1.Analysis of risk factors of cerebral infarction in young adults and its correlation with TOAST types

Objective To explore the risk factors of youth cerebral infarction and its correlation with clinical TOAST types.Methods 82 young patients with acute cerebral infarction(aged from 18 to 45 years old)were select-ed.The risk factors for youth cerebral infarction patients,and the proportion of TOAST subtype and related risk factors were analyzed.Results Risk factors for youth cerebral infarction were as follows:hyperlipidemia (χ2 =48.703,P <0.05),hypertension (χ2 =40.829,P <0.05),carotid sclerosis (χ2 =46.217,P <0.05),hyperhomocysteinemia (χ2 =40.255,P <0.05),smoking history (χ2 =7.853,P <0.05),diabetes (χ2 =18.256,P <0.05)and family history (χ2 =5.944,P <0.05),heart disease (χ2 =5.754,P <0.05).The proportion of their TOAST subtypes were as following:small artery occlusion lacunar(SAD)(39.0%),large artery atherosclerosis(LAA)(24.4%),stroke of other undetemined etiology(SUE)(17.0%),and acute stroke of other detemined etiology(SOE)(12.2%),cardio-embolism(CE)type(7.3%).Major risk factors for LAA subtype included hyperlipidemia,hypertension and carotid atherosclerosis;Major risk factor for SUE subtype was hyperhomocysteinemia;Major risk factors for SOE included hypertension,diabetes.Major risk factor for CE subtype was heart disease.Conclusion The highest proportion of TOAST types in youth cerebral infarction group is small artery occlusion lacunar.Major risk factors for this group of youth cerebral infarction are as follows:hypertension,carotid atherosclerosis,heart disease,diabetes,hyperhomocys-teinemia,obesity,smoking and family history,and these risk factors should be actively intervened.

Tumorigenesis is a complex process that is regulated by a variety of network signals. With the continuous development of the process, tumor cells gradually exhibit lots of hallmarks.Tumor cells have the characteristics of unlimited proliferation, resistance to apoptosis, evading immune surveillance, among others. As a unique organelles, mitochondria play an important role in cellular energy metabolism, reactive oxygen species producing and apoptosis process. Particularly, mitochondria have a close relationship with tumor development. In this review, we focus on the essential role of mitochondria in tumor cells development.

3.Association study on tryptophan hydroxylase 2 promoter region gene polymorphisms with alcohol dependence

Objective To investigate the association of single nucleotide polymorphisms (SNPs) of loci rs4570625,rs11178997 at promoter region of tryptophan hydroxylase 2 (TPH2) genes with alcohol dependence and depressive symptom in the alcohol-dependent subjects in north Chinese Han people.Methods Genotype and allele frequencies of the TPH2 rs4570625 and rs1 1178997 polymorphisms were examined in 60 alcohol-dependent subjects (patient group) and 62 normal controls (control group) by Polymerase Chain Reaction (PCR) amplification and DNA sequencing techniques.The alcohol-dependent subjects were evaluated by 24 Hamilton Depression Scale (HAMD).Association analysis was carried out between the polymorphism and symptom of depression in the alcohol-dependent subjects.Results There were significantly statistical differences in the rs4570625 alleles (G and T) (x2 =5.280,P<0.05),but no statistically significant differences in the genotypes (G/G,G/T,T/T) (x2 =5.078,P>0.05) between patient group and control group.Significant statistically difference could be found in the rs4570625 alleles (G and T) between the alcohol-dependent subjects who had depressive symptoms and the subjects who did not have depressive symptoms (P<0.05),but no significantly statistical differences could be found in the genotypes frequency (P>0.05).No significant statistically differences in the genotype and allele frequencies of the rs 11178997 were observed between the alcohol-dependent subjects and normal controls and the alcohol-dependent subjects who had depressive symptoms and the subjects who did not have depressive symptoms (all P>0.05).Conclusion The results suggest that the liability of alcohol dependence and depressive symptoms in alcohol-dependent subjects may be associated with the polymorphisms of TPH2 rs4570625 in north Chinese Han population,and the variant of the T allele may contribute to the morbidity.However,the polymorphisms of TPH2 rs1 1178997 may have no association with the susceptibility of alcohol dependence and the depressive symptoms accompanied with alcohol dependence.
4.Effect of Dexamethasone on the Expression of Interleukin?6 and Interleukin?8 in Human Periodontal Ligament Cells

