1.The impact of inflammatory cytokines and therapy effect of ecabet sodium quadruple therapy for patients with duodenal ulcer

Objective To study the effect of inflammatory cytokines and therapy effect of ecabet sodium quadruple therapy for patients with duodenal ulcer.Methods80 cases patients with H.pylori-positive ulcer duodenal in In our hospital In our hospital April 2015 to December 2016 were randomLy divided into observation group 42 cases and control group 38 cases, the observation group were treated with ecabet sodium quadruple therapy, the control group were treated with colloidal bismuth pectin quadruple therapy.Then therapy effect, H.pylori eradication rate, inflammatory cytokines, adverse reactions were compared between two groups.ResultsUlcer healing: the observation group total effective rate 97.62% was significantly higher than the control group 78.95% (χ2=6.966, P<0.05);Hp eradicate rate: the observation group Hp eradicate rate 95.24% was significantly higher than that of control group 76.32%(χ2=6.023, P<0.05);Serum inflammatory factors: observation group serum TNF-α, IL-6, IL-8 levels were significantly lower than the control group(t =4.272, 7.584, 3.095, P<0.05);Adverse reaction: observation group adverse reactions 7.14% was significantly lower than the control group 23.68%(χ2=4.281, P<0.05).ConclusionEcabet sodium quadruple therapy helps to reduce serum inflammatory factor expression level, then improve therapy effect and reduce adverse reactions.
2.Clinical efficacy of Fangfengtongsheng powder combined with Flupentixol and Melitracen tablets in treatment of anorectal neurosis

Objective To analyse clinical efficacy of Fangfengtongsheng powder combined with Flupentixol and Melitracen tablets in treatment of anorectal neurosis.Methods92 patients with anorectal neurosis from April 2014 to June 2016 were grouped two groups randomly, each with 46 cases.Two groups were treated with Flupentixol and Melitracen tablets and observation group was treated another with Fangfengtongsheng powder.Clinical efficacy of two groups were compared.Results①Total effective rate of observation group was 95.65%,higher than that of control group 78.26% (P<0.05).②After treatment,SAS and SDS score of observation group were lower than that of control group(P<0.05).③After treatment, anal bulge, anal pain, frequent defecation, anal foreign body sensation score and 8 dimensions of SF-36 scale score of observation group were better than that of control group(P<0.05).④The control group had 8.70% adverse reactions, compared with observation group 4.35%, the difference was not statistically significant.ConclusionFangfengtongsheng powder can improve the therapeutic effect of anorectal neurosis on the basis of Flupentixol and Melitracen tablets, relieve the anxiety level and improve the living standard of patients.
3.Clinical observing of advanced non-small cell lung cancer patients treated with CIK cell combined with chemotherapy

Objective To observe the efficacy of advanced non-small cell lung cancer patients treated with CIK cell combined with chemotherapy.Methods65 cases of advanced lung cancer patients admitted in our hospital from February 2013 to January 2015 were retrospectively analyzed.The patients were randomly divided into observation group (34 cases) and control group (n=31).The levels of vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) and carcinoembryonic antigen (CEA) in the two groups were observed before and after treatment.The patients in the control group were treated with chemotherapy on the basis of chemotherapy.Cytokeratin 19 (CYFRA21-1) levels, and clinical efficacy.Results①The patients with VEGF and MMP-9 values after treatment (298.67±36.49)ng/L, (1102.31±86.48)ng/mL, than the control group (459.19±45.28)ng/L, (1394.83±121.06)ng/mL, and the difference was significant (P<0.05).②CEA and CYFR21-1 value patients in the observation group were (14.16±1.08) ng/mL, (4.45±0.45) ng/mL, than the control group (17.86±1.84) ng/mL, (5.76±0.64) ng/mL, and the difference was significant (P<0.05).③The total observation group was significantly higher than patients 94.18% 74.19% efficiency, and the difference was statistically significant (P<0.05).ConclusionCIK cell biological therapy combined with chemotherapy for advanced lung cancer patients with a certain improvement is worthy of further research and application.
4.Activated Caspase-8 Regulates Bcl-2 Expression in Fas-and Actinomycin D-induced Apoptosis in Bel-7402 Cells

