1.Treatment of malignant gliomas with stem cells mediated oncolytic adenovirus

Oncolytic adenoviruses are genetically altered to replicate within tumor cella while exhibiting a high degree of infectivity,good safety and specificity.The use of oncolytic adenoviruses to treat malignant brain tumors has been proved to be a promising mode of cancer therapy.Local injection of adenoviral vectors fails to reach scattered infiltrative tumor cells within the brain parenchyma and the therapeutic effect of intracereberal injections of oncolytic adenoviruses is only seen in the vicinity of the injection site.To remedy this and prevent possible tumor recurrence,stem cells have been explored as vehicles for gene therapy in brain tumors given that they possess an intrinsic tropism for brain glioma.
2.Detection and significance of urokinase-type plasminogen activator and its receptor in synovial fluid and plasma in rheumatoid arthritis

Objective To measure the levels of urokinase-type plasminogen activator (uPA) and its receptor (uPAR) in synovial fluid and plasma of patients with RA and investigate its clinical significance.Methods By using ELISA sandwich method,the levels of uPA and uPAR in plasma from 46 patients with RA and 8 with osteoarthritis (OA) were measured.Those in synovial fluid (SF) from 14 patients with RA,and plasma from 12 healthy subjects were measured as controls.Results The concentration of uPA and uPAR in SF from patients was significant higher than that in their plasma (P<0.001,P<0.01),and the concentration of uPA and uPAR in plasma of RA was also much higher than that in plasma of OA patients (P<0.05,P<0.000 1) and healthy subjects (P<0.000 1,P<0.001).There was no significance between plasma levels of RF+ and RF- in RA patients.The concentration of uPA and uPAR of plasma in RA correlated positively with CRP,RF and the number of swelling joints.Conclusion The levels of uPA and uPAR in SF and plasma are useful parameters for monitoring disease activity of RA.These findings suggest that the uPA and uPAR genes may play an important role via proteolytic damage of the extracellular matrix during the development of RA.
3.Advances in oncolytic adenovirus-loaded mesenchymal stem cells for treating tumors

Oncolytic adenoviruses have been used in cancer gene therapy because of their ability to selectively transfect and repli-cate in tumor cells. However, systemically administering oncolytic adenoviruses is often inefficient because the host's immunity also at-tacks the viruses. Delivering oncolytic adenoviruses by loading them into mesenchymal stem cells effectively solves this problem. Mes-enchymal stem cells have been described to possess a tumor-homing ability, as well as the capacity to deliver oncolytic adenoviruses. This review discusses the current challenges faced by oncolytic adenovirus delivery via mesenchymal stem cells.
4.Studies on the relationship of CXCL1 and peripheral arthritis in spondyloarthropathy and its pathogenesis

Objective To investigate the role of chemokine in inducing peripheral arthritis in spondyloarthropathy(SpA).Methods Microarray was used to screen the candidates of inducing peripheral arthritis in SpA,and enzyme linked immunosorbent assay(ELISA)was used to verify the results.Peripheral blood mononuclear cells(PBMC)from 5 healthy subjects were co-incubated 1.5 hours with synovial fluid of 5 patients with SpA,total RNA were then extracted,and the samples were screened for the target genes by using superarray techniques.ELISA was used to determine CXCL1 protein levels of 54 ankylosing spondylitis(AS)patients and 30 healthy controls,also to determine the CXCL1 levels of synovial fluids from 33 SpA patients and 14 osteoarthritis(OA)patients,and to determine CXCL1 protein levels by ELISA before and after treatment with infliximab.Results The expression of CXCL1,CXCL2,CXCL3 and IL-8 were more than 2 fold higher after stimulated by synovial fluids.However,there was no change in other chemokines and cytokines,such as TNF?,IL-6 and IL-1 after stimulation.The protein level of CXCL1 was significant higher in serum of patients with AS than that of healthy control(P

7.The clinical characteristics of children with splenium of corpus callosum solitary lesions displayed on MRI

