1.Effects of mesenchymal stem cells transplantation on the brain-derived neurotrophic factor and growth associated protein-43 after the spinal cord injury of rats

[Objective]To observe the effects of mesenchymal stem cells(MSCs) transplantation on the brain-derived neurotrophic factor(BDNF) and growth associated protein-43(GAP-43) after the spinal cord injury(SCI) of rats, and to investigate the mechanism of repairing the SCI by MSCs transplantation. [Method]Seven days after the operation of SCI,the MSCs were transplanted into the injured site. Then GAP-43 and BDNF were tested by RT-PCR. By means of above,the mechanism of repairing the SCI after the cell transplantation was investigated.[Result]Compared with group B, transplantation of MSCs enhanced the expression of BDNF mRNA and GAP-43 mRNA in the spinal cord of group A, improved the neuron regeneration.[Conclusion] The transplantation of MSCs can repair the injuried site and promote the regeneration of axon by enhanced the expression of BDNF mRNA and GAP-43 mRNA. It is one of the mechanism of repairing the SCI by MSCs transplantation.
2.Investigation on Fluorine Contents of Well Water in Endemic Fluorosis Area of Hejian City

Objective To understand the relationship between the fluoride contents and the total hardness, pH value, the depth of well in the endemic fluorosis areas of Hejian city. Methods 40 villages in 2 towns of Hejian city were selected randomly and the well water samples in these villages were collected and determined. Results The contents of fluoride in the well water were higher than the limit(1 mg/L) in Sanitary Standard for Drinking Water Quality (China, 2001), ranged 1.2~4.4 mg/L, with a mean of 2.80 mg/L. ObjectiveTo detect the expressions of Toll-like receptor(TLR) 7 and 9 mRNA in peripheral blood mononuclear cells(PBMCs) from patients with vitiligo and their significance.Methods Real-time fluorescence-based quantitative reverse transcription-PCR was performed to detect the expressions of TLR7 and TLR9 mRNA in PBMCs from 50 patients with vitiligo and 25 normal human controls.Two-sample t test was conducted by using SPSS 11.5 software to compare the expression difference of TLR7 and TLR9 mRNA between the patients and controls.ResultsThe mRNA expression level (the ratio of the absolute copy number of a target gene to a control gene) of TLR7 and TLR9 in PBMCs were significantly higher in patients with vitiligo than in the normal human controls(0.85 ± 1.90 vs.0.44 ± 1.18,P < 0.05; 0.94 ± 2.25 vs.0.11 ±0.31,P < 0.01 ).No significant difference was observed in the expression level of TLR7 or TLR9 mRNA in PBMCs between patients with stable and active vitiligo,or between patients with localized and generalized vitiligo(both P > 0.05).ConclusionsThe expression level of TLR7 and TLR9 mRNA is elevated in PBMCs from patients with vitiligo,which may be involved in the pathogenesis of vitiligo.
4.Clinical effect of 125I radioactive seed implantation combined with intermittent hormonal therapy on prostate cancer

Objective:To explore clinical effect of 125I radioactive seed implantation combined with intermittent hormonal therapy (IHT) on prostate cancer. Methods:During the period from May 2016 to May 2017, 84 patients pathologically diagnosed with prostate cancer of T 3N 0M 0 by biopsy in Affiliated Hospital of Chifeng College were enrolled as study objects. They were divided into two groups by random number table method. 40 cases in control group were treated with intermittent hormonal therapy, while 44 cases in observation group were treated with 125I radioactive seed implantation combined with IHT. After the first IHT, time of prostate specific antigen (PSA) returning to normal and intermission time in both groups were observed. The dynamic changes of PSA level and prognosis in both groups during treatment were recorded. Results:After the first IHT, time of PSA returning to normal in observation group was shorter than that in control group, while intermission time was longer than that in control group ( P<0.05). At 3, 6, 12 and 24 months after treatment, PSA level in observation group was lower than that in control group ( P<0.05). The 2-year PSA-free progression survival (PSA-PFS) rate in observation group was higher than those in control group (77.27% vs 45.0%) ( P<0.05). There was no significant difference in 2-year total overall survival (OS) rate between the two groups ( P>0.05). There were no serious complications during treatment in observation group. Conclusions:125I radioactive seed implantation combined with IHT in treatment of prostate cancer can reduce PSA level more quickly, and increase 2-year PSA-PFS rate.
5.Metabolic syndrome in heart transplant recipients: prevalence and risk factors (a retrospective study)