Objective To investigate whether caspase-8 regulates Bcl-2 expression in Fas and Actinomycin D(AD) induced apoptosis in Bel-7402 cells.Methods Bel-7402 cells were divided into four groups randomly,that is:control,Fas group,Fas-AD group and Ac-IEFD-cho group,and treated with Fas-AD in the presence/absence of caspase-8 inhibitor Ac-IEFD-cho.The expressions of caspase-8 and Bcl-2 were determined with Western Blot and RT-PCR,respectively.Results Compared with control group and Fas group,after Bel-7402 cells were treated with Fas-AD,typical apoptosis morphology was clearly observed,the expression of activated caspase-8 increased significantly,and the Bcl-2 expression decreased apparently.However,after Bel-7402 cells were treated with Fas-AD in the presence of the caspase-8 inhibitor Ac-IEFD-cho,the expressions of activated caspase-8 decreased while the Bcl-2 expression increased.Conclusion Activated caspase-8 can regulate Bcl-2 in Fas-and AD-induced apoptosis in Bel-7402 cells.
5.Matrine Compoand Injection Interrening the Liver Fibrosis Rat Induced by Tetrachloride.

Objective To examine the change of TGF-?1 in the bloods of rats which were induced by tetrachoride during the different times and to explain the mechanism of the change.MethodsThe experimental liver fibrosis induced by tetrachloride in rats was treated by matrine compound injection.Serum TGF-?1 and pathological changes in liver tissue were observed in control group,model group and therapy group during the time of the 4th,6th and 9th week.ResultsThere was an obvious change of TGF-?1 in the blood in different time and different groups.ConclusionMatrine compound injection has an antifibrogenetic action and TGF-?1 can reflect the change of liver fibrogenesis of different time as an index.
6.Characteristics and Kinetic Study of 2-Hydroxypyridine Degradation by a Novel Bacterium Arthrobacter sp.2PR

A novel strain,which could use 2-hydroxypyridine (2HP) as the sole source of carbon,nitrogen,and energy,was isolated from petroleum-contaminated soil at the Liaohe estuarine wetland.Strain 2PR was identified as Arthrobacter based on the morphology and 16S rRNA gene sequence.The optimum growth and degradation condition upon 2PR is 30℃ and pH 7.0,respectively.Under this condition,2HP degradation rate were 97.34%,94.95%,94.48% and 89.21% with 2,4,6 and 8 mg/ml initial concentration of 2HP at 42,96,120 and 156 h,respectively.Strain growth and 2HP degradation kinetics studies indicated that the strain followed Logisitic model,which could provide a theoretical and technical reference for the biodegradation of 2HP.The color of strain 2PR culture upon 2HP-MSN changed from colorless to blue,and then turned to brown.The blue pigment,which was observed at the culture of strain 2PR,was identified as 4,5,4',5'-tetrahydroxy-3,3'-diazadiphenoquinone-(2,2') by high performance liquid chromatography (HPLC) and high-performance liquid chromatography-mass spectrometry (LC-MS) analysis.The LC-MS signal with m/z =249.1 was observed in resting cells reaction sample with 2HP as the substrate.The degradation of 2HP might be achieved by a dioxygenase to produce 2,3,6-trihydroxypyridine,which could transformed to the blue pigment spontaneously,and then 2,3,6-trihydroxypyridine was converted with an pyridine-ring cleavage reaction.Among all the reported strains,strain 2PR has the strongest tolerance and the highest 2HP degradation efficiency at present.The strain has a promising application potential for 2HP waste treatment.
7.Value of virtual touch tissues quantization in the stages of type 2 diabetic nephropathy

