1.Antitumor Effects of Lentinan Combined with Cisplatin on the Apoptosis Induction in mice bearing Lewis Lung Carcinomas

Objective To observe the effects of Lentinan(LNT) combined with Cisplatin (DDP) on the apoptosis induction in mice bearing Lewis lung carcinomas (LLC). Methods Lewis lung carcinoma cells (1?107) were subcutaneously inoculated into BALB/c mice. Mice were randomized into control groups and treatment groups. Observe the morphologic views under light microscope (LM) and electron microscope (EM). Results Compared with the control groups, the treatment groups were morphologic characteristic by relatively smaller in size, cell atypia were not evident, pathalogical karyokinesis were less interstitial blood vessel were fewer, while lymphocyte infiltrate into the interstitial. Apoptotic bodies could be seen under EM. Within all treatment groups, LNT+DDP group showed significant difference. Conclusions Lentinan combined with Cisplatin showed significant antitumor effects, and mechanism maybe inducing the tumor cell apoptosis and stimulate the proliferation of lymphocyte.
2.Comparison between enteral nutrition and parenteral nutrition support in aged ventilated patients

Objectives: To observe the effect of total enteral nutrition (TEN)and total parenteral nutrition (TPN)in aged patients with mechanical ventil ation. Methods: 60 cases of aged patients with mechanical ventilation were randomly divided to TEN group and TPN group.All patients accepted the same amount of calorie and nitrogen. Results: with regard to albumin and hemoglobin levels and nitrogen equilibrium TEN group was superior to parenteral nutrition group ( P 0.05). Conclusions: Enteral nutrition can reduce mechanical ventilation time and lower treatment cost in aged patients.
3.Clinical study of lactobacillus acidophilus combined with glutathione in the treatment of nonalcoholic fatty liver disease

Objective To investigate the clinical effects of lactobacillus acidophilus combined with glutathione in the treatment of nonalcoholic fatty liver disease.Methods 120 patients with nonalcoholic fatty liver disease were selected,and they were randomly divided into control group (60 patients used glutathione alone) and observation group (60 patients received lactobacillus acidophilus on the basis of control group).The clinical efficacy,liver function index,blood lipid index and the ultrasonography grading of fatty liver before and after treatment of the two groups were compared.Results The clinical total effective rates of the control group and the observation group were 76.67%,91.67%,respectively.The total effective rate of the observation group was significantly higher than that of the control group(x2 =10.52,P < 0.05).The levels of AST,ALT and GGT of the observation group after treatment were significantly lower than those of the control group and before treatment [(36.89 ± 7.14) U/L,(36.89 ± 7.14) U/L,(36.89 ±7.14) U/L vs.(49.36 ± 11.08) U/L,(45.91 ± 10.24) U/L,(90.28 ± 20.70) U/L;(36.89 ± 7.14) U/L,(36.89 ± 7.14) U/L,(36.89 ± 7.14) U/L vs.(92.90 ± 24.3) U/L,(81.11 ± 17.37) U/L,(147.27 ± 34.19) U/L,t =2.88,2.54,2.91;3.01,3.36;3.18,3.48;3.41,3.87;all P <0.05).The levels of TG,TC,HDL-C and LDL-C of the observation group after treatment were significantly better than those of the control group and before treatment [(1.66 ± 0.42) mmol/L,(4.05 ± 0.77) mmol/L,(1.69 ± 0.60) mmol/L,(2.44 ± 0.38) mmol/L vs.(2.13 ± 0.80) mmol/L,(4.64 ± 0.94) mmol/L,(1.45 ± 0.48) mmol/L,(2.97 ± 0.57) mmol/L;(1.66 ± 0.42) mmol/L,(4.05 ± 0.77) mmol/L,(1.69 ± 0.60) mmol/L,(2.44 ± 0.38) mmol/L vs.(2.71 ± 1.33) mmol/L,(5.42 ± 1.27) mmol/L,(1.08 ± 0.36) mmol/L,(3.49 ± 0.71) mmoL/L,t =2.43.2.13,2.55,2.07;3.02,3.41;3.11,3.65;2.81,3.30;2.87,3.15;all P < 0.05).In the control group,normal in 6 cases,mild in 24 cases,moderate in 21 cases and severe in 9 cases as fatty liver degree;in the observation group,normal in 13 cases,mild in 30 cases,moderate in 13 cases and severe in 4 cases as fatty liver degree.The ultrasonography grading of fatty liver of the observation group after treatment were significantly better than those of control group and before treatment (U =3.74,5.20,all P < 0.05).Conclusion Lactobacillus acidophilus combined with glutathione in the treatment of nonalcoholic fatty liver disease can efficiently promote the recovery process of liver function,regulate blood lipid levels and is helpful to improve the imaging grading.
4.Effects of Cognitive Behavioral Therapy and Citalopram on Regional Cerebral Blood Flow in Patients with Depression Disorder

