1.The effects of mycophenolate mofetil on renal interstitial fibrosis and epithelial-myofibroblast transiation in adenine-induced renal failure rats

Objective The aim of this study is to examine the effect of myoophenolate mofetil (MMF) on epithelial-myofibroblast transiation(EMT) in adenine-induced chronic renal failure (CRF) rat model and the role of vascular endothelial growth factor(VEGF) and inhibitor of differentiation (Id2 and Id3) in EMT in the rat kidney. Methods Sixty-four male Wistar rats were randomly assigned to the following groups: normal control (n=16), CRF (n=24) and MMF(n=24). CRF was induced by gastric gavage of adenine (125 mg·kg-1·d-1) to rats for eight weeks. CRF rats were treated with MMF (15 mg·kg-1·d-1) as "MMF" group. The rats were sacrificed at week 2, 4, 6 and 8, respectively.Urinary protein and serum ereatinine levels were measured, and the histopathologic degrees of interstitial fibrosis were evaluated in Massen-stained sections. Expressions of a-smooth muscle actin (α-SMA),transforming growth factor β1 (TGFβ1), VEGF and Id (Id2 and Id3) in the kidney tissue were assessed by immunohistochemistry, RT-PCR and/or Western blot methods. Results The urinary protein level in MMF group was evidently lower than that in CRF group (P<0.01), whereas no statistically significant difference was observed in serum creatinine level between the two groups. Renal interstitial fibrosis was reduced significantly with MMF treatment (P<0.01). Expression of α-SMA in MMF group was lower than that in CRF rats at week 6, 8 (P<0.01), while expression of TGFβ1 was decreased markedly at week 2, 4,6 (P<0.01). The expressions of VEGF in MMF rats were increased significantly at week 6,8 (P<0.01),and Id2,Id3 in MMF rats were increased significantly at week 4,6 (P<0.05). Conclusions MMF may ameliorate chronic renal fibrosis and EMT in adenine-induced CRF rats. This effect of MMF on EMT is probably related to upregulation of VEGF, Id2 and Id3 expressions and suppressing overexpression of TGFβ1 in renal tissue. The exact mechanism needs to be studied further.
2.Effect of Electro-Acupuncture Combined with Rehabilitation Training to Shorten Period of Flaccid Paralysis after Cerebral Infarction

Objective To observe the curative effect of electro-acupuncture combined with rehabilitation training to shorten the period of flaccid paralysis after cerebral infarction.Methods 78 victims were divided into treatment group and control group at random. The treatment group received electro-acupuncture combined with rehabilitation training, and the control group received rehabilitation training only. Muscular tension was evaluated on the 15th day and 30th day after admission according to the modified Ashworth spasm rating scale.Results The number of flaccid paralysis victims in the treatment group is significantly less than that in the control group (P<0.05) on the 15th day and 30th day after treatment.Conclusion Electro-acupuncture combined with rehabilitation training can shorten the period of flaccid paralysis after cerebral infarction significantly.
3.Inhibitory effect of vascular endothelial growth factor on transforming growth factor β1-induced epithelial-mesenchymal transition of HK2 cells and its relationship with connective tissue growth factor and PI3K-Akt pathway

Objective To examine the relationship of the inhibitory effect of vascular endothelial growth factor(VEGF) on epithelial-mesenchymal transition (EMT) induced by TGF-β1 in HK2 cells with the expression of connective tissue growth factor (CTGF) and PI3K-Akt pathway. Methods The cultured HK2 cells were divided into the following groups: normal control group, TGF-β1 (5 μg/L) group, VEGF (100 μg/L) group, TGF-β1 plus VEGF group. LY294002 (25 μmol/L), the blocker of PI3K-Akt pathway, was added to each of above-mentioned groups for the second part of the study, α-smooth muscle actin (α-SMA) and E-cadherin expressions of HK2 cells were assessed with double-stain immunocytochemistry method. The mRNA and protein expressions of α-SMA and CTGF of cells were assessed with RT-PCR and Western blot. The expressions of fibronectin (FN) and collagen Ⅰ (Col Ⅰ) in medium were assessed with ELISA. Results The expressions of α-SMA and CTGF significantly increased in HK2 cells treated with TGF-β1 compared with those in normal control (P<0.05), while significantly decreased in cells co-treated with TGF-β1 and VEGF compared with those treated with TGF-β1 alone (P<0.05, respectively). The expression of E-cadherin was exactly opposite to that of α-SMA. When LY294002 was added to TGF-β1 and VEGF co-treated cells, the expressions of α-SMA, CTGF, FN and Col Ⅰ were markedly up-regulated, when compared with those without LY294002 treatment (P<0.05). Conclusion Inhibitory effect of VEGF on TGF-β1-induced EMT of HK2 ceils in vitro may be related to down-regulation of CTGF expression and reduction of FN and Col Ⅰ, which may be partly dependent on PI3K-Akt pathway.
4.Treatment of acute gastric mucosal lesion after pericardial devascularization of advanced schistosomiasis:a report of 15 cases

