1.The Reverse Effect of Progesterone on a Human Tongue Cancer Drug-resistance Cell Line Tca8113/BLM

Objective To explore the reverse effect and mechanism of progesterone(Prog) on a human tongue cancer drug-resistant cell line Tca8113/BLM in vitro. Methods The reverse effect of Prog on Tca8113/BLM was determined by MTT. The effects of Prog on the intracellular ADM congregation, cell cycle distribution and the expression of P-gp in the cell membrane were detected by flow cytometry (FCM). Results The value of IC50 of Tca8113 and Tca8113/BLM cells without Prog was 9 56 and 120 1,respectively. After treated with Prog(2umol/L),The value of IC50 of Tca8113 and Tca8113/BLM cells was 9 56 and 12 1,respectively. The potent fold of BLM in Tca8113/BLM cells was 9 92, the concentration of intracellular ADM in Tca8113/BLM cells significantly increased(P
2.Comparison of narcotrend index and bispectral index during propofol-remifentanil anesthesia administered by TCI

Objective To compare bispectral index (BIS) with narcotrend index (NI) during propofol-remifentanil anesthesia administered by target-controlled infusion (TCI).Methods Ten ASA Ⅰ or Ⅱ pafients aged 18-56 yr weighing 52-67kg undergoing abdominal surgery lasting>1h were included in this study.BIS and NI were monitored simultaneously.Anesthesia was induced with TCI of propofol with target plasma concentration (Cp) of 3~4μg/ml and remifentanil (Cp 3-4ng/ml).Tracheal intubation was facilitated with cis-atracurium 0.3 mg/kg.The patients were mechanically ventilated.PETCO2 was maintained between 30-35 mm Hg.Anesthesia was maintained with TCI of propofol and remifentanil by the anesthesiologist bhnded to BIS and NI values.according to hemedynamic parameters.BIS and narcotrend values were recorded every minute and compared by another anesthesiologist.All data were compared by Bland-Altman analysis and with Kappa coefficient for agreement.The correlation between BIS and NI was tested by Spearman correlation analysis.The number of error ofjudgement (Type Ⅰ was defined as BIS<40 and NI>62;Type Ⅱ was defined as BIS>60 and NI<20)Was counted.Results The correlation and agreement between BIS and NI during maintenance of propofol-remifentanil anesthesia administered by TCI showed good consistency.Conclusion Both NI and BIS Can help anesthesiologist control the depth of anesthesia during TCI of propofol-remifentanil.
3.Clinical analysis of otogenic extracranial and intracranial complications.

OBJECTIVE:
To study the clinical feature and treatment of extracranial and intracranial complications caused by otitis media.
METHOD:
Three hundred and twenty patients of acute and chronic otitis media were admitted to our department between 2005 and 2014. Among them, 34 patients were diagnosed with extracranial and intracranial complications. The clinical features and treatment outcome were retrospectively studied. Of the 34 patients associated with complications, 25 had a single complication,8 had two complications and 1 had three complications. Complications included labyrinthitis in 14 cases, facial paralysis in 11, postauricular subperiosteal abscess in 6, Bezold abscess in 1, thrombophlebitis of sigmoid sinus in 2, otitis meningitis in land otogenic brain abscess in 8.
RESULT:
Thirty-three patients were cured or improved and 1 patient died.
CONCLUSION
Due to the widespread use of antibiotics, the clinical manifestations of extracranial and intracranial complications of otitis media become more hidden and atypical. The surgery is the primary treatment method.

5.Effects of Wenshen Jianpi Recipe on chronic wound healing in rats

OBJECTIVE: To study the effects of Wenshen Jianpi Recipe (WSJPR, a traditional Chinese medicine for warming kidney and invigorating spleen) on chronic wound healing and the mechanism. METHODS: Ninety-six SD rats were randomly divided into 4 groups, with 24 rats in each group, and back wound was made in the rats. For rats in 3 of the 4 groups, hydrocortisone injection was administered to induce chronic wound. Rats in 2 of the 3 groups were treated with WSJPR and Xinpukang Granules (XPKG) respectively, and the rats in the other group were untreated. The rats in the fourth group were taken as control. The wound healing time and the width of new epidermis were observed, and the histomorphological changes and cell cycle of the granulation tissue, and the protein expressions of epidermal growth factor (EGF), transforming growth factor-beta(1) (TGF-beta(1)) and fibronectin (FN) in the granulation tissue were tested with immunohistochemical technique and flow cytometry. RESULTS: The wound healing time of the WSJPR-treated and XPKG-treated groups was (17.0+/-1.9) and (18.8+/-1.9) d respectively, much shorter than that of the untreated and control groups (P<0.05). On the 14th experiment day, the width of new epidermis of the WSJPR-treated and XPKG-treated groups was (3.73+/-0.19) and (3.21+/-0.15) mm respectively, much wider than that of the untreated and control groups (P<0.05). The numbers of angiogenesis, fibroblasts and cells in the S phase in WSJPR-treated and XPKG-treated groups were much higher than those in the untreated and control groups (P<0.05). Compared with the untreated and control groups, the protein expressions of EGF, TGF-beta(1) and FN in WSJPR-treated and XPKG-treated groups were higher (P<0.05). CONCLUSION: WSJPR can enhance the wound healing. It was likely through accelerating the cell proliferation and up-regulating the expressions of EGF, TGF-beta(1) and FN.
7.The effects of mycophenolate mofetil on renal interstitial fibrosis and epithelial-myofibroblast transiation in adenine-induced renal failure rats

