1.Experimental Studies on Ulinastatin in the Treatment of Ulcerative Colitis

Objective To investigate the therapeautic effects of ulinastatin on ulcerative colitis(UC) and it's possible mechanism.Methods Forty adult Wistar rats were randomly divided into four groups;normal group(group A),UC group(group B),SASP(salicylazosulfapyridine) therapeautic therapeutic group(group C),ulinastatin(UTI) therapeautic group(group D). UC models were induced by trinitrophenylmethylnitramine(TNBS), Group A and Group B received 0 9% saline solution instead of ulinastatin and SASP as a control.Serum level of tumor necrotic factor-?(TNF-?),malondialdehyde(MDA),superoxide dismutase(SOD) and pathological change were measured respectively after drugs treatment for two weeks.Results Both UTI and SASP decreased the level of serum TNF-?,MDA ,increased the level of serum SOD ,and relieved mucosa lesion and pathological change in UC rats . UTI was better than SASP in the treatment of UC.Conclusions UTI has apparent therapeautic effects.The mechanism may be associated with inhibition of pro-inflammatory cytokines and clearance of free radicals.
2.Effect of dog-day moxibustion on changes of soluble cellular adhesion molecules in patients with bronchial asthma

BACKGROUND: It is thought in traditional Chinese medicine that dogday moxibustion (DDM) has good clinical effect in treating bronchial asthma. Study of its influence on soluble cellular adhesion molecules in patients with bronchial asthma can help to well understand its mechanism.OBJECTIVE: To observe the effect of dog-day on soluble cellular adhesion molecules in patients with bronchial asthmaDESIGN: Self-control, case-control observationSETTING: College of Acupuncture and Manipulation and Clinic of the Department of Acupuncture and Moxibustion of the First Affiliated Hospital, Guangzhou University of Traditional Chinese Medicine.PARTICIPANTS: Totally 28 patients with bronchial asthma receiving dog-day moxibustion in the Clinic of the Department of Acupuncture and Moxibustion, First Affiliated Hospital of Guangzhou University of Traditional Chinese Medicine from July to August 2004 were set as dog-day moxibustion group, and 10 concurrent health volunteers were set as normal control group.METHODS: According to the principle of "treating winter diseases in summer", the Chinese medicine which consists of such herbs as Baijiezi(Semen Sinapis Albae ), Mahuang(Herba Ephedrae ), Xixin (Herba Asari ),Gans ui (Radix Kansui ), Yanhu (Rhizoma Corydalis ) was stuck on the points such as Feishu, Dingchuan, Fengnen, Bailao, Shenshu. Point selection was at the two sides, and a lumpy salve was stuck on the point with adhesive tape. Three to six hours for adult and one to two hours for children in each sticking duration; and the operation was respectively done on the first (July20), second (July 30) and third (August 9) dog-day.MAIN OUTCOME MEASURES: Short-term effect 3 months later was observed, the levels of soluble cellular adhesion molecules and IgE were assayed before and 10 days after treatment, and comparison with normal control group was performed. The indexes of the patients before treatment were performed Pearson correlation analysis.RESULTS: Twenty-eight patients and 10 healthy persons all entered result analysis. [1] Intercellular adhesion molecule-1 level was significantly higher before treatment in dog-day moxibustion group than that after treatment in the dog-day moxibustion group and that in the normal control group [(281.39±64.58 ), (256.95±44.22), ( 194.30±39.05 ) μg/L,P < 0.01].[2]Vaso-cellular adhesion molecule-1 level was significantly higher before treatment in the dog-day moxibustion group than that after treatment in the dog-day moxibustion group and that in the normal control group [(234.79±89.75), (185.80±84.41), (128.85±51.28)μg/L,P < 0.01]. [3]IgE level was significantly higher before treatment than that after treatment in the dog-day moxibustion group and that in the normal control group [(482.61±123.43), (418.96±134.18), (161.53±43.64)μg/L,P< 0.01]. [4]Pearson correlation analysis result: Intercellular adhasion molecule-1 level is significantly positively correlated with IgE and vascular intercellular adhesion molecule-1 level (r =0.641,0.449 ,P < 0.01,0.05). [5]Effect: Total effective rate was 93% (26/28)in the treatment of bronchial asthma with dogday moxibustion . The type of the symptoms, with blister or not, course of disease and condition of disease had no significant influence on effect (P> 0.05).CONCLUSION: Dog-day moxibustion may effect through inhibiting the expression of adhesion molecule or inhibiting leukocyte to adhere endothelial cell directly so as to reduce the infiltration of inflammatory cells.
3.Relationship Between Syndrome Patterns of Traditional Chinese Medicine and Apoptosis- Related Oncogene mRNA Expressions in Precancerous Lesions of Gastric Cancer

