To cure one patient with spinal cord injury (SCI), who hospitalized for both legs couldn't moving for 21 years after external injury, by using the olfactory ensheathing cells (OECs) transplant and acupuncture treatment. The patient was diagnosed as ischemic SCI (below the T11) according to the neurological test and MRI for dorsal vertebra. T11 spinous process was made to be the center mark through operation with the length of about 3 cm. The area between the atrophique spinal cord and normal spinal cord without vessel was selected and injected with OECs 0.5?109 L-1. Hemostasis, stitching, binging and fixation were performed respectively, and normal treatment was given by using antibiotics. At 14 days after operation, the temperature on skin increased a little, and acupuncture treatment and rehabilitation therapy were performed. The acupoints were as following: Baihui (DU21), Yintang (EX-HN3), Hegu (LI4), Zhongji (RN3), Guanyuan (RN4), Biguan (ST31), Fengshi (GB31), Futu (ST32), Dubi (EX-LE5), Zusanli (ST36), Xuehai (SP10), Yinlingquan (SP9), Sanyinjiao (SP6), Jiexi (ST41), Taixi (KI3), Fuliu (KI7), Taichong (LR3), and Shenmai (BL62). The treatment was given by mild reinforcing and attenuating with 30-minute retention. The rehabilitation: The needle pressing, manipulation for muscular atrophy, bicycle practicing, andimperative standing were performed. After 2 weeks, the skin temperature of the patient increased with obvious improvement in his right leg. The physical strength of the patient was reinforced with less perspiration, and the patient could walk 3 km without fatigue, which indicated that after OECs transplant, treatment with acupuncture, massage and other rehabilitation trainings could facilitated the improvement of SCI of the advance stage as well as the amelioration of muscular atrophy.

OBJECTIVE To study the optimal reagent for regeneration of aptamer-coated piezoelectric quartz crystal chips and detect storage ability of aptamer-coated chips. METHODS Anti-IgE aptamers were immobilized on the gold surface of piezoelectric quartz crystal biosensor with biotin-avidin method. The reaction between anti-IgE aptamers and IgE was monitored in time on piezoelectric sensor. We tried to regenerate the sensor surface after binding IgE by rinsing the aptamer-coated chips with HCl, NaOH, EDTA, urea and formamide individually. Aptamer-coated chips were stored in PBS with 0.1% sodium azide and the stored chips were used to detect IgE in 35 days. RESULTS Of the five reagents, EDTA was the best one for regeneration of aptamer-coated chips and the sensor could retain 91.5% of the original detecting signals after five regeneration cycles. Moreover, the aptamer-coated chips could be stored in the binding buffer for 21 days without obvious loss of activity. CONCLUSIONS Compared with antibody-based piezoelectric sensor, aptamer-based sensor has lower cost, more regeneration cycles and longer time for the storage of chips. This series of experiments shows the superiority of aptamer detection.

OBJECTIVE To establish an aptamer-based piezoelectric sensor assay to detect human immunoglobulin E (IgE) and acquire the parameters about this kind of biosensor. METHODS Anti-IgE aptamers were immobilized on the gold surface of piezoelectric quartz crystal sensor with biotin-avidin method. The reaction between anti-IgE aptamers and IgE was monitored in time on piezoelectric sensor. Different concentrations of IgE were detected and the calibration curve of IgE was drawn. Non-specific response signals of BSA, IgG and IgE were monitored to show the detecting specificity of aptamer-based sensor. RESULTS For the aptamer-based sensor, the lowest limit of detection was 0.055mg/L and the linear range was 0.1-2.5mg/L. Non-specific signals of this sensor were less than 5% of specific signals of IgE at the same concentration. CONCLUSIONS The aptamer-based piezoelectric sensor can detect 5.5 ng IgE while the non-specific signals are very low. So this aptamer-based sensor is hopeful to be applied to clinical laboratory diagnosis.

Objective To investigate the causes of a suspected outbreak of lower respiratory tract infection (LR-TI)caused by carbapenem-resistant Acinetobacter baumannii (CRAB)in the general intensive care unit(ICU)of a hospital, and provide scientific evidence for effective control of healthcare-associated infection(HAI).Methods Epi-demiological investigation on patients infected with CRAB and on-site monitoring on ICU environmental hygiene from March 4 to 23,2014 were performed, active prevention and control measures were taken. Results A total of 7 patients developed CRAB infection, the total length of stay during epidemic period were 160 days, the infection density of LRTI was 43.75‰(7/160),4 of 5 patients at No.02 bed had CRAB LRTI.Univariate analysis revealed that adopting conventional sputum suction procedure was a risk factor;the qualified rate of bacterial count on the object surfaces was 31 .75% (20/63).CRAB was also isolated from outer frame of suction device of No.02 bed, which had the same antimicrobial pattern as CRAB from 7 patients’sputum. After taking a series of controlling measures, there was no CRAB infection case after March 24,2014,this outbreak was effectively controlled. Conclusion This suspected HAI outbreak may be caused by inadequate disinfection of outer frame of suction device contaminated by CRAB,and horizontally transmitted through manipulation of sputum suction by health care workers.Clean and disinfection of ICU environmental object surfaces is of great importance for preventing HAI.

Aiming at the single treatment and the design separation between treatment and assessment in electrotherapy equipment, a kind of system including low-intermediate frequency treatment and efficacy evaluation was developed. With C8051F020 single-chip microcomputer as the core and the circuit design and software programming used, the system realized the random switch of therapeutic parameters, the collection, display and data storage of pressure pain threshold in the assessment. Experiment results showed that the stimulus waveform, current intensity, frequency, duty ratio of the system output were adjustable, accurate and reliable. The obtained pressure pain threshold had a higher accuracy (< 0.3 N) and better stability, guiding the parameter choice in the precise electrical stimulation. It, therefore, provides a reliable technical support for the treatment and curative effect assessment.