Objective To explore the effect of dexamethasone(Dex)treatment on the expression of interleukin(IL)?6 and IL?8 in human peri?odontal ligament cells(hPDLCs). Methods hPDLCs were subjected to one of the following treatments for 24 h or 48 h:10-9 mol/L Dex;10-6 mol/L Dex;10μg/mL Porphyromonas gingivalis(P. g)?lipopolysaccharide(LPS);10-9 mol/L Dex+10μg/mL P. g?LPS;10-6 mol/L Dex+10μg/mL P. g?LPS;or 0.1%absolute ethyl alcohol(control). Protein and mRNA expression was detected using ELISA and real?time PCR ,respectively. Results At 24 h and at 48 h,IL?6 and IL?8 protein expression in the 10-9 mol/L Dex?and 10-6 mol/L Dex?treated groups was significantly lower than that in the control group(P<0.05). At 48 h,IL?6 mRNA expression in the 10-9 mol/L Dex?and 10-6 mol/L Dex?treated groups was signifi?cantly lower than that in the control group(P<0.05),while IL?8 mRNA expression in the 10-6 mol/L Dex?treated group was significantly higher than that in the control group(P<0.05). At 24 h and at 48 h,IL?6 protein and mRNA expression in the 10-9 mol/L Dex+10μg/mL P. g?LPS?treated group and the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly lower than that in the 10μg/mL P. g?LPS?treated group (P<0.05). At 24 h,IL?8 protein expression in the 10-9 mol/L Dex+10μg/mL P. g?LPS?treated group and the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly lower than that in the 10μg/mL P. g?LPS?treated group(P<0.05),while no such significant difference exist?ed at 48 h. At 48 h,IL?8 mRNA expression in the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly higher than that in the 10μg/mL P. g?LPS?treated group(P<0.05). Conclusion Dex inhibits the innate and P. g?LPS?induced expression of IL?6 in hPDLCs. However, Dex exerts profound effects on IL?8 expression,and treatment with high doses of Dex may promote IL?8 expression over an extended period.
5.Influence on the ovarian reserved function of Gn-RH injection in the bilateral laparoscopic cystectomy

Objective To investigate the influence on the ovarian reserved function of Gn-RH injection in the bilateral laparoscopic cys-tectomy. Methods The clinical data of 100 patients with endometriosis undergoing bilateral laparoscopic cystectomy were included in this study. 50 patients treated by routine bilateral laparoscopic cystectomy were set as control group. 50 patients treated by Gn-RH injection addi-tionally during the surgery were set as study group. The operative situation,serum FSH level,FSH/LH,E2 level,ovarian reserved function, pregnancy situation and recurrent rate were compared between two groups. Results The operative duration and electrocautery times in study group was significantly less than those in control group(P<0. 01). The residual ovarian tissue capsule wall thickness in study group was sig-nificantly more than that in control group(P<0. 01). At different time points,the serum FSH level,FSH/LH and E2 level in study group were significantly less than those in control group(P<0. 01,P<0. 05). The incidence of ovarian reserved function damage in study group was significantly less than those in control group(P<0. 05). The pregnancy rate was significantly higher and the time between surgery and pregnancy was significantly shorter in study group than those in control group(P<0. 05,P<0. 01). The recurrent rate in study group was sig-nificantly less than those in control group(P<0. 01). Conclusion There is a influence on ovarian reserved function of bilateral laparoscopic cystectomy. By inject Gn-RH during surgery,the ovarian reserved function could be protected,which increase the pregnancy rate and reduce recurrence.
6.The determination of perphenazine by a new flow-injection chemiluminescence method

Objective To establish a rapid and precise continuous flow-injection chemiluminescence (CL) method for the determination of perphenazine. Methods In HNO_3 medium, perphenazine could be oxidated by ceriuim (IV) and CL was proportional to the perphenazine concentration without any sensitizers. Thus, a new flow-injection CL method was developed. Results Under the optimized conditions, the proposed method allowed the determination range within 1.0?10~ -7 -7.0?10~ -5 g/mL with the detection limit of 8.0?10~ -8 g/mL. Eleven parallel assays were conducted on perphenazine of 1.0?10~ -6 g/mL, with the relative standard deviation of 1.8%. Conclusion The method is simple, rapid, precise, and sensitive, and has broad linear range; therefore, it has been applied to the the determination of the perphenazine in tablets with satisfactory results.
7.Disfunction of the dendritic cells from the aquaporin 5 knockout mice

