1.Newborn screening of inherited metabolic diseases by tandem mass spectrometry

Application of TMS technology in newborn screening has resulted in major expansion of disorder panel for metabolic diseases in recent years. This automated, multiplex testing methodology detects multiple analytes from single analysis of one blood spot, which leads to detection of 30-35 disorders of amino acids, organic acids, and fatty acids metabolism. The early identification of persons affected with inborn errors of metabolism has led to unexpected discoveries related to the natural history of the disorder or options for therapy. This article summarized (1) the basic principles of this technology and methodology. (2) Current status of application of this methodology in the United States, European countries and in China. (3) The positive impacts on the public health and advances in medical genetics. Finally (4) Challenges, issues and possible solutions. The purpose of this article aimed at introducing new technology and exploring the possibilities of implementing into developing countries where medical genetics is not developed and foreseeing the possible problems and obstacles.
2.Study on Insulin Resistance of Preventing and Treating Experimental Diabetic Rats by Powder of Detoxicating and Activating the Collaterals and Regulating Livers

Objective To research the effects of powder of detoxicating and activating the collaterals and regulating livers on diabetic rats models caused by high fat feed and streptozotocin, to provide scientific basis for clinical dosage. Methods Wistar rats were feeded by high fat feed for one month and forbidden to eat for twelve hours, then injected through abdomen with 30 mg/kg of 1.2% streptozotocin. Blank group was injected with the same amount of lemon buffer solution. One week later, sugar tolenrance test was conducted. Rats affected by diabetics were divided into model group, dimethyldiguanide group, Pyrrole group and group of detoxicating and activating the collaterals and regulating livers. Treatment group was perfused into stomach with different medicine by the dosage of 0.5, 5, 0.5 g/(kg?d) respectively. Blank group and model group were perfused with the same amount of physiological saline. Two months after the dosage, indexes such as saccharogenic hemoglobian and blood lipid were detected, and the sensitive indexes of insulin was calculated. Seventeen physiological,biochemicaland morphological indexes consistently showed that Astragalus polysaccharide was able to enhance the myocardia contractility,attenuate the area of myocardial infarction, and reduce the degree of myocardial damage of myocardially infarcted dog heart in vivo.The mechanism might be related to the inhibition of Na+-Kr+ATPase activity & the anti-free-radical-damage action.
4.Exploration and Practice of Industry-university-institute Unification Mode for Pharmacy Professionals

OBJECTIVE:To explore the new mode of industry-university-institute unification for pharmacy professionals. METHODS:The research-teaching interactive platform was established through the principles of“complementary advantages, shared interests and mutual benefits”. RESULTS:It changed the traditional teaching mode,and it ensured the professional plan-ning,curriculum development and training objectives of students based on“4 analysis,2 setting and 1 implementation”. CONCLU-SIONS:The opportunities for cooperation between students and employers were broadened and students’awareness of innovation and entrepreneurship was enhanced. It cultivates project-based pharmaceutical,technical and skilled personnel,solves the problem of industry-university-institute unification and has certain significance for the development of China’s pharmaceutical education.
5.Research progress on biochemical risk factors of stroke in young and middle-aged patients

The incidence of stroke is increasing year by year,about 75% of the patients with different levels of sequelae,and the age of onset also tends to be younger,to the community and the family to bring a heavy burden.Therefore,it is urgent to strengthen the study of the related risk factors of stroke in young people,so as to reduce the incidence of stroke.Traditional recognized risk factors such as hypertension,diabetes,high cholesterol,heart disease,obesity,and smoking can explain part of the stroke events,of these risk factors intervention treatment significantly reduced the incidence and mortality of stroke.However,cardio cerebral vascular system is not fully protected.In view of the biochemical indexes of blood of patients with specimens by convenience,detection index simple and precise,further studies of the relationship between biochemical risk factors and young stroke has made clear the necessity of and have become major challenges in current epidemiological study of cerebral stroke.
6.Relationship of hypothyroxinemia to gestational diabetes mellitus during early pregnancy

