1.Expression of Human Papillomavirus in Mammary Carcinoma and its Possible Mechanism in Carcinogenesis

To explore the role of Human papillomavirus (HPV) in mammary carcinogenesis, the expression of the HPV-16, iNOS, P53 and hTERT proteins in breast carcinomas and their relationships were investigated. 52 samples of breast cancer and 16 samples of benign breast tumors were assayed using the immunohistochemical SP method for detection of protein expression levels. The expression of HPV-16, iNOS, P53 and hTERT proteins in a mammary carcinoma was 44.2%, 57.7%, 63.5% and 59.6% respectively, which was significantly greater than the corresponding levels in the benign group. The expression of iNOS, P53 and hTERT was correlated with the presence of an HPV-16 infection in a mammary carcinoma (P<0.05). The connection between these events might also involve the iNOS, mutated type P53 and the hTERT protein.
2.Association study on tryptophan hydroxylase 2 promoter region gene polymorphisms with alcohol dependence

Objective To investigate the association of single nucleotide polymorphisms (SNPs) of loci rs4570625,rs11178997 at promoter region of tryptophan hydroxylase 2 (TPH2) genes with alcohol dependence and depressive symptom in the alcohol-dependent subjects in north Chinese Han people.Methods Genotype and allele frequencies of the TPH2 rs4570625 and rs1 1178997 polymorphisms were examined in 60 alcohol-dependent subjects (patient group) and 62 normal controls (control group) by Polymerase Chain Reaction (PCR) amplification and DNA sequencing techniques.The alcohol-dependent subjects were evaluated by 24 Hamilton Depression Scale (HAMD).Association analysis was carried out between the polymorphism and symptom of depression in the alcohol-dependent subjects.Results There were significantly statistical differences in the rs4570625 alleles (G and T) (x2 =5.280,P<0.05),but no statistically significant differences in the genotypes (G/G,G/T,T/T) (x2 =5.078,P>0.05) between patient group and control group.Significant statistically difference could be found in the rs4570625 alleles (G and T) between the alcohol-dependent subjects who had depressive symptoms and the subjects who did not have depressive symptoms (P<0.05),but no significantly statistical differences could be found in the genotypes frequency (P>0.05).No significant statistically differences in the genotype and allele frequencies of the rs 11178997 were observed between the alcohol-dependent subjects and normal controls and the alcohol-dependent subjects who had depressive symptoms and the subjects who did not have depressive symptoms (all P>0.05).Conclusion The results suggest that the liability of alcohol dependence and depressive symptoms in alcohol-dependent subjects may be associated with the polymorphisms of TPH2 rs4570625 in north Chinese Han population,and the variant of the T allele may contribute to the morbidity.However,the polymorphisms of TPH2 rs1 1178997 may have no association with the susceptibility of alcohol dependence and the depressive symptoms accompanied with alcohol dependence.
3.Relationship among mTOR levels,regulatory T cells and cytokins in patients with acute coronary syndrome

Objective To assess whether mTOR was activated in peripheral T lymphocytes of acute coronary syndrome (ACS)patients and to investigate the relationship among mTOR,regulatory T cells and cytokins. Methods Patients with acute coronary syndrome (n=32) were selected in ACS group. Meanwhile, patients who complainted of chest pain but were proven to be normal in ECG and in coronary arteriography, were excluded as ACS patients but were diagnosed as Chest Pain Syndrome (CPS) and selected in CPS group (n=28). The expression levels of mTOR, phospho-p70S6KT389 (indicative of mTORC1 activity) and phospho-AKTS473(indicative of mTORC2 activity) were investigated in T cells, which were isolated from peripheral blood of patients in ACS group or CPS group, using Western blot. The proportion of CD4+CD25+Foxp3+Tregs over CD4+cells were evaluated by FACS. And T cell subset related cytokines such as IFN-γ, IL-4, IL-17 and TGF-β1 were exam?ined by ELISA. Then we investigated the relationship among mTOR,regulatory T cells and cytokins by Spearman analysis. Results mTOR and p-p70S6KT389 expression were significantly enhanced in ACS group as compared with those in patients from CPS group (P<0.01). Higher levels of IFN-γ, IL-17 but lower level of TGF-β1 cytokines as well as decreased propor?tion of Tregs were observed in ACS group than those in CPS group (P<0.05). There was no significant difference in the level of IL-4 and p-AKTS473 between the two groups (P>0.05). By correlation analysis, p-p70S6KT389 expression level positively correlated with IFN-γ, IL-17(rs=0.91,092,P<0.01)and negatively correlated with Tregs and TGF-β1(rs=-0.85,-0.80, re?spectively, P<0.01). Conclusion mTORC1 pathway was activated in peripheral T lymphocytes of ACS patients, and Tregs insufficiency and cytokins imbalance both contribute to the activation of mTORC1 pathway and pathological process of ACS.

