Objective To study the role of human leukocyte antigen-G (HLA-G) in the carcinogenesis and development of choriocarcinoma. Methods We designed and synthesized a double strand small interference RNA (siRNA) of HLA-G, then transfected it into a HLA-G overexpressed choriocarcinoma cell line, JEG-3 by lipofectine; HLA-G mRNA level was detected by real time RT-PCR; HLA-G protein level was detected by western blot. The living cells of JEG-3 were counted under microscope after transfection by siRNA. Results Double strand siRNA of HLA-G effectively downregulated the mRNA level and the protein level of HLA-G. The mRNA levels by Ct value at different concentration (1.0, 2.5, 5.0 ?g/L) were 20.67?0.02, 21.37?0.03,21.43?0.02, respectively. Objective Study on the best drug compatibility for the double-lumen endobronchial intubation anesthesia. Methods Eighty ASA Ⅰ - Ⅱ patients undergoing selective thoracis surgery requiring intubation with double-lumen tubes were randomly divided into A and B group, with 40 cases in each group.The systolic blood pressure (SBP), diastolic blood pressure (DBP), heart rate (HR), were recorded before induction (T0) , after drug injection (T1), during intubation (T2), and at 1 min (T3), 3 min (T4)and 5 min (T5) after intubation. Objective To explore the effects of dexmedetomidine on recovery quality of hypertension patients under general anesthesia,so as to provide reference for its clinical application.Methods 31 patients who underwent abdominal surgery were chosen as study subjects.They were randomly divided into observation group(15 patients) and control group(16 patients) according to the digital table.At the time of 15 min before the end of surgery,the patients in the observation group were injected with dexmedetomidine of 0.5g/kg,and the patients in the control group were injected with the same volume of normal saline instead in 15 min.At the time of before anesthesia(T0),before injection(T1),3 min after injection (T2),just before extubation(T3),1 min(T4),5 min(T5),15 min(T6) after extubation,the hemodynamic parameters,the sedation scores at the time of T5 and T6,and the occurrence of cough during extubation were recorded and compared between the two groups.Results During operation and extubation,the levels of SBP,DBP,HR and their fluctuations in the observation group were lower than those in the control group.At the time of T5 and T6,the sedation scores in the observation were lower than those in the control group.The mean rank and scores of the two groups [at the time of T5 were [(12.2±24.6) and (19.5±24.6),respectively,Z=2.27,P=0.022].The mean rank and scores of the two groups at the time of T6 were T6[(12.3±24.7) and (19.5±24.7),respectively,Z=2.25,P=0.024].During the time of extubation,the occurrence of cough in the observation group was lower than that in the control group.The mean rank of the cough score in the observation group and the control group was[(12.2±24.1) and (19.5±24.1),Z=2.33,P=0.020].Conclusion Small dose of dexmedetomidine can reduce the incidences of intraoperative cerebral vascular accident risk,improve the quality of general anesthesia recovery in hypertension patients,and thus should be recommend in clinical application.

Objective To investigate the different expressions of various subtypes of human leukocyte antigen-G(HLA-G)mRNA in placentas among early and late-onset severe preeclamptic and normal pregnant women. Methods Eleven placental samples from early-onset severe preeclamptic pregnancies and fourteen from late-onset severe preeclamptic pregnancies were collected as study group;eight placental samples from normal pregnancies were collected as control group.The levels of HLA-G mRNA in the three groups were measured by nested RT-PCR. Results Compared with the counterparts in the control group,the leveI of HLA-G1 subtype was significantly decreased (median:Early-onset group:1.37,P<0.05;Late-onset group:24.90,P<0.05;Control group:46.67,P<0.05)and the level of HLA-G5 subtype increased(median:Early-onset group 19.23;Late-onset group:3.65;Control group:1.33)in both early and late-onset severe preeclamptic group.The level of HLA-G2 subtype increased(median:Early-onset group:0;Late-onset group:21.59,P<0.05;Control group:5.39)in the late-onset severe preeclamptic group. Conclusion The reduced expression of HLA-G membrane-bound subtype mRNA in the placenta and the increased expression of HLA-G soluble subtype mRNA in the placenta may be related tO the pathogenesis of preeclampsia.