Objective To investigate the effect of aquaporin 5(AQP5)gene to the differentiation and development of bone marrow-deriued dendritic cells(BMDC).Methotis The DCs obtained from the bone marrow of the AQP5-/- and the wild type(WT)mice were cultured and induced to maturation by LPS.The phenotype of the two types DCs were assayed by FACS,and the phagocytosis ability Was assayed by co-incubating with FITC-conjucted ovalbumin.Results Contrast to the DCs from the WT mice,the DCs from the A9P5-/-mice showed reducing positive rate of expression of the co-stimulating molecular such as CD40,CD80.CD86.And the phagoeytosis ability of DCs from the AOP5-/-mice was lower than that of the WT mice.The relationship of the phagncytosis rate to the time from the AOP5-/- mice was different from that of the WT mice.The stimulating ability of the AOP5-/-DCs was lower than that of the WT mice.Conclusion Water transmembrane moving mediated by AQP5 iS very important to the normal function of DCs.The certain mechanism of the signal moleeular is to be determined. 9.Prognosis and risk factors of neonatal cerebral infarction

Objective To determine the prognosis and risk factors of neonatal cerebral infarction.Methods From January 2002 to December 2010,44 newborn infants were diagnosed with cerebral infarction by imaging examinations at Peking University First Hospital.The neurodevelopmental outcomes of these newborn infants were followed up and evaluated by clinical manifestations,Gesell development scale,cranial imaging,electroencephalogram and auditory evoked potential.Factors related to prognosis were analyzed with single and multi-factor Logistic regression analysis.Results Thirty-eight (86%) cases were followed up,and of these cases,five children died and the results of three were inconclusive due to small age (less than 6 months old).Among the remaining 30 children,neurodevelopmental outcome was normal in 15 cases and abnormal in the remaining 15 cases,thus,the incidence of sequelae was 50% (15/30) and the mortality rate was 13% (5/38).Of the 15 abnormal cases,all had cerebral palsy and movement retardation,eight cases had cognitive impairment,eight cases had epilepsy and five had visual impairment.The incidence of large cerebral infarction (more than one lobe) was 14/15,worse cranial imaging outcome (one month after treatment,cerebral infarction lesion still present or had expanded)was 13/15,and severe complications was 8/15 in the newborns with sequelae,which were higher than in those without sequelae (4/15,5/15 and 1/15,respectively) (x2=13.889,8.889 and 7.778,all P<0.05).Logistic regression analysis showed that large cerebral infarction was a risk factor for sequelae (OR=38.500,95%C1:3.749-395.407,P=0.002),however,worse cranial imaging outcome (OR=8.563,95%CI:0.909-80.683,P=0.061) and severe complications (OR=18.024,95%CI:0.516-630.163,P=0.111) were not risk factors for sequelae.Cerebral infarction with middle cerebral artery injury had a high risk of movement retardation (OR=6.000,95%CI:1.172-3.725,P=0.025),and those with a large cerebral infarction were more likely to have epilepsy (x2=7.273,P=0.010).The incidence of large cerebral infarction in the newborn infants with cognitive impairment was 8/8,which was much higher than in those without cognitive impairment (46%,10/22),thus,infarct area may be related to cognitive ability (x2=7.273,P=0.010).Conclusions Neonatal cerebral infarction might result in many types of sequelae,with motor impairment being the most common form.A large cerebral infarction is more likely to result in abnormal neurodevelopmental outcome.
10.DETERMINATION OF TETRACYCLINE AND OXYTETRACYCLINE BY FLOW-INJECTION CHEMILUMINESCENCE METHOD

Objective To establish a rapid and precise continuous flow-injection chemiluminescence method for the determination of tetracycline and oxytetracycline. Methods In NaOH solution, tetracycline and oxytetracycline can sensitize obviously the chemiluminesence (CL) intensity of the reaction of luminol with KIO4, the sensitized CL intensity is proportional to the concentration of tetracycline and oxytetracycline. So, a new flow-injection CL method has been developed. The optimum chemical conditions for the CL reaction were investigated. Results Under the optimized conditions (KIO4 concentration: 1.0×10-5 mol/L; NaOH concentration: 0.1mol/L; luminol concentration: 1.0×10-4mol/L), tetracycline and oxytetracycline were determined. The linear range of the working curves was 1.0×10-7 -1.0×10-4g/mL, the detection limits was 1.0×10-8g/mL and 1.1×10-8g/mL, and the relative standard deviation was 2.6% (CS=1.0×10-6g/mL; n=11) and 2.0% (CS=1.0×10-6g/mL; n=11) respectively. Conclusion The method is simple, rapid, and sensitive, and it has been successfully applied to the the determination of tetracycline and oxytetracycline tablets, the mean recoveries being 99.7% and 98.8% respectively.
