1.The expression of p53, bcl-2, and growth factors and cell cycle arrest of retinal vascular endothelial cells of diabetic rats

Objective To observe the expression of p53, bcl 2 genes, vascular endothelial cell growth factor (VEGF), basic fibroblast growth factor (bFGF), insulin like growth factor I (IGF I), and the receptors of these factors of retinal vascular endothelial cells (VECs) of 1 to 20 week diabetic rats, and the relationship between the expressions and cell cycle arrest. Methods Retinal sections of diabetic rats induced by alloxan were immunohistochemically stained and observed by light microscopy (LM) and electron microscopy (EM). Dot blotting and Western blotting were used to determine the expression of mRNA, proteins of p53 and bcl 2. Results Under LM, immunohistochemical positive expression of p53 and bcl 2 were found on the vessels of ganglion cell layer and inner nuclear layer of retinae of 8 to 20 week diabetic rats; under EM, these substances were observed depositing in VECs. The retinal VECs also expressed VEGF, bFGF, IGF I and their receptors. There was no positive expression of other cell types in these retinae, all cell types of retinae in control group, or all cells of retinae of diabetic rats with the course of disease of 1 to 6 weeks. The result of dot blotting revealed that retinal tissue of 20 week diabetic rat expressed p53 and bcl 2 mRNA, and the result of Western blotting revealed that they also expressed p53 and bcl 2 proteins. But retinal tissues of control group did not. Positive expression of bax was not found in the retinae in control group or 1 to 20 week diabetic rats. Conclusion p53, bcl 2 may introduce cell cycle arrest of VECs of retinae in 8 to 20 week diabetic rats. High glucose might stimulate the expression of VEGF, bFGF, IGF I and their receptors, and the growth factors may keep VECs surviving by self secretion.
2.Influence of Acupuncture on Insulin Resistance in Simple Obesity Patients

To investigate the regulatory effect of acupuncture on insulin resistance in simple obesity patients and its clinical effect. Methods: By the method to benefit qi, strengthen the spleen, dissolve phlegm and remove turbidity, eight periumbilical acupoints, Tianshu (ST 25), Huaroumen (ST 24), Wailing (ST 26), Yinjiao (CV 7) and Shuifen (CV 9), and Guanyuan (CV 4) and Zusanli (ST 36) were used, in cooperation with ear-points-embedding method, and together with 30 healthy persons for comparison, in order to observe the changes in FBG, INS,ISI, TC, TG, LDLC and HDLC and to observe the changes in the waist circumference, hip circumference, waist-hip ratio, body weight and BMI in simple obesity patients before and after acupuncture treatments, and to observe the differences of the therapeutic effects between grade Ⅰ and grade Ⅱ obesity. Results: Hyperinsulinemia (HI) existed in the patients with simple obesity before the treatment, and ISI significantly decreased (P<0.01) in comparison with the healthy group, and waist circumference, hip circumference, waist-hip ratio, body weight and BMI significantly decreased (P<0.05, P<0.01), and FBG, INS, TC, TG and LDLC also significantly decreased (P<0.01, P<0.05), and ISI and HDLC significantly increased (P<0.05) after the treatment. Conclusion: Acupuncturecan not only effectively improve the body fat parameters but also regulate the abnormal fat metabolism and improve insulin resistance status in simple obesity patients.
3.The clinical features of 15 cases with severe enterovirus 71 ( EV71 ) infection and the genotype of EV71

