1.Study on Anti-tumor Activities of Physalins B in Vitro

Objective To study the in-vitro anti-tumor activities of physalin B (PhB) extracted from Physalis angulata L.. Methods Th e effect of PhB on the cell proliferation and the morphology changes of tumor cell strains HepG2 and SGC7901 was observed by methyl thiazolyl tetrazolium (MTT) assay and by phase contrast microscopy respectively, and the results were compared to positive control anti-cancer drug hydroxycamptothecine (HCTP) . Apoptosis of HepG2 and SGC7901 cells induced by PhB was determined by 4’ , 6-diamidino-2-phenylindole ( DAPI) staining method. Results MTT results showed that the growth of HepG2 and SGC7901 was significantly inhibited by PhB in dose-and time-dependent manner, and the inhibitory effect of high concentration of PhB was better than the positive control medicine HCTP. Phase contrast microscopy results showed that the cellular morphology was improved by PhB, which was accorded with the results showed by MTT assay. DAPI staining results showed that the apoptosis of HepG2 and SGC7901 was enhanced by PhB. Conclusion PhB extracted from Physalis pubescens L. exerts certain inhibitory effect on the proliferation of HepG2 and SGC7901 cells.

Objective:To observe the effect of Xinmailong injection (XI) on free calcium ( Ca2+) content in rat myocardial cells. Methods:Free Ca2+content in rats treated with various dosages of XI were measured by fluorospectrophotometry. Results:Xinmailong injection significantly increased free Ca2+content and the content was 169.18?11.64, 233.26?10.69 and 164.25?10.34 nmol?L-1 in 0.19 g?L-1 , 0.38 g?L-1 and 0.76 g?L-1 XI group respectively, the differences being significant as compared with 120.64?3.02 nmol?L-1 in the control group (P
3.The results with pleural effusion of lung cancer treated with brucea javanica oil emulsion

Objective To evaluate the curative effects and side effects of burcea javanica oil emulsion(BJOE) in the treatment of pleural effusion of lung cancer by thoracic injection.Methods 40 cases with pleural effusion of lung cancer were treated by using BJOE.A dose of 50~100ml BJOE was injected intrapleurally,once every 5~7 days,4 time for a course of treatment,then the effects and side effects were observed after one month.Results The effective rate was 90%(36/40).The side effects were fever(n=4) and digestive sickness(n=2) as well thoracalgia(n=1).Conclusion Brucea javanica oil emulsion has good curative effect and low side effect in the treatment of pleural effusion of lung cancer.
4.Protective effect of Swertia Punicea on chronic liver injury

AIM:To observe the protective effect of Swertia Punicea on chronic liver injury induced by CCl 4. METHODS: To measure levels of serum ALT、AST and the total protein, and to observe changes of the liver index、the spleen index as well as the liver pathological change. RESULTS: Swetia Punicea obviously lowered the levels of serum ALT and AST, and increased the content of the total protein. The drug decreased the liver index, but it had no obvious effect on the spleen index. It also improved the liver fibrosis. CONCLUSION: Swertia Punicea can protect liver and decrease enzymatic activity, and can retard or ease up the formation of the liver fibrosis.
5.Protective Effect of Swertia Punicea on Experimental Hepatic Injury

Objective: to observe the protective effect of Swertia Punicea on experimental hepatic injury induced by CCl4 and BCG/LPS respectively. Methods: Mouse models of hepatic injury were induced by CCl4 and BCG/LPS. Serum levels of ALT and AST, MDA content and the pathological changes of hepatic tissue were observed. Results: Swertia Punicea obviously inhibited the rising levels of serum ALT and AST, improved the histological features and relieved hepatic edema induced by CCl4 in mice. However, it has no obvious action on the rising levels of ALT and AST induced by BCG/LPS. It also reduced MDA content and decreased the hepatic index and the splenic index. Conclusion: Swertia Punicea has antioxidation and a protective action on the chemical and immunological hepatic injury, especially on the chemical hepatic injury.