[Objective] To study the relationship between syndrome patterns of traditional Chinese medicine and bcl- 2 oncogene, p53 suppression gene mRNA expressions in precancerous lesions of gastric cancer (PIGC). [Methods] Forty PLGC cases were endoscopically and pathologically confinned, including 24 cases of moderate dysplasia of gastric mucosa, 9 cases of severe dysplasia, 7 cases of incomplete colonic intestinal metaplasia. Of the 40 cases, 10 were complicated with Qi stagnation, 12 with stomach-heat, and 18 with blood stasis, mRNA expression of bcl- 2 oncogene and p53 suppression gene were detected in situ by molecular hybridization method. [Results] The mRNA overexpression of bcl- 2 oncogene and p53 suppression gene were found in PLGC, and the expression was gradually increased with the progress of lesions. In the complicated cases, the mRNA expression of bcl- 2 oncogene and p53 suppression gene were the least in the cases with Qi stagnation and less in the cases with stomach- heat than in the cases with blood stasis. [Conclusion] Abnormal mRNA expression of bcl- 2 oncogene and p53 suppression gene were found in PLGC cases and related with different complicated cases, indicating the specificity of different syndrome patterns.
4.Analysis of the Influencing Factors of Post-stroke Depression: Based on Machine Learning

Objective To determine the influencing factors of post-stroke depression by machine learning.Methods Stroke patients' medical records (688 cases eligible) were extracted from record system,including age,gender,pulse manifestation,complexion,tongue quality,fur,Chinese medicine intervention,body mass index (BMI),blood pressure,blood glucose,blood triglyceride,blood total cholesterol,smoking history,drinking history,depression family history,stroke lesion site in imaging,as well as the final depression judgment.Single rule algorithm (1R) was adopted to learn.The risk factors influencing post-stroke patients' depression in extracted information were determined.Then the cases collected were divided into the training dataset (500 cases) and the test dataset (188cases).Optimal discriminant results were obtained by random forest model.Results Single rule algorithm showed that the most important influencing factor of post-stroke depression was stroke lesion site.By computer speculation,stroke lesions in the frontal and temporal lobes were most prone to post-stroke depression.Basal ganglia,brain stem,cerebellum,medulla oblongata and occipital lobe lesions were less likely to cause depression.The accurate classification rate could amount to 88.95% (612/688 cases).Random forest model determination was made in the former 500cases in the training dataset.The total correct rate of determining depression was 98.2%.The total correct rate of determination in 188 cases of the test dataset was 99.47%.Six hundred and eighty-eight patients' data were learnt by random forest model.The total correct rate was 98.84%.The importance measure results showed that top 3 important indexes of post-stroke depression were lesion site,Chinese medicine intervention and depression family history.Conclusion Patients with lesions in the frontal & temporal lobes and depression family history were most prone to post-stroke depression.

6.Combined open prostatectomy and preperitoneal inguinal herniorrhaphy: a 21-case report.

OBJECTIVETo evaluate the effect of simultaneous open prostatectomy and preperitoneal inguinal herniorrhaphy.
METHODSTwenty-one patients with benign prostatic hyperplasia(BPH) and inguinal hernia, underwent simultaneous suprapubic or retropubic prostatectomy and preperitoneal inguinal herniorrhaphy through Pfannenstiel's incision. In 4 cases with bigger inguinal mass, hernia repair was performed with polypropylene meshwork (PPM).
RESULTSThe 21 patients were followed up for 6 months to 7 years. There were 2 cases of recurrence (9.5%), with no morbidity and mortality in the other 19 cases.
CONCLUSIONSOpen prostatectomy and preperitoneal inguinal herniorrhaphy can be performed during the same session without increasing infection and recurrence. The operation method is simple and the effect is positive, especially for older men.