Objective To explore the effects of cognitive behavioral therapy and citalopram on regional cerebral blood flow(rCBF)in the patients with depression disorder.Methods 38 patients with depression disorder were divided into the cognitive behavioral therapy group(n=18),and citalopram group(n=20).The rCBF changes and scores of Hanmilton Depression Scale(HAMD)of all patients in two groups were evaluated with 99Tcm-ECD single photon emission computed tomography(SPECT)and HAMD before and after treatment.Results After treatment,the symptoms of all patients in two groups improved,but the cases of the citalopram group got a rapid improvement compared with those of the cognitive behavioral therapy group.Both of two methods could increase blood flow in right basal ganglion;while,the blood flow also increased in right limbic system in the cognitive behavioral therapy group,and in right temporol lobule in the citalopram group.Conclusion Both cognitive behavioral therapy and citalopram are effect on symptoms of depression disorder,but affect blood flow in different parts.
5.ELK-3 interference inhibits the epithelial-mesenchymal transition of human hepatocellular carcinoma cells

Objective To investigate the relationship of ELK-3 and epithelial-mesenchymal transition ( EMT) for ex-ploring its possible mechanism .Methods The human hepatocellular carcinoma cells ( HCC) were divided into small interference RNA transfection group and Ras-ELK-3 pathway inhibitor group .The protein level of ELK-3 target gene EGR-1 E-cadherin ,vimentin and p38 in HCC were determined by Western blot analysis .Results The protein level of ELK-3 and its target gene EGR-1 in treated human hepatocellular carcinoma cells significantly decreased as compared with the negative control group (P<0.01).The protein level of E-cadherin was significantly increased (P<0.01), while vimentin and p38 were decreased in HCC cells with ELK-3 interference (P<0.01).Conclusions ELK-3 in-terference can inhibit the epithelial-mesenchymal transition of HCC cells by down-regulating p38.

ObjectiveTo investigate whether CD4+ CD25+ CD127dim/- regulatory T lymphocytes (Treg) can induce the proliferation of hepatic stellate cells (HSC) and expression of fibrosis-related factors on HSC in vitro and further to explore the mechanism of Treg inducing fibrogenesis. MethodsHSC LX-2 cells were subcultured.CD4+ CD25+ CD127dim/- cells were purified using magnetic cell separation. The HSC were co-cultured with Treg by direct contact or by Transwell system in vitro. The HSC cultured alone was used as control. Cell proliferation was measured by CCK-8 assay.The expression of transforming growth factor (TGF)-β1 was detected by enzyme-linked inmunosorbent assay (ELISA), and the expressions of hyaluronic acid (HA) and precollagen Ⅲ (PC Ⅲ ) were detected by radio immunoassay (RIA). The data were analyzed by LSD-t test. ResultsHSC proliferation was strongest when Treg∶ HSC= 1.5∶ 1. The absorbance in direct contact co-culture group and Transwell system co-culture group was (0. 713±0. 032) cpm and (0. 735±0. 028) cpm, respectively, both of which were higher than that in control group [(0. 677 ± 0. 029) cpm](t = 5. 4003 and 8. 7878,respectively; both P<0. 01). The concentrations of TGF-β1 in the supernatant were (781. 59 ±76.45) pg/mL and (813. 53±60. 62) pg/mL, respectively in direct contact co-culture group and Transwell system co-culture group, which were significantly higher than that in control group [(722.51±59. 66) pg/mL](t = 4.0014 and 6. 1653, respectively; both P<0.01).The concentrations of HA were (433. 575±27.90) ng/mL and (445.40±23.73) ng/mL, respectively in direct contact co-culture group and Transwell system co-culture group, which were higher compared to that in control group [-(415. 83±19.44) ng/mL](t =3. 3124 and 5. 5231, respectively; both P<0.01). Likewise, the concentrations of PCⅢ were (21. 93± 1.71) and (23. 125± 1.87) ng/mL in direct contact group and Transwell group, respectively compared to (20. 10± 1.49) ng/mL in control group (t = 4. 8082 and 7. 9436, respectively; both P < 0.01).Furthermore, the absorbance,concentrations of TGF-β1, HA and PC Ⅲ in Transwell co-culture group were all higher compared to direct contact group (t = 3. 3875, 2.1639, 2. 2107 and 3.1354, respectively; all P<0. 05).ConclusionsThe cell proliferation and the expressions of fibrosis-related factors in HSC increase greatly after co-cultured with CD4+ CD25+ CD127dim/-Treg. Therefore, Treg may play an important role in inducing liver fibrogenesis.
7.Expression of miR-486-3p in psoriatic lesions and its effect on keratin 17 expression

