1.Preparation and Quality Evaluation of Vardenafil Hydrochloride Orally Disintegrating Tablets

Objective To prepare vardenafil hydrochloride orally disintegrating tablets and evaluate their quality. Methods The tablets were prepared by direct power compression method, using crosslinking povidone ( PVPP ) as disintegrants. The preparation method was optimized by response surface test using amount of PVPP, menthol and taste-masking agents as factors with disintegrating time and distance of bitterness as index. The results of taste of orally disintegrating tablets were determined by electronic tongue, comparing to the results of taste tests. At the same time, the properties of the tablets were evaluated using appearance, content uniformity, disintegrating time, et al. as index. Results The optimal formula was as follows:PVPP 13. 26%, menthol 0. 43%, taste-masking agent SGxj 1. 26%. The results on evaluation of electronic tongue were consistent with the results of taste tests. The quality of the prepared tablets was in line with standard. The disintegrating time was (22. 34 ± 0. 34 ) s. Conclusion The preparation technology of orally disintegrating tablets is simple, and controllable in quality.
2.Study on Influential Factors for the Stability of Gardenia Yellow Solution

OBJECTIVE:To investigate the influential factors for the stability of gardenia yellow solution. METHODS:Using pigment loss rate as index,the stability of gardenia yellow solution was investigated within 12 h under different light(strong light, natural light,dark place),temperature(4,25,60,80 ℃),pH(3.0,5.0,7.0,9.0,11.0),oxidant concentration(hydrogen per-oxide solution,0,0.1%,0.2%,0.3%) conditions. The effects of 3 natural antioxidants as tea polyphenol,rosmarinic acid and grape seed extract on the stability of gardenia yellow solution were investigated within 12 h under different light(strong light,natu-ral light,dark place) and temperature (25,60,80 ℃) conditions;the effects of different concentrations of tea polyphenol (0, 0.05%,0.1%,0.2%)on the stability of gardenia yellow solution were also investigated within 12 h. RESULTS:The pigment loss rates were 20%,10% and 10% within 12 h under 3 light conditions;5%,5%,30% and 60% under 4 temperature conditions;12%,6%,6%,6% and 16% under 5 pH conditions;4%,12%,15% and 18% under 4 oxidant concentrations. After adding 3 antioxidants,pigment loss rate decreased by 10% under different light and temperature conditions except for 80 ℃,and the de-crease of tea polyphenol was most significant;among 4 concentrations of tea polyphenol,pigment loss rate was the lowest in 0.1%group. CONCLUSIONS:Gardenia yellow solution can't keep stable under strong light and high temperature;3 antioxidants can im-prove the stability of gardenia yellow solution,especially 0.1%tea polyphenol.
3.Juvenile onset respiratory papillomatosis: risk factors for severity.

OBJECTIVE:
This study was designed to explore the risk factors associated with severity of juvenile onset recurrent respiratory papillomatosis.
METHOD:
A retrospective study was conducted to study determinants of severe forms of juvenile recurrent onset respiratory papillomatosis. The patients were separated into different groups based on the onset age, the first recurrence of age, the first recurrence of period, gender and incision of tracheal respectively. The relationship among the lesion severity score,the involvement of the subregion, operation period and the next operation period were also explored.
RESULT:
It was observed that some children who recurred before 4 years old required more surgery, shorter operation period(the average, longest or shortest operation period) than those elder children, the differences were statistically (P=0. 029, 0. 003, 0. 010, 0. 039, respectively). The severity score of lesion was correlated positively with the involvement of the subregion and negatively with operation period (r=0. 914, -0. 451, respectively). Some children who diagnosed before 4 years old had to endure more severity score and shorter operation period than those older children, the differences were statistically (P= 0. 036, 0. 000, respectively). 8 cases accepted incision of tracheal, they accepted more surgery too. But the differences in the onset age, the first recurrence of age, and the operation period were not statistically.
CONCLUSION
The results showed that the clinical course of juvenile onset recurrent respiratory papillomatosis was closely related to the first recurrence age and period, while the severity of disease was associated to the onset age and the involvement of the subregion.

4.Levels of serum IL-4, IFN-gamma, IL-32 in children with juvenile-onset recurrent respiratory papillomatosis.

