2.Application and development of quantitative nuclear magnetic resonance technology

Quantitative nuclear magnetic resonance (qNMR) technology has significant advantages in quantification due to its simple sample processing and high reproducibility. Two-dimensional qNMR analysis, which can solve the quantification problem of different components in complex systems, has gradually been applied in medicine, food, metabonomics, chemical engineering, and other fields. This paper reviews the analysis methods, influencing factors, experimental optimization, application fields, and other aspects of qNMR to promote its wide and effective application.
3.EFFECTS OF FREE FATTY ACID ON GENE, PROTEIN EXPRESSION AND TYROSINE PHOSPHOLATION OF LEPTIN RECEPTOR IN RAT ISLET CELLS

Objective The effects of free fatty acid on leptin receptor in rat islet cells were investigated. Methods Islet cells of newborn sprague Dawley rats were incubated with palmitate acid (0 25mmol/L) or oleic acid (0 125mmol/L) for 12 hours, 24 hours and 36 hours respectively. Western blotting was used to assess the protein content of leptin receptor in rat islet cells. The RNA expression level of leptin receptor was assayed by RT PCR. Tyrosine phospholation of leptin receptor was determined by method of immuno precipitation. 4.Research progress of role of innate immunity in infection of leptospira

Leptospirosis is an important zoonosis with worldwide distribution. As the first procedure in immunity system, the role and significance of innate immunity in controlling infection at the early stage of the disease have gradually been emphasized. Macrophages can phagocytize and kill leptospira, while the pathogenic leptospira can evade the killing by macrophages. In addition, neutrophils, complement system and cytokines also contribute to the defence of leptospira infection.
5.Development of Protective Mask with Dual Canisters & Big Eyes Window for Liquid Propellant

Objective To develop MFT-2S protective mask with dual canisters and big-eye-window and to improve the three-grade system of protective equipment,so as to solve the problems on protective mask in environment with gas of high concentration.Methods MFT-2S protective mask was derived from MFT-2,with the advantages of MFT-2.The fixation intensity was effectively guaranteed by installing two sets of joint components and stainless steel clamps in the lower part of mask bilaterally.The sealing component was installed in communication component.Each canister was fixed at one side respectively.The fogproof lenses were reinstalled and the length of canister was increased properly.Results The results showed that,compared with MFT-1 mask,the inspiration resistance of MFT-2S mask decreased by a half,its total weight became about 50% of MFT-1 mask,and the leakage coefficient of MFT-2S mask was 0.005 percent.Its antidim character was improved and MFT-2S mask had better protective performance than MFT-2 by twice.Conclusion According to the transformed protection indices,MFT-2S mask,with the advantages of being light,simplified and standardized,can take the place of MFT-1 mask.
6.Study on the Formation Technology and Dissolution in vitro of Ibuprofen Dropping Pills

ABSTACT OBJECTIVE:To study the formation technology of Ibuprofen dropping pills and determine the dissolution in vitro. METHODS:Based on single factor test and with the spherical degree,drug-loading rate and pill weight variation as the evaluated indexes,response surface methodology was employed to screen the ratios of drug to matrix,drug solution temperatures and cryo-genic temperatures in the formation technology,and verification tests were conducted. The dissolution in vitro of the dropping pills prepared by the optimal technology was investigated and compared with that of the preparations sold in the market. RESULTS:The optimal formation technology of Ibuprofen dropping pills was as follows as the ratio of drug to matrix of 1∶6,drug solution temper-ature of 83 ℃ and cryogenic temperature of 7.3 ℃. In the verification tests,the self-made dropping pills demonstrated a spherical degree of 0.945 9,drug-loading rate of 99.82%,pill weight variation of 0.040 28 and drug-loading capacity of 30 mg/pill,with the actual comprehensive score of 0.972 5,deviating from the theoretical one (0.980 0) by 0.771 2%(RSD<1.5%,n=3). The dropping pills prepared had a dissolution rate of 25.36%at 5 min and an accumulated dissolution rate up to 90.12%at 30 min,sim-ilar with the Ibuprofen tablets sold in the market in drug release in vitro (f2=54.91),conforming to a first-order kinetic equation. CONCLUSIONS:The optimized formation technology is simple,stable,feasible and well reproducible,and the dropping pills which are prepared by such technology can release drug quickly.
7.Efficiency of Compound Active Carbon Filter in Reducing Free Radicals in Cigarettes Smoke

Objective To study the efficiency of the compound active carbon filter in reducing the content of free radicals in the smoke of cigarettes. Methods 9 trademarks of cigarettes installed the compound active carbon filters were collected and ESR was used to analyze the content of free radicals in the particle and gas phases. Results The results showed that the efficiency of the compound active carbon filters to reduce the content of free radicals in the cigarette smoke was not identical,some samples even showed an inverse result. Conclusion Based on the results of the present paper,it is not considered that the compound active carbon filter can always reduce the free radicals in the smoke of cigarettes.
8.Relationship Between Free Radical Content and Tar Content in Cigarettes

Objective To study the methods of preparing samples, analyse free radical in smoke of cigarette and the relationship between free radical and the other harmful components in the cigarette smoke. Methods The contents of two types of free radical in smoke of 13 kinds of cigarettes were determined with electron spim resonance(ESR) and the self-made equipment. Results The analytic results showed that the main component was the free radical in gas state in all cigarette samples and it was about 10-30 times higher than the content of free radical in solid state. The content of free radical in gas state of cigarette made in China was lower than that made in foreign country. Conclusion The analysis method using ESR and the self-made equipment will be easily used in the related experiment; The level of free radical in gas and solid state is positively related to the level of tar and nicotine in the cigarette sample of same trademark but it is not the same in the samples of different trademark. Therefore, we can not to infer the content of free radial in cigarette smoke by the content of tar and nicotine in it,especially for the content of free radial in gas state. Using charcoal filter is benefit to reduce the level of free radical in cigarette smoke.

10.Effects of free fatty acid on gene expression and protein tyrosine phosphorylation of leptin-receptor in rats

Objective:To investigate the effect of free fatty acid on leptin receptor in rats.Methods:High plasma free fatty acid rat model was created by intravenous injection of free fatty acid. Western blot was used to assess the protein level of leptin-receptor in rat liver, muscle and pancreas tissues. The RNA expression level of leptin-receptor was examined by RT-PCR .The tyrosine phosphorylation of leptin-receptor was examined by immuno-precipitation. Results:The protein and RNA levels of leptin-receptor in liver,muscle and pancreas tissues of high plasma free fatty acid rats were similar to those of normal group(P>0.05).But the tyrosine phosphorylation of leptin-receptor significantly downregulated in liver and muscle tissues as compared with control group(P<0.05). Conclusions:High plasma free fatty acid may affect the tyrosine phosphorylation of leptin-receptor in rat liver and muscle.