Objective To explore the risk factors of youth cerebral infarction and its correlation with clinical TOAST types.Methods 82 young patients with acute cerebral infarction(aged from 18 to 45 years old)were select-ed.The risk factors for youth cerebral infarction patients,and the proportion of TOAST subtype and related risk factors were analyzed.Results Risk factors for youth cerebral infarction were as follows:hyperlipidemia (χ2 =48.703,P <0.05),hypertension (χ2 =40.829,P <0.05),carotid sclerosis (χ2 =46.217,P <0.05),hyperhomocysteinemia (χ2 =40.255,P <0.05),smoking history (χ2 =7.853,P <0.05),diabetes (χ2 =18.256,P <0.05)and family history (χ2 =5.944,P <0.05),heart disease (χ2 =5.754,P <0.05).The proportion of their TOAST subtypes were as following:small artery occlusion lacunar(SAD)(39.0%),large artery atherosclerosis(LAA)(24.4%),stroke of other undetemined etiology(SUE)(17.0%),and acute stroke of other detemined etiology(SOE)(12.2%),cardio-embolism(CE)type(7.3%).Major risk factors for LAA subtype included hyperlipidemia,hypertension and carotid atherosclerosis;Major risk factor for SUE subtype was hyperhomocysteinemia;Major risk factors for SOE included hypertension,diabetes.Major risk factor for CE subtype was heart disease.Conclusion The highest proportion of TOAST types in youth cerebral infarction group is small artery occlusion lacunar.Major risk factors for this group of youth cerebral infarction are as follows:hypertension,carotid atherosclerosis,heart disease,diabetes,hyperhomocys-teinemia,obesity,smoking and family history,and these risk factors should be actively intervened.

Objective To explore the impact of family history of hypertension and diabetes mellitus on the phenotype of insulin resistance and the ?-cell function in normoglycemic subjects. Methods 275 normoglycemic subjects were divided into four groups according to family history of hypertension and diabetes,namely group combined with family history of hypertension and diabetes (H+D+group),group without family history of hypertension and diabetes (H-D-group),group without family history of hypertension but with diabetes (H-D+group),group with family history of hypertension but without diabetes (H+D-group). Objective:To explore the utility of psychiatric service of clients having online counseling for anxiety or depression. Methods:The anxiety group was consisted of clients (n=107) who were positive in the Self-rating Anxiety Scale (SAS) online, similarly the depression group was consisted of clients (n=102) with positive results in Self-rating Depression Scale (SDS) online screening. Both groups were investigated with the questionnaires include issues related to the clinical features, diagnostic categories and treatment vs professional advice on clients. Results:In the anxiety group (male 69/female 38), the average age was 28+8, 88(82.2%) with more than 15 years education. In the depression group (male 57 vs. female 45), the average age was 29+8, 81(79.4%) with more than 15 years education. None was illiterate. For the age, sex, education and occupation, there was no significant difference between the two groups. 81(79.4%) clients in the anxiety group and 89(87.2%) clients in the depression group were advised to visit psychiatrists or psychologists. A few were advised to counseling online only and there was no significant difference between the two groups. The main diagnoses were mood disorders, neurosis and psychogenic disorders. Actually, 29(27.1%) in the anxiety group were treated with anxiolitics, 31(30.2%) in the depression group were treated with antidepressants at local clinics. Conclusion:Most of the clients should visit psychiatrists or psychologists, and a few may have their psychological counseling online only. Health education and psychological counseling online may be helpful to improve the rate of diagnosis and treatment of mental disorders in local clinics.