Objective To investigate the relationship of hypothyroxinemia to gestational diabetes mellitus during early pregnancy. Methods A total of 11 365 cases of women with early singleton pregnancies were collected from Chongqing Health Center for Women and Children. The screening of thyroid function was performed. The postload glucose concentrations and the risks of gestational diabetes mellitus in hypothyroxinemia were investigated. The relationship of thyroid hormones to postload glucose concentration was evaluated. The association between thyroid peroxidase antibodies(TPOAb) and gestational diabetes mellitus was analyzed. Results Early pregnancy women with hypothyroxinemia had a higher postload glucose concentration. Comparing to normal pregnancy women, the prevalence of gestational diabetes mellitus in hypothyroxinemia was higher(42.2%vs18.4%, P<0.05). Free thyroxine(FT4) was significantly negatively correlated with postload glucose concentration. FT4 was associated with gestational diabetes mellitus. The risk of gestational diabetes mellitus was greater when FT4 was lower. The prevalence of gestational diabetes mellitus in early pregnancy women with TPOAb levels ≥500 IU/ml was significantly higher than that in normal pregnancy women (31.2%vs 18.4%, P<0.05). Conclusion Early pregnancy women with hypothyroxinemia and TPOAb≥500 IU/ml were related to the risk of gestational diabetes mellitus. Lower FT4 was the risk factor of gestational diabetes mellitus.
7.The relationship between accompanying infection and immunologic function changes in type 2 diabetes melli-tus patients

Objective To investigate the relationship between accompanying infection and the change of immunologic function in type 2 diabetes mellitus ( T2DM) patients.Methods One hundred and fifteen T2DM patients with infection and 95 T2DM patients with no infection were selected,and 102 subjects with no history of dia-betes were selected as no diabetetes with infection group.The venous blood of all groups were sampled after an over-night fast of 12h.Glycosylated hemoglobin a1c(HbA1c) level was tested by glycosylated hemoglobin automatic analy-zer.The levels of T cell subsets including CD3 ,CD4 ,CD8 ,NK cell and B cell ratio were tested by flow cytometry,Ig and complement level was tested by immune nephelometry.Results The level of body mass indices(BMI) in T2DM patients with infection group[(27.39 ±9.18) kg/m2 ] and with no infection group[(26.15 ±7.39) kg/m2 ] were higher than no diabetes with infection group (24.21 ±5.37)kg/m2 (t =2.548,4.702,all P <0.05).The levels of IgG,IgA,IgM,C3 ,C4 in T2DM with infection group were (11.83 ±3.92)mg/mL,(3.02 ±0.96)mg/mL,(3.38 ± 0.82)mg/mL,(1.70 ±0.38)mg/mL,(0.52 ±0.18)mg/mL,which in T2DM with non infection group were (12.46 ± 2.47)mg/mL,(2.63 ±1.37)mg/mL,(2.91 ±1.79)mg/mL,(1.58 ±0.43)mg/mL,(0.46 ±0.31)mg/mL,which in no diabetetes with infection group were (13.26 ±3.74)mg/mL,(2.06 ±1.86)mg/mL,(2.49 ±1.01)mg/mL, (1.19 ±0.82)mg/mL,(0.30 ±0.05)mg/mL.In T2DM with non infection group and T2DM with infection group, humoral immunity index including the level of IgG was lower and IgA,IgM,C3 ,C4 levels were higher than no dia-betetes with infection group,the differences were statistically significant(t =7.052,23.059,12.617,18.326,8.730, all P <0.05).The levels of CD4 ,CD8 ,NK,CD4 /CD8 ,CD3 were (37.68 ±8.39)%,(31.58 ±6.98)%,(10.76 ± 6.49)%,(1.19 ±0.75),(62.83 ±5.28)% in T2DM with infection group,which in T2DM with non infection group were (39.23 ±10.28)%,(27.61 ±5.65)%,(14.89 ±7.12)%,(1.39 ±1.01),(64.19 ±6.46)%,which in no diabetes with infection group were (42.91 ±5.67)%,(25.17 ±7.25)%,(16.39 ±6.24)%,(1.86 ±0.82), (73.65 ±9.10)%.Cellular immunity index containing CD4 ,CD8 ,NK,CD4 /CD8 ,CD3 levels decreased more signifi-cantly than no diabetic group,T2DMI group compared with T2DM group,the levels of IgG,CD4 ,CD8 ,NK,B cell ratio, CD4 /CD8 ,CD3 decreased obviously,while the levels of IgA,IgM,C3 declined greatly,there were significant differences (t =11.038,8.237,18.549,25.871,2.436,all P <0.05).Conclusion Patients with T2DM have humoral and cellular immunity abnormalities,T2DM patients with infection is closely related with the imbalance of immunologic function.
8.Relationship between the expression of long-non-coding RNA ( lncRNA ) HOTAIR and cellular radiosensitivity in esophageal squamous cell carcinoma