Objective To explore the effects of zinc finger E-box binding protein (ZEB)2 3′UTR gene transfection on proliferation, invasion and migration in human gastric epithelial cell line GES-1. Methods The synthetic ZEB2 3′UTR and miR-200b micmics were transfected into GES-1 cell line by lipofectamine 2000. We set up control grop, the mutation group and ZEB2 3′UTR group. Real-time quantitative PCR was performed to evaluate the expression levels of miR-200a/b/c and ZEB1/ZEB2 mRNAs after transfection.And then we set up control group, ZEB2 3′UTR group, ZEB2 3′UTR+negative control group and ZEB2 3′UTR+miR-200b micmics group. The protein expression levels of ZEB1, ZEB2, matrix metallopro-teinases (MMP) 2/9 and proliferating cell nuclear antigen (PCNA) were detected by Western blot assay. The invasion and mi-gration capability were analyzed by transwell assay and wound healing test. MTT assay was used to detect the proliferation ability. Results Compared with control group and mutation group, the expressions of miR-200a/b/c were significantly de-creased, especially for miR-200b. And the expressions of ZEB1/ZEB2 were significantly increased at both mRNA and pro-tein levels after transfected with the ZEB2 3′UTR, enhancing the capability of migration,invasion,and proliferation (P <0.05). Compared with ZEB2 3′UTR group, the capabilities of proliferation,invasion and migration were significantly lower in combined group. Conclusion ZEB2 3′UTR can increase the ability of cell proliferation, invasion and metastasis through regulating the levels of miR-200a/b/c, and then influence the regulation of transcription of the target gene, which could lead to malignant transformation of GES-1 cells.
8.Clinical analysis of acute kidney injury in 1113 patients after cardiac valve replacement surgery

Objective To investigate the incidence and risk factors of acute kidney injury(AKI)after different types of cardiac valve replacement surgery. Methods A single cohort of 1113 patients who received cardiac valve replacement surgery from April 2009 to March 2010 in Zhongshan Hospital,Fudan University were prospectively analyzed.Multivariate Logistic regression analysis was used to evaluate possible risk factors associated with post-operative AKI.Akl was defined as a relative 50% increase or an absolute increment of 26.4 μmol/L in Scr within 48 hours and/or urine volume <0.5ml·kg-1·h-1 up to 6h.Results Of the 1113 patients, the incidence of AKI was 33.24%.In-hospital mortality of AKI patients was 6.49%,which was 5.373 times higher than that of non-AKI patients(P<0.01).The incidence of AKI in patients who simultaneously received cardiac valve replacement and coronary artery bypass grafting was 75.00%,which was significantly higher as compared to other types of valve replacement surgery(P<0.01).Unconditional multivariate Logistic regression analysis revealed that male,old age,long extracorpeal circulation (CPB)time(≥120 min)and combined with coronary artery bypass grafting surgery were the independent predictors of AKI episodes,and the corresponding OR values were 1.455,2.110,1.768 and 2.994 respectively. Conclusions AKI is a common and serious complication after cardiac valve replacement surgery.Patients who received combined cardiac surgery as valve replacement and coronary artery bypass grafting have higher incidence of AKI.Old age,male,long CPB time(≥120 min)and combined with coronary artery bypass grafting surgery are the independent risk factors of post-operative AKI for patients undergoing cardiac valve replacement surgery.
9.Assessment of shape and function of right ventricle in heart transplantation patients by single beat real-time three-dimensional echocardiography

Objective To investigate the shape and function of right ventricles in patients received heart transplantation(HT). Methods Sixty healthy volunteers(control group) and 31 HT patients(HT group) were enrolled, and the HT group was further divided into the nonrejected(HTn) group and the rejected(HTp) group based on the endomyocardial biopsy(EMB) results. All the participants received routine echocardiography. Single beat real-time three-dimensional echocardiography (sRT-3DE) was performed in all to evaluate parameters concerning modality and systolic function of participants' right ventricles. Results 1) Right ventricular stroke volume (RVSV) and right ventricular ejection fraction (RVEF) were significantly different among the groups[RVSV: control group vs HTn group vs HTp group was (56.18 ± 23.72)ml vs (36.08 ± 10.94)ml vs (26.22 ± 9.84)ml, P <0.0001; RVEF:control group vs HTn group vs HTp group was (62.09± 7.18)% vs (51.04 ± 7.58) vs (35.86 ± 9.86)%, P <0.00001=.2= When taking the influence of rejection into consideration,RVEF was proved to be a stable and sensitive indicator. Conclusions sRT-3DE can quickly assess shape and systolic function of right ventricle.RVEF was the most stableand sensitive among all the RV-related indicators and is a promising indicator in the clinic follow-up of HT patients.
10.Protective Effects of Luotai Injection Powder on Cerebral Ischemia

Objective:To study the protecting effects of Luotai Injection Powder on cerebral ischemia.Methods: The protecting effects of Luotai Injection Powder on cerebral ischemia were studied on animal models of cerebral ischemia induced by ligaturing bilateral common carotid artery and decressing blood pressure. Results:Luotai Injection Powder (at doses of iv. 60mg/kg and 120mg/kg) significantly decreased the water content, capillary permeability and ischemia injury in brain of rats. In addition, it significantly increased the tolerance in mice at a dose of iv. 120mg/kg. Conclusions: Luotai Injction Powder has a protective effect on acute cerebral ischemia
