1.Effect of Extract of Chrysanthemum Indicum on Tumor Necrosis Factor-α Expression and Neutrophil Function in Chronic Bronchitis Rats

Objective To study the effect of the extract of Chrysanthemum Indicum(CI)on the expression of tumor necro-sis factor-α (TNF-α) and neutrophil function in chronic bronchitis (CB)rats and to explore the therapeutic mechanism for chronic bronchitis. Methods The extract of CI was prepared. Wistar rats were randomly divided into blank control group, model group, Chuanxiling group, and low-, middle-and high-dose CI extract groups. Rat model of CB was established by intratracheal injection with low-dose lipopolysaccharide. After drug intervention, the expression of TNF-α in rat serum and bronchoalveolar lavage fluid was detected by ELISA method. Phagocytic function of the neu-trophil and respiratory burst of the rats were measured by flow cytometry (FCM). Results Compared with the blank con-trol group, the phagocytic function of the neutrophil, respiratory burst of the rats, and the expression of TNF-α in rat serum and bronchoalveolar lavage fluid of the model group were increased significantly. CI extract significantly decreased the abnormal rising of the above indexes. Conclusion Down-Regulation of TNF-α expression, and decrease of the neutrophil phagocytic function and rat respiratory burst may be one of the therapeutic mechanisms of Chrysanthemum In-dicum extract for the treatment of CB.

AIM: To study the therapeutic effects and the related mechanism of Pingchuanling(PCL) on bronchial asthma in rats. METHODS: The rats asthma model was established by ovalbumin(OVA) sensitization.The SD rats were divided into the normal control group,asthma model group,dexamethasone group,low and higher dose of PCL group.Eos in serum and BALF were taken count of and tissue slice dyed with HE were observed.In addition Fas,Bcl-2 expression in lung tissue were examined by immunotisssuchemical technology. 3.Effects of Zhengan Xifeng Decoction on Blood Pressure and Gastrointestinal Motility in Spontaneously Hypertensive Rats

Objective To explore the influencing mechanism of Zhengan Xifeng Decoction on blood pressure and gastrointestinal motility in SHR. Methods After 15 WKY and 90 SHR rats were were randomly divided into normal control group, model group, Benner Pury group, amlodipine group, Zhengan Xifeng Decoction high, middle, and low dose groups. The normal control group and model group were fed with distilled water daily. Rats in treatment group were administered with corresponding drugs daily. Blood pressure, gastric residual and intestinal propulsive ratio of rats was detected after eight-week intervention. Results Compared with the blank control group, systolic pressure, diastolic pressure, and mean arterial pressure were significantly higher than WKY rats of same age (P<0.05). Compared with the model group, the systolic pressure, diastolic pressure, and mean arterial pressure of treatment groups were significantly reduced (P<0.05). Compared with the blank control group, gastric residual rate of rats in the model group significantly decreased (P<0.05). Compared with the model group, the gastric residual rate in Zhengan Xifeng high dose group increased significantly, and intestinal propulsive ratio significantly increased (P<0.05), without statistical significance in the other treatment groups. Conclusion Zhengan Xifeng Decoction can reduce blood pressure in SHR, and regulate gastrointestinal motility.
4.Early orogastric feeding compared with nasoenteric tube enteral nutrition in postoperative pancreaticoduodectomy patients

Objective To study the postoperative nutritional effects of early orogastric feeding compared with nasoenteric tube enteral nutrition within postoperative 24-48 h in pancreaticoduodenectomy (PD) patients.Methods The clinical data of 87 PD patients from June 2013 to January 2015 in Huaxi Hospital was analyzed retrospectively.Patients were divided into orogastric enteral nutrition group and nasoenteric tube enteral nutrition group.Patients' tolerance,hospital stay,mortality,and major complications associated with PD were compared.Results The average time of bowel function recovery (P <0.001) and average hospital stay (P=0.017) in the nasoenteric tube enteral nutrition group were significantly shorter than those in the orogastric enteral nutrition group.The differences of patients' tolerance were not statistically different (P =0.089).The differences of major complications and mortality were not statistically different (P =0.745,P =1.00).Conclusions Nasoenteric tube enteral nutrition in after PD patients improves nitrogen balance,promotes bowel function recovery and makes hospital stay shorter.
5.Research progress of osteoporosis based on Th17/Treg balance

Osteoporosis(OP)is a chronic systemic metabolic bone disease.In recent years,with development of population aging,incidence of OP is also increasing,bringing huge economic burden to family and society.There are many causes of OP,such as aging,oxidative stress,epigenetic and many other aspects,and with further development of research,inflammatory factors and other bone immunology into people's vision.Th17 cells are effector T cell subpopulation that induce development of inflammation and promote bone loss.On the contrary,Treg can maintain their own tolerance and further suppress their own immunity,thus playing a bone protective role.Under physiological conditions,Th17/Treg balance can maintain bone homeostasis,while under pathological condi-tions,disruption of Th17/Treg balance can lead to bone loss.Based on this,this paper will review pathways and cytokines mediating Th17/Treg balance in OP,in order to provide new ideas and methods for treatment of OP.
6.Postnatal feeding with high-fat diet induces obesity and precocious puberty in C57BL/6J mouse pups: a novel model of obesity and puberty