Objective To investigate the application value of virtual touch tissues quantization(VTQ)in the stages of type 2 diabetic nephropathy.Methods 120 cases of type 2 diabetes were divided into 3 groups according to urinary albumin excretion rate (UAER):diabetes without urinary albumin,diabetes with trace albuminuria and diabetes with mass albuminuria.The control group consisted 30 normal cases.VTQ was used to measure the renal shear wave velocity (SWV),then compare were made between the 4 groups.Besides,correlation analysis was made between SWV and UAER in diabetic nephropathy group.Results When compared SWV,there was no significant difference between diabetes without urinary albumin and control group (P =0.144),but there was significant difference between each 2 group of diabetes with trace albuminuria,diabetics with mass albuminuria group and control group (P < 0.05).There was also significant difference between each 2 group of diabetes (P < 0.05).The correlation coefficients for SWV and UAER in diabetes were 0.697(P <0.05).ROC analyses showed the area under curves for SWV diagnosis for diabetic nephropathy was 0.93.The best cutoff point,sensitivity and specificity were 2.43 m/s,85.7% and 84.5 %,respectively.Conclusions The renal SWV increases as the renal injury progresses,which also plays an important role in diabetic nephropathy stages.
8.Content Determination of Heavy Metals in Xiaohuoluo Pills by Atomic Absorption Spectrometry

Objective:To determine the content of lead, cadmium, arsenic, mercury and copper in Xiaohuoluo pills produced by 34 enterprises. Methods:Using microwave digestion, the contents of lead, cadmium and arsenic were determined by graphite furnace atomic absorption spectrometry;the content of mercury was determined by hydride generation atomic absorption spectrophotometry;the content of copper was determined by air-acetylene flame atomic absorption. Results: Lead, cadmium, arsenic and mercury in Xiao-huoluo pills from 34 different manufacturers were beyond the limits in various degrees. Conclusion: The method is simple and accu-rate, and can be used in the heavy metal determination of Xiaohuoluo pills.
9.Determination of Dracorhodin and Curcumin in Dieda Pills by UPLC

Objective:To determine the content of dracorhodin and curcumin in Dieda pills by UPLC .Methods:A UPLC method was adopted.The determination was performed on a Waters Acquity UPLC BEH C 18 column(100 mm ×2.1 mm,1.7 μm) with the mobile phase consisting of acetonitrile-0.05 mol· L-1 NaH2PO4(50 ∶50).The detection wavelength was 440 nm for dracorhodin and 431 nm for curcumin, the column temperature was 30℃and the flow rate was 0.1 ml· min-1 .Results:There was a good linear range of 0.001 8-0.036 4 μg(r=0.999 9)for dracorhodin and 0.000 8-0.015 6 μg(r=0.999 9) for curcumin.The average recovery for dracorhodin was 97.94%(RSD=0.89%) and that for curcumin was 98.45%(RSD=0.91%).Conclusion:The method is simple, rapid and reproducible ,which can be used for the determination of dracorhodin and curcumin in Dieda pills .
10.Effects of 5-Aza-CdR on cell apoptosis in lung cancer A549/DDP cell line and expression of tumor suppressor gene hMLH1

Objective To investigate the effects of 5-Aza-2'-deoxycytidine (5-Aza-CdR) on the apoptosis of A549/DDP cells and the expression of hMLH1 gene.Methods A549/DDP cells were treated with 5-Aza-CdR at 0.5,5,50 μmol/L.The growth curve of A549/DDP cells was investigated by MTT assay.The methylation status of hMLH1 gene was detected by methylation specific PCR (MSP).The expression of hMLH1 mRNA was evaluated by FQ-PCR.The apoptosis rate of A549/DDP cells was analyzed by flow cytometry.Results A549/DDP cells treated with 5-Aza-CdR showed a slow growth in comparison with the control cells,and the growth rates were decreased with the increasing of 5-Aza-CdR concentration.The apoptosis rate after treatment was higher than that before treatment in A549/DDP cells (P < 0.05),and had a positive correlation with 5-Aza-CdR dose (P < 0.001).hMLH1 mRNA expression level was increased in a 5-Aza-CdR concentration dependent manner (P < 0.05).hMLH1 promoter in A549/DDP cells was methylated and hMLH1 mRNA was negatively expressed before treatment,but the mRNA was positively expressed after treatment with 5-Aza-CdR.Conclusions 5-Aza-2'-CdR can induce apoptosis of A549/DDP cells by inducing demethylation of hMLH1 promoter and thereby enhancing hMLH1 gene expression and its tumor suppressor function.