5.Study on the activation of mTOR/p70S6K signaling pathway in peripheral T lymphocytes of the CHD patients

Objective To investigate whether mTOR/p70S6K signal was activated in peripheral T lymphocytes of the coronary heart disease (CHD) patients and to investigate its possible role in the pathogenesis of coronary atherosclerosis. Methods The mRNA expressions of mTOR andp70S6K were detected by RT-PCR and protein expressions of mTOR, p70S6K and phosphorylated-p70S6K (p-p70S6K) were detected by Western blot assay in T cells isolated from peripheral blood of the CHD patients (including AMI, UAP, SAP) and the CPS patients. Results Levels of mTOR and p-p70S6K were significantly enhanced in the AMI and UAP groups as compared with those in the SAP and CPS groups (P < 0.05, respectively). The mRNA expressions of mTOR and p70S6K were significantly increased in the AMI and UAP groups as compared with those in the SAP and CPS groups (P < 0.05). Conclusion The mTOR/p70S6K pathway is activated in the peripheral T lymphocytes of CHD patients with AMI and UAP , which may participate in the pathological process of coronary atherosclerosis.
6.Evaluation of people's acceptable range for posed smile characteristic index

Objective To obtain the acceptable range for posed smile characteristic index,in order to supply diagnostic and therapeutic basis for orthodontic treatment.Methods The 200 subjects included in the study consisted of 100 experienced orthodontists and 100 laypersons.Both two frontal posed smile photographs of man and woman were changed by several smile characteristic indexes,including the amount of incisor exposure,amount of gingival display,smile arc,buccal corridor fill,horizontal inclination of maxillary occlusal plate and distance from lower lip to maxillary incisor.All the subjects were desired to evaluate each images according to their own aesthetic standard.Results Each acceptable range for the amount of incisor exposure,amount of gingival display,smile arc,buccal corridor fill,horizontal inclination of maxillary occlusal plate and distance from lower lip to maxillary incisor,was 75 %-100 % (male and female),0-2 mm (male) and 0-3 mm (female) ; 50 %-100 %(male and female) ; 0 %-15 % (male) and 0 %-20 % (female) ; 0-6° (male and female) ; 0 mm (male and female)(P<0.05),respectively.And there was perception difference between the orthodontists and the laypersons on smile evaluation (P<0.05).Conclusions Posed smile analysis should be an im portant aspect of orthodontic diagnosis and treatment planning.Orthodontists should not disturb con sonant smiles but create them with proper bracket positioning.
7.Expression of novel apoptosis-related protein PDCD5 in granulosa cells of polysystic ovary syndrome

Objective: To detect the novel apoptosis-related protein PDCD5 expression in granulosa cells of polysystic ovary syndrome(PCOS) and normal ovary, and explore the pathogenesis of PCOS. Objective:To study the difference in anti-hepatic fiborsis components between pre-and post-processed Trionyx Sinensis Carapace to guide the clinical application of Trionyx Sinensis Carapace. Methods:SDS-PAGE gel electrophoresis was used to compare the constituents in pre-and post-processed Trionyx Sinensis Carapace, and the inhibitory effect on the proliferation of HSC-T6 was deter-mined by MTT. Results:The processed Trionyx Sinensis Carapace had much more components than the crude Trionyx Sinensis Cara-pace,and the relative molecular mass in the first-level band decreased resulting in the generation of micromolecular polypeptides. Both pre-and post-processed Trionyx Sinensis Carapace had anti-hepatic fibrosis, while the vinegar-processed Trionyx Sinensis was more ef-fective. Conclusion:The difference in the active components in pre- and post-processed Trionyx Sinensis Carapace is obvious, which provides foundation for the clinical application and further researches.
9.A survey on topical medication compliance in patients with glaucoma

A survey on compliance with topical medication and the relevant factors were conducted in 190 patients with glaucoma in Miaohang Township, Baoshan County of Shanghai. All patients used 1 or more topical ocular hypotensive medications for at least 6 months and a special questionnaire was designed for the survey. The survey revealed that the overall non-compliance rate was 54. 7% (104/190) in this group of patients; which was closely correlated with age, medication application time and the extent of visual defect ( OR = 2. 550, 0. 225 and 0. 342, P < 0. 05 ). Education levels, gender of patients, glaucoma type, number of daily medication, medication types and the stability of diseases were not correlated with the compliance.

Staphylococcus aureus is a major cause of hospital-acquired infection. Because the bacteria are very easy to become resistant to antibiotics, vaccination is a main method against S. aureus infection. Clumping factor B (ClfB) is an adhesion molecule essential for S. aureus to colonize in the host mucosa and is regarded as an important target antigen. In this study, we successfully used Escherichia coli to express a segment encoding the N1-N3 regions of ClfB protein (Truncated-ClfB) cloned from S. aureus. The protein was purified by affinity and ion exchange chromatographies and gel filtration. Rabbits were immunized three times with purified Truncated-ClfB. After that, blood was collected to prepare serum which were then used for measurement of antibody level. Phagocytosis of S. aureus opsonized by the serum was determined by a flow cytometry. Results show that the serum IgG titer reached 1:640 000. Phagocytosed S. aureus by polymorphonuclear leukocytes were significantly more when the bacteria were opsonized by the serum from Truncated-ClfB immunized rabbits than those from no immunized group (P < 0.01). Therefore, the results indicated that Truncated-ClfB could be a promising vaccine candidate against S. aureus infection.