Objective To explore the clinical feature of severe enterovirus 71 (EV71) associated hand foot and mouth disease (HFMD),and genotype of EV71.Methods Fluorescent quantitation PCR was done for detecting EV71.RT-PCR was performed to amplify VP1 for sequencing and identifying genotype.A retrospective analysis was performed based on the clinical data of 15 cases with severe EV71 infection.Results EV71 nucleotide was positive in all 15 cases.The genotype of EV71 was C4.All cases had abnormal temperature and followed with nervous symptoms in the early stage.Average time was 1.26 days from onset to severe complications appearance.Eleven cases progressed to neurogenic pulmonary edema.Four cases accepted nasal continuous positive airway pressure.Eleven cases accepted oral trachea cannula mechanical ventilation.Except for 3 cases died,one case abandoned,others 11 cases were cured.Conclusion The isolated strains of EV71 in this study are all C4 genotype.All cases with severe EV71 infection were followed with nervous symptoms in the early stage,most of whom would progress to neurogenic pulmonary edema.The mortality would be cut down by using mechanical ventilation in early stage.
4.Mechanism of Banxia Houpo Decoction in the treatment of tension-type headache based on network pharmacology and molecular docking

Objective:To explore the mechanism of the active compounds in Banxia Houpo Decoction treating tension type headache through network pharmacology and molecular docking.Methods:The potentially effective components and targets of Banxia Houpo Decoction were screened by TCMSP, and the action targets of tension type headache were obtained by GeneCards, PharmGKB, TTD, Drugbank and OMIM. The intersection target of "Banxia Houpo Decoction - tension type headache" was obtained by Perl software. The protein interaction network was uploaded to STRING database and topological analysis was carried out. With the help of Cytoscape 3.8.0 software, the visualization network of "Banxia Houpo Decoction - medicine ingredient-Target-tension type headache" was constructed, and the GO enrichment analysis and KEGG pathway enrichment analysis of the intersection targets were carried out by using R 4.1.0 language and related programs. The AutoDockTools-1.5.6 software was used to complete the molecular docking analysis.Results:There were 33 intersection targets in Banxia Houpo Decoction and tension type headache. Topological attribute analysis suggested that MAPK1, TP53, ESR1, PTGS2, MYC, CYP1A2, CYP3A4 and GSTP1 might be important potential targets of Banxia Houpo Decoction in the prevention and treatment of tension type headache. GO enrichment analysis showed 516 cell biological processes (BP), 62 cell components (CC) and 149 molecular functions (MF). KEGG pathway enrichment analysis showed that there were 94 related signal pathways, such as cGMP-PKG signaling pathway, Cholinergicsynapse, Serotonergic synapse and TNF signaling pathway.Conclusions:Banxia Houpo Decoction has multi-component, multi-target and multi-pathway characteristics in the prevention and treatment of tension-type headache. It mainly acts on 5-HT synaptic pathway, TNF signal pathway, cholinergic synaptic pathway, G protein coupled receptor pathway and other pathways through ESR1, TP53, PGTS2 and other multi target.
5.Effect of human papillomavirus 16 E6 gene on the expression of E-cadherin in human cervical cancer cell line SiHa

Objective To investigate the effect of human papillomavirus (HPV) 16 E6 gene on the expression of E-cadherin (E-cad) in cervical cancer cell line SiHa. Methods HPV16 E6 expression system was established in the cervical cancer cell line SiHa by using transient transfection system, MTT method was used to detect SiHa cell proliferation activity, and reverse transcription-polymerase chain reaction (RT-PCR) and western blot method was used respectively to detect E-cad mRNA and protein expression level in cells after HPV16 E6 transfection. Results Compared with the blank control group (non-transfected plasmid) and the vector group (the addition of pcDNA3.1), the cell viability rate of the E6 group (pcDNA3-1-HPV16 E6) was significantly increased (P<0 .05), while there was no significant difference between the vector group and the blank control group (P>0.05). The relative expressions of E-cad mRNA in the E6 group, the vector group and the blank control group were 0.26±0.12, 0.82±0.14, 0.83±0.21 respectively, then the protein relative expressions in the three groups were 0.62±0.02, 1.33±0.04, 1.31±0.05 respectively. The expressions level of E-cad mRNA and protein in E6 group were significantly lower than those in the other two groups (all P<0.05). However, there was no significant difference between the vector group and the blank control group (both P>0.05). Conclusion The instantaneous transfection of HPV16 E6 gene can reduce the expression of E-cad in cervical cancer cells, which is closely related to the occurrence and development of cervical cancer.