Objective To discuss the therapy for acute gastric mucosal lesion(AGML)after pericardial devascularization of advanced schistosomiasis. Methods Fifteen advanced schistosomiasis patients with AGML after pericardial devascularization re-ceived supporting therapy,reducing stomach acid and protecting gastric mucosa therapy,reducing gastric blood flow therapy,en-doscopic spraying topical hemostatic agents or electric coagulation therapy,etc. Results After pericardial devascularization of ad-vanced schistosomiasis,15 patients occurred AGML,and among which,10 cases occurred from 8 to 14 days,and the shortest pe-riod was 5 days and the longest was 23 days. Among the 15 patients,13 patients were cured(86.7%),and 2 died. Conclusion The comprehensive therapy including supporting therapy,reducing gastric acid and protecting gastric mucosa,and endoscopic he-mostasis is effective in the treatment of AGML.
5.The study of classification for breast solid nodular lesions using an ultrasonographic characteristic diagnostic score system

Objective To study the accuracy of an ultrasonographic characteristic diagnostic score system(UCDSS) which was used to classify the breast solid nodular lesions. Methods UCDSS were established by analyzing the ultrasonographic sign of the 205 breast solid nodular lesions. These lesions were classified according to the total scores of each ultrasonographie sign. ROC curve was used to evaluate the value of UCDSS. Results The area under ROC curve of UCDSS was 0. 977, the sensitivity and specificity was 92.0%, 92.3% respectively. Conclusions The classification based on the UCDSS may increase the diagnostic accuracy of the differentiation between benign and malignant breast solid nodular lesions.
7.Effects of Model of Rehabilitation Education on Recovery of Stroke

Objective To observe the effects of different models of rehabilitation education on stroke rehabilitation. Methods There were 35 stroke patients in group Ⅰ,40 patients in group Ⅱ. All the patients received the same rehabilitation treatment, but different model of rehabilitation education. They were evaluated with Functional Independence Measure (FIM) before and after treatment, and the incidence of complications were investigated. Results The FIM grade in all patients increased significantly after treatment (P<0.05), and increased more in the group Ⅱ (P<0.05). The incidence of shoulder pain/ shoulder subluxation, edema in hemiplegic extremity, muscle atrophy were a lower in the group Ⅱ than that in the groupⅠ (P<0.05). Conclusion Improving the model of rehabilitation education can improve the efficacy of stroke rehabilitation.
8.Effect of Integrated Traditional Chinese and Western Medicine on Primary Trigeminal Neuralgia

ObjectiveTo observe the effect of Integrated Traditional Chinese and Western Medicine on primary trigeminal neuralgia. Methods126 cases with primary trigeminal neuralgia were divided into treatment group (63 cases) and control group (63 cases), who accepted Integrated Traditional Chinese and Western Medicine and western medicine only. They were assessed with Chinese version McGill Pain Questionnaire before and after treatment, and followed up for 1 year. ResultsThe incidence of cure in treatment group was higher than that of control group (P<0.05), and the relapse was reduced (P<0.05). ConclusionThe Integrated Traditional and Western Medicine is more effective on primary trigeminal neuralgia, and reduce the relapse.
9.Cloning and Identification of the cDNA and Genomic DNA Sequences of the Defensin Gene of Anopheles sinensis

Objective To clone and identify the cDNA sequence and genomic DNA sequence of Anopheles sinensis defensin gene. Methods Referring to the published defensin gene sequences of Aedes aegypti and Anopheles gambiae, pairs of primers were designed to amplify the cDNA sequence and genomic DNA sequence of Anopheles sinensis defensin gene with template of Anopheles sinensis total RNA by RT-PCR and genomic DNA by nested PCR, respectively. These amplified fractions were cloned and sequenced, and were further identified and analyzed by relevant bioinformatics softwares. Results Complete genomic DNA sequence was cloned, including 5′ and 3′ UTR fraction and two exons separated by a 85 bp intron. Besides, the whole cDNA sequence of Anopheles sinensis defensin gene with 324 bp was also cloned, and its ORF encoded 107 amino acids. The mature peptide had 40 amino acids residues. Conclusion The whole cDNA sequence and complete genomic DNA sequence of the defensin gene of Anopheles sinensis have been cloned and identified for the first time.