Objective To determine,in our experience,the prevalence and presence of risk factors for metabolic syndrome (MS) in heart transplant recipients.Methods We studied 135 patients who underwent heart transplantation in our hospital from September 2000 to December 2011,According to the diagnostic criteria of MS,they were divided into two groups.All variables that could be related to the development of MS during the follow-up period were analyzed.The prevalence and presence of risk factors for MS in our experience were determined.Student t-test,Wilcoxon rank sum test and x2 test were used for univariate statistical analysis and logistic regression for multivariate analysis.Results (1) Of the 135 patients,63 developed MS (46.67%) during a mean follow-up period of 47.3 months.The multivariate analysis identified the follows as predictive factors for the development of MS:age (OR=1.09,95% CI 1.01-1.22,P<0.05),body mass index (BMI) (OR=1.65,95% CI 1.16-1.87,P<0.05),history of DM (OR =3.27,95% CI 1.12-34.21,P<0.05).Conclusion In our population,the prevalence of MS after heart transplant is 46.67% during a mean follow-up period of 47.3 months.Age,pre-operative overweight especially BMI,and history of DM were significant and independent risk factors for the development of MS during the follow-up period.For these patients,dose of immunosuppressant and diet control are important.
6.Reliability of the Boston bowel preparation scale

Objective To analyze the reliability of the Boston bowel preparation scale (BBPS) for the assessment of bowel preparation quality. Methods Demonstration video of Boston bowel preparation scale provided by Boston Medical Center were viewed by 49 clinicians,who came from several hospitals in Guangdong province and participated in an education conference.Then 3 testing colonoscopy videos were scored with BBPS and inter-observer reliability was assessed. 13 participants repeated the above test 3 months later,and intra-observer reliability was assessed.Results The correlation coefficient of inter-observer for BBPS scores was 0.987 (95% CI:0.949 - 1.000 ),and that of intra-observer was 0.713(95 % CI:0.452 -0.849).Conclusion BBPS is a reliable measure of bowel preparation,while the validity of BBPS deserves further analysis.
7.Construction of TRAIL eukaryotic expression vector driven by the hTERT promoter and apoptotic effect on nasopharyngeal carcinoma cell line CNE-2

OBJECTIVE To explore the effect of gene therapy on the nasopharyngeal carcinoma (NPC) cell line CNE-2 by using the combination of TRAIL gene with hTERT promoter. METHODS Total RNA was ex-tracted from PBL (peripheral blood lymphocytes)whose proliferation had been stimulated. TRAIL gene with interleukin 2 signal peptide gene was amplified by RT-PCR and cloned into the vector pGL3-181hTE down-stream to the RT promoter to form an eukaryotic vector. The vector was transfected into CNE-2 cells and HL-7702 cells through lipofection. Flow cytometry (FCM), agarose gel electrophoresis and cell morphology were used to examine the cell apoptosis. RESULTS In tranfected nasopharyngeal carcinoma cells CNE-2, FCM analysis showed that apoptotic peak appeared before G1 phase. A ladder-like pattern of DNA fragmentation appeared upon agarose gel electrophoresis. Many cells exhibited apoptotic changes such as cell shrinkage , nulear condensation , and nuclear fragmenta-tion under transmission electron microscope (TEM). CONCLUSION The recombinant eukaryotic ex-pression vector for TRAIL gene driven by hTERT pro-moter was successfully constructed and shown to induced apoptosis in CNE-2 cells. The results suggest that TRAIL may be a promising target for gene therapy of NPC.
8.Normal range of serum uric acid in quails of different strains and genders