7.Rapid health technology assessment of dienogest in the treatment of endometriosis

AIM:To rapidly evaluate the effective-ness,safety and economy of dienogest in the treat-ment of endometriosis so as to provide evidence-based reference for clinical drug selection and deci-sion.METHODS:Retrieved from PubMed,Embase,Cochrane Library,CNKI,SinoMed,Wanfang and health technology assessment(HTA)organization websites,HTA report,systematic review/Meta-anal-ysis and pharmacoeconomic study were included during the inception to Sep 2023.Data extraction and quality evaluation were carried out for the lit-erature that met the inclusion and exclusion crite-ria,and the research results were summarized and analyzed qualitatively.RESULTS:Nine systematic re-view/Meta-analyses,and 2 pharmacoeconomic studies were included.The quality of Meta-analysis literatures was low,and the quality of economic re-search was good.In terms of effectiveness,com-pared with no treatment,dienogest can significant-ly reduce postoperative recurrence rate,VAS score,and improve pregnancy rate and effective rate;Di-enogest has similar therapeutic effects in terms of recurrence rate as GnRHa and COC;With regard to recurrence rate and pregnancy rate,dienogest is superior to danazol,gestrinone,and mifepristone.In terms of safety,compared with placebo,the inci-dence of vaginal bleeding and headache was signifi-cantly increased with dienogest,and there was no difference in bone loss;The incidence of vaginal bleeding caused by dienogest was significantly high-er than that of GnRHa,but the incidence of hot flashes and bone loss was lower.In terms of econo-my,dienogest has a more cost-effective advantage comparing with GnRHa,but does not have econom-ic benefits comparing with COC.CONCLUSION:Di-enogest has good effectiveness and safety in the treatment of endometriosis,and economically su-perior to GnRHa,but inferior to COC.
8.Drug treatment options and research progress of adolescent endo-metriosis

Endometriosis is a chronic,relapsing disease that requires long-term management.Ado-lescent endometriosis is a group that can be ig-nored,and the drug treatment is an important method for long-term management and fertility preservation.There are many kinds of drugs to treat endometriosis,such as NSAIDs,combined oral contraceptives(COC),progesterone,GnRHa.For the drug treatment selection,the characteris-tics of adolescents should be carefully considered and individualized treatment should be provided.In order to better long-term management,it is nec-essary to develop new treatments,which can effec-tively relieve endometriosis without damaging fer-tility.This review focuses on the drug treatment op-tions and research progress of basic drug treat-ment for adolescent endometriosis,so as to delay disease progression,reduce adverse reactions,pre-serve fertility,and improve quality of life.
9.Apoptosis inducing effect of ponicidin in leukemia K562 cells and its mechanisms of action.

OBJECTIVETo investigate the apoptosis inducing effects of ponicidin (PON) on leukemic K562 cells and its mechanisms of action.
METHODK562 cells in culture medium in vitro were given different concentrations of PON (10-50 micromol x L(-1)) for 24, 48 and 72 h. The inhibitory rate of the cells was measured by MTT assay, cell apoptotic rates were detected by flow cytometry (FCM) using Annexin V staining after K562 cells were treated with different concentrations of PON for 72 hours, and cell morphology was observed by Wright-Giemsa staining. Western blot was used to detect caspase-3 and poly(ADP-ribose) polymerase (PARP) expression, and the protein levels in mitogen-activated protein kinase signaling pathways (MAPKs, p-P38, p-ERK and p-JNK) as well as p-AKT and p-P85 in PI3K/AKT signaling pathways were also detected.
RESULTPON (over 30 micromol x L(-1)) could inhibit the growth of K562 cells in both time- and dose-dependent manner. FCM analysis revealed that apoptotic cells were gradually increased in a dose-dependent manner after treatment for 72 hours, and that marked morphological changes of cell apoptosis such as condensation of chromatin was clearly observed by Wright-Giemsa staining after treatment by 50 micromol x L(-1) PON. Western blot showed cleavage of the caspase-3 zymogen protein (32 kD), with the appearance of its 17 kD subunit, and a cleaved 89 kD fragment of 116 kD PARP was also found. Furthermore, Western blotting also showed that expression of p-AKT and p-P85 in PI3K/AKT signaling pathways was downregulated dramatically whereas the expression of p-P38 as well as p-ERK and p-JNK remained unchanged after the cells were treated by PON for 48 h.
CONCLUSIONThe results demonstrate that PON exhibits in vitro anti-leukemia effect by induction of apoptosis in K562 cells, and that PON induced apoptosis in K562 cells mainly related to activation of caspase-3 as well as inactivation of PI3K/AKT signaling pathway via down regulation of the expression of p-AKT and p-P85 protein levels. These results provide strong laboratory evidence for further anti-leukemia trials of PON.
