV had a high efficacy that kills over 90% of 2 dengue vector species, namely Aedes aegypti and Aedes albopicctus after 24 hours exposure at the distance of 30 meters from the brass nozzle of a STIHL SR 400 sprayer. In the field trials, Cynoff 25 ULV also had high efficacy in killing Aedes species, e.g. 100% of Aedes species were killed after spraying and the effects lasted for 3 months for Aedes aegypti and one month for Aedes albopictus species. The insecticide had no side-effects or any other adverse effects to humans, livestock and the environment in the intervention areas. \r\n', u'

10.Evaluation of the efficacy of Bistar 80SC by residual application in dengue vectors control in the North of Vietnam

Background: Two species, Aedes aegypti and Aedes albopictus are the main intermediate vectors of transmission of the dengue viruses in Vietnam. Insecticide applications by different methods that may help interrupt the spread of dengue outbreaks. Many different groups of insecticides have been used for dengue vector control. Some recent studies revealed that Aedes vectors obscuring their resistance to insecticides at different levels. Therefore, a new insecticide formula is required for effective dengue vectors control. Objective: To evaluate the efficacy of Bistar 80SC by residual application in suppression of Aedes species in a northern province of Vietnam. Subject and methods: Bistar 80 SC with a component of Bifenthrin 80g/L was evaluated by residual application in suppression of 2 Aedes species in vitro and in an intervention commune of Hien Giang, Thuong Tin district and a control commune of Van Mo, Ha Dong town, Ha Tay province from March, 2004 to June, 2004. Results: WHO Bifenthrin paper test kit was effective at the concentration of 37.5mg/m2 and over with Aedes aegypti vectors ranged from 80% to 100% mortality in the North, In other words, Aedes vector remains susceptible to Bifenthrin in the region. The mortality on wood surface after 60 minutes of exposure to Bistar 80SC at concentrations of 25mg/m2 by using residual application was higher than those on brick walls. Up to 100% Aedes aegypti was killed after 60 minutes in exposure and remains in effect until the end of the third months and one month with Aedes albopictus. No side effects, to the sprayer and humans, livestock and environment caused by Bistar 80SC were reported in the intervention area. Conclusion: Bistar 80SC is suggested as a nominated alternative to effectively control the dengue outbreak by residual application.