OBJECTIVE:
To explore the relationship between serum IL-4, IFN-gamma, IL-10 levels and the aetiology of juvenile-onset recurrent respiratory papillomatosis.
METHOD:
Serum IL-4, IFN-gamma, IL-32 levels of 15 JORRP children were detected by use of enzyme-linked immunosorbent assay (ELISA) and compared with those of healthy control group.
RESULT:
Serum IL-4 levels were significantly higher in the JORRP children (P<0.01): (524.65 +/- 147.77)pg/ml in the JORRP children and (213.27 +/- 87.48) pg/ml in the healthy control group. Serum IFN-gamma levels were significantly lower in the JORRP children (P<0.01): (2.87 +/- 0.84) pg/ml in the JORRP children and (10.63 +/- 5.09) pg/ml in the healthy control group. Serum IL-32 levels were significantly lower in the JORRP children (P< 0.01): (2.47 +/- 1.60) pg/ml in the JORRP children and (9.08 +/- 2.66) pg/ml in the healthy control group.
CONCLUSION
1) While the concentration of Th2 like cytokine IL-4 in children with JORRP was higher than that in control group, the concentration of Th1 like cytokine IFN-gamma in children with JORRP was lower than that in controls, indicating that the polarization of Th1 /Th2 T cell in children with JORRP; 2) The polarization of Th1/Th2 T cell may cause the reduction of the serum IL-32 as a proinflammatory role in host immunity system that could not eradicate HPVs because of lacking enough inflammatory stimulation.

5.Simultaneous Determination of Four Related Substances in Olmesartan Medoxomil Hydrochlorothiazide Tablets by HPLC-Principal Component Self-Control with Correct Factor

OBJECTIVE: To establish the method for simultaneous determination of four known related substances (olmesartan,olmesartan ester dimer ,olmesartan ester alkene ,benzothiadiazine impurity ,called impurity A ,B,C,D for short )in Olmesartan medoxomil hydrochlorothiazide tablets. METHODS :HPLC-principal component self-control with correction factor were adopted. The determination was performed on YMC-Triart C 8 column with mobile phase A consisted of acetonitrile- 0.015 mol/L potassium dihydrogen phosphate solution (pH adjusted to 2.8 with phosphoric acid )(70 ∶ 30,V/V),mobile phase B consisted of acetonitrile-0.015 mol/L potassium dihydrogen phosphate solution (pH adjusted to 2.8 with phosphoric acid )(15 ∶ 85,V/V)at the flow rate of 0.8 mL/min(gradient elution ). The detection wavelength was set at 265 nm,and column temperature was 25 ℃. The temperature of injector was 4 ℃;the injection volume was 10 μL. RESULTS:The correction factors of impurity A ,B,C,D were 1.42,1.17,0.89,0.92,respectively. The linear range of olmesartan medoxomil ,hydrochlorothiazide and impurity A ,B,C,D were 0.252 7-7.580 0,1.152 1-4.562 9,0.244 0-18.299 0,0.244 7-3.670 8,0.265 2-3.978 3 and 0.149 9-4.497 3 μg/mL(r≥ 0.999 7),respectively. The limits of detection were 0.084 2,0.050 7,0.081 3,0.081 6,0.088 4,0.050 0 μg/mL,respectively. The quantitative limits were 0.252 7,0.152 1,0.244 0,0.244 7,0.265 2 and 0.149 9 μg/mL,respectively. The results of intermediate precision ,stability(24 h)and repeatability tests all met the relevant requirements. The average recovery rates were 104.00%-108.04%,102.00%-104.94%,100.99%-106.89%,92.00%-95.18%,102.00%-105.06%,103.90%-107.00%(n=3), respectively. The contents of impurity A ,B and D in 3 batches of Olmesartan medoxomil hydrochlorothiazide tablets were 0.90% -1.00% ,0-0.11% ,0.16% -0.24% ,respectively. The impurity C and other impurities were not detected. There is no significant difference between the results measured by the established method and by the external standard method. CONCLUSIONS:The method has been proved to be highly sensitive and reproducible. It can be used to simultaneously determine four known substances in Olmesartan medoxomil hydrochlorothiazide tablets.
6.Study on Vascular Protection Effect and Mechanism of Timosaponin B Ⅱ Based on Zebrafish Model