Objective To investigate the relationship between the expression level of HOTAIR and cellular radiosensitivity in esophageal squamous cell carcinoma (ESCC). Methods Four ESCC cell lines ( K150, K450, TE-1, and Eca109 ) were used in this study. Quantitative real-time polymerase chain reaction was applied to measure the expression level of HOTAIR in the above cell lines;colony-forming assay was applied to measure the survival fraction of different cells irradiated by different doses of X-ray. The t-test or analysis of variance was applied for analysis of differences. The correlation analysis was used by Pearson methods. Results The four cell lines all showed high expression levels of HOTAIR and radioresistance. Compared with the other three cell lines, Eca109 had a lower expression level of HOTAIR, a lower survival fraction at each radiation dose point, and significantly lower D0 and Dq . 9.Effects of simvastatin and amlodipine on heme oxygenase/carbon monoxide system in atherosclerosis model of rabbit

Objective To investigate the change rule and correlation of heme oxygenase-1(HO-1)/carbon monoxide(CO)and the influence of simvastatin and amlodipine in athemsclemtic progress.Methods The rabbits received 1%cholesterol diet(n=24)for eight weeks.After eight weeks,rabbits were fed with normal diet for eight weeks.The rabbits in model group(n=8)were administrated with cholesterol diet.The rabbits in simvastatin group(n=8)were administrated with simvastatin.The rabbits in amlodipine group(n=8)were administrated with amlodipine.The levels of serum lipids and plasma carbon monoxide were obtained at the beginning,the 8th week and the 16th week.The expression of heme oxygenase-1 in the thoraoia aortic tissue were observed with immunohistochemistry technique.Results By the end of 16th week,the levels ofserum lipids and plasma carbon monoxide in model group were obviously increased,however,the expression of heine oxygenase-1 were markedly decreased.Compared with model group.The levels~rurfl lipi&and plasma carbon monoxide in simvastatin group were significantly decreased,while the expression of heme oxysenase-1 in aortic great reduced.The levels flerum lipids in amlodipine group were not significant ckmged,the levels of plasma carbon monoxide were obviously decreased,while tlle expression ofheine oxygenase-1 in aortic great reduced.Conclusions In atheresclerofic progress,heme oxygenase-1(HO-1)/carbon monoxide(CO)appared the reciprocal relationship,and amlodipine may suppress athemsclemtie progress by decreasing the system.
10.Combined Therapy of Ultrapulse CO_2 Laser with Autologous Epidermal Grafting in Vitiligo

Objective To observe the clinical efficacy and safety of ultrapulse CO 2 laser-assisted au-tologous epidermal grafting for the treatment of vitiligo.Methods1605patients with30660skin grafts were treated by CO 2 laser-assisted autologous epidermal grafting.Among them,782patients had localized vi-tiligo,319segmental,226general and278scattered.Epidermal grafts were harvested with negative pressure blistering method from normally pigmented skin,and recipient areas were prepared with a ultrapulse CO 2 laser.The curative effect was evaluated8weeks later.Results79.04%of the lesions were cured complete-ly,and total response rate was92.21%,with better effect in localized and scattered lesions than in other le-sions.The best results were shown in face,while the worst results shown in brow and hairline lesions.Con-clusions The advantage of combined therapy of CO 2 laser with autologous epidermal grafting in vitiligo is of easy performance,convenience,high response rate,which is worthy of large scale application.