Objective To explore the prominent clinical features and the prognosis of corpus callosum solitary lesions cases in children.Methods There were 16 cases with corpus callosum solitary lesions receiving treatment at our hospital from January to December 2013.All of the clinical features were collected and retrospectively analyzed.Results The 16 cases aged 1 year to 13 years with an average age of (3.44 ± 3.65) years.The causes of the disease included the viral encephalitis,convulsions,and rotavirus infection.The most prevalent disorders were convulsions (10 cases),gastrointestinal symptoms (8 cases),fever (8 cases),positive cervical resistance (6 cases),fatigue (5 cases),altered states of consciousness (2 cases),headache (1 case),and dizziness (1 case).Corpus callosum lesions lasted for 13-50 days,with an average of (28.8 ± 10.2) days.Cranial magnetic resonance showed the splenium of the corpus callosum with isolated oval lesions,low signal on T1 weighted images and apparent diffusion coefficient images,high signal on T2 weighted images and diffusion weighted images.Conclusions The splenium of the corpus callosum lesions of children occur in a lower rate than adults with the etiology including intracranial infection,convulsions and rotavirus infection.Main clinical manifestations include fever,convulsions and gastrointestinal symptoms (nausea,diarrhea,etc).The cases followed-up showed the lesions and symptoms are reversible,and the prognosis is good.
8.MR Appearances of Anterior Cruciate Ligament Injury of Knee Joint

Objective To evaluate MR appearances of normal and injured anterior cruciate ligament(ACL).Methods The MR findings of normal ACL in 16 cases and ACL tear in 15 cases were reviewed retrospectively.Results Normal ACL appeared as a flat band with low signal intensity on T1 and T2-weighted imaging. The injured ACL can appeared as interruption in continuation(n=5), irregular shape(n=1),abnormal direction(n=2) and increased in signal intensity(n=7).Conclusion MRI is a accurate and noninvasive diagnostic method for ACL tear.
9.Preventive and therapeutic effects of Yishen Huanji Decoction on kidney injury in rats induced by simulation of military overtraining

OBJECTIVE: To observe the preventive and therapeutic effects of Yishen Huanji Decoction (YSHJD), a compound traditional Chinese herbal medicine, on military overtraining-induced kidney injury in a rat model. METHODS: Thirty SD rats were randomly divided into normal control group, untreated group and YSHJD-treated group. The military overtraining-induced kidney injury in rats was established by forcing to run on the treadmill for 8 weeks. The rats in YSHJD-treated group were administered with YSHJD at the same time. The 24-hour urines were collected every weekend for detecting the contents of urinary sediment, 24-hour urine total protein, 24-hour urine albumin and activity of N-acetyl-beta-D-glucosaminidase (NAG). The blood and renal tissues were collected after 8-week training, and the levels of serum urea nitrogen (BUN) and creatinine (SCr) were detected. Angiotensin II (Ang II) was detected by radioimmunoassay and activity of Na(+), K(+)-ATPase in kidney was analyzed by chemical colorimetric method. RESULTS: Compared with the normal control group, after 8-week training, the contents of 24-hour urine protein, activities of NAG in urine, and the levels of BUN and SCr in rats in the untreated group and YSHJD-treated group were obviously increased (P<0.05), and these parameters of the YSHJD-treated group were lower than those of the untreated group (P<0.05). The contents of Ang II in blood plasma and kidney of rats in the untreated group and YSHJD-treated group were higher than those of the normal control group (P<0.05), and had no statistical difference between the untreated group and YSHJD-treated group. Compared with the normal control group, the activity of Na(+), K(+)-ATPase in kidney of rats in the untreated group was obviously decreased (P<0.05), and had no statistical difference between the normal control group and YSHJD-treated group. CONCLUSION: YSHJD can protect against military overtraining-induced kidney injury in rats by decreasing the contents of 24-hour urine protein, BUN and SCr, and the activity of NAG, and increasing the activity of Na(+), K(+)-ATPase.
10.Nutritional support post-layngectomy

Objective:To provide the basis for postoperative nutritional support,exp lore the appropriate support methods and its influnce on rehabilitation in postoperative patients with laryngea l carcinoma.Methods:285 patients with laryngeal carcinoma were randomly divided into therapy group and control group.The therapy group (161 patients)was given homogenate diet and the control group(124 patients) was given mixed mi lk by nasal feeding 5-6 times per day.The nutritional index and occurr ence of pharyrgeal fistula were monitored at preoperation,7th and 14th days postopera tion r espectively.The duration of hospitalization of each patient was recorded at the same time. Results:The decreasing of the nutritional index w as significantly lower in the therapy group than in the control group(P<0.0 5).The duration of hospitalization was shortented to about 12 days, and the incidence of pharyngeal fistul a was reduced to 11%.Conclusion:The appropriate nutrition may decrease the incidence of pharyngeal fistula,shorten the mean duration in hospital and reduce the economic burden of postoperative patients with laryngeal carcinoma.