OBJECTIVE:To study the protective effect of timosaponin BⅡ(TB-Ⅱ)on blood vessels and explore its possible mechanism. METHODS :Using aquaculture water as blank control ,the effects of 100,200 and 400 μg/mL TB-Ⅱ treatment for 48 h on the situation of subintestinal veins (SIVs)in normal zebrafish embryos 24 h after fertilization (24 hpf)were investigated. PTK787(0.06 μg/mL),a tyrosine kinase inhibitor ,was used to induce the model of zebrafish intestinal vascular injury ;using combing with 0.1% dimethyl sulfoxide but no PTK 787 as blank control ,combing with PTK 787 but no drug as model control ,the effects treatment of 100,200 and 400 μg/mL TB-Ⅱ for 48 h on the SIVs of zebrafish model with vascular injury were investigated. Relative expressions of fam-like tyrosine kinase 1(Flt-1),kinase insert domain containing receptor (Kdr),kinase insert domain containing receptor l (Kdr-l),vascular endothelial growth factor A (VEGF-A),tumor necrosis factor α(TNF-α)and interleukin 6 (IL-6)mRNA were detected by RT-PCR. RESULTS :100 μg/mL TB-Ⅱ could significantly increase the sprouting vessel of normal zebrafish SIVs sprouting vessel number (P<0.05),and 200 μg/mL TB-Ⅱ could significantly increase SIVs number of normal zebrafish (P<0.05). Compared with blank control , SIVs treatment (P<0.01),and the relative expressions of Flt-l , Kdr,Kdr-l,VEGF-A,TNF-α and IL-6 mRNA were alse decreased significantly (P<0.05 or P<0.01). There was no significant difference in SIVs number and the expression of Flt-l ,TNF-α mRNA in zebrafish treated with 100 μg/mL TB-Ⅱ and the expression of TNF-α mRNA in zebrafish treated with 400 μg/mL TB-Ⅱ, but there was statistical significance in other indexes (P<0.05 or P<0.01). CONCLUSIONS :TB-Ⅱ has a certain function of promoting angiogenesis and repairing damaged blood vessels ,and its mechanism is related to the up-regulation of vascular endothelial growth factor receptor and pro-inflammatory cytokine expression.
7.Study on the Mechanism of Bambuterol Hydrochloride in the Improvement of Chronic Obstructive Pulmonary Disease Model Rats

OBJECTIVE:To study the mecha nism of Bambuterol hydrochloride in the improvement of chronic obstructive pulmonary disease (COPD)model rats ,and to find the potential biomarker. METHODS :Totally 30 rats were randomly divided into normal group ,model group and bambuterol hydrochloride group (3.3 mg/kg),with 10 rats in each group ;COPD model was established by lipopolysaccharide (LPS)infusion combined with smoking in model group and bambuterol hydrochloride group. After modeling ,bambuterol hydrochloride group was given relevant medicine intragastrically ,normal group and model group were given constant volume of normal saline intragastrically ,once a day ,for consecutive 45 d. After last medication ,the serum sample and alveolar lavage fluid of rats were collected. The levels of interleukin- 6(IL-6)and tumor necrosis factor-α(TNF-α)in alveolar lavage fluid were detected by ELISA. The serum metabolites were detected by LC-MS and analyzed by metabolomics. Orthogonal partial least squares discriminant analysis (OPLS-DA)was used to screen out the differential metabolites. The potential biomarkers were identified based on the related literature ,and the metabolic pathway enrichment analysis was carried out by MetPA analysis platform. RESULTS :Compared with normal group ,the levels of IL- 6 and TNF-α in alveolar lavage fluid of rats were increased significantly in model group (P<0.05). Compared with model group ,the levels of IL- 6 and TNF-α in alveolar lavage fluid of rats were decreased significantly in bambuterol hydrochloride group (P<0.05). Results of metabolomics and OPLS-DA showed that 21 differential metabolites and 12 potential biomarkers were found (including maleylpyruvate , hydroxypyruvate, tartronate semialdehyde,etc.). Bambuterol hydrochloride can significantly reduce the levels of maleylpyruvate ,methylselenocysteine and 5-deoxy-D-glucuronic acid (P<0.05), while increase the levels of hydroxypyruvate , tartronate semialdehyde and. These biomarkers were mainly @163.com concentrated in pentose phosphoric acid pathway ,glyoxyli acid and tricarboxylic acid metabolism pathway ,followed by 开发。E-mail:pn333@163.com inositol phosphoric acid metabolism pathway ,arginine and tyrosine metabolism pathway ,glycine,serine and threonine metabolism pathway. CONCLUSIONS :The mechanism of bambuterol hydrochloride improving COPD may be associated with the decrease of the levels of TNF-α and IL-6,as well as the pathway of amino acid metabolism ,energy metabolism and lipid metabolism.
