1.Progress on the mechanism of iron overload in myelodysplastic syndrom
International Journal of Pediatrics 2017;44(8):551-553
Myelodysplastic syndrome (MDS) is a group of origin of hematopoietic stem/progenitor cells of myeloid clonal disorders.Studies have shown that MDS patients are with varying degrees of abnormal iron metabolism in the course of diagnosis and supportive care.Iron overload can damage the body's iron balance.Excess free iron can sink in plasma and tissue, especially in the body's major organs such as heart, liver and endocrine gland, resulting in structural damage and organ dysfunction, accelerating transformation to acute myeloid leukemia, and increasing the death rate.Iron chelation can improve the prognosis of patients, prolong overall survival and leukemia-free survival. The mechanism of iron overload in MDS research will be described in this paper.

International Journal of Surgery 2009;36(1):60-63
Cancer stem cells are defined as the unique subpopulation in the tumors that possess the abili-ty to initiate tumor growth and sustain self-renewal as well as metastatic potential.Accumulating evidences in recent years strongly indicate the existence of cancer stem cells in solid tumors of a wide variety of organs. In recent years, many cancer stem cells have been found, including leukemia, breast cancer, brain ttunor,liv-er cancer, colon carcinoma, etc. In this review,we will discuss the conception and origin of the gastric cancer stem cells, and their role in the development of multi-drug resistance.

Objective To investigate the main causes and strategies for the difficulties in ureteroscopic lithotripsy. Methods From March 2004 to December 2006,19 cases of ureteral calculi, who experienced difficulties during holmium laser lithotripsy or pneumatic lithotripsy under a rigid ureteroscope, were analyzed retrospectively.Among the cases,3 had difficulties in ureteroscope placement due to the narrow ureteral ingress,6 owing to calculus obstruction complicated with ureteral inflammatory polypi,4 resulted from twisted ureter, and 6 because of stenosis of the ureter. Results In 15 of the patients, the operation was successfully performed by changing surgical approach, controlling the hydraulic irrigation, and incising the stenotic segments, etc. Two patients, who had stenotic ureter, received ESWL with double-J catheter dwelling.Lithotripsy failed in 2 cases, and PCNL was used to remove the calculi. The postoperative complications occurred in 4 cases, including 2 with mucosal laceration and 2 ureteral perforations. One of the 4 cases was transferred to an open surgery, and the other 3 were cured by conservative treatments. Conclusions Holmium Laser lithotripsy and pneumatic lithotripsy under a rigid ureteroscope are safe and effective in treating ureteral calculi.

Purpose The aim is to study the transduction of m urine stem cell factor(SCF) into umbilical blood cells by LipofectinRMmediated and expression in them. Methods Murine SCF cDNA encoding extra cellu lar domain was isolated using PCR from plasmid pRC/CMV(containing SCF gene), and then recombined into the expression vector pcDNA3,and transferred into enrichme nt cultural umbilical blood cells. Semi-quantitative RT-PCR was used to examin e the expression on mRNA level. And the effects of supernatant of transfected umb ilical blood cells were investigated alone or in coordinate with GM-CSF by colo ny formation test of human bone marrow cells in semisolid culture. Results SCF mRNA was expressed in transfected umbilical blood cells. The su pernat ant of transfected umbilical blood cells could increase the number of CFU-GM, s ynergizing with GM-CSF.Conclusion The supernatant of umbil ical b lood cells transfected with vector containing mSCF gene can stimulate the colony formation of human bone marrow cells in combination with GM-CSF.

Virologica Sinica 2008;23(3):183-188
The baculoviral inhibitors of apoptosis play a significant role in infectivity and viral host-range, which make them potential candidates for the engineering and improvement of baculovirus insecticidal. The iap3 gene of Spodoptera exigua nucleopolyhedrovirus (SeMNPV), amplified by PCR, was 939 bp encoding IAP3. The PCR product was cloned into EcoR I/Bam H I of the plasmid pEGFP-C1. GFP was fused to the N-terminaus of IAP3 to study distribution in HEK293. It was observed that the plasmid expressing IAP3 significantly inhibited apoptosis induced by cisplatin in HEK293 cells. We conclude that the IAP3 of SeMNPV is functional in mammalian cells.

Tethered cord syndrome is a severe and congenital disease. Resecting the diseased ilium terminal(FT), eystis, lipoma is the main treatment for the disease, relieving the injury to the eonus medullaris. Why the FT is diseased and thickened? Is it necessary to eut internal FT or external FT? What is the injury mechanism of the low-positional or normal positional oonus medullaris? To study the component and structure of the conus medullaris and FT may give definite answers.

Objective To study the risk factors of central lymph node (CLN) metastasis in papillary thyroid carcinoma (PTC),to provide the basis for clinical treatment. Methods A total of 407 patients with PTC in thyroid surgery, the Third Affiliated Hospital of Harbin Medical University from January 2010 to June 2012 who had undergone at least ipsilateral CLN were studied. These factors included normal situation, thyroidglobulin antibody or thyroid microsomal antibody, family history, pathological features (the size of primary, tumor multifocal, tumor location and capsular invasion),TNM staging and CLN metastasis.Univariate analysis and multivariate analysis were performed using x2 test and binary Logistic regression test, respectively. Results The CLN metastasis rate was 51.351% (209/407) of the 407 patients with PTC.The CLN metastasis was significantly associated with age, tumor multifocal, the size of primary tumor, capsular invasion and TNM (x2 =21.080,14.974,47.671,12.858,8.765,all P < 0.01).Sex,thyroid globulin antibody or thyroid microsomal antibody, family history and tumor location were not associated with CLN metastasis (x2 =1.457,1.106,0.000,0.260,all P > 0.05).Multivariate analysis indicated that age (< 45 year),the size of primary tumor (≥ 1.0 cm),tumor-muhifocal,and TNM (T3 + T4) were the independent risk factors for CLN metastasis [odds ratio (OR) =0.937,2.347,0.380,0.389,all P < 0.01].Conclusions Age (< 45 year),the size of primary tumor (≥ 1.0 cm),tumor-multifocal and TNM (T3 + T4) are risk factors for CLN metastasis in PTC.Dissection of CLN should be considered for PTC patients with these factors.