Objective To explore the normal range of serum uric acid in quails.Method The data were collect-ed from male and female quails of different strains in 11 experiments, and analyzed by the way of medical reference value. Result The mean value of serum uric acid of male Difake strain quails was (221.06 ±79.59)μmol/L,and the normal range was (87.32 to 382.34)μmol/L.The mean value of serum uric acid of female Difake strain quails was (189.85 ± 68.58)μmol/L, and the normal range was ( 72.72 to 369.73 )μmol/L.The mean value of serum uric acid of male Longcheng strain quails was (217.22 ±72.91)μmol/L,and the normal range was (82.92 to 360.24)μmol/L.The mean value of serum uric acid of female Longcheng strain quails was ( 197.27 ±66.84 )μmol/L, and the normal range was (95.36 to 348.73)μmol/L.The serum uric acid value of female quails was significantly lower than that of male quails. There was no significant difference in the value of serum uric acid between female and male quails from each day-age groups.Conclusions This study has established the normal ranges for serum uric acid in quails of different strains and genders.
9.Study on serum vitamin D,immunoglobulin and alkaline phosphatase levels in children with recurrent community acquired pneumonia

Objective To determine the clinical significance of measuring the levels of 25-(OH) vitamin D3 [(25-(OH) D3)],immune globulins and serum alkaline phosphatase(AKP) in children with recurrent community-acquired pneumonia(CAP).Methods Serum samples were collected in children with recurrent CAP (recur-rent CAP group,n =38),first CAP (first CAP group,n =123),and age matched normal children (heath control group,n =39).The 25-(OH) D3 level,IgG,IgM,IgA and AKP levels were measured.Results The serum 25-(OH)D3 concentration of the recurrent CAP group was (47.87 ± 2.28)nmol/L,that of the first CAP group was (84.43 ± 4.12) nmol/L,and that of the heath control group was (91.2 ± 7.85) nmol/L,the differences among the three groups were statistically significant (t =3.716,8.881,all P <0.05).The differences of the serum IgA concen tration among the three groups had statistically significant differences (t =4.596,5.319,9.230,all P < 0.05).But the serum IgG concentration among the three groups had no statistically significant differences(t =1.115,1.887,1.090,all P > 0.05).The serum IgM concentration among the three groups had no statistically significant differences (t =0.804,0.942,0.918,all P > 0.05).The serum AKP concentration among the three groups had no statistically signifi cant differences(t =0.277,0.661,0.866,all P > 0.05).The serum 25-(OH) D3 levels in 161 cases of CAP patients were not correlated to any of the levels of IgM,IgG,IgA,or AKP.Conclusion The levels of 25-(OH) D3 and IgA are significantly lower in children with recurrent CAP,indicating that vitamin D deficiency may play a role in the pathogenesis of recurrent CAP.Prophylactic use of vitamin D supplementation may improve the immune function and reduce the development of recurrent CAP in children.
10.Expression of Toll-like receptors 7 and 9 as well as their regulatory molecules myeloid differentia-tion factor 88 and nuclear factor-κB in peripheral blood mononuclear cells of patients with vitiligo

Objective To compare expression of Toll?like receptors 7 and 9(TLR7, TLR9)as well as their regulatory molecules myeloid differentiation factor 88(MyD88)and nuclear factor?κB(NF?κB)in peripheral blood mononuclear cells(PBMCs)between patients with vitiligo and healthy individuals, and to explore their significance. Methods Flow cytometry was performed to measure expression of TLR7 and TLR9 in PBMCs among 36 patients with vitiligo and 22 healthy controls, and real?time fluorescence?based quantitative PCR(RT?PCR)was conducted to determine mRNA expression of MyD88 and NF?κB in the above blood samples. Results Compared with healthy controls, patients with vitiligo showed higher expression of TLR7 and mRNA expression of MyD88 and NF?κB, but lower expression of TLR9. However, significant differences were only observed in the mRNA expression of NF?κB(t=2.814, P=0.008), but not in the expression of TLR7 and TLR9 or the mRNA expression of MyD88 between patients and controls (t = 1.477, 1.761, 0.058, all P > 0.05). Conclusion NF?κB, as a key signaling molecule of TLR7 and TLR9 regulation pathways, increases obviously in patients with vitiligo, suggesting that NF?κB may be involved in the pathogenesis of vitiligo.
