Objective:To observe changes of cerebral nerve cell apoptosis of the rat after acute stroke and the preventive and treating effect of Jiannao Decoction on the apoptosis.Methods:SHRsp rats were used to establish the model of acute stroke,which were treated with Jiannao Decoction.Symptoms and signs of the nervous system and changes of pathological structures of the hipocampus were investigated before and after establishment of the model,and changes of cerebral nerve cell apoptosis before and after establishment of the model and medication were investigated with TUNEL,immunohistochamical and Western blot methods. Results:The incidence rate of stroke in the prevention group was significantly lower than that of the model group(P0.05).Conclusion:Jiannao Decoction has significant protective action on brain in the rat with stroke,which is related with inhibiting the effect of nerve cell apoptosis and conduction of apoptosis-related signals.

Objective To investigate the effect of human hepatocellular carcinoma HepG2 cell-derived Exosome on the differentiation of mesenchymal stem cells(MSC)into cancer-associated myofibroblasts(CAF)and the impacts of CAF on liver cancer cell proliferation,migration,and invasion. Methods The protein expression of HepG2 cell-derived Exosome was detected by Western blotting. MSCs were separated from human adipose tissue and cultured with HepG2 cell-derived Exosome(100 ng/nl)to initiate differentiation. The expressions of mesenchymal markers and several interleukins were also detected by Western blotting. HepG2 cells were co-cultured with the conditioned media(CM),in which HepG2 Exosome induced the differentiation of MSC into CAF. The expressions of epithelial and mesenchymal markers were detected by real-time polymerase chain reaction(PCR)and Western blotting. Cell proliferation was assessed using MTS assay. Transwell chambers were used in the in vitro migration and invasion assay. Results HepG2 cell-derived particles expressed CD63,70 kilodalton heat shock proteins,and 90 kilodalton heat shock proteins. With the treatment of HepG2 cell-derived Exosome,the expressions of mesenchymal marker α-smooth muscle actin,fibroblast activation protein α,interleukin(IL)-6,IL-8,and IL-1β were up-regulated,while vascular endothelial growth factor had no significant change. The conditioned media which HepG2 Exosome induced MSC differentiation CAF(CAF-CM)could significantly promote HepG2 cells proliferation(1.075±0.104),compared to BSA control(0.874±0.066,P=0.023)and MSC-CM(0.649±0.034,P=0.0005). CAF-CM could significantly enhance cell migration [(42.5±9.1) cells vs.(18.5±3.1) cells,P=0.001] and invasion [(29.0±3.5) cells vs.(13.1±3.7) cells,P=0.009] compared to its control group. Moreover the conditioned medium which HepG2 Exosome induced MSC to differentiate into CAF could also promote the expressions of mesenchyme-related genes Smad interacting protein 1(P=0.040),β-catenin(P=0.038),fibronectin(P=0.029),and Vimentin(P=0.013)and inhibit the expression of epithelial related genes zonula ocdudens-1(P=0.010).Conclusions Exosome extracted from HepG2 cells can induce human adipose-derived MSC to differentiate into cancer-associated myofibroblasts. CAF-like cells can promote the migration of the liver cancer cell line HepG2.

Objective To investigate the effect of aquaporin 5(AQP5)gene to the differentiation and development of bone marrow-deriued dendritic cells(BMDC).Methotis The DCs obtained from the bone marrow of the AQP5-/- and the wild type(WT)mice were cultured and induced to maturation by LPS.The phenotype of the two types DCs were assayed by FACS,and the phagocytosis ability Was assayed by co-incubating with FITC-conjucted ovalbumin.Results Contrast to the DCs from the WT mice,the DCs from the A9P5-/-mice showed reducing positive rate of expression of the co-stimulating molecular such as CD40,CD80.CD86.And the phagoeytosis ability of DCs from the AOP5-/-mice was lower than that of the WT mice.The relationship of the phagncytosis rate to the time from the AOP5-/- mice was different from that of the WT mice.The stimulating ability of the AOP5-/-DCs was lower than that of the WT mice.Conclusion Water transmembrane moving mediated by AQP5 iS very important to the normal function of DCs.The certain mechanism of the signal moleeular is to be determined. Objective To establish the resist-fatigue animal test through using Maca, in order to provide reference for the relevant departments to exploit new sports nutrition . Methods Thirty male KM mice were randomly divided into Quiet control group (A),Exhaustive group (B,high altitude),Maca exhaustive group (C), there were 10 mice in each group. Drinking water containing Maca (Maca =500 mg/L) was supplied in group C. Additional exhaustive group (D,low altitude) was set.Thirty days later, all mice were killed after one exhaustive test, then we tested the exhaustive time, the level of T-SOD and liver glycogen. Results The exhaustive time of group C was significantly longer than group B (P<0.05), the T-SOD level of group C was significantly higher than group A and group B (P<0.05), and the liver glycogen content of group C was significantly higher than group B (P<0.05) . Conclusion Maca can raise T-SOD and liver glycogen level of the mice after exhaustive swimming, and has function of resist-fatigue.

Objective To explore the effects and indications of physical cooling in fever patients after bone fracture surgery.Methods 450 fever patients after bone fracture surgery were selected.They were divided into different groups according to ~e main influencing factors which affected the effect of physical cooling,including the temperature of sponge bathing,the degree of fever,clinical phases of fever.The influence of above factors on physical cooling was observed.Results The temperature for sponge bathing maintaining at 38～40 ℃(the experimental group) achieved better results than that at 32～34 ℃(the control group).Physical cooling at constant peak period of fever had better effect than that during the fervescence period,and the effect of sponge bathing treatment was better in mild fever patients than in high fever patients.Conclusions The single physical hypothermia only fit the patients following bone fracture surgery who had a fever below 38.5 ℃.Patients who had an infectious high fever should receive drug combination.Temperature of sponge bathing maintaining at 38～40 ℃,bathing timing at constant peak period of fever may get best cooling effect.