Objective The aim of this study is to examine the effect of myoophenolate mofetil (MMF) on epithelial-myofibroblast transiation(EMT) in adenine-induced chronic renal failure (CRF) rat model and the role of vascular endothelial growth factor(VEGF) and inhibitor of differentiation (Id2 and Id3) in EMT in the rat kidney. Methods Sixty-four male Wistar rats were randomly assigned to the following groups: normal control (n=16), CRF (n=24) and MMF(n=24). CRF was induced by gastric gavage of adenine (125 mg·kg-1·d-1) to rats for eight weeks. CRF rats were treated with MMF (15 mg·kg-1·d-1) as "MMF" group. The rats were sacrificed at week 2, 4, 6 and 8, respectively.Urinary protein and serum ereatinine levels were measured, and the histopathologic degrees of interstitial fibrosis were evaluated in Massen-stained sections. Expressions of a-smooth muscle actin (α-SMA),transforming growth factor β1 (TGFβ1), VEGF and Id (Id2 and Id3) in the kidney tissue were assessed by immunohistochemistry, RT-PCR and/or Western blot methods. Results The urinary protein level in MMF group was evidently lower than that in CRF group (P<0.01), whereas no statistically significant difference was observed in serum creatinine level between the two groups. Renal interstitial fibrosis was reduced significantly with MMF treatment (P<0.01). Expression of α-SMA in MMF group was lower than that in CRF rats at week 6, 8 (P<0.01), while expression of TGFβ1 was decreased markedly at week 2, 4,6 (P<0.01). The expressions of VEGF in MMF rats were increased significantly at week 6,8 (P<0.01),and Id2,Id3 in MMF rats were increased significantly at week 4,6 (P<0.05). Conclusions MMF may ameliorate chronic renal fibrosis and EMT in adenine-induced CRF rats. This effect of MMF on EMT is probably related to upregulation of VEGF, Id2 and Id3 expressions and suppressing overexpression of TGFβ1 in renal tissue. The exact mechanism needs to be studied further.
8.Effect of hyperthermia in combination with progresterone on a human tongue cancer drug-resistance cell line Tca8113/BLM

Objective; To explore the possible effect of hyperthermia combined with Prog on a human tongue cancer cell line Tca8113 and drug-resistance cell line Tca8113/BLM in vitro. Methods;The effect of hyperthermia combined with Prog on Tca8113 and Tca8113/ BLM cells were determined by MTT. The concentration of ADM, the expression of P-gp and MRP were detected by flow cytometry (FCM). Results;Compared with Prog or hyperthermia, the hyperthermia at 41 X. for 1 h combined with Prog showed obvious inhibitory effect in Tca8113 and Tca8113/BLM cells(P<0.01). The concentration of ADM in Tca8113 and Tca8113/BLM cells increased obvi-ously(P<0.01). Expression levels of P-gp and MRP in Tca8113 and Tca8113/BLM cells decreased significantly. Conclusion:The hyperthermia combined with Prog can increase the intracellular concentration of ADM in Tca8113 and Tca8113 /BLM cells, which reverse the drug-resistance of Tca8113/BLM cells to BLM and down-regulate expressions of P-gp and MRP.
9.Application of scene Imitation in the education of patients before fiberoptic bronchoscopy

Objective To explore the application and effect of scene imitation method in the education of patients before fiberoptic bronchoscopy.Methods 264 elective bronchoscopy patients between March 2007 and Decembet 2007 were randomly divided into control group(120 patients)and the experiment group(144 patients)Traditional health education was applied in the control group before bronehoscopy.Scene imitation method in health education was applied in the experiment group before bronchoscopy.Results The examination succes rate in the experiment group was significantly improved(P<0.05).The medical order obeying behavior(MOOB)of the patients was significantly different between the two group(P<0.01).Conclusion Before the use of fiberoptic bronchoscopy,the new scene simulation method Can help patients on the list obtain more cognitive ability about the testing.Comparing the traditional education,it establishes a sound relationship between doctors and patients,reduces complication and leads to more accurency of testing,thus helping patients to recover soon.
10.Inhibitory effect of vascular endothelial growth factor on transforming growth factor β1-induced epithelial-mesenchymal transition of HK2 cells and its relationship with connective tissue growth factor and PI3K-Akt pathway

Objective To examine the relationship of the inhibitory effect of vascular endothelial growth factor(VEGF) on epithelial-mesenchymal transition (EMT) induced by TGF-β1 in HK2 cells with the expression of connective tissue growth factor (CTGF) and PI3K-Akt pathway. Methods The cultured HK2 cells were divided into the following groups: normal control group, TGF-β1 (5 μg/L) group, VEGF (100 μg/L) group, TGF-β1 plus VEGF group. LY294002 (25 μmol/L), the blocker of PI3K-Akt pathway, was added to each of above-mentioned groups for the second part of the study, α-smooth muscle actin (α-SMA) and E-cadherin expressions of HK2 cells were assessed with double-stain immunocytochemistry method. The mRNA and protein expressions of α-SMA and CTGF of cells were assessed with RT-PCR and Western blot. The expressions of fibronectin (FN) and collagen Ⅰ (Col Ⅰ) in medium were assessed with ELISA. Results The expressions of α-SMA and CTGF significantly increased in HK2 cells treated with TGF-β1 compared with those in normal control (P<0.05), while significantly decreased in cells co-treated with TGF-β1 and VEGF compared with those treated with TGF-β1 alone (P<0.05, respectively). The expression of E-cadherin was exactly opposite to that of α-SMA. When LY294002 was added to TGF-β1 and VEGF co-treated cells, the expressions of α-SMA, CTGF, FN and Col Ⅰ were markedly up-regulated, when compared with those without LY294002 treatment (P<0.05). Conclusion Inhibitory effect of VEGF on TGF-β1-induced EMT of HK2 ceils in vitro may be related to down-regulation of CTGF expression and reduction of FN and Col Ⅰ, which may be partly dependent on PI3K-Akt pathway.