Childhood obesity and obesity-related metabolic complications are induced by a high-fat postnatal diet.The lack of a suitable animal model,however,remains a considerable challenge in obesity studies.In the current study,we provided high-fat diet (HFD) to dams during lactation and to pups after weaning.We also developed a novel model of C57BL/6J mouse pups with HFD-induced postnatal obesity.Results showed that feeding with HFD induces fat deposition and obesity in pups.Furthermore,HFD more potently increased the body weight (BW) of male than female pups.HFD-fed female pups were obese,underwent precocious puberty,and showed increased kisspeptin expression in the hypothalamus.However,parental obesity and precocious puberty exerted no synergistic effects on the HFD-induced postnatal weight gain and puberty onset of the pups.Interestingly,some HFD-fed litters with normal BW also exhibited precocious puberty.This finding suggested that diet composition but not BW triggers puberty onset.Our model suggests good construction validity of obesity and precocious puberty.Furthermore,our model can also be used to explore the mutual interactions between diet-induced postnatal childhood obesity and puberty.
7.Effects of Astragalus polysaccharides on adipogenic differentiation of bone marrow mesenchymal stem cells in low oxygen environment.

To study the effect of Astragalus polysaccharide (APS) on adipogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) cultured in hypoxic environment. The optimal APS concentration, which could promote the proliferation of BMSCs, was screened by methyl thiazolyl tetrazolium method. The concentration was used to intervene in BMSCs-induced by adipogenic differentiation fluid growing in different oxygen concentrations (3%, 6%, 10% and 20%). The formation of lipid droplets in the BMSCs-intervened was observed by oil red O staining under the optical microscope. The mRNA and protein levels of the lipid relating genes peroxisome proliferator activated receptor gamma 2 (PPAR-γ₂) and lipoprotein lipase (LPL) were detected by Real-time PCR and Western blotting, respectively. The results showed that, comparing with the control group, 40 μg/mL APS could significantly promote the proliferation of BMSCs under low oxygen concentration. A large amount of lipid droplets existed in BMSCs growing in the adipogenic inducing fluid containing 40 μg/mL APS and the hypoxic environment, and the protein and mRNA levels of PPAR-γ₂ and LPL also raised. It was worth noting that the phenomenon was more significant in 10% oxygen concentration, and the difference was statistically significant (P<0.05). 40 μg/mL APS had effect on promoting the proliferation and adipogenic differentiation of BMSCs cultured in hypoxic environment, and the effect was related to the concentration of oxygen of BMSCs-cultured.

Osteoporosis can be induced by various factors including prolonged glucocorticoid usage, diminished estrogen levels, secondary hyperparathyroidism, and alterations in the microenvironment of bone tissue. The bone metabolism imbalance(osteogenic-lipogenic imbalance) plays a crucial role in the development of osteoporosis. This imbalance is primarily driven by an increase in the differentiation of bone marrow mesenchymal stem cells into adipocytes and a decrease in their differentiation into osteoblasts, thus forming the core of the osteogenic-lipogenic imbalance observed in osteoporosis. The bone morphogenesis protein(BMP) plays a crucial role in the regulation of the osteogenic-lipid balance in osteoporosis. This regulatory function is accomplished through both the Smad-dependent and Smad-independent pathways. This review centers on the Smad-dependent and Smad-independent pathways facilitated by BMP, offering a comprehensive overview of the potential mechanisms through which BMP-2, 4, 6, 7, and 9 contribute to the regulation of osteogenesis and lipid metabolism in osteoporosis via these pathways. In order to present novel insights for the identification of efficacious targets for clinical anti-osteoporosis medications.
9.Research Progress on the Mechanism of Non-coding RNA Regulation of Bone Reconstruction in Osteoporosis and the Therapeutic Mechanism of Traditional Chinese Medicine for Tonifying Kidney and Strengthening Bone

Non-coding RNAs(ncRNAs) are special RNAs that they don't have protein coding function, but they can affect chromosome structure, gene transcription and participate in the processes of epigenetic modifications. ncRNAs include long non-coding RNAs, microRNA, etc. In recent years, it has been found that these ncRNAs can maintain bone remodeling by adjusting bone resorption and formation in osteoporosis(OP). In the future, it may be a key target of the drug action screening which is clarifying the regulatory mechanism of ncRNAs in the occurrence and development of OP. OP belongs to bone rheumatism category in traditional Chinese medicine, according to the theory of “the kidney generating marrow and dominating bone” in traditional Chinese medicine, kidney tonifying and bone strengthening formulas are used to treat the OP in clinic, and the curative effect is remarkable. It has been found that kidney tonifying and bone strengthening prescriptions can enhance the proliferation of osteoblasts or inhibit the differentiation of osteoclasts by up-regulating or down-regulating the expression of ncRNA, and finally maintain OP bone homeostasis, thus exerting therapeutic effect. However, the specific molecular mechanism is still in its exploratory stage. Therefore, this paper summarized the molecular mechanism of kidney tonifying and bone strengthening prescriptions regulating ncRNAs in the treatment of OP in recent years, in order to provide the new ideas for the screening of the key therapeutic targets of OP drugs and the prevention and treatment of OP with traditional Chinese medicine.
