1.Comparison of Depression and Anxiety of Korean and Han Climacteric Residents in Community

Objective:To understand the depressive and anxiety condition of Han and Korean climacteric residents in community.Methods:1262 climacteric residents including 639 Korean and 623 Han residents selected from eight neighborhood communities were investigated with a Health-Investigating Scale(self-made),the Center for Epidemiological Survey Depression Scale(CES-D) and Self-rating Anxiety Scale(SAS).Results:According to CES-D,there were totally 127(10.12%) met the criteria of depression,in which 69(54.3%) were Korean and 58 were Han(45.7%).By SAS,391(32.24%) residents in which 240(61.4%) Korean and 151(38.6%) Han had anxiety symptoms.Stepwise multi-factorial regression analysis revealed the major factors related to depression were:nationality,income,drink,personality and employed,those related to anxiety were nationality,personality,drink,income and health.Conclusions:There are more depression and anxiety in Korean climacteric people than in Han.
2.Codonopsis lanceolata saponin preconditioning alleviates hepatic ischemia/reperfusion injury in rats

Objective To investigate the protective effects of Codonopsis lanceolata saponin (CLS) on hepatic ischemia-reperfusion injury (HIRI) and its mechanism in rats.Methods The rats were divided randomly into 3 groups:sham-operated (SO) group, HIRI group and CLS group.HIRI model was made by Pringle method.CLS group was pretreated with CLS (200 mg · kg-1 · d-1) by intragastric administration for 7 days before operation, while SO group and HIRI group were given with distilled water 10 ml · kg-1 · d-1 for 7 days.ALT, AST, SOD, MDA in serum, and SOD and MDA in hepatic tissue were detected by colorimetric method.IL-18 in serum and hepatic tissue was detected by ELISA.Results The levels of serum ALT, AST, MDA, IL-18, TNF-α and tissue MDA, IL-18 in HIRI group were significantly higher than those in SO group and CLS group (all P < 0.05), respectively.The serum and hepatic tissue SOD activity in HIRI group was significantly lower than that in SO group and CLS group (all P < 0.05).Histological observation revealed that the obvious cell necrotic foci with a large number of inflammatory cells infiltration in the HIRI group, while the lesion was obviously alleviated in CLS group.Conclusions Codonopsis lanceolata saponin preconditioning alleviates hepatic ischemia/reperfusion injury.The mechanism may be correlated with inhibiting inflammatory factors and enhancing anti-oxidation.
3.Influence of Rhodiolasachalinensis and Codonopsispilosula mixture in immune function of mice

Objective Using serum pharmacology method to assess the optimal dose of Rhodiolasachalinensis extract (RSE)and Codonopsispilosula extract (CPE)combination as well as the effects of the combination on the immune function of mice.Methods Using the 3×3 facterial design,nine medicated serum of CPE and RSE mixture were made to assess the optimum combinational dose of CPE and RSE by detecting the T,B lymphocyte proliferation abilities and NK cell activity in vitro .Using the optimum combinational dose and reducing 2.5,5 and 10 times as high,middle and low doses of RSE+CPE groups,and the T,B lymphocyte proliferation abilities and the activity of NK cells in the mice were detected in vivo .Results The result of serum pharmacology indicates that compared with control group,the proliferation abilities of T,B lymphocytes and the activity of NK cells inducing by ConA and LPS in 200 mg· kg-1 RSE + 790 mg· kg-1 CPE group were increased (P < 0.05).The result of in vivo experiment indicated that compared with cyclophosphamide group,the spleen indexes (SI)in middle and high doses of RSE+CPE groups were significantly increased (P <0.05);compared with low dose of RSE+CPE group,the WBC number in middle dose of RSE + CPEgroup was significantly increased (P < 0.05 ). Compared with cyclophosphamide group,the T and B cell proliferation abilities induced with ConA and LPS and killing activities of NK cells in low,middle and high doses of RSE+CPE groups and positive drug CVT-200 group were significantly increased (P <0.05).Compared with CVT-200 group,low and high doses of RSE+CPE groups,the T and B cell proliferation abilities induced with ConA and LPS and the activity of NK cells in middle dose of RSE+CPE group were significantly increased (P <0.05).Conclusion RSE and CPE mixture can enhance the immune function of mice;RES 200 mg·kg-1 and CPE 790 mg·kg-1 are the optimal doses.
4.Monitoring of chimerism and fusion gerne by STR-PCR and RT-PCR in relapse chronic myeloid leukemia patients after hematopoietie stem cell transplantation

Objective To investigate the value of the multiple short tandem repeat (STR)amplification by fluorescence labeling polymerase chain reaction (PCR) combined with fusion gene bcr-abl mRNA expression for quantitative determination of chimerism and qualitative detection of bcr-abl transcripts,and to evaluate the status of engraftment and predict the outcome of allogeneic hematopoietic stem cell transplantation (allo-HSCT). Methods 5 relapse patients with CML after alIo-HSCT were dynamically investigated. Quantitative analysis of donor chimerism was performed by multiplex PCR amplification of STR markers and capillary electrophoresis with fluorescence detection, qualitative detection of bcr-abl transcripts was performed by RT-PCR. Results The donors alleles appeared in all of 5 patients on day 28 post transplant, and bcr-abl expression was negative. But 5 patients had unstable mixed ehimerism. (DC: 0~80.4 %) at the different time points after aIIo-HSCT and bcr-abl was positive. One of them kept eontinuely the mixed chimerism in the relapse of disease, and died after one year, and the other changed from MC to CC by intervention of clinical treatment. Reduction of donor chimerism were detected prior to the occurrence of graft rejection and disease relapse, while bcr-abl gene expression was positive. Conclusion The results of STR-PCR in the range of its sensitivity fully correspond with bcr-abl tests in patients with CML. The combination of STR-PCR with RT-PCR provides a highly sensitive and valuable tool for engraftment evaluation, graft rejection, relapse and predicting GVHD. Furthermore it can provide a basis for early intervention of clinical treatment, and can identify these patients at high risk with molecular or cytogenetic relapse after allo-HSCT.
5.The efficacy and safety of PAD and VAD regimens for untreated multiple myeloma

Objective To compare the efficacy and adverse effects of bortezomib+adriamycin+dexamethasone (PAD) and vincristine + adriamycin + dexamethasone (VAD) regimens in untreated multiple myeloma (MM). Methods There were 26 and 28 new diagnosed MM patients in PAD and VAD groups. Both clinical effects and adverse effects were observed. Patients accepted VAD or PAD regimens for 2-4 cycles and followed up for 7-27 months. Results There were 10, 5 and 11 patients accepted 2, 3 and 4 cycles in PAD group, and 6, 11 and 11 in VAD group. In PAD group, there were 2, 14, 9, 1 and 0patients achieved complete remission (CR), very good partial remission (VGPR), partial remission (PR),stable disease (SD) and progressive disease (PD); in VAD group, the number were 0, 4, 12, 10 and 2.The rate of patients who achieved good efficacy (CR+VGPR) in PAD group was 61.5%, which was higher than that in VAD group (14.3%).The incidences of infection and gastrointestinal symptoms were similar in the two groups, while the incidences of peripheral neuropathy, thrombosis and Herpes Zoster infection in PAD group were higher than those in VAD group. Conclusions Compared with the conventional VAD chemotherapy, PAD may improve CR and VGPR rates in new diagnosed MM, while it may bring more and severer toxicities in peripheral neuropathy, thrombosis and Herpes Zoster infection. Preventive medical care is necessary in PAD protocol.