Objective To determine the expression of miR-486-3p in psoriatic lesions and healthy human skin and to estimate its effect on keratin 17 (K17) expression in HaCaT human keratinocytes.Methods Bioinformatics was used to predict microRNAs that may affect the expression of K17.Tissue samples were obtained from the lesions of 10 patients with psoriasis and normal skin of 10 healthy human controls.RNA was extracted from these tissue samples and reversely transcribed into cDNA with the addition of a Poly (A) tail.Then,real time quantitative PCR was performed to measure the expression of miR-486-3p.Cultured keratinocytes were transfected with miR-486-3p mimics or negative control,and Western blot was performed to determine K17 expression at 48 hours after the transfection.To evaluate the inhibitory effect of miR-486-3p on K17 expression,cultured 293T cells were transfected with the plasmid containing K17 3' untranslated region (UTR) seed sequence,three plasmids containing the complete deletion,interval mutation or double repeats of the seed sequence,or negative control plasmid.At 24 hours after the transfection,a dualluciferase reporter (DLR) assay was performed to quantify the expression of K17.Results Real time PCR showed that the expression level of miR-486-3p was significantly lower in psoriatic lesions than in the normal skin (0.211 ± 0.120vs.0.555 ± 0.425,t =2.62,v =9,P < 0.05).The HaCaT cells transfected with the mimics of miR-486-3p exhibited decreased expression of K17 compared with those transfected with the negative control.DLR assay revealed that the expression level (fluorescence intensity) of K17 in the negative control group was significantly higher than that in the 293T cells transfected with the seed sequence and those with the double repeats of the seed sequence (100.00% vs.65.31% ± 6.32% and 54.18% ± 10.01% respectively,both P < 0.05),but did not differ from that in the 293T cells transfected with the complete deletion and interval mutation of the seed sequence (100.00% vs.114.77% ± 16.14% and 110.21% ± 12.99% respectively,both P > 0.05).Conclusions The expression of miR-486-3p,which may inhibit K17 expression by binding to the seed sequence of K17 3'UTR,is lower in psoriatic lesions than in normal skin.The decreased expression and inhibitory effect of miR-486-3p may be implicated in the initiation and progression of psoriasis.
8.Efficacy of alendronate sodium combined with dietary and exercise intervention in postmenopausal osteoporosis

Objective To evaluate the efficacy of alendronate sodium combined with dietary and exercise intervention in postmenopausal osteoporosis. Methods 88 postmenopausal osteoporosis patients were enrolled and treated with alendronate sodium and Vitamin D combined with dietary and exercise intervention for 12 months. Bone mineral density (BMD) of lumber spine and femur neck were measured before and after the treatment. Results Medication combined with dietary and exercise intervention significantly increased bone mineral density of postmenopausal osteoporosis patients, and the serum levels of bone alkaline phosphatase (BLAP), alkaline phosphatase (AKP) and tartrate resistant acid phosphatase (TRACP) were significantly higher than those pre-treatment, with significant differences (P<0.05). Conclusion Compared to simple medication treatment, medication combined with dietary and exercise intervention is more effective to enhance medication efficacy and BMD level.
9.Effect of Scalp Acupuncture on Cognitive and Motor Functions of Stroke Patients in Recovery Stage

Objective To observe the effect of scalp acupuncture on cognitive and motor functions of stroke patients in recovery stage.Methods80 stroke patients with cognitive disorder were randomly divided into the scalp acupuncture group (n=41) and control group (n=39). All patients of two groups were treated with routine therapy and rehabilitation training, but the cases of the scalp acupuncture group were added with scalp acupuncture. The cognitive function, motor function and activities of daily living (ADL) of all patients were assessed by delitescence and amplitude of event-related potential (ERP) P300, Fugl-Meyer Assessment (FMA), Barthel Index (BI) respectively before and 3 months after treatment.ResultsAfter treatment, P300 delitescence of the patients in the scalp acupuncture group shorten to 38 ms in average while in the control group shorten only to 17 ms, the therapeutic effect of the scalp acupuncture group was superior to that of the control group ( P<0.05). The scores of FMA and BI of the patients in the scalp acupuncture group were also better than those in the control group ( P<0.05).ConclusionScalp acupuncture can efficiently improve the cognitive function of stroke patients in the recovery stage, and promote the recovery of motor function and ADL.
10.Efficacy of edaravone dexborneol combined with alteplase in treatment of acute ischemic stroke

Objective To investigate the efficacy and safety of edaravone dexborneol combined with alteplase in the treatment of acute ischemic stroke (AIS). Methods The data were collected from 124 patients with AIS who were admitted to our hospital from November 2020 to April 2022. The patients were randomly divided into experimental group (intravenous thrombolysis with alteplase + treatment with edaravone dexborneol) and control group (intravenous thrombolysis with alteplase), and the two groups were compared for efficacy. Results The overall response rate in the experimental group was significantly higher than that in the control group (82.3% vs 64.5%, P < 0.05). The National Institutes of Health Stroke Scale scores at different stages after thrombolysis were significantly lower in the experimental group (5.40 ± 3.82, 4.14 ± 3.44, and 0.57 ± 0.99) than in the control group (P < 0.05). No adverse drug reactions were observed in the two groups during the treatment. Conclusion Edaravone dexborneol combined with alteplase has definite clinical efficacy in the treatment of AIS.