In total, 4006 women aged 25 -65 years at Qiaozi Town, Huairou District, Beijing were screened for cervical cancer during April to June, 2009. Results showed that 54. 52% of the women screened suffered from varied gynecological illness, highest in those aged 30 - 34 years ( 80. 90% ) and lowest in those aged 50 -54 years. The top three gynecological morbidity among them were cervicitis (34. 77% ), virginities ( 12. 88% ) and uterine leiomyoma (8. 34% ). Precancerous lesions in 12 women,endometrial cancer in one woman and cervical cancer in one woman were detected by cervical cytological examinations.

Objective To explore the effect of dexamethasone(Dex)treatment on the expression of interleukin(IL)?6 and IL?8 in human peri?odontal ligament cells(hPDLCs). Methods hPDLCs were subjected to one of the following treatments for 24 h or 48 h:10-9 mol/L Dex;10-6 mol/L Dex;10μg/mL Porphyromonas gingivalis(P. g)?lipopolysaccharide(LPS);10-9 mol/L Dex+10μg/mL P. g?LPS;10-6 mol/L Dex+10μg/mL P. g?LPS;or 0.1%absolute ethyl alcohol(control). Protein and mRNA expression was detected using ELISA and real?time PCR ,respectively. Results At 24 h and at 48 h,IL?6 and IL?8 protein expression in the 10-9 mol/L Dex?and 10-6 mol/L Dex?treated groups was significantly lower than that in the control group(P<0.05). At 48 h,IL?6 mRNA expression in the 10-9 mol/L Dex?and 10-6 mol/L Dex?treated groups was signifi?cantly lower than that in the control group(P<0.05),while IL?8 mRNA expression in the 10-6 mol/L Dex?treated group was significantly higher than that in the control group(P<0.05). At 24 h and at 48 h,IL?6 protein and mRNA expression in the 10-9 mol/L Dex+10μg/mL P. g?LPS?treated group and the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly lower than that in the 10μg/mL P. g?LPS?treated group (P<0.05). At 24 h,IL?8 protein expression in the 10-9 mol/L Dex+10μg/mL P. g?LPS?treated group and the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly lower than that in the 10μg/mL P. g?LPS?treated group(P<0.05),while no such significant difference exist?ed at 48 h. At 48 h,IL?8 mRNA expression in the 10-6 mol/L Dex+10μg/mL P. g?LPS?treated group was significantly higher than that in the 10μg/mL P. g?LPS?treated group(P<0.05). Conclusion Dex inhibits the innate and P. g?LPS?induced expression of IL?6 in hPDLCs. However, Dex exerts profound effects on IL?8 expression,and treatment with high doses of Dex may promote IL?8 expression over an extended period.

OBJECTIVE:To investigate the effect of salesclerk factors on the consumers'purchasing behavior in drugstores in Beijing citizens,and provide reference for better satisfaction of consumers'requirements. METHODS:Clustering random sampling method was launched among the drugstore consumers in 16 districts of Beijing,the results were statistically analyzed,and then the suggestions were provided. RESULTS:Totally 180 valid questionnaires were collected from the 208 questionnaires with effective re-covery rate of 86.5%. 70.6% consumers preferred the salesclerk to ask actively when they entered a drugstore;moderate depen-dence on the salesclerk was found mainly in the aspect ofmedicine selection,medication methods guidingandhelping disease judgment;moderate trust to the salesclerk was found,andthe poor professional knowledge,poor service attitudeandpoor professional ethicswere the main reasons for the distrust;the professional moralandknowledgeof salesclerks were the most-ly expected qualities;the licensed pharmacist in the drugstore was not recognized by 66.1% consumers. CONCLUSIONS:Drug-store should understand the consumer's expectations and requirements to its salesclerk and improve accordingly in the respect of strengthening the train of professional quality,enhancing the ability of pharmaceutical care,increasing the propaganda of licensed pharmacist,conducting medication consultation actively,exploring the diversity of consumer's requirements,and meeting the con-sumer's requirements more extensively.
