1.Suppression of ABCE1 Leads to Decreased Cell Proliferation and Increased Apoptosis in 95-D/NCI-H446 Lung Carcinoma Cells

ATP-binding cassette protein E (ABCE1) has been annotated as an Rnase L inhibitor in eukaryotes. Previous study showed that the overexpression of ABCE1 was related with the occurrence and clinical stage of lung adenocarcinoma. As an initial investigation into the novel functions of ABCE1, siRNA-expressing vectors targeting sites of the ABCE1 gene were constructed from RNAi-Ready pSIREN-DNR-DsRed-Express vector. Cultured 95-D and NCI-H446 lung carcinoma cells were transfected with the siRNA-expressing vectors using FuGENE 6 and transfection efficiency was determined by using fluorescence microscopy. The expression level of ABCE1 protein was determined by Western blot and immunofluorescence staining. Cell viability was determined by MTT, cell cycle was analysed by flow cytometry.The apoptotic rate was observed by ELISA. Fluorescence microscopy showed a satisfactory transfection efficiency which was about 42.70%. Cell viability and the growth fraction were markedly suppressed, whereas the apoptosis was significantly increased in SiRNA-95-D and SiRNA-NCI-H446 cells than controls(P< 0.05). It can be concluded that the siRNA targeting ABCE1 gene shows a dramatic inhibitory effect on RNA transcription and protein expression and a promoting effect on the apoptosis in 95-D/NCI-H446 cells, which offers a reliable base for the further in vivo experiment.

5.Analysis of psychological resilience level and its influencing factors inpatients of postpartum pelvic floor dysfunction

Objective To analyze the patients′ psychological resilience level of postpartum pelvic floor dysfunction and to explore the influencing factors, in order to provide the scientific basis for patients′psychological intervention. Methods A cross-sectional survey was conducted on 208 patients of postpartum pelvic floor dysfunction by using the Connor-Davidson Resilience scale(CD-RISC), Positive and Negative Affective scales(PANAS) and Medical Coping Modes questionnaire(MCMQ, which includes "confrontation","acceptance-resignation"and"avoidance"). Results The score of psychological resilience of 208 patients was (53.64 ± 12.29)points, differences in age, family monthly income, living locations, delivery methods and delivery times reached a statistically significant level(F=15.287,5.731,t=-2.571,-15.911,-2.922,all P<0.05). Patients′psychological resilience level had significant positive correlation with positive emotions(P<0.01)and"confrontation",and it had negative correlation with"acceptance-resignation"(P<0.01).Age,living locations, delivery methods, positive emotions and "acceptance- resignation" are the forecast factors of psychological resilience, which can explain 69.6% of total variation. Conclusions Patients of postpartum pelvic floor dysfunction have low levels of psychological resilience, which associated with positive emotions and medical coping modes. Therefore, nursing staffs should pay attentions to the patients′ psychological status, develop individualized psychological guidelines,implement psychological intervention and health education,in order to improve the level of patients′resilience and improve their living quality.
6.Effect of nursing intervention on women with overactive bladder:a systematic review

Objective To systematically evaluate the effectiveness of different nursing interventions for female overactive bladder, and to provide evidence- based evidence for nursing intervention for overactive bladder in women. Methods PubMed, Cochrane Library, Embase, ScienceDirect and CNKI,Wanfang database and CBM comprehensively by computer and included in the literature of nursing intervention for female overactive bladder patients. Results Four randomized controlled trials were included, a total of 426 participants were in, the intervention group was 214 participants, control group was 212 participants, the results of the study showed that pelvic floor muscle training,health education,psychological nursing intervention can improve patients with overactive bladder symptoms and enhance the quality of life. Conclusion Nursing intervention for women with overactive bladder is an effective way to control symptoms such as frequent micturition and urgency,improve bladder overactivity symptoms and enhance quality of life.

9. Establishment of self-management behavior scale for lung cancer patients after radiotherapy

Objective:
To preliminarily explore the distribution of factors in the self-management behavior scale for lung cancer patients after radiotherapy, and analyze the reliability and validity. In order to provide scientific quantitative tools for the evaluation and determination of self-management behavior of patients with lung cancer.
Methods:
Research group made the initial scale through literature methodology, semi-structured interviews method and expert consultation. A total of 189 inpatients were admitted. The reliability and validity of the scale were examined by exploratory factor analysis.
Results:
The self-management behavior scale for lung cancer patients after radiotherapy included 4 dimensions and 25 entries. The coherence value of this scale was 0.78 and the average scale-level content validity index was 0.95. Principal component analysis and maximum variance orthogonal rotation analysis were used to extract 5 common factors, and the cumulative variance contribution rate was 60.162%. Factor 4 and factor 5 were similar in meaning and contained only two entries, so merged the two factors. Criterion related degree was 0.579, the standard total Cronbach α coefficient was 0.828, the split-half reliability was 0.778.
Conclusions
The self-management behavior scale for lung cancer patients after radiotherapy has good reliability and validity, and the scale can be preliminarily applied to the self-management behavior comprehensive evaluation of patients with lung cancer radiotherapy.
