Objective To investigate the effects of Raddeanin A(RA)on apoptosis and cell cycle of hu-man colon cancer HT29 cells,and discuss the possible mechanism.Methods Different concentration of RA 1,2, 4,8,16μmol/L disposed HT29 cells 12,24,48 hour,3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bro-mide(MTT)assay was used for the cell proliferation;Hoechst33258 staining was performed to observe the effects of RA on apoptosis morphology;Flow cytometry(FCM)was devoted to calculate apoptosis rate and detect cell cycle;The expression of Cleaved-caspase-3,Cleaved-PARP,CyclinB,CDK1 proteins and other proteins were tested by Western blot(WB). Results RA could inhibit the proliferation of HT29 cells,and associated with concentration and time(P<0.05);The IC50of 12 h,24 h,48 h were(9.31 ± 0.63)μmol/L,(4.88 ± 1.02)mol/L,(3.60 ± 0.89) μmol/L.The nucleus pycnosis,fracture were detected by Hochest33258 staining,RA induced apoptosis in HT29 cells;Western blot(WB)assay showed that the expression of Cleaved-caspase-3,Cleaved-PARP rised.HT29 cells were arrested at the period of G2/M,and the CyclinB,CDK1 proteins down-regulated.WB result showed that PI3K, AKT proteins up-regulated,while p-PI3K,p-AKT protein down-regulated.Conclusion RA can inhibit the prolifer-ation of colon cancer HT29 cells and its mechanism is probably by regulating the PI3K/AKT signaling pathways which through inducing apoptosis and blocking the cell cycle.
4.Effect of raddeanin A on autophagy and apoptosis of colon cancer HCT116 cells through regulating mTOR pathways

Objective To explore raddeanin A(RA) inhibiting the proliferation of HCT116 cells and its related mechanism.Methods The MTT method was used to observe the RA inhibition on the proliferation of gastric cancer cells HCT116.Flow cytometry was used to detect the RA effects on cell apoptosis.Transmission electron microscope(TEM) was used to observe RA induced autophagy.Western blot was used to test expression of apoptosis and autophagy related proteins.Results RA could significantly inhibit the proliferation of HCT16 cells with concentration and time dependence;double layer membrane of autophagosome was detected by TEM;FCM showed that RA induced apoptosis in HCT116 cells,moreover the apoptosis rate was increased with the concentration increase;Western blot showed that the expression of autophagy related proteins(Beclin1 and LC3) was increased,the expression of apoptosis inhibition protein Bcl-2 was decreased.On the contrary,the expression of apoptosis promotion proteins(Bax,Cleaved-caspase-3,Cleaved-PARP) was increased,the expression of mTOR protein in the mTOR signal pathway was increased,while the p-mTOR protein expression was decreased.When the mTOR pathway inhibitor rapamycin(RAPA) was added to cells,Beclin-1,LC3,Cleaved-caspase-3,Cleaved-PARP proteins were increased,and apoptosis rate was also increased.Conclusion RA can inhibit the proliferation of HCT116 cells and can induce cellular autophagy and apoptosis,its mechanism may be realized by regulating the mTOR signal pathway.

Objective To understand the compliance of highly active anti-retroviral therapy (HAART) and influencing factors in people living with HIV/AIDS (PLWHA) in Nanjing. Methods PLWHA receiving HAART in No. 2 Hospital of Nanjing during May-June 2014 were recruited in this study. Self-administrated questionnaire was used to collect the data about HAART compliance and socio-demographic characteristics of PLWHA surveyed. Descriptive and multivariate statistical analysis were conducted to examine the effects of the factors on self-reported HAART adherence. Results A total of 276 PLWHA were surveyed,According to the evaluation criterion of Center for Adherence Support Evaluation (CASE),252 cases showed good compliance (91.3%). logistic regression analysis revealed that smoking,progress of the disease and side effects,reminding of taking drug and age were correlated with self-reported HAART adherence. Conclusion It is suggested to strengthen the education about antiviral therapy compliance in PLWHA with mild infection and those who are smokers and young,suffer from side effects,have no reminding methods for taking drug.
6.Rapid Identification of Gastrodiae Rhizoma with Different Sulfur Fumigation Levels Based on Ultra-fast Gas Phase Electronic Nose

ObjectiveIn order to find a fast odor-based method for the identification of sulfur fumigated Gastrodiae Rhizoma, an ultra-fast gas phase electronic nose technology was used to identify the odors of different degrees of sulfur fumigated Gastrodiae Rhizoma decoction pieces. MethodHeracles NEO ultra-fast gas phase electronic nose was employed to collect gas chromatograms of unsulfured and sulfured with different degrees of Gastrodiae Rhizoma decoction pieces, gas chromatograms were performed under programmed temperature (initial temperature of 40 ℃, 0.2 ℃·s-1 to 60 ℃, and then 4 ℃·s-1 to 250 ℃), the sample volume was 5 mL, the incubation temperature was 65 ℃ and incubation time was 35 min. Kovats retention index and the AroChemBase database were used for qualitative analysis, and stoichiometric analysis was performed on this basis. Principal component analysis (PCA), discriminant factor analysis (DFA) and partial least squares-discriminant analysis (PLS-DA) models were established to identify the Gastrodiae Rhizoma decoction pieces with different degrees of sulfur fumigation. ResultAccording to the comparative analysis of AroChemBase database, there were significant differences in the odor characteristics of sulfur fumigated and non-sulfur fumigated Gastrodiae Rhizoma, cyclopentane, acetone and heptane might be the odor components to distinguish the degree of sulfur fumigation in Gastrodiae Rhizoma decoction pieces. The identification index of PCA model was 81, the accumulative discriminant index of the discriminating factors was 92.09% in DFA model, the supervisory model interpretation rate of PLS-DA model was 0.963 and the predictive ability parameter was 0.956, indicating that PCA, DFA and PLS-DA models could well distinguish Gastrodiae Rhizoma decoction pieces with different sulfur fumigation degrees. ConclusionHeracles NEO ultra-fast gas phase electronic nose can be used as a rapid method to identify and distinguish Gastrodiae Rhizoma decoction pieces with different levels of sulfur fumigation. Meanwhile, it can provide a rapid, simple and green method and technology for identification of traditional Chinese medicine decoction pieces by sulfur fumigation.

OBJECTIVETo understand the economic burden caused by antiretroviral treatment for people living with HIV/AIDS and influencing factors in Nanjing.
METHODSPeople living with HIV/AIDS were divided into two groups, HIV infected patients and AIDS patients. The data about their direct medical expenditure, direct non-medical expenditure and indirect economic loss in the past year were collected. Univariate and multivariate analysis were conducted to identify the potential influencing factors.
RESULTSThe median of direct medical expenditure and total costs for 133 HIV infected patients were 1,200 yuan RMB and 1,972 yuan RMB respectively. The median of direct medical expenditure and total costs for 145 AIDS patients were 1 060 yuan RMB and 2 826 yuan RMB respectively. The differences in direct medical expenditure and total costs between HIV infected patients and AIDS patients showed no statistical significance. The results from univariate analysis indicated that the sample source influenced total costs significantly. Multivariate analysis showed that onset time and CD level were negatively correlated with direct medical expenditure. The patients infected through heterosexual contact had more direct medical expenditure than those infected through homosexual contact. The patients receiving HIV test in hospitals had more direct medical expenditure than those receiving volunteer counseling and testing.
CONCLUSIONFurther efforts should be made to expand HIV testing and treatment coverage in order to detect HIV infections as early as possible. Early antiretroviral treatment should be given to HIV infected patients to maintain their immunity and reduce their medical expenditure.
